Melania snail(Semisulcopira bensoni) is used as ingredient in Korean traditional soup and nutritional foods. Generally, lipoxygenase in several food products may produce off-flavors during their processing and storage. Therefore, the inactivation of lipoxygenase is required to make the better extracts from Melania sanil. Also, the quality on freshness of Melania snail may be evaluated by lipoxygenase activity. The lipoxygenae activity was the highest at 40~60% saturation among several concentrations in salting-ouot saturated solution of ammonium sulfate. The partial purification of lipoxygenase was successfully obtained by Sephacryl S-200 gel chromatography. The first peak among three peaks for protein determination showed the highest activity of lipoxygenase in 13~16 fractions among 100 fractions. The highest peak of lipoxygenase activity by ion exchange chromatography was shown at 0.1M NaCl. In the purification step, the specific activity was 20.8U/mg and activity yield was 19.8%. The optimum pH and temperature were pH6.0~8.0 and 3$0^{\circ}C$, respectively. Molecular weight of the lipoxygenase was estimated about 35kDa by SDS-PAGE.

The proximate, fatty acid and dimethyl acetal (DMA) compositions of domestic and imported melania snails Semisulcospira sp. being sold commercially were studied. The protein content ranged from 8.09% (China) to 11.3% (North Korea). The lipid content ranged from 0.84 (China) to 2.36% (North Korea). Domestic melania snail had protein (10.5%) and lipid (1.02%) contents intermediate between those of the two imported melania snails. The prominent fatty acids were 16:0 and 18:0 saturated fatty acids, 16:1n-7, 20:1n-11, 18:1n-9, 18:1n-7 and 16:1n-9 monounsaturated fatty acids, and 20:4n-6, 20:5n-3 (eicosapentaenoic acid, EPA), 22:2 non-methylene-interrupted diene (NMID), 18:3n-3, 18:2n-6 and 22:5n-3 (docosapentaenoic acid, DPA) polyunsaturated fatty acids (PUFA). The percentage of n-3 PUFA was higher in domestic (19.6%) and North Korean (imported, 19.5%) snails than in that from China (imported, 16.8%), while the n-6 PUFA level was higher in snails from China (21.2%) and domestic snails (14.7%) than in that from North Korea (9.87%). The total DMA, i.e., 18:0 DMA, 16:0 DMA and 18:1 DMA was 7.30, 7.27, and 4.86% domestic snail and those from China and North Korea, respectively. These results suggest that melania snail is a source of n-3 PUFA, and DMA derived from plasmalogen.

We investigated the nutritional status of the melania snail (Semisulcospira gottschei) using RNA/DNA ratios to evaluate the effect of feeding conditions (artificial versus natural) on the reaction times of the snails in a time course following starvation. In the short experiments (48 h), the RNA/DNA ratios of the artificial feeding groups were significantly higher than those of the natural groups. While two RNA/DNA ratio peaks were observed in the artificial food group during daytime, the natural food group showed a higher ratio at night. Under starvation conditions, the RNA content decreased whereas the DNA content was constant. The RNA/DNA ratios of the freshwater snail in both groups dramatically decreased after starvation and remained constant until the end of the experiment. We verified that the RNA/DNA ratio serves as an index of nutritional condition with respect to the effect of dietary differences. These results are important for understanding optimized aquaculture rearing conditions for this important commercial freshwater snail.
Melania snail (Semisulcospira libertina) was traditionally used as the healthy food in Korea. It was generally known to improve liver function and heal a diabetes. The aim of this study was to elucidate the anti-diabetic mechanism of melanian snail hydrolysates treated with protamex (MPH) by investigating the inhibitory action on protein tyrosine phosphatase 1B (PTP1B), the improving effect on the insulin resistance in C2C12 myoblast and the protective effect for pancreatic beta-cell (INS-1) under the glucose toxicity. The melania snail hydrolysates treated with protamex (MPH), which showed the highest degree of hydrolysis (43%), and inhibited effectively PTP1B activity ($IC_{50}=15.42{\pm}1.1{\mu}g/mL$), of which inhibitory effect was higher than usolic acid, positive control ($IC_{50}=16.65{\mu}g/mL$). MPH increased the glucose uptake in C2C12 myoblast treated with palmitic acid. In addition, MPH increased insulin mRNA expression level by over 160% with enhanced cell viability in INS-1 cell under the high glucose concentration (30 mM). These results suggest that MHP may improve the diabetic symptom by the inhibiting the PTP1B activity, increasing the glucose uptake in muscle cell and protecting the pancreatic beta-cell from glucose toxicity.

To clarify reproductive ecology of the melania snail Semisulcospira coreana (v. Martens, 1886) in Bukhan River, gonad development, fatness, gonad index, sex ratio, first sexual maturity of population, monthly change of larvae number and developmental stages in brood pouches were investigated by six identification methods. As maturation progrsses, the sex of the snali can be distinguishable easily by color:: the ovary being blue-green and testis light yellow. The sex ratio of female to male individuals over 13.95 mm shell height was significantly different from 1:1 (${\chi}^2$ = 38.45, p < 0.05). The sex ratio of female to male individuals changed drastically according to the season, Based on the monthly variations of fatness, gonad index and histological analysis, spawning occurred twice a year (spring and autumn) and the mean size of matured eggs was $450{\mu}m$ in diameter. The monthly change of larval number in brood pouch showed also two distinct peaks in March and September during the year. The average number of larvae in brood pouches was 286 - 862 individuals. In this study, the number of larvae in the brood pouches were a minimum in December and a maximum in March (975 larvae). The biological minimum size (the size at 50% of group sexual maturity) of the melania snail was 13.95 mm in shell height in females and males. All females over 15 mm in shell height possesed brood pouches.
To identify the food characteristics of seven melania snail species in Korea, the detailed proximate, fatty/amino acid, mineral composition and chlorophyll contents were investigated. The content ranges of moisture, crude protein, crude lipid and ash were 70.1-81.0%, 11.6-16.9%, 0.4-1.6% and 1.9-3.4%, respectively. K. nadifila showed the significant mineral contents Fe, P, Ca than other species. Total Chlorophyll level was highest in K. globus ovalis ($1.2\pm0.2$ mg/g). The major fatty acids were 16:0, 22:1, 20:1, 18:1, 20:5 and 22:6 were identified as small amounts (1.0-3.3%) of total fatty acids. Citrulline was shown to be the major free amino acids (20.4-53.1%).
The reproductive cycle of ovoviviparous melania snail, Semisulcospira libeytina libertina was studied in the valley near Naewon Temple in Yangsan, Korea. Water temperature (WT) of the habitat ranged from 1.3$^{\circ}C$ to 22.5$^{\circ}C$ The meat weight rate (MWR) reached the maximum with the beginning of WT increase in March and the minimum in August. Monthly changes in average oocyte diameter showed the maximum (249.6$\pm$2.6 ${\mu}{\textrm}{m}$) in July and the minimum (134.3$\pm$2.8 ${\mu}{\textrm}{m}$) in December S. libertina libertina seemed to be a year-round breeder except for mid-summer and mid-winter. Two main reproductive cycle of the species could be divided into five successive stages: multiplicative (March, October), growing (April and May, November), mature (June and July, December), ovulation (August, January), parturition (September and October, March to May) and resting (September, February) stages in female and multiplicative (March, October), growing (April, November), mature (March to June, December), copulatory (July and August, January), resting (September, February) stages in male.

This study was conducted to compare the biological activities of 7 melania snails from the family Pleuroceridae (Semisulcospira coreana, Koreanomelania nodifila, Semisulcospira forticosta, Koreoleptoxis globus ovalis, Semisulcospira libertina, Semisulcospira tegulata and Semisulcospira gottschei) in Korea. Among the 7 species, S. coreana, Korean. nodifila, S. forticosta and S. gottschei showed over 80% cytotoxicities on three cancer cell lines (SNU-1, A549 and Hep 3B) compared to the non-treatment, whereas S. libertina and S. tegulata showed almost no growth inhibition activities on the same cancer cell lines. In relation to ACE inhibition activity, only S. coreana, Korean. nodifila, and S. forticosta showed over 60% ACE inhibition activities, whereas other melania snails exhibited inhibition activities of lower than 25%. DPPH radical scavenging activities were also determined, and used to categories melania snails into three groups based on Duncan's multiple range test at P<0.05. The amount of TNF-${\alpha}$ produced by in vitro mouse peritoneal macrophage was determined according to bioactivity on L-929 cells. Three melania snails, S. coreana, Korean. nodifila and S. gottschei, exhibited 95.2%, 89.7% and 93.7% cell death(%) on L-929 cells, respectively. Glucose-6-phosphate dehydrogenase inhibitory activity was also obtained in the extract of S. coreana (31.9%) and Korean. nodifila (28.1%), showing that these extracts can be used as supplemental dietary health foods. In conclusion, we believe that the extracts of melania snails should be given due consideration in functional health food development.
The present study was conducted to find out effect of PFOS on survival, activity, growth and organ structure of the melania snail, Semisulcospira gottschei. Experimental groups were composed of one control condition and four PFOS exposure condition (0.1, 0.5, 1.0 and 5.0 mg/L). After 16 week exposure, survival rate and activity were not significantly influenced at the two lower exposure groups, 0.1 and 0.5 mg/L, but they were significantly reduced in 1.0 and 5.0 mg/L groups. Total weight and meat weight rate (MWR) were reduced in PFOS exposure group in comparison to control group. Also, histological degenerations such as acidification of mucous, necrosis and split of muscular fiber bundle, atrophy of anterior pedal gland were recognized in the foot. Hepatopancreas showed the atrophy and degeneration of the digestive cell, vacuolation of digestive gland and closure of lumen in digestive gland.
The in vitro antioxidant activities of five melania snails in the genus Semisulcospira (S. coreana, S. forticosta, S. libertina, S. tegulata and S. gottschei) were tested in detail. The total phenolic contents of the snails ranged from $32.3{\pm}1.0$ to $87.9{\pm}6.9$ mg gallic acid equivalent (GAE)/g dry sample. The $EC_{50}$ values for the 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities were $2.245{\pm}0.179$ mg/mL for S. coreana and $9.319{\pm}1.276$ mg/mL for S. gottschei and differed significantly (P<0.05) among the tested species. The superoxide dismutase (SOD)-like activity was highest for S. gottschei at 67.2% and lowest for S. forticosta at 4.7%. However, no significant differences among the species were recognized for the peroxynitrite anion scavenging activity. Comparing the correlation coefficients between the total phenolic contents and the DPPH radical and peroxynitrite anion scavenging activities, there was a low level relationship between each activity.
