Optimization of Medium Composition for Production of the Antioxidant Substances by Bacillus polyfermenticus SCD Using Response Surface Methodology

  • Production of the antioxidant substances by Bacillus polyfermenticus SCD was investigated using shake-flask fermentation. The one-factor-at-a-time method was first employed to determine the key ingredients for optimal medium composition, then further investigation of the medium composition was performed using response surface methodology (RSM). The antioxidant activity was measured using 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) assays. After screening various elements, fructose, tryptone, and $MgSO_4\;7H_2O$ were chosen as the main factors for study in the statistical experimental design. Central composite design (CCD) was then used to determine the optimal concentrations of these 3 components. Under the proposed optimized medium containing 2.8% fructose, 1.34% tryptone, 0.015% $MgSO_4\;7H_2O$), 0.5% NaCl, and 0.25% $K_2HPO_4$, the model predicted an antioxidant activity of 80.5% ($R^2=0.9421$. The actual experimental results were in agreement with the prediction.

Statistical Approach to Development of Culture Medium for Ansamitocin P-3 Production with Actinosynnema pretiosum ATCC 31565

  • The Plackett-Burman design and the response surface method (RSM) with a central composite design (CCD) were employed to develop a culture medium for ansamitocin P-3 production using Actinosynnema pretiosum ATCC 31565. Among the 11 nutrients tested using the Plackett-Burman design, two carbon sources, sucrose and dextrin, and two nitrogen sources, polypeptone and yeast extract, were selected. Optimization of the concentrations of the selected nutrients was then performed using RSM with CCD. After two rounds of RSM, the optimum concentrations ($\%w/v$) of sucrose, dextrin, polypeptone, and yeast extract were identified as 4.5, 4.5, 0.16, and 0.89, respectively. The maximum ansamitocin P-3 titer was 45.2 mg/l with the optimized medium, which was about 6 times higher than that (7.315 mg/l) obtained with an $R_{2}YE$ medium before optimization.

Construction of asm2 Deletion Mutant of Actinosynnema pretiosum and Medium Optimization for Ansamitocin P-3 Production Using Statistical Approach

  • Ansamitocin P-3 is a potent antitumor agent produced by A. pretiosum. A deletion mutant of A. pretiosum was constructed by deleting the asm2 gene, a putative transcriptional repressor. The deletion mutant showed a 9-fold enhanced ansamitocin P-3 productivity. The response surface method with central composite design was employed to further optimize the culture medium composition for ansamitocin P-3 production by the deletion mutant. The concentrations of four medium ingredients, dextrin, maltose, cotton seed flour, and yeast extract, which have been reported as major components for ansamitocin production, were optimized through a series of flask culture experiments. The optimum concentrations of the selected factors were found to be dextrin 6.0%; maltose 3.0%; cotton seed flour 0.53%; and yeast extract 0.45%. The maximum titer of ansamitocin P-3 was 78.3 mg/l with the optimized composition, about 15-folds higher than the unoptimized titer of 5.0 mg/l obtained with YMG medium.

Bioprocess Development for Production of Alkaline Protease by Bacillus pseudofirmus Mn6 Through Statistical Experimental Designs

  • A sequential optimization strategy, based on statistical experimental designs, is employed to enhance the production of alkaline protease by a Bacillus pseudofirmus local isolate. To screen the bioprocess parameters significantly influencing the alkaline protease activity, a 2-level Plackett-Burman design was applied. Among 15 variables tested, the pH, peptone, and incubation time were selected based on their high positive significant effect on the protease activity. A near-optimum medium formulation was then obtained that increased the protease yield by more than 5-fold. Thereafter, the response surface methodology(RSM) was adopted to acquire the best process conditions among the selected variables, where a 3-level Box-Behnken design was utilized to create a polynomial quadratic model correlating the relationship between the three variables and the protease activity. The optimal combination of the major medium constituents for alkaline protease production, evaluated using the nonlinear optimization algorithm of EXCEL-Solver, was as follows: pH of 9.5, 2% peptone, and incubation time of 60 h. The predicted optimum alkaline protease activity was 3,213 U/ml/min, which was 6.4 times the activity with the basal medium.

Biosurfactant Production from Novel Air Isolate NITT6L: Screening, Characterization and Optimization of Media

  • In this paper, an air isolate (NITT6L) has been screened based on hemolytic activity, emulsification activity, drop collapsing test, and oil displacement test, as well as lipase activity. It was found that strain NITT6L was able to reduce the surface tension of the medium from 61.5 to 39.83 mN/m and could form stable emulsions with tested vegetable oils. Morphological, biochemical, 16S rRNA sequencing analyses, and fatty acid methyl ester analysis using gas chromatography confirmed that the air isolate under study was Pseudomonas aeruginosa. Characterization of the biosurfactant using agar double diffusion assay revealed that the biosurfactant was anionic in nature, and CTAB-methylene blue assay and Molisch test revealed its glycolipid nature. The FT-IR spectrum confirmed that the crude biosurfactant was a rhamnolipid. Using unoptimized medium containing sucrose as the carbon source, the isolate was found to produce 0.3 mg/ml of rhamnolipid in batch cultivation (shake flask) at $37^{\circ}C$ and pH 7. Optimization of the medium components was carried out using design of experiments and the yield of rhamnolipid has been enhanced to 4.6 mg/ml in 72 h of fermentation.

Differential Evolution Algorithms Solving a Multi-Objective, Source and Stage Location-Allocation Problem

  • The purpose of this research is to develop algorithms using the Differential Evolution Algorithm (DE) to solve a multi-objective, sources and stages location-allocation problem. The development process starts from the design of a standard DE, then modifies the recombination process of the DE in order improve the efficiency of the standard DE. The modified algorithm is called modified DE. The proposed algorithms have been tested with one real case study (large size problem) and 2 randomly selected data sets (small and medium size problems). The computational results show that the modified DE gives better solutions and uses less computational time than the standard DE. The proposed heuristics can find solutions 0 to 3.56% different from the optimal solution in small test instances, while differences are 1.4-3.5% higher than that of the lower bound generated by optimization software in medium and large test instances, while using more than 99% less computational time than the optimization software.

Optimization of Extracellular Production of Recombinant Human Bone Morphogenetic Protein-7 (rhBMP-7) with Bacillus subtilis

  • Extracellular production of recombinant human bone morphogenetic protein-7 (rhBMP-7) was carried out through the fermentation of Bacillus subtilis. Three significant fermentation conditions and medium components were selected and optimized to enhance the rhBMP-7 production by using the response surface methodology (RSM). The optimum values of the three variables for the maximum extracellular production of rhBMP-7 were found to be 2.93 g/l starch, 5.18 g/l lactose, and a fermentation time of 34.57 h. The statistical optimization model was validated with a few fermentations of B. subtilis in shake flasks under optimized and unoptimized conditions. A 3-L jar fermenter using the shake-flask optimized conditions resulted in a higher production (413 pg/ml of culture medium) of rhBMP-7 than in a shake flask (289.1 pg/ml), which could be attributed to the pH being controlled at 6.0 and constant agitation of 400 rpm with aeration of 1 vvm.

  • In order to maximize the riboflavin production by a mutant strain Ashbya gosspyii, the optimization of medium and fed-batch fermentation were performed. As carbon sources, glucose and soybean oil were necessary for the riboflavin overproduction. Optimal concentrations of glucose and soybean oil in the flask cultures were found to be 3.0% and 0.5%, respectively, in a complex medium containing corn steep liquor(CLS) 1%. Among the various organic nitrogen sources tested, CSL was the most effective one both for the cell growth and riboflavin overproduction.

Optimization of Mass cultivation Media for the Production of Biomass and Natural Colourants from Two Marine Cyanobacteria by a Mathematical Design of Experiments

  • Optimization of chemicals in the large scale sea water medium and inoculum for biomass and natural colourants production in the marine cyanobacteria, Phomidium tenue BDU 46241 (phycoerythrin producer) and P.valderianum BDU 30501 (phycocyanin producer) was carried out by experiments in L8 orthogonal array. Mathematical analysis revealed the significance of these factors. The factor(s) that critically control the yield varied with the organism and the end-product further, the desirable level of these factors between the normal and a higher level tested was identified and improved media were evolved. In both cyanobacteria, higher level of $K_2$$HPO_4$, $NaNO_3$ and inoculum with normal level of ferric ammonium citrate was found to be desirable for biomass production and additionally, higher level of $MgSO_4$ for pigment production. The level of other factors varied with the organism and the end-product. Confirmation experiments showed that the clues obtained based on mathematical experimentation are valid. In P.tenue, the medium optimized for biomass production increased the yield of biomass by 495% and the medium optimized for phycoerythrin production increased the yield of biomass by 408% with 30% increase in phycoerythrin content of the biomass. Similarly in P.valderianum, the medium optimized for biomass production increased the yield of biomass by 224% and the medium optimized for phycocyanin production increased the yield of biomass by 143% with 44% increase in phycocyanin content of the biomass.

혼합영양 조건하에서 Haematococcus sp.의 배지 최적화 및 대사산물 생산 (Medium Optimization for Cell Growth and Metabolite Formation from Haematococcus sp. under Mixotrophic Cultivation)

  • Haematococcus sp.를 대상으로 mixotroph 형태에서 배양을 위한 배지 최적화 연구를 수행하였다. 기본 배지로는 modified MS 배지가 적절했으며, 탄소원으로는 glucose가, 그리고 농도는 10 g/l가 적합하였다. 질소원으로는 KNO3를 선정하였으며, 농도는 1.9 g/l이 최적이었다. 최적의 배지조건에서 Haematococcus sp.를 초기 접종량(0.18 g/l)로 접종하여 14일 후에 5.58 ± 0.25 g/l로 성장하였으며, 이는 건조 세포중량 기준으로 약 31배의 성장한 것이다. 이때 생성된 클로로필은 172.16 ± 7.79 mg/l였으며, 카로티노이드는 42.33 ± 1.91 mg/l이었다. 본 연구의 결과는 추후 미세조류 대량 배양과 대사산물의 생산에 이용가능한 기초자료가 될 수 있을 것이라 판단된다.
