Growth Inhibition of Food-borne Bacteria by Juice and Extract of Ginger and Garlic (생강과 마늘 즙 및 추출물의 식중독 세균에 대한 증식저해작용)

  • This study was conducted to understand the inhibitory garlic and ginger against the growth of food born pathogenic bacteria. Juice was prepared from the raw spices by using an electric homogenizer and membrane filter. Dry-powdered spices were treated with double distilled water and 70% ethanol to extract the antibacterial substances, respectively. Growth inhibitory effects of juice and extracts of the spices were monitored by using bacterial strains such as B. subtilis, L. moncytogenes, S. aureus,E. coli O157 : H7, P. aeruginosa, and S. typhimurium. On a solid medium where E. coli and S. aureus cells were grown, ginger juice formed inhibitory zone at the concentrations of 2-10% by paper disc test. The Bone formed by ginger juice was wider and more transparent than that formed by garlic juice on the same concentration.1. monocytogenes and B. subtilis were more sensitive to garlic juice than others, and stopped growing at 2% garlic juice. Ginger juice showed the growth inhibition by 30-50% at 1.0% concentration. On the contrast, P. aeruginosa which resisted to the garlic juice was the most sensitive to ginger juice. Water extract of garlic was not effective to inhibit the bacterial growth, while 2% ginger extract completely inhibited the growth of E. coli and S. aureus. Alcohol extract of ginger inhibited the growth of bacteria at the concentration of 0.3%. This growth inhibition is almost 10 times lower than that of the garlic extract. It was clear that ginger had more potential than garlic as an inhibitor to control the growth of the indicator organisms.

Filtration with Sedimentation II: New Analysis and Solid Mass Fraction of Suspension Entering Cake (침전이 수반되는 케이크 여과 II: 새로운 분석과 케이크에 도입되는 현탁액의 고체 질량분율)

  • Filtration on horizontal filter medium facing upward is accompanied by sedimentation. When sedimentation phenomenon is not considered, the average specific cake resistance by filtration could be in error more than two times. In the previous paper on this subject, it was insisted that the solid mass fraction of suspension changes in filtration period by sedimentation. But we could not find out the degree of change. At the first half of this paper, the average specific cake resistance measured by permeation will be proved to be exact in several means. The average specific cake resistance conventionally measured by filtration does not give correct results because it uses initial solid mass fraction of suspension. Then we calculated the changed solid mass fraction of suspension due to sedimentation during filtration by the experimental values obtained "filtration-permeation" method. We also determined the solid mass fraction at a certain moment of filtration from the suspension height and a new equation derived in this study. The two results were compared and proven that they almost matched.

Development of the continuous-wave $Cr^{4+}$: YAG laser with a broad tunability in the infrared region (적외선 영역에서 광대역으로 파장 가변되는 연속 발진 $Cr^{4+}$:YAG 레이저 개발)

  • We have investigated the output power performance of a continuous-wave chromium-doped YAG laser ($Cr^{4+}$:YAG laser), pumped by a Nd: YAG laser. With an output coupler of 1% transmission, we obtained as much as 400 mW of the average output power at 1.450 ${\mu}{\textrm}{m}$. The power slope efficiency of 8%, when the gain medium was cooled to 2$0^{\circ}C$. We tuned the wavelength of the $Cr^{4+}$:YAG laser, at pump power of 6 W, by using birefringent filter. As a result, lasing wavelength was tuned from 1.399 ${\mu}{\textrm}{m}$ to 1.532 ${\mu}{\textrm}{m}$, with the maximum output power of 340 mW at 1.4903 ${\mu}{\textrm}{m}$ and the linewidth was 0.21 nm.

Homogeneous Shape Forming of Alumina by Pressure-Vacuum Hybrid Slip Casting (가압-진공 하이브리드 주입성형에 의한 알루미나의 균질 성형)

  • Conventional methods for preparing ceramic bodies, such as cold isostatic pressing, gypsum-mold slip casting, and filter pressing are not completely suitable for fabricating large and thick ceramic plates owing to disadvantages of these processes, such as the high cost of the equipment, the formation of density gradients, and differential shrinkage during drying. These problems can be avoided by employing a pressure-vacuum hybrid slip casting approach that considers not only by the compression of the aqueous slip in the casting room (pressure slip casting) but also the vacuum sucking of the dispersion medium (water) around the mold (vacuum slip casting). We prepared the alumina formed bodies by means of pressure-vacuum hybrid slip casting with stepwise pressure loading up to 0.5 MPa using a slip consisting of 40 vol% solid, 0.6 wt% APC, 1 wt% PEG, and 1 wt% PVA. After drying the green body at $30^{\circ}C$ and 80% RH, the green density of the alumina bodies was about 56% RD. The sintered density of an alumina plate created by means of sintering at $1650^{\circ}C$ for 4 h exceeded 99.8%.This method enabled us to fabricate a $110{\times}110{\times}20$ mm alumina plate without cracks and with a homogeneous density, thus demonstrating the possibility of extending the method to the fabrication of other ceramic products.

  • MIRIS (Multipurpose Infra-Red Imaging System), is a small infrared space telescope which is being developed by KASI, as the main payload of Science and Technology Satellite 3 (STSAT-3). Two wideband filters (I and H) of the MIRIS enables us to study the cosmic infrared background by detecting the absolute background brightness. The narrow band filter for Paschen ${\alpha}$ emission line observation will be employed to survey the Galactic plane for the study of warm ionized medium and interstellar turbulence. The opto-mechanical design of the MIRIS is optimized to operate around 200K for the telescope, and the cryogenic temperature around 90K for the sensor in the orbit, by using passive and active cooling technique, respectively. The engineering and qualification model of the MIRIS has been fabricated and successfully passed various environmental tests, including thermal, vacuum, vibration and shock tests. The flight model was also assembled and is in the process of system optimization to be launched in 2012 by a Russian rocket. The mission operation scenario and the data reduction software is now being developed. After the successful mission of FIMS (the main payload of STSAT-1), MIRIS is the second Korean space telescope, and will be an important step towards the future of Korean space astronomy.

Gene filtering based on fuzzy pattern matching for whole genome micro array data analysis (마이크로어레이 데이터의 게놈수준 분석을 위한 퍼지 패턴 매칭에 의한 유전자 필터링)

  • Microarray technology in biological science enables molecular level observations and analyses on the biological phenomina by allowing to measure the RNA expression profiles in cells. Microarray data analysis is applied in various purposes such as identifying significant genes which react to drug treatment, understanding the genome scale phenomina. In drug response experiments, the microarray-based gene expression analysis could provide meaningful information. It is sometimes needed to identify the genes which shows different expression behavior for treatment group and normal group each other. When the normal group shows the medium level expression, it is not easy to discriminate the group just by expression level comparison. This paper proposes a method which selects group-wise representative values for each gene and sets the value range of the groups in order to filter out the genes with specific pattern. It also shows some experiment results.

Isolation, Purification, and Characterization of a Thermostable Xylanase from a Novel Strain, Paenibacillus campinasensis G1-1

  • High levels of xylanase activity (143.98 IU/ml) produced by the newly isolated Paenibacillus campinasensis G1-1 were detected when it was cultivated in a synthetic medium. A thermostable xylanase, designated XynG1-1, from P. campinasensis G1-1 was purified to homogeneity by Octyl-Sepharose hydrophobic-interaction chromatography, Sephadex G75 gel-filter chromatography, and Q-Sepharose ion-exchange chromatography, consecutively. By multistep purification, the specific activity of XynG1-1 was up to 1,865.5 IU/mg with a 9.1-fold purification. The molecular mass of purified XynG1-1 was about 41.3 kDa as estimated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Sequence analysis revealed that XynG1-1 containing 377 amino acids encoded by 1,134 bp genomic sequences of P. campinasensis G1-1 shared 96% homology with XylX from Paenibacillus campinasensis BL11 and 77%~78% homology with xylanases from Bacillus sp. YA-335 and Bacillus sp. 41M-1, respectively. The activity of XynG1-1 was stimulated by $Ca^{2+}$, $Ba^{2+}$, DTT, and ${\beta}$-mercaptoethanol, but was inhibited by $Ni^{2+}$, $Fe^{2+}$, $Fe^{3+}$, $Zn^{2+}$, SDS, and EDTA. The purified XynG1-1 displayed a greater affinity for birchwood xylan, with an optimal temperature of $60^{\circ}C$ and an optimal pH of 7.5. The fact that XynG1-1 is cellulose-free, thermostable (stability at high temperature of $70^{\circ}C{\sim}80^{\circ}C$), and active over a wide pH range (pH 5.0~9.0) suggests that the enzyme is potentially valuable for various industrial applications, especially for pulp bleaching pretreatment.

A Study on The Correction of The Channel Equalizer Decision Error Using Channel Estimator (채널추정기를 이용한 등화기 결정오류 정정 알고리즘에 관한 연구)

  • The process of transmitting messages through a medium with a limited bandwidth or channel dispersion inevitably involves signal distortion and noise influxes, resulting in the degradation of transmission quality due to the inter-symbol interference and additional noise, which increases the error rate of the received symbols. The main role of the equalizer is to remove the channel distortion and noise from the received signal to recover the transmitted messages. A number of studies on the equalizer composed of a combination of linear filter and error control coding have shown that they played a key role in enhancing the transmission efficiency, which is essential for digital communication. This paper proposes a new algorithm to correct the residual symbol errors in the message signal. In general, equalizer performance improvement algorithms were developed to improve the initial convergence speed or steady-state error. In this paper, however, the equalizer input signal was reconstructed using the equalizer decision symbols and the channel estimates to directly correct the decision errors by analyzing the statistical characteristics of the difference signal between the actual received signal and the reconstructed signal.

Valuation of Molecular Weight Distribution Charteristics of Soluble Microbial Products(SMP) Using the Batch Filtration Test (회분여과 방식을 통한 생물대사산물의 분자량 분포 특성 평가)

  • The formation of soluble microbial products(SMP) and molecular weight distribution on loading rate were observed in batch-type culture medium, which phenol was fed as a substrate. The molecular weight destribution was obtained by using 30K, 100K dalton and $0.45{\mu}$ membrane filters. When the phenol concentrationas a substrate was 120, 230 and 440 mg/L , the specific substrate utilization rate(q) showed 0.639, 1.281 and 1.744 mgTOC/mgMLSS/day, respectively. The endogenous biomass decay rate constant($K_d$) at each substrate concentration was 0.00536, 0.0661 and 0.0749($day^1$), respectively. The $SMP_e$ product rate constant($k_{SMP}_ e$) showed 0.006, 0.0058 and 0.0057($day^1$), respectively. The initial influent substrate during the course of time degraded and produced $SMP_s$. The $SMP_s$ was converted to the $SMP_{nd}$ and endogenous phase converted to the $SMP_e$ ingredients. The molecula weight distribution on loading rate was converted to a higher MW during the course of time.

Tunable Mechanically Formed Long-Period Fiber Gratings using Periodically Arrayed Metal Wires (금속선의 주기적인 배열을 이용하여 기계적으로 형성한 파장 가변 장주기 광섬유 격자)

  • In this paper, we have presented mechanically formed long-period fiber gratings using periodically arrayed brass wires with a $250-{\mu}m$ diameter and realized the function of current-controlled wavelength-tuning. With the thermo-optic effect of the surrounding medium around the fiber cladding, the continuous displacement of the resonance wavelengths is achieved through the resistant heat of the wire which changes the refractive index of surrounding material. The tunability for each mode as a function of an applied electrical power is investigated. When the glycerin is used as a thermo-optic material, the measured tuning ranges of $LP_{03}$ and $LP_{04}$ within electrical power of 20 W reach to 14 nm and 48 nm, respectively. The experimental results are in good agreement with the theoretical that which is analyzed by a geometric-optics approximation.
