The present study aimed at determining the effective dose of Folltropin, a follicle timulating hormone (FSH), on superovulation in indigenous cows of Bangladesh. Fifteen regularly cycling 5~7 years old dry cows, weighing 200~250 kg with 2.5~3.0 body condition scores (BCS) were divided into three groups (n=5). Individual groups were superovulated with 100, 200 or 300 mg of Folltropin per animal. The superovulation treatment was initiated at Day 10 or Day 11 of the estrous cycle (Day 0=day of estrus). Alfaprostol (6 mg) was injected to each cow 72 h after the initiation of superovulation treatment to induce eestrus. After confirming standing estrus, the cows were inseminated 2~3 times, 12 h apart, depending on the duration of estrus. At Day 6 or Day 7, individual horns of the uterus were flushed with 150~200 $m\ell$ of phosphate buffered saline supplemented with BSA (0.2%), penicillin (100 IU/$m\ell$) and streptomycin (100 $\mu\textrm{g}$/$m\ell$) using a two-way foley catheter. The embryos were concentrated, removing the excess medium through an embryo filter, and identified under a stereomicroscope. The identified embryos were collected, washed four times, evaluated and graded as excellent, good, fair or poor. The excellent, good and fair embryos were considered as transferable quality embryos. The mean (range). numbers of embryos collected vs. transferable quality embryos far 100, 200 and 300 mg of Folltropin were 4.5 (1~10) vs. 3.5 (1~8); 2.5 (1~4) vs. 1 (0~2) and 0.0 (0~0) vs. 0.0 (0~0), respectively, Folltropin at a dose of 100 or 200 mg produced suitable ovarian stimulation for superovulation in indigenous zebu cows of Bangladesh. A dose of 300 mg or more Folltropin consistently caused preovulatory corpora lutea formation in the ovaries and resulted in zero embryo recovery.
The World Wide Web has become an entrenched global medium for storing and searching information. Most people begin at a Web search engine to find information, but the user's pertinent search results are often greatly diluted by irrelevant data or sometimes appear on target but still mislead the user in an unwanted direction. One of the intentional, sometimes vicious manipulations of Web databases is Web spamming as Google bombing that is based on the PageRank algorithm, one of the most famous Web structuring techniques. In this paper, we regard the Web as a directed labeled graph that Web pages represent nodes and the corresponding hyperlinks edges. In the present work, we define the label of an edge as having a link context and a similarity measure between link context and the target page. With this similarity, we can modify the transition matrix of the PageRank algorithm. A motivating example is investigated in terms of the Singular Value Decomposition with which our algorithm can outperform to filter the Web spamming pages effectively.
Embryonic stem (ES) cells, derived from preimplantation embryo, are able to differentiate into various types of cells consisting the whole body, or pluripotency. In contrast, terminally differentiated cells do not usually alter their nature but frequently die or transform if they are exposed to inappropriate external stimulations. In addition to the plasticity, ES cells are expected to be different from terminally differentiated cells in very many ways, such as patterns of gene expressions, ability and response of the cells in confronting environmental stimulations, metabolism, and growth rate. As a model system to differentiate these two types of cells, human ES cells (MB03) and terminally differentiated cells (HeLa), we examined the ability of these two types of cells in confronting a severe oxidative insult, that is $H_2O$$_2$. Approximately 1$\times$10$^4$ cells were plated in 96 well plate and serum starved for overnight. The conditioned cells were exposed to a various concentration of $H_2O$$_2$ fur 24 hrs and loaded with neutral red (50$\mu\textrm{g}$/ml) for 4 hrs, washed with PBS for 2 min three times, and entrapped dye was dissolved out using acetic ethanol. Cytotoxicity was determined by reading the amount of dye in the medium using microplate reader. equipped with 575 nm filter. Relative amount of the dye entrapped within MB03 or HeLa were not significantly different when cells were exposed up to 0.4 mM $H_2O$$_2$. However, this sharply decreased down to 0.12% in HeLa cells when the cells were exposed to 0.8 mM $H_2O$$_2$, while it was approximately 54% in MB03 suggesting that this concentration of $H_2O$$_2$ is the defensive threshold for HeLa cells. The resistance to oxidative stimulation reversed, however, when cells were co-treated with BSO (L-buthionine- 〔S, R〕-sulfoximine) which chelates intracellular GSH. This result suggests that cellular GSH is the major defensive mechanism of human ES cells. Induction of enzymes involved in GSH metabolism and type of cell death is currently being studied.
A two stage ac drive configuration consisting of a single-phase line commutated rectifier and a three-phase voltage source inverter (VSI) is very common in low and medium power applications. The deterministic pulse width modulation (PWM) methods like sinusoidal PWM (SPWM) could not be considered as an ideal choice for modern drives since they result mechanical vibration and acoustic noise, and limit the application scope. This is due to the incapability of the deterministic PWM strategies in sprawling the harmonic power. The random PWM (RPWM) approaches could solve this issue by creating continuous harmonic profile instead of discrete clusters of dominant harmonics. Insufficient filtering at dc link results in the amplitude distortion of the input dc voltage to the VSI and has the most significant impact on the spectral errors (difference between theoretical and practical spectra). It is obvious that the sprawling effect of RPWM undoubtedly influenced by input fluctuation and the discrete harmonic clusters may reappear. The influence of dc link fluctuation on harmonics and their spreading effect in the VSI remains invalidated. A case study is done with four different filter capacitor values in this paper and results are compared with the constant dc input operation. This paper also proposes an ingenious RPWM, a ripple dosed sinusoidal reference-random carrier PWM (RDSRRCPWM), which has the innate capacity of suppressing the effect of input fluctuation in the output than the other modern PWM methods. MATLAB based simulation study reveals the fundamental component, total harmonic distortion (THD) and harmonic spread factor (HSF) for various modulation indices. The non-ideal dc link is managed well with the developed RDSRRCPWM applied to the VSI and tested in a proto type VSI using the field programmable gate array (FPGA).

This study was carried out to investigate the influence of different sampling methods on the measured concentrations of polycyclic aromatic hydrocarbons (PAH) both in the vapor and particulate phases, and to evaluate the effects of ambient temperature and sampling duration on the losses of PAH associated with particle samples due to volatilization. The experimental protocol of this study is consisted of two parts. The first part is related to the comparison of PAH concentrations measured by 4 different sampling systems, each of which involves different sampling principles for comparison purposes, including a medium-volume sampler with XAD-2 adsorbent, a high-volume sampler with polyurethane foam (PUF), two identical low-volume samplers: one with XAD-2 and the other with PUF, respectively. The second part of this study is to quantitatively estimate the losses of particulate PAH samples by volatilization during sampling, using two identical low-volume samplers: one was used for changing the filters every 3 hrs, 6 hrs, 12 hrs, and 24 hrs sampling, while the other was maintained for continuous 48 hours sampling without changing the filter. The concentrations of volatile PAH including 2-3 rings appeared to be significantly affected by the type of adsorbent. Measured levels of these lower-molecular weight PAH by XAD-2 adsorbent were much higher than those by PUF for both high-volume and low-volume sampling. PUF was found to give rise to unknown components that interfered with the PAH analysis, even after extensive clean-up. In addition, the retention efficiency of PUF for lower molecular weight PAH was subject to a large variation, being significantly influenced by sampling conditions such as ambient temperature. However, the effect of sampling methods with different adsorbents on the measured levels of semi-volatile compounds including 4 rings PAH such as fluoranthene, pyrene, BaA and chrysene, was not so much significant as more volatile PAH compounds. It was also clear from this study that volatilization losses of the semi-volatile PAH collected on the filters were inevitably occurred during prolonged sampling, and hence the results obtained from conventional sampling methods may not be expected to yield an accurate distribution of PAH between the vapor and particulate phases.

Enzyme extracts of cellulase [filter paper cellulase (FPase) and carboxymethyl cellulase (CMCase)], chitinase, and chitosanase produced by Aspergillus niger NRRL-567 were evaluated. The interactive effects of initial moisture and different inducers for FP cellulase and CMCase production were optimized using response surface methodology. Higher enzyme activities [FPase $79.24{\pm}4.22$ IU/gram fermented substrate (gfs) and CMCase $124.04{\pm}7.78$ IU/gfs] were achieved after 48 h fermentation in solid-state medium containing apple pomace supplemented with rice husk [1% (w/w)] under optimized conditions [pH 4.5, moisture 55% (v/w), and inducers veratryl alcohol (2 mM/kg), copper sulfate (1.5 mM/kg), and lactose 2% (w/w)] (p<0.05). Koji fermentation in trays was carried out and higher enzyme activities (FPase $96.67{\pm}4.18$ IU/gfs and CMCase $146.50{\pm}11.92$ IU/gfs) were achieved. The nonspecific chitinase and chitosanase activities of cellulase enzyme extract were analyzed using chitin and chitosan substrates with different physicochemical characteristics, such as degree of deacetylation, molecular weight, and viscosity. Higher chitinase and chitosanase activities of $70.28{\pm}3.34$ IU/gfs and $60.18{\pm}3.82$ to $64.20{\pm}4.12$ IU/gfs, respectively, were achieved. Moreover, the enzyme was stable and retained 92-94% activity even after one month. Cellulase enzyme extract obtained from A. niger with chitinolytic and chitosanolytic activities could be potentially used for making low-molecular-weight chitin and chitosan oligomers, having promising applications in biomedicine, pharmaceuticals, food, and agricultural industries, and in biocontrol formulations.

Objectives We investigated the genotoxic effects of 40-59 nm silver nanoparticles (Ag-NPs) by bacterial reverse mutation assay (Ames test), in vitro comet assay and micronucleus (MN) assay. In particular, we directly compared the effect of cytochalasin B (cytoB) and rat liver homogenate (S9 mix) in the formation of MN by Ag-NPs. Methods Before testing, we confirmed that Ag-NPs were completely dispersed in the experimental medium by sonication (three times in 1 minute) and filtration ($0.2{\mu}m$ pore size filter), and then we measured their size in a zeta potential analyzer. After that the genotoxicity were measured and especially, S9 mix and with and without cytoB were compared one another in MN assay. Results Ames test using Salmonella typhimurium TA98, TA100, TA1535 and TA1537 strains revealed that Ag-NPs with or without S9 mix did not display a mutagenic effect. The genotoxicity of Ag-NPs was also evaluated in a mammalian cell system using Chinese hamster ovary cells. The results revealed that Ag-NPs stimulated DNA breakage and MN formation with or without S9 mix in a dose-dependent manner (from $0.01{\mu}g/mL$ to $10{\mu}g/mL$). In particular, MN induction was affected by cytoB. Conclusions All of our findings, with the exception of the Ames test results, indicate that Ag-NPs show genotoxic effects in mammalian cell system. In addition, present study suggests the potential error due to use of cytoB in genotoxic test of nanoparticles.

The main purposes of this study are to investigate the distributional characteristics of polycyclic aromatic hydrocarbons (PAH) in the vapor and particulate phases in the ambient atmosphere, and to evaluate the effect of ambient temperature on the vapor-particle partitioning during the sampling period. A total of 64 samples were collected during a period of 1995 to 1996, using a medium-volume sampler with XAD-2 adsorbents and quartz fiber filters. Analyses of PAH were carried out using HPLC with UV and Fluorescence detections. In this study, a significant seasonal variation in the distributions was observed, reflecting the effect of ambient temperature on the vapor-particle partitioning of PAH. The relationship between the vapor-particle distributions of the 3 to 5 rings PAH and ambient temperature is considered to be well described using the Langmuir adsorption concept. The estimated empirical constants for each PAH in the relationship, particularly for the more volatile compounds, were also comparable with results from other studies. However, it is still difficult to accurately estimate the initial vapor-particle distribution of PAH in the ambient air, since it is not known to what extent the trapped vapours originated from the particles laden in the filter by being volatilized or from the air samples initially present in the vapour phase. The distribution factors for volatile PAH with 3 to 4 rings appeared to be comparable with those in the literature. It should be noted, however, that these distribution factors give information only about the distribution of PAH between the two phases under a specific sampling condition, and hence may provide only semi -quantitative information on the vapor-particle distributions in the atmosphere.
Topex/Poseidon sea surface heights are compared to tide gauge sea levels in the South Indian Ocean in the period of January 1993 to December 1995. A user's handbook (AVISO) for processing sea surface height data was used in this study. Topex/Poseidon sea surface heights were obtained from satellite data at the proximity of tide gauge stations. These data were reproduced by a linear interpolation with the interval of 10 days and were processed by the Gaussian filter with a 60-day window. The tide gauge sea levels were obtained in the same manner as the satellite data. The main results on RMS (Root-Mean-Square) and CORR (CORRelation coefficient) in our study were shown as follows: 1) on the characteristics between two data (in-situ and model data), the results (RMS=2.96 cm & CORR=$92\%$ in the Amsterdam plateau, and RMS=3.45 cm & CORR=$59\%$ in the Crozet plateau) of the comparison of Topex/Poseidon sea surface heights with tide gauge sea levels, which was calculated by in-situ data of obsewed station showed generally low values in RMS and high values in CORR against to the results (RMS=4.69 cm & CORR=$79\%$ in the Amsterdam plateau, and RMS= 6.29 cm & CORR= $49\%$ in the Crozet plateau) of the comparison of Topex/Poseidon sea surface heights with tide gauge sea levels, which was calculated by model data of ECMWF (European Center for Medium-range Weather Forecasting), and 2) on the characteristics between two areas (Kerguelen plateau and island), the results (RMS=3.28 cm & CORR= $54\%$ in the Kerguelen plateau) of open sea area showed low values in RMS and high values in CORR against to the results (RMS= 5.71 cm & CORR=$38\%$ in the Kerguelen island) of coast area, respectively.

"Wind and Ports" is a European project that has been carried out since 2009 to handle wind forecast in port areas through an integrated system made up of an extensive in-situ wind monitoring network, the numerical simulation of wind fields, the statistical analysis of wind climate, and algorithms for medium-term (1-3 days) and short term (0.5-2 hours) wind forecasting. The in-situ wind monitoring network, currently made up of 22 ultrasonic anemometers, provides a unique opportunity for detecting high resolution thunderstorm records and studying their dominant characteristics relevant to wind engineering with special concern for wind actions on structures. In such a framework, the wind velocity of thunderstorms is firstly decomposed into the sum of a slowly-varying mean part plus a residual fluctuation dealt with as a non-stationary random process. The fluctuation, in turn, is expressed as the product of its slowly-varying standard deviation by a reduced turbulence component dealt with as a rapidly-varying stationary Gaussian random process with zero mean and unit standard deviation. The extraction of the mean part of the wind velocity is carried out through a moving average filter, and the effect of the moving average period on the statistical properties of the decomposed signals is evaluated. Among other aspects, special attention is given to the thunderstorm duration, the turbulence intensity, the power spectral density and the integral length scale. Some noteworthy wind velocity ratios that play a crucial role in the thunderstorm loading and response of structures are also analyzed.
