A Study on Optical Design Factors by Artificial Recharge Performance (인공함양 주입성능평가에 의한 설계요소 산정 연구)

  • The design factors of artificial recharge are determined by considering the hydrogeological characteristics of the aquifer. The optimal design factors for artificial recharge were derived after performing the injection tests step by step for each injection type (vertical well, ditch and mixed type), which were built in the test site of the study area. It was analyzed that the difference in the injection effect according to the diameter of the injection well was not large, and the 100 mm well was evaluated as appropriate in consideration of the availability and economy of land use. Since the injection effect was well maintained even in the upper rock, the depth of the injection well was proposed for the alluvial layer and the upper rock layer. On the other hand, in four cases of filter media in the ditch, it was analyzed that the penetration efficiency and the hydraulic interference effect indicated excellent injection performance when a filter medium of 10 to 30 mm diameter was filled in the ditch. In addition, the proper spacing of the injection wells was analyzed as 9~12 m considering the interference efficiency. The interference efficiency attenuation coefficient per 1 m of hole spacing was calculated to be 1.75% in this area. In the future study, the artificial recharge design factors obtained in this stage are applied and verified on site construction and operation. Also it is expected to contribute to securing water in areas where there is always a lack of water.

Growth Acceleration and Acclimatization of In Vitro Plantlets derived from Apical Meristem of Sweet Potato (고구마의 경정조직 유래 기내 소식물체의 생장촉진과 순화)

  • The single node cuttings of sweet potato (cv. Mokpo #29) plantlets maintained in vitro were cultured with (MF+) or without membrane filter (MF-) under photomixotrophic (PM), hetrotrophic (HT) and autotrophic (AT) conditions. Shoot length was the greatest (11.9cm) in 3$0^{\circ}C$ (HT) treatment and it was the shortest (3.4 cm) in $25^{\circ}C$ (PM) treatment. Nodal explants cultured in 3$0^{\circ}C$ treatment looked more vigorous than those of $25^{\circ}C$ in appearance, and node number was the greatest (10.5 per plantlet) among the treatments. But plantlets grew in 3$0^{\circ}C$ (HT) treatment were observed all overgrown. The size in leaf area was about 2 times greater and shoot length was about 2 times shorter in PM than in HT condition. Percent dry matter of shoots was 5.9% (HT) and 7.4% (PM) in $25^{\circ}C$ treatment and 6.1% (HT) and 7.4% (PM) in 3$0^{\circ}C$ treatment. Plantlets cultured in the MF+ treatments were less succulent than those cultured in the MF- treatment. Vitrified plantlets were examinated 14.8% (both $25^{\circ}C$ and 3$0^{\circ}C$) in PM condition and 22.2% ($25^{\circ}C$) and 31.5% (3$0^{\circ}C$) in HT condition. Sucrose was necessary for the survival of in vitro plantlets. In the sucrose-free medium, explants cultured in the MF- had turned yellow and were dead after 30 days of culture. But explants cultured in the MF+ were alive and produced plantlets with shoot and root (AT). On the other hand, the survival of explants on the MS basal medium (sucrose-free and hormone-free) depended entirely upon the MF attachment.

Studies on the Submerged Culture of Lentinula edodes Mycelia in Brewer's Yeast Extract Medium (맥주효모 추출물을 이용한 표고버섯 균사체의 심부배양에 관한 연구)

  • Brewer's yeast extract can be used as a good substrate for the culture of Lentinula edodes Mycelia(LEM). We found that it was better to filter the extract through three kinds of sieves and then to heat for hydrolysis and concentration at $90^{\circ}C$ prior to use. Also the maximum condition for the growth of LEM was investigated. We found that addition of inorganic salts such as calcium enhanced the growth of LEM. On the other hand, addition of carbon and nitrogen sources to the medium did not affect, and even inhibited under certain conditions, the growth of LEM. The maximum temperature for the growth of LEM was around $25^{\circ}C$. Also, it grows better when agitated by shaking at 100 rpm for airration. The appropriate concentration of the extract to use was 10%. Under these conditions, LEM could reach to the confluency after cultivation of 12 days. Our extract formula seems better than other available media for LEM growth, producing higher crude protein content and better taste.

In vitro Development of Somatic Cell Nuclear Transferred Bovine Embryos Following Activation Timing in Enucleated and Cryopreserved MII Oocytes (탈핵 후 동결한 MII 난자의 활성화 시기가 체세포 핵치환 이후 소 난자의 체외발달에 미치는 영향)

  • This study was to evaluate the in vitro survival of bovine enucleated MII (eMII) oocytes according to minimum volume cooling (MVC) freezing method and activation timing, and their in vitro development after somatic cell nuclear transfer (SONT). in vitro matured bovine oocytes for 20 h were stained with 5 $\mu\textrm{g}$/$m\ell$ Hoechst, and their 1st polar body and MII plate were removed by enucleation micropipette under UV filter. Also, eMII oocytes were subjected to activation after (group II) and before (group III) vitrification in 5 ${\mu}{\textrm}{m}$ ionomycin added CRlaa medium for 5 min. For vitrification, eMll oocytes were pretreated with EG10 for 5 min, exposed to EG30 for 30 sec and then directly plunged into L$N_2$. Thawing was taken by 4-step procedures at 37$^{\circ}C$. Survived eMII oocytes were subjected to SONT with cultured adult bovine ear cells. Reconstructed oocytes were cultured in 10 $\mu\textrm{g}$/$m\ell$ of cycloheximide and 2.5 $\mu\textrm{g}$/$m\ell$ of cytochalasin D added CRlaa medium for 1 h, and then in 10 $\mu\textrm{g}$/$m\ell$ of cycloheximide added CRlaa medium for 4 h. Subsequently, the reconstructed oocytes were incubated for 2 days and cleaved embryos were further cultured on cumulus-cell monolayer drop in CRlaa medium for 6 days. Survival rates of bovine vitrified-thawed eMII oocytes in group II (activation after vitrification and thawing) and III (activation before vitrification) were 81.0% and 84.9%, respectively. Fusion rates of cytoplasts and oocytes in group II and III were 69.0% and 70.0%, respectively, and their results were not different with non-frozen NT group (control, 75.2%). Although their cleaved rates (53.4% and 58.4%) were not different, cytoplasmic fragment rate in group II (32.8%) was significantly higher than that in group III (15.6%)(P<0.05). Also, subsequent development rate into >morula in group II (8.6%) was low than that in group III(15.6%). However, in vitro development rate in group III was not different with that in control (24.8%). This result suggested that MVC method was appropriate freezing method for the bovine eMII oocytes and vitrified eMII oocytes after pre-activation could support in vitro embryonic development after SONT as equally well as fresh oocytes.

Feasibility Tests for Treating Fine Suspended Solids from Mining Drainage, using Various Media by Column Methods - A Case from H Coal Mine (광산배수 부유물질 저감을 위한 다양한 여과 매질의 특성 및 적용성 평가 - H 석탄광산 배수)

  • Fine suspended solids from mine drainage draw attentions due to their potential adverse influences on the water quality, such as increasing turbidity and degrading aesthetic landscape. Currently, sand filter beds are adapted in some mine drainage treating systems. However, more efficient system is in demand, as the existing sand beds reveal some problems, such as frequent maintenance intervals. Various filtering mediums including fly ash, mine tailing aggregates and the sand were tested for improving the current system, using column experimental set-up. Mine drainage samples were collected from the current treating systems in the abandoned H coal mine. The experiment was run for 7 days. Suspended solids recorded as 100.9 mg/L and the value exceeds the current standard, 30 mg/L. Sand was proved to still be the optimum medium for the fine suspended solids, compared to fly ash and fly ash + sand. Mine tailing aggregates were placed at the exit of the columns, substituting gravels. The tailing aggregates is made by mine tailings and clay. Sand bed filters can also be improved by mixing granular activated carbon, which was found to be economical and efficient in the batch experiment, conducted at the same time.

  • This study was carried out in order to reduce the installation expense of heating system for greenhouse comparing to geothermal heat pump and develope the coefficient of performance (COP) for a heat pump. For getting plenty of heat flux from geothermal energy. Surface water in river channel was used for getting a lots of geothermal heat by penetrating water through underground soil layer of the river bank that make heat transmission to passing water. The range of water temperature after the process of Ground filtration is 13~18 degrees celsius which is very similar to low heat source of geothermal heat pump system and the plenty amount of heat source from that make the number of geothermal heat exchanging hole and the expense for geothermal heat exchanger construction reduced. Drainage well is also used for returning filtration water to the aquifer that keep the water good recirculation from losing geothermal heat and water resource. For the COP improvement of Heat pump, thermal storage tank with separating insulation plate according to the temperature difference make the COP of Heat pump that is similar to thermal storage tank with diffuser. Developed thermal storage tank make construction expense cheaper than customarily used one's. and that sand filter and oxidation sand (FELOX) are going to be used for improving ground filtration water quality that make heat exchanger efficiency better. All above developed component skill are going to be set on the Ground filtration water source heat pump system and applied for medium, large scale for protected greenhouse in riverside area and on-site experiment is going to do for optimizing the heating system function and overcome the problem happening in the process of on-site application afterward.

Rapid Detection of Escherichia coli in Fresh Foods Using a Combination of Enrichment and PCR Analysis

  • The objective of this study was to determine the minimum enrichment time for different types of food matrix (pork, beef, and fresh-cut lettuce) in an effort to improve Escherichia coli detection efficiency. Fresh pork (20 g), beef (20 g), and fresh-cut lettuce (20 g) were inoculated at 1, 2, and 3 Log CFU/g of Escherichia coli. Samples were enriched in filter bags for 3 or 5 h at $44.5^{\circ}C$, depending on sample type. E. coli cell counts in the samples were enriched in E. coli (EC) broth at 3 or 5 h. One milliliter of the enriched culture medium was used for DNA extraction, and PCR assays were performed using primers specific for uidA gene. To detect E. coli (uidA) in the samples, a 3-4 Log CFU/mL cell concentration was required. However, E. coli was detected at 1 Log CFU/g in fresh pork, beef, and fresh-cut lettuce after 5, 5, and 3-h enrichment, respectively. In conclusion, 5-h enrichment for fresh meats and 3-h enrichment for fresh-cut lettuce in EC broth at $44.5^{\circ}C$, and PCR analysis using uidA gene-specific primers were appropriate to detect E. coli rapidly in food samples.

조류를 이용한 유기성 폐수 처리 시스템과 물벼룩 성장 조건

  • Food wastewater eluted from the three-stage methane fermentation system developed in this lab showed high concentrations of TCOD, BOD, T-N and T-P. Because the effluent of biological filter chamber (BFC) still had high concentration of nitrogen and organic material, the effluent was treated with algal periphyton system using algae. The removal rates of COD, T-N and T-P wer 96, 98 and 91%, respectively, in this system. The grown algae could digested byy waterfleas using the ecological food chain system. Food wastewater is better than algal culture medium for growth of waterflea, Moima Macrocopa. During 12days, the individual of waterflea increased to 180 in the food wastewater containing a T-N concentration of 150 mg/ ${\ell}$.

  • Human chorionic gonadotropin (hCG) is a heterodimeric glycoprotein hormone consisting of non-covalently linked two subunits, the ${\alpha}$ and ${\beta}$ subunit. It has been used as a infertility drug for ovulation to mimic luteinizing hormone $(LH).^{1)}$ A stable cell line was established by transfection of Rc/CMV-i-dhfr-hCG, expression vector containing hCG ${\alpha}-$ and ${\beta}-genes$, into dihydrofolate reductase-deficient CHO cells and subesquent methotrexate-mediated gene amplification. Anchorage-dependent CHO cells were adapted into a serum-free and/or animal component-free suspension medium through gradual serum weaning for the hCG production. The established cell line showed typical morphological characteristics and growth profile of CHO cells, and could produce FSH with passage-to-passage consistency. The high density perfusion culture of the CHO cells was carried out in Celligen Plus bioreactor equipped with a spin-filter as a internal cell retention device. The cell density reached up to $>1x10^{7}$ cells/ml in less than 7 days and a perfusion-control strategy based on cellular consumption rates of glucose was $established.^{2)}$ Biologically active recombinant hCG was purified by a series of chromatographic steps including anion exchange chromatography and hydrophobic interaction chromatography to homogeneity. The highly purified recombinant hCG was characterized for physicochemical, immunological and biological properties.

Effect of Step Pressure on Shape Forming of Alumina by Pressure-Vacuum Hybrid Slip Casting (가압-진공 하이브리드 주입 성형에 의한 알루미나의 성형에 미치는 다단 가압의 영향)

  • Conventional cold isostatic pressing, slip casting, and filter pressing are not completely suitable for fabricating large plates because of disadvantages such as the high cost of equipment and formation of density gradient. These problems could be avoided by employing pressure-vacuum hybrid slip casting (PVHSC). In the PVHSC, the consolidation occurs not only by the compression of the slip in casting room, but also by vacuum sucking of the dispersion medium around the mold. We prepared the alumina bodies by the PVHSC in a static- or stepwise-pressure manner for loading up to 0.5 MPa using an aqueous slip. The green bodies were dried at $30^{\circ}C$ with 40 ~ 80% relative humidity. Under static pressure, casting induced a density gradient in the formed body, resulting in cracking and distortion after the firing. However, the stepwise pressure loading resulted in green bodies with homogeneous density, and the minimization of the appearance of those defects in final products. Desirable drying results were obtained from the cast bodies dried with 80% RH environment humidity. When sintered at $1650^{\circ}C$ for 4 h, the alumina plate made by stepwise-pressure casting reached full density (> 99.7% relative density).
