Effects of Chrysanthemum indicum L. Extract on the Growth and Differentiation of Osteoblastic MC3T3-E1 Cells (감국(Chrysanthemum indicum L.) 추출물이 MC3T3-E1 조골세포의 증식 및 분화에 미치는 영향)

  • Chrysanthemum indicum L. (Asteraceae) is a common traditional herbal medicine used for the treatment of inflammation, hypertension, and respiratory diseases due to its strong antagonistic function against inflammatory cytokines. In this study, the effects of Chrysanthemum indicum L. extract (CIE) on the function of osteoblastic MC3T3-E1 cells and the production of local factors in osteoblasts were investigated. CIE (100 ${\mu}g/mL$) significantly increased the growth of MC3T3-E1 cells and caused a significant elevation of alkaline phosphatase (ALP) activity, and the deposition of collagen and calcium in the cells (p<0.05). The effect of CIE in increasing cell growth, ALP activity, and collagen content was completely prevented by the presence of 1 ${\mu}M$ tamoxifen, suggesting that CIE's effect might be partly involved in estrogen-related activities. These results indicate that the enhancement of osteoblast functionality by CIE may prevent osteoporosis and inflammatory bone diseases.

Effect of Bu-Zhong-Yi-Qi-Tang on Proliferation of T Cells (보중익기탕의 T세포 증식 유도 효과)

  • Bu-Zhong-Yi-Qi- Tang extracts is a traditional oriental medicine in a mixture type exhibiting strong anti-bacterial, analgesic, and chemopreventive activities. In this study, we have evaluated effects of the total and polysaccharide fraction of Bu-Zhong-Yi-Qi- Tang extracts on the T cell proliferation, cytokine production, and induction of IL-2 receptor and MHC class n. For this experiment, we established CD4$^{+}$ CD8$^{[-10]}$ T cell line producing IL-2 and IFN-${\gamma}$ when stimulated with ovalbumin antigen in the presence of antigen presenting cells. The significant effect of Bu-Zhong-Yi-Qi-Tang on antigen-induced T cell proliferation in the presence of antigen presenting cells was observed. The proliferation and IFN-${\gamma}$ production of T cells was increased in a dose dependent manner, and expression of IL-2 receptor on T cells and MHC class n molecule on antigen presenting cells was also induced in the presence of Bu-Zhong-Yi-Qi-Tang polysaccharide fraction. It was demonstrated that polysaccharide fraction of Bu-Zhong-Yi-Qi-Tang stimulates the antigen-induced T cell proliferation and the production of IFN-${\gamma}$ possibly through the increase of IL-2 receptor and MHC class n expression. Therefore Bu-Zhong-Yi-Qi-Tang can be regarded as a natural and useful immunomodulator having a relatively nonotoxic property. Further studies are needed to better characterize the nature of Bu-Zhong- Yi-Qi-Tang extract.

Anti-inflammatory effects of Rubus coreanus Miquel through inhibition of NF-${\kappa}B$ and MAP Kinase

  • BACKGROUND/OBJECTIVES: Rubus Coreanus Miquel (RCM), used as a traditional Korean medicine, reduces chronic inflammatory diseases such as cancer and rheumatoid arthritis. However, its mechanism has not been elucidated. In this study, we examine the anti-inflammatory effects of RCM and their possible mechanisms using RAW 264.7 cells. MATERIALS/METHODS: Unripe RCM ethanol extract (UE), unripe RCM water extract (UH), ripe RCM ethanol extract (RE), and ripe RCM water extract (RH) were prepared. Inflammatory response was induced with LPS treatment, and expression of pro-inflammatory mediators (iNOS, COX-2, TNF-${\alpha}$, IL-$1{\beta}$, and IL-6) and NO and $PGE_2$ productions were assessed. To determine the anti-inflammatory mechanism of RCM, we measured NF-${\kappa}B$ and MAPK activities. RESULTS: UE and UH treatment significantly reduced NF-${\kappa}B$ activation and JNK and p38 phosphorylation and reduced transcriptional activities decreased iNOS, COX-2, and pro-inflammatory cytokines expressions, and NO and $PGE_2$ productions. RE and RH treatments reduced IL-$1{\beta}$ and IL-6 expressions through suppressions of JNK and p38 phosphorylation. CONCLUSIONS: In this study, we showed that RCM had anti-inflammatory effects by suppression of pro-inflammatory mediator expressions. Especially, unripe RCM showed strong anti-inflammatory effects through suppression of NF-${\kappa}B$ and MAPK activation. These findings suggest that unripe RCM might be used as a potential functional material to reduce chronic inflammatory responses.

Effects of Baicalein on Picryl Chloride-induced Contact Dermatitis in BALB/c Mice (Baicalein의 BALB/c Mice에서의 접촉성 피부알레르기 예방효과)

  • Contact dermatitis is induced by allergy response from a lot of chemical substances brought on by environmental contamination. In this study, we investigated allergy prevention effect of baicalein which is Scutellaria radix extract. Scutellaria radix is the dried roots of Scutellaria baicalensis Georgi (Labiatae). It has been used in oriental traditional medicine for treatment of fever, diarrhea and inflammation. We have induced allergy by PCL after oral administration of 0 mg/kg, 50 mg/kg or 100 mg/kg baicalein dose for 8 times during 8 days in BALB/c mice and measured ear thickness, IgE level, histamine level, mast cell number and histopathological changes. The ear thickness of the 100 mg/kg baicalein group was thinner than that of the 0 mg/kg baicalein group. IgE level and histamine level of the 100 mg/kg baicalein group were lower than those of 0 mg/kg baicalein group. The number of mast cells of 0 mg/kg baicalein group more than that of 100 mg/kg baicalein group in the ear tissue. In addition, intradermal edema with prominent inflammatory cell infiltration was observed at 0 mg/kg baicalein group more than 100 mg/kg baicalein group in the ear tissue. These results suggest that baicalein has allergy prevention effect in BALB/c mice.

The Comparison between Hot-Water Extracts and Microwave Extracts of Scutellaria radix for Antioxidant and Neuroprotective Effects (가열 추출법과 마이크로웨이브 추출법을 이용한 황금 물 추출물의 뇌세포 보호 및 항산화 효과 비교 연구)

  • Microwaves are non-ionizing electromagnetic waves of frequency between 300MHz to 300GHz and positioned between the X-ray and infrared rays in the electromagnetic spectrum. In recent years, the use of microwave for extraction of ingredient from plant material has shown remarkable research interest and potential. Scutellaria radix has been used as a traditional medicine for a variety of diseases. It has been reported to exert beneficial health effects, such as anti-bacterial, antiviral, anti-inflammatory, and free-radical scavenging. Oxidative stress or the accumulation of reactive oxygen species (ROS) leads neuronal cellular death and dysfunction, and it contributes to neuronal degenerative disease such as Alzheimer's disease, Parkinson's disease and stroke. In this study, we aimed to compare the neuroprotective and antioxidant effect of Scutellaria radix extracted by different methods using hot-water extraction (SBE-DW) or microwave extraction (SBE-DW-MW). As a result, we first examined HPLC analysis of hot-water and microwave extracts of Scutellaria radix. The hot-water and microwave extracts of Scutellaria radix showed the discernible difference patterns of HPLC analysis. Microwave-water extracts of Scutellaria radix increased DPPH radical scavenging activity more than hot-water extraction. Microwave-water extracts of Scutellaria radix also showed neuroprotective effects and ROS inhibition against glutamate-induced oxidative stress in mouse hippocampal HT22 cells, but hot-water extraction not showed. In addition, the phosphorylation of MAPKs induced by glutamate insult was prevented by microwave-water extracts of Scutellaria radix. Thus, these results suggested that microwave extraction can be utilized for improving the extraction efficiency and biological activity of Scutellaria radix.

Protective Effects of Pyrrosiae Folium on the 2% Glucose-Induced Toxicity in Caenorhabditis elegans (석위가 예쁜꼬마선충에서 Glucose로 유도된 독성에 미치는 영향)

  • Pyrrosia lingua which belongs to Polypodiaceae has been used as a traditional medicine for the treatment of urinary system inflammation, urination disorder, and bronchitis. However, there are not enough phytochemical and pharmacological studies of P. lingua up to now. Here in this study, the protective effect of MeOH extract of whole plant of Pyrrosia lingua (MPL) against 2% glucose-induced toxicity was investigated using Caenorhabditis elegans (C. elegans) model system. We found that MPL significantly extended the lifespan of wild-type nematode under normal culture condition. MPL also effectively recovered the decreased lifespan caused by 2% glucose-toxicity. In addition, MPL efficiently attenuated the increased glucose concentration inside of nematode. Further studies evaluating diabetes-related factors revealed that MPL reduced both intracellular ROS and lipid accumulation which were up-regulated under 2% glucose supplement condition. Our data also showed that MPL improved the 2% glucose-induced shortened body movement of nematode. Lastly, we carried out genetic studies using several single gene knockout mutants to establish the possible target of MPL. Our results demonstrated that genes such as daf-2 and daf-16 were responsible for the protective activity of MPL against 2% glucose-induced toxicity. These results indicate that MPL exerts protective action against 2% glucose via regulation of insulin/IGF-1 sinaling pathway and FOXO activation.

Developing Digital Library Collection Using Citation and Homepage Information (인용정보와 연구자 홈페이지를 이용한 디지털 도서관 장서개발 방안 연구)

  • Nowadays the information environment enables users to access the traditional library collection as well as various digital information resources. In this rapidly changing environment, the use of digital information resources such as web sites, data, and homepages has increased. In this research, highly-cited authors' research outcomes as well as the research outcomes of the people, who cited the highly-cited authors' works, were extracted then compared with information stored in the medical colleges' digital libraries and the academic information portals in the clinical medicine area by using the citation information provided by Essential Science Indicators from ISI Web of knowledge. Out of 10,000 authors,146 people's homepages, which present research outcomes, were analyzed. The research outcomes listed in the homepages included journal papers, monographs, conference proceedings, and lecture notes. About 15% of the journal papers, 32% of the monographs, 48% of the conference proceedings, and 100% of lecture notes were accessible only through the homepages. The research outcomes accessible from the homepages were almost analogous to the ones available through the medical college's digital libraries and the academic information portals. Therefore, the digital library collection will be improved and expanded quantitatively and qualitatively by collecting and using the information in the homepages of the prestigious researchers.

Comparison of the Anti-inflammatory Effects of the Extracts from Rubus coreanus and Rubus occidentalis (토종 복분자와 외래종 복분자 추출물의 항염증효과 비교)

  • The dried fruit of the Rubus coreanus, which is well-known in Korea and referred to as 'Bokbunja,' has been employed as a traditional medicine for centuries. This crude drug has been utilized in Korea for the management of impotence, spermatorrhea, enuresis, asthma, and allergic diseases. Our previous study demonstrated that the ethanol extracts of R. coreanus have anti-inflammatory effects. The principal objective of the present study was to conduct a comparison of the anti-inflammatory effects of the ethanol extracts of R. coreanus and R. occidentalis; here, we tested the unripe (URCE), half-ripened (HRCE), and ripened fruits (RCE) of R. coreanus, and the unripe (UROE), half-ripened (HROE), and ripened fruits (ROE) of R. occidentalis. We found that URCE, UROE, HRCE, and HROE reduced the production of nitric oxide and prostaglandin $E_{2}$ as well as pro-inflammatory cytokines in lipopolysaccharide-stimulated RAW264.7 murine macrophages. Interestingly, the R. coreanus extracts showed stronger inhibitory effects on the production of these inflammatory mediators than the R. occidentalis extracts.

Experimental studies on antitumor effects and immune responses of Eunwhasagantang and Eunwhasagantangganokyong (은화사간탕(銀花瀉肝湯)과 은화사간탕가녹용(銀花瀉肝湯加鹿茸)의 항암효과(抗癌效果)와 면역반응(免疫反應)에 관(關)한 연구(硏究))

  • This study was performed to investigate the effects of Eunwhasagantang and Eunwhasagantangganokyong on the viability of tumor cells in vitro(MTT assay), on antitumor effects after Sarcoma-180 cells transplantation into the peritoneal cavity or left groin, and on decreased immune responses in mice induced by methotrexate. The extracts of its herbal medicines were orally administered for 14 or 21 days. To evaluate the effects of the Eunwhasagantang and Eunwhasagantangganokyong many items such as 50% inhibitory concentration($IC_{50}$), mean survival days, tumor and body weight for antitumor effects, and delayed type hypersensitivity, hemagglutinin titer, hemolysin titer, rosette forming cells, natural killer cell activity, lymphocyte transformation, productivity of interleukin-2 and phagocytic activity for immune responses were measured in ICR mice. The results were obtained as follows; 1. $IC_{50}$ of Eunwhasagantang treated group was 0.000204mg/ml on SNU-396 and that of Eunwhasagantangganokyong treated group was 0.000103mg/ml on SNU-1, those results indicate that the medicine has high antitumor activity. 2. Mean survival times in Eunwhasagantang and Eunwhasagantangganokyong treated groups were slightly increased with no significance, as compared with the control group. 3. Tumor weight in Eunwhasagantang and Eunwhasagantangganokyong treated group was depressed, as compared with the control group(p<0.01). 4. Body weight in Eunwhasagantang and Eunwhasagantangganokyong treated group was significantly increased, as compared with the control group(p<0.05). 5. Delayed type hypersensitivity in Eunwhasagantang and Eunwhasagantang-ganokyong treated group was slightly decreased with no significance, as compared with the control group. 6. Hemagglutinin titer in Eunwhasagantang and Eunwhasagantangganokyong treated group was slightly increased with no significance, as compared with the control group. 7. Hemolysin titer only in Eunwhasagantang treated group was significantly increased, as compared with the control group(p<0.01). 8. Rosette forming cells only in Eunwhasagantangganokyong treated group was slightly increased with no significance, as compared with the control group. 9. In the NK cell activity, the ratio of effector cells and target cells of the Eunwhasagantang treated group was significantly increased(p<0.01) in case which the ratio was 100: 1, and that of the Eunwhasagantangganokyong treated group was significantly increased(P<.01, p<0.05) in case which the ratio was 100:1, 50:1, as compared with the control group. 10. Lymphocyte trasnformation in Eunwhasagantang and Eunwhasagantangganokyong treated group was significantly increased, as compared with the control group(p<0.01). 11. Interleukin-2 in Eunwhasagantang and Eunwhasagantangganokyong treated group was significantly increased, as compared with the control group(p<0.05, p<0.01). 12. Phagocytic activity in Eunwhasagantang and Eunwhasagantangganokyong treated group was significantly increased, as compared with the control group(p<0.05). According to the above results, it could be suggested that Eunwhasagantang and Eunwhasagantangganokyong have prominent antitumor effects, and enhance both cellular and humoral immunity.

Hematological Effects of Water Extracts of Cham-Dang-Gui on Dietary Induced Iron Deficient Anemia Rat (식이로 유도된 철 결핍성 빈혈 흰쥐에서 참 당귀 열수추출물이 혈액학적 빈혈지표에 미치는 영향)

  • Cham-dang-gui (Angelica gigas nakai) has been used in traditional Korean medicine to treat hemotological disorders. The purpose of this study is to investigate the hematological effects of water extracts of Cham-dang-gui in rats with anemia induced by iron-free diets. Rats were divided into two groups, a regular diet group (RD) and an iron-free diet group (FeD). The iron-free diet group was then subdivided into the following three treatment groups: saline (1.0 ml/kgBW/day, FeDS), Cham-dang-gui (1.0 g/kgBW/day, FeDA), and iron (iron succinylate 14 mg/kgBW/day, FeDFe) groups. Rats were fed an iron-free diet for 6 weeks to induce iron-deficient anemia, and subsequently underwent the treatments, during which they were fed an iron-free diet for 3 weeks followed by a regular diet for 3 weeks. Body weights of the iron-diet groups (FeDS, FeDA, FeDFe) were lower than that of RD group. The blood levels of Hgb, Hct, MCV, MCH and TIBC were significantly higher in RD and FeDFe groups than those of other groups. The water extract of Cham-dang-gui with iron-free diet has no hematological effects. A regular diet followed by iron-free diets significantly increased blood levels of Hgb, Hct, and MCH.
