Validation Study for Image Performance of I-131 Using GATE Simulation Program (GATE 시뮬레이션 프로그램을 이용한 I-131의 영상 특성의 타당성에 관한 연구)

  • The purpose of this study was to validate for GATE (Geant4 Application for Tomographic Emission) simulation by comparing the results of GATE simulation and experiment in real SPECT system. Futhermore, we want to prove that it is possible that the quantitative research of gamma camera/SPECT imaging for therapeutic radio isotope by using GATE simulation. In this study, the SPECT system on simulation referred to the parameters of Stream-R Forte version 1.2 (Philips Medical System, Best and Heerlen, Netherlands). To understand the I-131 image of gamma camera/SPECT system, we acquired the energy spectrum and measured the full width at half maximum (FWHM) which comes from line spread function (LSF) with and without scatter material in real SPECT system. And to compare with experiment, we also measured the FWHM and acquired the energy spectrum without scatter material in GATE simulation. As a result, without scatter material, the energy peak was almost same location, which are located nearby 364 keV, and other spectrum factors are same tendency in both cases. The FWHM was increased by increasing the distance of source to detector, and the error rate was approximately 3.8%. When we used the line source with scatter material, energy spectrum also indicated similar tendency in both cases. As you confirmed earlier, GATE simulation included real instrument and radioisotope characters for therapeutic radioisotope. Therefore this result that it was possible that various quantitative study for therapeutic radioisotope imaging in gamma camera/SPECT using GATE simulation.

  • PET/CT is a machine for imaging in vivo functions or metabolic activities after the administration of radiopharmaceuticals labeled with radioisotope emitting positrons in the body. Recently the number of PET/CT installed in Korean medical institutions is increasing rapidly. In response, the number of PET/CT tests to be used in the diagnosis and treatment of tumors is also increasing every year, and this is increasing the necessity for developing the methods of PET/CT performance evaluation and quality control. Among the test items for the performance evaluation and quality control of PET/CT suggested in NU 2-2007, this study examined spatial resolution test, sensitivity test, image quality, attenuation accuracy & scatter correction test, scatter fraction, count losses and randoms test and accuracy( correction for count losses and randoms).

Proposal of a New Estimation Method of Colonization Rate of Arbuscular Mycorrhizal Fungi in the Roots of Chengiopanax sciadophylloides

  • This study proposed a rapid method to quantify the colonization rate of arbuscular mycorrhizal fungi (AMF) in plant roots. The method involved the use of an image analysis software (WinRHIZO Pro). The colonization rate is defined as the ratio of the fungal body to the plant root area in a micrograph. Three seedlings of Chengiopanax sciadophylloides, a woody species that accumulates radiocesium, were collected from a secondary forest in the Yamakiya district of Kawamata, Fukushima Prefecture during May-September 2014. The colonization of AMF structures was examined under a light microscope, and the percentage of colonization was determined using the WinRHIZO method. The superiority of the new method was verified by comparing with a modified grid-line intersect method. The colonization of AMF was confirmed in all the seedlings, and a significant coefficient of determination ($R^2=0.94$) was found with both the methods. The results suggested that the WinRHIZO method is reliable for estimating the colonization of AMF in C. sciadophylloides.

  • The evaluation of radioactivated components of heavy-ion accelerator facilities affects the safety of radiation management and the exposure dose for workers. and this is an important issue when predicting the disposal cost of waste during maintenance and dismantling of accelerator facilities. In this study, the FLUKA code was used to simulate the proton treatment device nozzle and classify the radio-nuclides and total radioactivity generated by each component over a short period of time. The source term was evaluated using NIST reference beam data, and the neutron flux generated for each component was calculated using the evaluated beam data. Radioactive isotopes caused by generated neutrons were compared and evaluated using nuclide information from the International Radiation Protection Association and the Korea Radioisotope association. Most of the nuclides produced form of beta rays and electron capture, and short-lived nuclides dominated. However, In the case of 54Mn, which is a radioactive product of iron, the effect of gamma rays should be considered. In the case of tritium generated from a material with a low atomic number, it is considered that handling care should be taken due to its long half-life.

  • Microsurgical vascularized bone transfer has the disadvantages of limitation of available donor sites, loss of donor function, and the possibility of donor site defects or deformity. To overcome these shortage of current microsurgical tissue transfer, the method of creating the neovascularized free flap has been introduced. Potentially, this technique must be an innovation in providing the free vascularized bone grafts that are not limited by natural vascular anatomy. But, as could be imagined technique resulted in unavoidable donor bone defect and additional operation for harvesting the autologous bone. The purpose of this study was to evaluate the efficacy of demineralized allogeneic bone as a possible substitute for autologous bone in fabricating the neo-osseous flap. By histologic, microangiographic and radioisotope method, the viability and vascularity of neo-osseous flap, which has been fabricated using allogeneic bone or autologous bone, was assessed in rat model. After 6 weeks, demineralized allogeneic bone showed consistent bone formation and neovascularization. The clinical and microscopic findings of demineralized allogeneic bone group were inferior to those of autogenous bone with regard to bone regeneration. The amount of bone blood floow per dry weight of demineralized allogeneic bone group was significantly higher than that of autogenous bone, even higher that of control intact iliac bone. In conclusion, findings supported that allogeneic bone could be the potential substitute for autologous bone source in creating a prefabricated neo-osseous flap.

  • The aim of this work is to study the radiation shielding properties of cement samples with waste glass incortated into its composition. The mass attenuation coefficient (MAC) of the samples were experimentally determined to evaluate their radiation shielding ability. The experimental coefficient was evaluated using NaI detector for gamma energies between 59.53 keV and 1408.01 keV using different radioactive point sources Am-241, Eu-152, Co-60, and Cs-137, and the gamma transmission parameters half-value layer, mean free path, and transmission factor were calculated. The theoretical coefficient of the composites was determined using Geant4 and XCOM software. The results were also compared against Geant4 and XCOM simulations by calculating the relative deviation between the values to determine the accuracy of the results. In addition the mechanical properties (including Compressive and porosity) as well as the thermogravimetric analysis were tested for the present samples. Overall, it was concluded that the cement sample with 50% waste glass has the greatest shielding potential for radiation shielding applications and is a useful way to reuse waste glass.

Cu-64 as a Cancer Theranostics Agent

  • Theranostics, a composite word of therapy and diagnosis, is known as personalized medicine and the concept of diagnosis and treatment at the same time. In nuclear medicine, it means performing both therapeutic and diagnostic radioisotope therapy using the same target molecule. The increased production and utilization of 64Cu opens a new era of theranostics. The studies introduced here have shown that 64CuCl2 and various compounds or biomolecules labeled with 64Cu are unique radiopharmaceuticals with physiological properties suitable for use as diagnostic and therapeutic agents. So far, these two abilities have been described only for radioactive iodine. Although 64Cu has complex chemical properties compared to other PET radioisotopes such as 68Ga, it has an appropriate half-life and enables high-quality PET images similar to 18F, which is an advantage in terms of diagnosis. In addition, since it also has therapeutic properties through the release of β- particles and Auger electrons by electron capture, radiopharmaceuticals using 64Cu stand for innovative radiopharmaceuticals for theranostic purposes. Therefore, based on the initial results obtained using 64Cu as a therapeutic agent, it is expected that additional research on the application of 64Cu will lead to a new era in the theranostics field.

  • PIB is the first amyloid plaque PET image tracer reported for the first time in 2003, and is considered to be the best and is still being utilized due to its very high uptake and kinetic properties. Initially, it was synthesized by radioisotope labeling using a precursor containing a methoxy methyl protection group, but now it is synthesized using a 6-OH precursor that can be easily synthesized in one step using [11C]methyl triflate. Carbon-11 has several limitations in clinical studies using PET because its half-life is as short as 20 minutes. In this study, in order to overcome the difficulty of this half-life, a rapid method using Sep-Pak was adopted instead of HPLC purification to significantly reduce the burden of the purification process and attempted synthesis. As a result, the synthesis time was shortened by more than 50%, and the yield of the final compound was higher than the previous result and showed relatively high specific radioactivity, confirming that it is a strategic method with high applicability for various precursors having primary amines.

Quantitation of Plasma Apolipoprotein A-I with a Sandwich Type Enzyme-Linked Immunosorbent Assay Using Monoclonal Antibodies

  • A sandwich-type enzyme-linked immunosorbent assay (ELISA) for the quantification of human apolipoprotein A-I (apoA-I) was developed using monoclonal antibodies. For this assay, we used three monoclonal antibodies to trap and detect apo A-I. HDAI16 and HDA15 monoclonal antibodies were used for trapping apoA-I and HDAI8 monoclonal antibody was for detecting apoA-I. These three monoclonal antibodies were produced by immunizing mice with high density lipoprotein (HDL) isolated from human plasma. By immunoblot analysis, these three monoclonal antibodies were specific to apoA-I and showed no cross-reactivities with other plasma proteins. The results of competition assays for epitope cross-reactivity test also verified that these monoclonal antibodies identified separate and distinct epitopes on HDL and apoA-I. Affinity constants of monoclonal antibodies were measured by ELISA. Their association constants ranged from $10^7$ to $10^8$ $M^{-1}$. For this assay, pure apoA-I was isolated by affinity chromatography using monoclonal antibodies. In this sandwich assay, the amount of HRP-labeled HDAI8 bound to apoA-I trapped by HDAI16 and HDAI5 was proportional to apoA-I concentration in the range of 0 to 500ng/ml. ApoA-I concentration in plasma was calculated from the linear regression equation of standard curve. The precision and reliability of the assays are reflected in the low intra-and interassay coefficients of variation that averaged 3.25% and 4.30%, respectively. This assay is sensitive, simple, reproducible, convenient in incubation interval, and does not use radioisotope: thus it can be widely applied in clinical laboratories.

Study on the Compatibility for an Ir-192 Source Manufactured by Korea Atomic Energy Research Institute (KAERI) in GammaMed Brachytherapy Machine (한국원자력연구소에서 개발한 Ir-192 선원의 감마메드 치료기 호환성 연구)

  • The compatibility with GammaMed-12i brachytherapy machine for an Ir-192 encapsulated source (IRRS20, KAERI, Korea) manufactured by Korea atomic energy research institute (KAERI) has been investigated. As a mechanical testing of compatibility, precise measurement of step movement with channels, measurement of curvature of radius for wire, and emergency return testing were performed. Periodic measurements of air kerma strength for 45 days were carried out to evaluate decay characteristics of Ir-192 radioisotope and comparison of dose distributions in phantom between KAERI and old sources previously used were performed by film dosimetry. KAERI source has a good compatibility with GammaMed12i machine as a result of mechanical testing. There are in good agreement with calculated values in activity characteristics and there were small differences in dose distributions around the source in comparison between KAERI and old source.
