Effects of Oocyte Maturational Age and Activation Conditions on the Development of Porcine Parthenogenetic Embryos

  • This study was conducted to investigate the effects of oocyte maturational age and activation condition on in vitro development of porcine parthenogenetic embryos (parthenotes). Porcine follicular oocytes were matured in vitro for 30 to 44 hr. Maturation rate was examined during in vitro maturation (IVM) every 2 hr interval. The cdc2 kinase activity was measured at 36 and 44 hr of IVM. Some oocytes were activated at 36 or 44 hr of IVM by three different conditions; 1) single electric stimulation (1.5 kV/cm for $30{\mu}sec$; ES), 2) double electric stimulations (1.5 kV/cm for $30{\mu}sec$, followed by 1.0 kV/cm for $50{\mu}sec$ after 1 hr; ES+ES) or 3) ES+ES followed by culture in 6-dimethlyaminopurine (6-DMAP) for 4 hr (ES+ES+D), and cultured for 6-7 days. Maturation rate was significantly increased as culture period was increased to 36 hr (66.9%, p<0.05), and then gradually increased to 87.1% at 44 hr of IVM. The cdc2 kinase activity was decreased (p<0.05) with culture period prolonged from 36 hr to 44 hr. Lower blastocyst formation rate (4.3%, p<0.05) were obtained by ES in 36 hr-matured oocytes compared to other treatments (16.5 and 20.5%) in the same age and the same treatment in 44 hr-matured oocytes (15.0%). High blastocyst formation rate (23.6%) was obtained by ES+ES+D in 44 hr-matured oocytes (p<0.05). These results demonstrate that porcine oocyte activation and in vitro development of parthenotes can be affected by interactions between oocyte maturational age and activation condition.

Skeletal Age Assessment of SMI and MP3 Stages to Predict the Pubertal Growth Spurt (사춘기 최대 성장 단계 예측을 위한 SMI 및 MP3 단계별 골연령 평가)

  • This study aimed to assess the skeletal age of skeletal maturational indicator (SMI) and middle phalanx of the middle finger (MP3) stages and to predict the SMI and MP3 stages corresponding to pubertal growth spurt in boys and girls respectively. The skeletal age was assessed from hand-wrist radiographs of 363 children (182 boys, 181 girls) aged 7 to 16 years by radiologists using the Korean standard bone age chart. Also, SMI and MP3 stages were evaluated from the radiographs. From these records, the mean skeletal age of SMI and MP3 stages was calculated. The stages including pubertal growth spurt were SMI 4 - 5, MP3 FG - G stages in boys and SMI 3 - 4, MP3 F - FG stages in girls.

Assessment of Midpalatal Suture Maturation by Skeletal Maturity on Hand Wrist Radiographs (성장기 아동의 수완부 골 성숙도를 이용한 정중구개봉합 성숙도 평가)

  • The aim of this retrospective study was to evaluate the correlation between the midpalatal suture maturation and skeletal maturation in growing children aged 7 - 15 years and predict the maturational stages of the midpalatal suture corresponding to skeletal maturity assessed by the skeletal maturation indicators (SMI) and middle phalanx of the third finger (MP3) stages. The group of this retrospective study was consisted of randomly selected 132 male and 135 female in age from 7 - 15 years. The maturation of the midpalatal suture was evaluated by using images from cone-beam computed tomography (CBCT) while the skeletal age was assessed by hand-wrist radiography. CBCT images and hand-wrist radiographs used in this study were obtained from all subjects for orthodontic diagnosis before orthodontic treatment. The maturational stages of the midpalatal suture showed strong correlations with both SMI and MP3 stages. The correlation between the midpalatal suture maturation and SMI (Spearman's correlation coefficient, ϒS = 0.905, p < 0.05) was slightly greater than that of MP3 stages (ϒS = 0.830, p < 0.05). There was a positive significant correlation between the midpalatal suture maturation and chronological age (ϒS = 0.868,p < 0.05). CBCT for evaluation of the midpalatal suture maturational stages may be unnecessary in every pediatric patients because SMI and MP3 stages were both replaceable useful methods for assessing maturation of the midpalatal suture before orthopedic treatment. In this retrospective study, the diagnostic reliability of the SMI method for estimating midpalatal suture maturation showed better reliability than the MP3 method.

Nuclear Maturation and Pronuclei Formation in Bovine Oocytes Matured In Vitro for Prolonged Period (체외 성숙 시간에 따른 소 난자의 처녀 발생)

  • Response of the oocytes to parthenogenetic activation is one of the indice for cytoplasmic maturation. Maturational age-dependent parthenogenetic activation was examined in bovine oocytes. Follicular oocytes recovered from the slaughter house ovaries were matured in vitro in TCM 199+15% FCS+1Oiu/ml PMSG +10 iu/ml hCG from 24 to 48 h at 6 h intervals. The in vitro matured oocytes were activated by 7% ethanol for 7 min. The nuclear maturation and the cytoplasmic maturation were analysed by the nuclear configuration and pronuclei formation stained by rapid staining method. Cumulus oophori expansion increased as the maturation time increased. Proportions of the nuclear maturation were 81, 89, 72, 60 and 60% in IVM 24, 30, 36, 42 and 48 h groups, respectively. Abnor¬mality in metaphase II chromosome increased sharply from 36 h IVM. The rates of the pronuclei formation and diploid upon ethanol activation were 67, 68, 73, 84 and 87%, and 4, 5, 10, 16 and 20% in IVM 24, 30, 36, 42 and 48 h groups, respectively. It was suggested that maturational age increased the formation of the pronuclei and diploid, and that cytoplasmic maturation require longer maturation period than normal nuclear maturation. These results should be useful for determination of an appropriate time for fertilization in mammalian eggs matured or preincubated in vitro.

Risk Factors of Primary Nocturnal Enuresis (일차성 야뇨증의 위험 요인)

  • Purpose: The Pathogenesis of primary noctllrnal enuresis(PNE) is still controversial. Genetic factor and maturational delay of micturition reflex including arousal disorder, lack of nocturnal Arginine Vasopressin(AVP) release and functional bladder capacity have been suggested. We analyzed the risk factors of PNE. Methods: Fifty five children with PNE (20 enuretics diagnosed at school physical examination of the first grade students at Mok-Dong Elementary School and 35 enuretics (Age 6-7 year) diagnosed at Ewha Womans University Mok Dong Hospital) and 221 control students without PNE at school physical examination were included. Genetic, stress and developmental factors, arousability, water intake, urine volume, maximun voiding volume and daytime voiding dysfunction were compared. Results: 1) There was no significant difference between PNE and control group in sex ratio, birth order, percentage of working mothers, parental and child personality, age to start walking, school record and duration of sleep. 2) Family history in the PNE group was significantly higher than control group ($20.0\%\;vs\;2.7\%$)(P<0.05). 3) The difficulty in arousal in the PNE group was significantly more common than the control group ($70.9\%\;vs\;54.3\%$)(P<0.05). 4) Nocturnal water intake in the PNE group was significantly greater than the control group ($330{\pm}158.2\;mL\;vs\;235{\pm}129.5\;mL$). Nocturnal urine volume in the PNE group was significantly greater than the control group ( $390{\pm}61.5\;mL\;vs\;140{\pm}43.2\;mL$)(P<0.05). Daily water intake and daily urine volume did not significantly differ between the two groups. 5) Maximum urine volume per void in the PNE group was significantly lower than the control group ($107{\pm}35.9\;mL\;vs\;236{\pm}41.3\;mL$). Daytime voiding dysfunction in the PNE group was significantly more common than the control group ($80.0\%\;vs\;57.9\%$). The voiding frequency in the PNE group was significantly greater than the control group ($7.0{\pm}3.6\;vs\;5.4{\pm}1.6$)(P<0.05). Conclusion: In addition to genetic factors and maturational delay of micturition reflex (difficulty in arousal, nocturnal polyuria and decreased functional bladder capacity) nocturnal polydypsia was found to be the important risk factors fur PNE. So nocturnal fluid restriction should be encouraged as the first-line management of PNE.
