Diversity and distribution of mating types in Lentinula edodes and mating type preference in domesticated strains

  • Mating type of Lentinula edodes is determined by two unlinked genetic loci, A and B. To better understand mating behavior of L. edodes, we investigated variations in mating type genes in129 dikaryotic strains collected from East Asia. Through sequence analysis of A locus, we discovered that hypervariable region spanning N-term of HD2-intergenic region-N-term of HD1 could represent A mating type. Mating and hypervariable region analyses revealed 70 unique A mating types: 27 from 98 cultivated strains, 53 from 31 wild strains, and 10 commonly found. It was also revealed that only a few A mating type alleles such as A1, A4, A5, and A7 were prevalent in cultivated strains. Contrarily, A mating type in wild strains was highly diverse: 23 unique A alleles were discovered in small mountainous area in Korean peninsula, suggesting rapid evolution of A mating type in nature. The B locus was assessed by allelic variations in pheromone (PHB) and pheromone receptor (RCB) pairs which constituted subloci Ba and Bb. Sequence analyses and mating assay revealed 5 alleles of RCB1 with 9 associated PHBs in Ba sublocus and 3 alleles of RCB2 with 5 associated PHBs in Bb sublocus. Each RCB was primarily associated with two PHBs. Each PHB-RCB pair was always discovered as a distinct unit. This allowed us to propose 15 B mating types via combinations of five Ba and three Bb subloci. Further investigation on 129 strains confirmed that the B locus, unlike the A locus, was indeed restricted to 15 mating types. Thus, the total number of mating types became 1,050 in L. edodes through a combination of 70 A and 15 B. This number will further increase because of rapid diversification of A mating type. Our findings provide a comprehensive and practical knowledge on mating behaviors of L. edodes.

Phylogenetic Analysis of Phaeosphaeria Species Using Mating Type Genes and Distribution of Mating Types in Iran

  • Phaeosphaeria species are pathogenic on wheat, barley and a wide range of wild grasses. To analyze mating type loci of the Phaeosphaeria species and investigate mating type distribution in Iran, we sequenced mating type loci of 273 Phaeosphaeria isolates including 67 isolates obtained from symptomatic leaves and ears of wheat, barley, and wild grasses from two wheat growing region in Iran as well as 206 isolates from our collection from other regions in Iran which were isolated in our previous studies. Mating type genes phylogeny was successfully used to determine the species identity and relationships among isolates within the Phaeosphaeria spp. complex. In this study, we reported seven new host records for Phaeosphaeria species and the Phaeosphaeria avenaria f. sp. tritici 3 group was first reported from Iran in this study. Mating type distribution among Phaeosphaeria species was determined. Both mating types were present in all sampling regions from Iran. We observed skewed distribution of mating types in one region (Kohgiluyeh va Boyer-Ahmad) and equal distribution in the other region (Bushehr). However, when considering our entire dataset of 273 Iranian Phaeosphaeria isolates, the ratio of mating types was not deviated significantly from 1:1 suggesting possibilities for isolates of opposite mating type to interact and reproduce sexually, although the sexual cycle may infrequently occur in some regions especially when the climatic conditions are unfavorable for teleomorph development.

표고 담자포자 유래 단핵균사의 A 교배형과 생장 속도 상관관계 (Correlation of A Mating Type with Mycelial Growth Rate in Basidiospore-derived Monokaryons of Lentinula edodes)

  • 표고는 사극성의 교배계를 갖는 담자균의 일종으로, 표고의 교배형은 A와 B라 불리는 서로 독립된 두 유전자좌에 의해 결정된다. 이론적으로 하나의 이핵균주는 네 개의 서로 다른 교배형을 갖는 담자포자를 1:1:1:1의 비율로 만들 수 있다. 과거 연구 결과에 따르면, 표고 담자포자에서 교배형이 편향된 분리비로 나타남이 보고되었다. 하지만 이러한 결과들은 꺽쇠연결과 같은 형태학적 특성만을 기반으로 교배형을 결정했다는 한계가 있다. 이에 본 연구에서는 교배형의 편향된 분리비가 표고에서 일반적인 현상인지 보다 명확하게 알아보기 위해서 최근에 보고된 DNA 마커를 활용하여 세 가지 표고 품종들의 담자포자에 대한 교배형 분석을 수행하였다. 그 결과 교배형의 편향된 분리비가 과거 보고와 일치하게 균주 특이적인 특성임을 확인하였다. 분석한 세 품종 중 한 품종을 제외하고 나머지 두 품종에서 편향된 분리비가 관찰된 것이다. 다음으로는 각 담자포자 유래 단핵균사들의 생장 속도와 교배형의 상관관계를 분석하였다. 그 결과 표고 단핵균사의 생장 속도는 B 교배형과는 관계가 없고, A 교배형과 관계가 있음을 확인하였다. 따라서 A 교배형 유전자좌에 존재하는 호메오도메인 전사인자 혹은 A 교배형 유전자좌와 연관된 유전자들이 단핵균사의 생장에 영향을 줄 것으로 보인다. 버섯 신품종 육성에서 교배형의 중요성을 고려할 때, 본 연구는 효율적인 신품종 육성 전략을 세우거나 단핵균사 생장 기작을 이해하는 데 도움을 줄 것으로 기대된다.

Activation of the Mating Pheromone Response Pathway of Lentinula edodes by Synthetic Pheromones

  • Pheromone (PHB)-receptor (RCB) interaction in the mating pheromone response pathway of Lentinula edodes was investigated using synthetic PHBs. Functionality of the C-terminally carboxymethylated synthetic PHBs was demonstrated by concentration-dependent induction of a mating-related gene (znf2) expression and by pseudoclamp formation in a monokaryotic strain S1-11 of L. edodes. Treatment with synthetic PHBs activated the expression of homeodomain genes (HDs) residing in the A mating type locus, and of A-regulated genes, including znf2, clp1, and priA, as well as genes in the B mating type locus, including pheromone (phb) and receptor (rcb) genes. The synthetic PHBs failed to discriminate self from non-self RCBs. PHBs of the B4 mating type (B4 PHBs) were able to activate the mating pheromone response pathway in both monokaryotic S1-11 and S1-13 strains, whose B mating types were B4 (self) and B12 (non-self), respectively. The same was true for B12 PHBs in the B4 (non-self) and B12 (self) mating types. The synthetic PHBs also promoted the mating of two monokaryotic strains carrying B4-common incompatible mating types ($A5B4{\times}A1B4$). However, the dikaryon generated by this process exhibited abnormally high content of hyphal branching and frequent clamp connections and, more importantly, was found to be genetically unstable due to overexpression of mating-related genes such as clp1. Although synthetic PHBs were unable to discriminate self from non-self RCBs, they showed a higher affinity for non-self RCBs, through which the mating pheromone response pathway in non-self cells may be preferentially activated.

Differential Growth Response of $A_1\;and\;A_2$ Mating Types of Phytophthora infestans on Rye A and V-8 Juice Agar Media Supplemented with Rhizome Powder of Cyperus rotundus

  • A new medium for studies of diversity among populations of $A_1\;and\;A_2$ mating types of Phytophthora infestans has been evolved. The rye A agar and V-8 juice agar media on which P. infestans grows well have been amended with rhizome powder of Cyperus rotundus. A total of 259 isolates of $A_1\;and\;A_2$ mating types representing Japan, Korea, India, Taiwan, Indonesia, Thailand, China, Nepal, U.K and Medico were screened for their growth response on these two media. Most of the A1 isolates did not grow well on them except Thailand while growth of $A_2$ mating types differed as some grew on it whereas others did not. It is quite likely that the populations of $A_2$ mating types that did not grow well on rhizome-amended medium are of different clonal lineage. This suggests that this medium can be used for the study of diversification among the isolates of the same or both the mating types as well as to detect the newly introduced genetically different isolates of P. infestans in a locality where it was not reported earlier.

한국에서 분리한 도열병균의 교배형 (Mating Type Alleles of Magnaporthe grisea in Korea)

  • Twenty six isolates of Magnaporthe grisea originated from rice and other gramineous hosts Korea were tested for mating type with MAT1-1 and MAT1-2 standard isolates. Ninety three and 64% of rice and grass isolated mated and produced perithecia with standard isolates, respectively. Both mating types were found from rice and non-rice isolates, but MAT1-1 allele of M. grisea was predominant in Korea.

Genetic DNA Marker for A2 mating type in Phytophthora infestans

  • The Phytophthora infestans requires two mating types for sexual reproduction. Amplified fragment length polymorphism (AFLP) was used to specifically detect different mating types of P. infestans. The AFLP primers E+AA (5'-GACTGCGTACCAATTCAA-3') and M+CAA (5'-GATGAGTCCTGAG-TAAC AA-3') detected a fragment that is specific in the A2 mating type of P. infestans. This fragment was cloned and sequenced. Based on the sequence data, PHYB-1 and PHYB-2 primer were designed to detect the A2 mating type of P. infestans. A single 347 bp segment was observed in the A2 mating type of P. infestans, but not in the A1 mating type of P. infestans or other Phytophthora spp. Identification of mating type was performed with phenotype (sexual reproduction) and genotype (CAPs marker) methods. Two factors, the annealing temperature and template DNA quantity, were investigated to determine the optimal conditions. Using mating type-specific primers, a unique band was obtained within annealing temperatures of 57$^{\circ}C$-62$^{\circ}C$ and DNA levels of 10pg-100 ng (data not shown).

Marker-Assisted Mating Applied in In-Situ Conservation of Indigenous Animals in Small Populations : (1) Choosing Mating Schemes for Maximum Heterozygosity

  • Maintaining maximum genetic variability is of critical importance with in-situ conservation of animal species in small populations. Marker-assisted mating (MAM) was suggested to achieve maximum heterozygosity in offspring populations. The aims of this research was to investigate and decide the effectiveness and promising types of MAM to achieve this goal. Analysis of variance with simulation data revealed that the heterozygosity in offspring populations was significantly determined by sire heterozygosity from mating of non-inbred parent animals, and significantly by sire heterozygosity and percent parental difference in offspring reproduced by inbred parents. Seven types of marker-assisted mating schemes were examined, in which offspring exhibited heterozygosity that was -0.01 to 7.37% below or above that from random mating of non-inbred parent animals, and 0.00 to 16.39% above that from random mating of inbred parent animals. The great increase in offspring heterozygosity was observed with mating by tandem maximizing sire heterozygosity, percent parental difference, and dam heterozygosity. Random mating resulted in fluctuation of offspring heterozygosity. These results suggested that MAM was a promising method for maintaining maximum offspring variability in in-situ conservation of animal species in small populations.

  • 1. 표고균(菌)의 교배계(交配系)에 대(對)해서는 기히 여러 연구자(硏究者)에 의해 4극성(極性)의 교배형(交配型)이 보고 되여 있는 바와 같이 본 시험에서도 동일(同一)한 결과(結果)를 보였다. 2. 본(本) 시험(試驗) 교배계간(交配系間)에는 어떤 계통(系統) 혹(或)은 단포자간(單胞子間)에든 Clamp 형성(形成)이 되었다. 3. 비교적(比較的) 근친계(近親系)라고 볼 수 있는 계통간(系統間)에는 (뉴기니아 계통간에) Clamp 형성량(形成量)도 적고 균계발육(菌系發育)도 부진(不進)하였다.

Mating Types and Optimum Culture Conditions for Sexual StateFormation of Fusarium fujikuroi Isolates

  • Twenty-five isolates of Fusarium fujikuroi acquired from rice seeds and rice plants evidencing symptoms of Bakanae disease were evaluated to determine their mating types and characterize the formation of their sexual state. The mating types of the isolates were evaluated via multiplex PCR with the diagnostic primers of the mating-type (MAT) region: GFmat1a, GFmat1b, GFmat2c, and GFmat2d. Among the 25 isolates, 11 were identified as MAT-1 (male), and 14 as MAT-2 (female). Four MAT-1 isolates and three MAT-2 isolates were mated and cultured to evaluate the optimal culture conditions for the production of their sexual states. Among four tested media, 10% V8 juice agar proved optimal for the perithecial production of the isolates. The isolates also generated the largest numbers of perithecia when incubated at 23oC in alternating cycles of 12 hr fluorescent light and NUV fluorescent light and 12 hr darkness.
