Determination of Liraglutide in Rat Plasma Using Selective Liquid Chromatography-Tandem Mass Spectrometry

  • Liraglutide is a medication prescribed for the management of type 2 diabetes and chronic obesity. A simple, sensitive, and selective liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for the quantitative analysis of liraglutide in rat plasma. After a simple protein precipitation step, liraglutide was chromatographically separated using the ACQUITY Premier Peptide BEH C18 Column with mobile phases comprising 50% acetonitrile and 50% methanol, and water with 0.3% FA. Positive ion electrospray ionization in multiple reaction monitoring mode was used to achieve detection. Good linearity was observed in the 5-600 ng/mL concentration range (R2 > 0.99). Liraglutide had intra- and inter-day precision values of 2.13%-9.86% and 4.14%-8.36%, respectively. The accuracy ranged from -2.36% to 2.58%. The recovery and matrix effect were within acceptable limits. This selective LC-MS/MS method was used to study the pharmacokinetic properties of liraglutide after subcutaneous administration in rats.

  • Analytical method of 4-nitrotoluene (4-NT) in water was developed by 9as chromatography/mass selective detector/selected ion monitoring (GC/MSD/SIM). 4-NT was extracted with diethyl ether. Organic layer was washed with 5 % sodium chloride solution. The influence of solvent and evaporation condition on extraction of 4-NT were examined. The retention time of 4-NT peak was 7.72 min. Coefficient of variation (CV) of 4-NT (ng) within day and day-to-day was ranged from 7.0 to 14.6% and from 7.7 to 20.8%, respectively. Recovery of 4-NT was ranged from 84 to 109%, and detection limit of 4-H was lese than 1 ng/㎖.

  • The use of the ammonia-water zeotropic mixture as a working fluid in the power generating system has been considered as a proven technology for efficient recovery of low-grade heat sources. This paper presents a thermodynamic performance analysis for ammonia-water evaporator using low-grade heat source, based on the exergy and entransy which has been recently introduced as a physical quantity to describe the heat transfer ability of an object. In the analysis, effects of the ammonia mass fraction and source temperature of the binary mixture are investigated on the system performance such as heat transfer, effectiveness, exergy destruction, entransy dissipation, and entransy dissipation based thermal resistance. The results show that the ammonia mass concentration and the source temperature have significant effects on the thermodynamic system performance of the ammonia-water evaporator.

  • This study explored the analytical conditions for 21 veterinary antibiotics which have been popularly sold in South Korea in 2014 but have not yet been targeted in EPA method 1694. Most of the selected antibiotics were separated by a reverse-phase C18 column with a combination of (buffered) water and organic polar solvent, which was commonly methanol and acetonitrile in the gradient elution mode. Volatile additives such as formic acid, ammonium acetate and ammonium formate were usually added to the mobile phases to minimize asymmetrical and tailing of antibiotics' peaks and to increase their ionization in mass spectrometry. The analytical methods of aminoglycoside antibiotics were distinct from those of the other antibiotics in terms of adoption of ion-pair chromatography (IPC) and hydrophilic interaction liquid chromatography (HILIC) capable of retaining and separating extremely polar compounds due to their hydrophilicity. Trifluoroacetic acid or heptafluorobutyric acid was frequently added to the mobile phase as an ion-pair reagent for the IPC. Tandem mass spectrometry was numerously applied to the detection of antibiotics using positive electrospray ionization (ESI) and the selected reaction monitoring (SRM) mode. All reviewed analytical methods had been/were validated by evaluating recovery, limits of detection and quantification, decision limit or detection capability of the methods.

  • The transient behavior of a passive air breathing direct methanol fuel cell (DMFC) operated on vapor-feeding mode is studied in this paper. It generally takes 30 minutes after starting for the cell response to come to its steady-state and the response is sometimes unstable. A mathematical dynamic one-dimensional model for simulating transient response of the DMFC is presented. In this model a DMFC is decomposed into its subsystems using lumped model and divided into five layers, namely the anodic diffusion layer, the anodic catalyst layer, the proton exchange membrane (PEM), the cathodic catalyst layer and the cathodic diffusion layer. All layers are considered to have finite thickness, and within every one of them a set of differential-algebraic governing equations are given to represent multi-components mass balance, such as methanol, water, oxygen and carbon dioxide, charge balance, the electrochemical reaction and mass transport phenomena. A one-dimensional, isothermal and mass transport model is developed that captures the coupling between water generation and transport, oxygen consumption and natural convection. The single cell is supplied by pure methanol vapor from a methanol reservoir at the anode, and the oxygen is supplied via natural air-breathing at the cathode. The water is not supplied from external source because the cell uses the water created at the cathode using water back diffusion through nafion membrane. As a result of simulation strong effects of water transport were found out. The model analysis provides several conclusions. The performance drop after peak point is caused by insufficiency of water at the anode. The excess water at the cathode makes performance recovery impossible. The undesired crossover of the reactant methanol through the PEM causes overpotential at the cathode and limits the feeding methanol concentration.

  • 대형 선박의 추진용 디젤엔진에서 버려지는 배기가스의 열을 회수하기 위한 랭킨사이클이 적용된 발전시스템에 대하여 R245fa 및 water의 작동유체에 따른 그 엑서지 특성을 분석하였다. 그 이론적인 계산 결과로, R245fa에 대하여 터빈입구의 압력이 증가할수록 엑서지 효율 및 시스템의 엑서지 효율이 증가하였고, 엑서지 파괴율은 주로 응축기 및 증발기에서 상대적으로 높게 나타났다. 그리고 질량유량의 증가에 따라 시스템의 엑서지 효율이 증가하는 특성을 보였다. Water의 경우에, 증발기에서의 엑서지 파괴율은 R245fa의 경우와 유사하게 나타났지만, 터빈입구의 압력 및 질량유량 비율의 변동에 대하여 열원에 대한 엑서지 손실률이 가장 큰 폭으로 변동하였다.

  • 본 연구는 GC/MS를 이용하여 수질, 토양 및 저질 시료 중의 benzophenone (BP)을 분석하는 방법을 확립하고자 하였다. BP는 수질 시료 (100 mL)에서 n-hexane으로 추출하였으며, 토양 및 저질 시료 (10 g)에서는 먼저 메탄올로 추출한 후 hexane으로 다시 추출하여 농축시켜 분석하였다. 수질 시료 중의 BP 회수율은 71.4% 이상이었으며 토양에서의 회수율은 86.5-94.7%를 보였고 재현성은 19.8% 이하였다. 검정곡선은 상관계수 ($r^2$) 값이 수질과 토양 모두에서 0.998이상의 좋은 직선성을 보여주었다. 수질 시료의 경우 43지역 중 3지역의 수질에서 30-200 ng/L 농도 범위로 BP가 검출되었으며 토양과 저질 시료에서는 모든 지역에서 검출되지 않았다. 이 분석방법은 환경 중에 미량으로 존재하는 BP의 분석과 모니터링에 유용하게 사용할 수 있는 적합한 방법이라 사료된다.

Simultaneous Quantitative Determination of Nine Hallucinogenic NBOMe Derivatives in Human Plasma Using Liquid Chromatography Tandem Mass Spectrometry

  • We developed a bioanalytical method for simultaneous determination of nine NBOMe derivatives (25H-NBOMe, 25B-NBOMe, 25E-NBOMe, 25N-NBOMe, 25C-NBOH, 25I-NBOH, 25B-NBF, 25C-NBF, and 25I-NBF) in human plasma using liquid chromatography tandem mass spectrometry (LC-MS/MS). Human plasma samples were pre-treated using solid-phase extraction. Separation was achieved on a C18 column under gradient elution using a mobile phase containing 0.1% formic acid in acetonitrile and 0.1% formic acid in water at a flow rate of 0.3 mL/min. Mass detection was performed in the positive ion mode using multiple reaction monitoring. The calibration range was 1-100 ng/mL for all quantitative analytes, with a correlation coefficient greater than 0.99. The intra- and inter-day precision and accuracy varied from 0.85 to 6.92% and from 90.19 to 108.69%, respectively. The recovery ranged from 86.36 to 118.52%, and the matrix effects ranged from 27.09 to 99.72%. The stability was acceptable in various conditions. The LC-MS/MS method was validated for linearity, accuracy, precision, matrix effects, recovery and stability in accordance with the FDA guidance. The proposed method is suitable for reliable and robust routine screening and analysis of nine NBOMe derivatives in forensic field.

작동매체에 따른 2중효용 흡수식 시스템의 성능해석 (Thermodynamic Analysis of Double-effect Absorption Heat Pump System with New Working Pairs)

  • Performance analysis of double-effect absorption heat pump system has been done to find improved working pairs (or mixture) by computer simulation. Based on the thermodynamic analysis, the coefficient of performance and mass flow ratio are investigated to compare three aqueous solutions [LiCl-water, LiSCN-LiBr-water, LiCl-$CaCl_2$-$Zn(NO_3)_2$-water] which was developed for only cooling, with conventional LiBr-water solution. It is found that the performances of the new aqueous solutions are better than that of LiBr-water solution not only in cooling systems, but also in heating systems. Theoretical thermodynamic performance data can be used in heat recovery systems by basic design data.

Gravity Recovery and Climate Experiment (GRACE) 중력자료 해석을 위한 자료 처리 및 응용 (Data Reductions of Gravity Recovery and Climate Experiment (GRACE) Gravity Solutions and Their Applications)

  • 2002년 4월에 발사된 Gravity Recovery and Climate Experiment(GRACE) 위성의 중력시간변화 측정을 통해, 기후 및 환경 변화에 의한 지구 내 질량 재배치 연구가 가능해 졌다. GRACE 중력 자료는 구면조화 함수의 계수인 중력 스펙트럼 형태로 제공이 되며, 이를 구면조화 함수를 이용하여 원하는 지역의 중력 변화 또는 질량 변화로 환산을 해야 한다. 하지만, GRACE 중력 자료는 측정 잡음 이외에도 공간적인 알리아스 에러가 존재하여, 질량 재배치 효과를 확인하기 위해서는 중력 스펙트럼의 처리 과정이 필요하다. 이 연구에서는 GRACE 자료를 처리하는 가장 일반적인 방법을 소개하고, 처리된 중력 자료를 이용한 연구 사례를 소개하였다. GRACE 중력 자료를 이용하여 광범위한 지구과학 연구가 진행 중이지만, 그 중 가장 활발한 연구 분야인 육지의 물수지 연구, 빙하 변화 연구 그리고 해수면 상승 연구 등을 중심으로 소개하였다. GRACE 위성과 유사한 인공위성 중력 관측 사업이 2020년까지 계획되어 있으며, 향후 수십년간 축적된 인공위성 중력 자료는 지구 환경 변화 연구에 핵심적인 자료로 활용될 것으로 기대된다.
