We have screened 766 strains of fungi from the BIOTEC Culture Collection (BCC) for xylanases working in extreme pH and/or high temperature conditions, the so-called extreme xylanases. From a total number of 32 strains producing extreme xylanases, the strain BCC7928, identified by using the internal transcribed spacer (ITS) sequence of rRNA to be a Marasmius sp., was chosen for further characterization because of its high xylanolytic activity at temperature as high as $90^{\circ}C$. The crude enzyme possessed high thermostability and pH stability. Purification of this xylanase was carried out using an anion exchanger followed by hydrophobic interaction chromatography, yielding the enzyme with >90% homogeneity. The molecular mass of the enzyme was approximately 40 kDa. The purified enzyme retained broad working pH range of 4-8 and optimal temperature of $90^{\circ}C$. When using xylan from birchwood as substrate, it exhibits $K_m$ and $V_{max}$ values of $2.6{\pm}0.6\;mg/ml$ and $428{\pm}26\;U/mg$, respectively. The enzyme rapidly hydrolysed xylans from birchwood, beechwood, and exhibited lower activity on xylan from wheatbran, or celluloses from carboxymethylcellulose and Avicel. The purified enzyme was highly stable at temperature ranges from 50 to $70^{\circ}C$. It retained 84% of its maximal activity after incubation in standard buffer containing 1% xylan substrate at $70^{\circ}C$ for 3 h. This thermostable xylanase should therefore be useful for several industrial applications, such as agricultural, food and biofuel.
Mukbang, which appeared through the new media platform in the early 2000s, reflected the life of the young generation who had to eat alone in the face of deteriorating socio-economic conditions. Many young people were able to watch Mukbang together, and gain comfort by establishing a virtual community of everyday life with those who suffered similar difficulties. Since then, Mukbang has evolved into various forms, reflecting the society and also accommodating the needs of viewers, and now it has become a global phenomenon, not just in Korea. Early research focuses on the phenomenon as a bizarre side that appears in the early stages rather than the growth and change of Mukbang, and deals with it in a limited way. This article critically examines these limitations, and examines how the growth and development of Mukbang interacted with the social change process, and how the uniqueness of Mukbang could emerge in the characteristics of the new media. Through this, we will be able to look at the context of the growth of Mukbang as a global phenomenon and the impact on the legacy media.
The purpose of the study is to examine how housing consciousness has been changed through various discourse on ideal housing from opening ports to the present. Reviews of literature including newspapers, magazines, novels, advertising materials, research papers, books, and so on are mainly utilized in accomplishing the purpose. This study covers four periods such as the enlightenment era, the Japanese colonial nile, before and after the Korean war, and after the economic development to present. During the enlightenment period in which foreign culture and housing were introduced, with reconsideration on traditional housing basic physiological housing value including health and hygiene was appeared. The desires of Munhwa housing with western styles and housing improvement were emphasized due to the new cultural improvement during the Japanese colonial rule. Before and after the Korean war which was chaotic times with rehabilitation, the provision of a large amount of public housing with minimum conditions for the basic human needs was the most important. After 1960s, due to the housing policy focusing on the mass-provision of apartments and the discourse on apartment as an ideal housing for the ordinary households in Korea, apartment dwellings become structure type norms in Korea.
Pinellia koreana K-H Tae & J-H Kim is a recently discovered Korea endemic medicinal plant species whose natural habitat is rapidly destroyed by industrial development. Described in this paper are culture conditions for high frequency plant regeneration via bulblet formation from leaf explant cultures of P. koreana. Leaf explants formed white nodular structures and off-white calluses at a frequency of 91.2% when cultured on MS medium supplemented with 2 mg/L BA and 0.5 mg/L NAA. However, the frequency of white nodular structures and off-white calluses formation was slightly decreased with an increasing concentration of NAA up to 4 mg/L, where the frequency reached 31.7%. Most petiole explants did not form white nodular structures and off-white calluses except the combination treatment of 2 mg/L BA and 2 mg/L NAA. Upon transfer onto MS basal medium, over 90% of nodular structures gave rise to numerous bulblets and developed into plantlets. Plantlets regenerated from bulblets were transplanted to potting soil and grown to maturity at a survival rate of over 95% in a growth chamber. Therefore, the in vitro plant regeneration system of P. koreana obtained in this study will be useful for mass propagation and long-term preservation of genetic resources of P. koreana.
Statistical experimental methods were used to optimize the medium for mass production of a novel laccase3 (Lac3) by recombinant Saccharomyces cerevisiae TYEGLAC3-1. The basic medium was composed of glucose, casamino acids, yeast nitrogen base without amino acids (YNB w/o AA), tryptophan, and adenine. A one-factor-at-a-time approach followed by the fractional factorial design identified galactose, glutamic acid, and ammonium sulfate, as significant carbon, nitrogen, and mineral sources, respectively. The steepest ascent method and response surface methodology (RSM) determined that the optimal medium was (g/L): galactose, 19.16; glutamic acid, 5.0; and YNB w/o AA, 10.46. In this medium, the Lac3 activity (277.04 mU/mL) was 13.5 times higher than that of the basic medium (20.50 mU/mL). The effect of temperature, pH, agitation (rpm), and aeration (vvm) was further examined in a batch fermenter. The best Lac3 activity was 1176.04 mU/mL at 25 ℃, pH 3.5, 100 rpm, and 1 vvm in batch culture.

β-Glucosidase production by the white rot fungus Flammulina velutipes CFK 3111 was evaluated using different carbon and nitrogen sources under submerged fermentation. Maximal extracellular enzyme production was 1.6 U/ml, corresponding to a culture grown in sucrose 40 g/land asparagine 10 g/l. High production yield was also obtained with glucose 10 g/land asparagine 4 g/l medium (0.5 U/ml). Parameters affecting the enzyme activity were studied using p-nitrophenyl-β-D-glucopyranoside as the substrate. Optimal activity was found at 50℃ and pHs 5.0 to 6.0. Under these conditions, β-glucosidase retained 25% of its initial activity after 12 h of incubation and exhibited a half-life of 5 h. The addition of MgCl2, urea, and ethanol enhanced the β-glucosidase activity up to 47%, whereas FeCl2, CuSO4, Cd(NO3)2, and cetyltrimethylammonium bromide inflicted a strong inhibitory effect. Glucose and cellobiose also showed an inhibitory effect on the β-glucosidase activity in a concentration-dependent manner. The enzyme had an estimated molecular mass of 75 kDa. To the best of our knowledge, F. velutipes CFK 3111 β-glucosidase production is amongst the highest reported to date, in a basidiomycetous fungus.

Although mud shrimp (Upogebia Major) provides benefits on the tidal flat ecosystem, it is causing the mass mortality of Manila clam (Tapes philippinarum) and large economic loss to the aquaculture farmer due to spreading in the entire coastal clam farm area of the West Sea of Korea. In this study, a mud shrimp blocking box for cultivating clam was tested to prevent the mud shrimp from invading the habitat of the clam. To determine the effect of the blocking box, the growth characteristics of the clams under various culture conditions were investigated. The shell length, total weight, condition index, the presence of mud shrimp in the box, and the soil decay in the box were examined according to whether the clams were raised in the box or in natural state, whether the box is covered with a net or not and the gap size of the box. As a result, no significant differences in shell length, total weight, and condition index were found between the clam grown in the box and the natural state (P>0.05) without any evidence of invading of mud shrimp to the box. In addition, the soil inside the box was in good condition, not decayed. Therefore, using the mud shrimp blocking box, mud shrimp can be prevented from propagating to the habitat of the clam, thereby contributing to the normal production of the clam and thus to restoring the clam farming.
This study investigated the ecology and growth of Monostroma nitidum Wittrock in both its natural habitat and the laboratory. The maximum length, width, and weight of M. nitidum in March were 9.0$\pm$4.7 cm, 9.6$\pm$3.6 cm, and 1.52$\pm$1.13 g, respectively. Yellowish-green or yellowish-brown reproductive thalli began to appear in January, and over 80% of the thalli matured by March. The male and female spores were ca. 6 $\mu$m long, and elongate and ovoid in shape. The spores had two flagella and one-eye spot, and tended to swim toward light. Maximum number of spores released from matured thalli was 236 cells/mL after 70 minutes at a light intensity of 100 $\mu$mol/m$^2$/s. The zygote diameter ranged from 3.4-6.0 $\mu$m (mean 4.2 $$m) and increased to 69.8 $\mu$m 14 weeks after culture. The mass release of zoospores was observed from thalli in the dark (3 to 12 days), after 30 min under dry conditions in the shade, at 25$^{\circ}C$, and a light intensity of 100 $\mu$mol/m$^2$/s. The maximum number of zoospores released was 109.8 cells/mL after 60 min of induction. M. nitidum fronds on the net increased to 6.8-7.2 cm in length, and 6.6-8.9 cm in width during the winter.

High-acidity vinegar was manufactured using pear concentrate by fed-batch fermentation without additional nutrients, and the physicochemical properties and volatile components were investigated at different fermentation stages (Stages 1-4) and at various initial alcohol concentrations (IAC; 6-9%). The levels of reducing sugar, free amino acids, total phenolic content, total flavonoid content, and radical scavenging ability increased slightly during Stage 4 (high-acidity vinegar), which was affected by alcohol feeding. The contents of approximately 20 types of volatile compounds differed between the moderate- and high-acidity vinegar samples, as determined by solid-phase microextraction/gas chromatography-mass spectroscopy. The level of acetic acid in high-acidity vinegar increased according to the initial alcoholic content applied. The high-acidity vinegar produced by fed-batch culture at an IAC of 6-7% showed improved physicochemical and volatile properties as compared to the moderate-acidity vinegar.

In addition, we can easily see increasing cases where what a designer intends is visualized using creative typography in diverse mass media such as TV commercials, movies or web. Thanks to the effects of this trend, various ways of manufacturing works have been proposed in the field of motion typography by applying diverse factors including verbal ones, time, form, motion, colors, and sound for the purpose of expressing formless semantic notions through visual form of typography. However, physiological features such as sex, age, health status, pathological conditions, and body size can have a bigger effect on the process of real communication. Therefore, if property of quasi-verbal sound can be reflected appropriately in motion typography where communication is expressed only by visual form, it can enable people to understand what a designer intends faster and more exactly.
