Effects of Packaging Methods on Quality of Marsupenaeus Japonicus Oranda (포장방법에 따른 보리새우 오란다의 품질특성 확인)

  • Packaging method and storage time play a significant role in maintaining the quality, safety, and shelf-life of food. In this study, the quality such as acid value, peroxide value, total bacterial count, color difference, and flavor of Marsupenaeus japonicus Oranda was evaluated depending on the five packaging methods (T-N, P-N, P-O2R, P-V, and P-N2) and storage periods. Changes of acid value and peroxide value strongly depend on the existing oxygen of headspace in the Marsupenaeus japonicus Oranda packages. Regardless of packaging methods, the change in acid value, peroxide value, microbial growth and flavor of the Marsupenaeus japonicus Oranda occurred depending on the storage time. Especially, it is identified the quality changes of Marsupenaeus japonicus Oranda stored with P-N2 package with less oxygen contents were retarded, compared to the them with T-N packaged with high oxygen contents.

Species Identification of Five Penaeid Shrimps Using PCR-RFLP and SSCP Analyses of 16S Ribosomal DNA

  • DNA-based molecular markers for differentiation of five penaeid shrimps (Penaeus monodon, P. semisulcatus, Feneropenaeus merguiensis, Litopenaeus vannamei and Marsupenaeus japonicus) were developed based on polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and single-stranded conformation polymorphism (SSCP) of 16S ribosomal (r) DNA. Differentiation of P. monodon, P. semisulcatus and L. vannamei can be unambiguously carried out by PCR-RFLP of 16S $rDNA_{560}$ whereas P. semisulcatus and M. japonicus shared a BABB mitotype. These shrimps were successfully discriminated by SSCP analysis of 16S $rDNA_{560}$. Nevertheless, the amplification success for L. vannamei and F. merguiensis was not consistent when tested against larger sample sizes. As a result, 16S $rDNA_{560}$ of an individual representing the most common mitotype of each species was cloned and sequenced. The new primer pair was designed and tested against the large sample sizes (312 bp product, N = 185). The amplification success was consistent across all species. PCR-RFLP of 16S $rDNA_{312}$ was as effective as that of 16S $rDNA_{560}$. Differentiation of all shrimp species were successfully carried out by SSCP analysis.

Development of a Single Tangle Net for the Brown Shrimp by Observation of Entanglement Behaviour

  • Three panel trammel nets were made illegal for the brown shrimp (Penaeus japonicus) or fishes by Korean fisheries law while trammel nets for fleshy prawn in the West sea remained legal. In this study a single panel tangle net with vertical loop lines rigged between the float line and sinker line was specially designed to catch brown shrimp. This net was developed for the first time after observation of the brown shrimp behaviour when reacting to a net in an observation tank. In field experiments these single tangle nets were compared with the traditional trammel nets in the coastal waters of the Keoje area. The mean number of the brown shrimp for 53 fishing operations was 1.13 per unit panel of the single tangle nets when fitted with the vertical loop lines. This was $84\%$ of the mean catch of 1.36 achieved with the trammel nets. These results of fishing experiments using single tangle nets in the field revealed a high fishing efficiency for the brown shrimp and showed little difference from trammel nets. The size of the brown shrimp or number of by-catch was not different between single tangle nets and trammel nets.
