Validation of an HPLC/UV-based method for Salicornia herbacea-derived isorhamnetin-3-O-glucoside and quercetin-3-O-glucoside quantification

  • Salicornia herbacea is a type of salt marsh plant that has been used in traditional medicine to treat several diseases. Isorhamnetin-3-O-glucoside (I3G) and quercetin-3-O-glucoside (Q3G) are major flavonoids in S. herbacea that are known to exert various pharmacological activities. Therefore, our study sought to validate and optimize an HPLC/UV-based analytical method for I3G and Q3G yield quantification, as well as to determine its limit of detection, limit of quantification, linearity, precision, and accuracy. Upon testing a concentration range of 31.5-1.9 ㎍/mL the results exhibited good linearity (r2 ≥0.9996 and r2 ≥0.9999 for I3G and Q3G, respectively), and the procedure was deemed precise (relative standard deviation of ≤3.19 and ≤3.85%, respectively), and accurate (102.6-105.0 and 92.9-95.2%, respectively). The results showed that our proposed method could be used for rapid I3G and Q3G evaluation in S. herbacea.

서해안 염생식물 퉁퉁마디의 발아지속과 복원 (Germination Continuity and Restoration of Salicornia europaea, Halophyte in West-coast of Korea)

  • Salicornia europaea (glasswort) is succulent, annual, halophytic plant mainly distributed throughout reclaimed land or salt marsh. It has strong tolerance to salt so that it plays the part of the pioneer species in the first succession. According to domestic and foreign studies, S. europaea contains plenty of minerals and antioxidant in the body. Since people take note of an availableness of this plant as health diet, the natural growth sites are threatened. In addition to development of salt marsh and sea shore, imprudent harvest has a bad effect to S. europaea population maintenance. To seek ways to preserve the population of this plant, we carried out the continuity of seed germination and restoration test. Seokmo Island, Daebu Island, Youngjong Island and Sudokwon landfill in Korea are selected sites for research. Result of germination continuity shows that most S. europaea seeds germinate on March but no more after July. However the germination was occurred after that time in the greenhouse. So we concluded that no germination after July is a matter of environmental condition not the number of remaining seeds. Also germination was seldom occurred in the spot where seeds production was not happened. In result of continuity test of seed germination by soil depth, germination was occurred vigorously only in top soil. From these results, we note that most S. europaea germinate in the early spring, and germination is finished by July. And this rapid germination speed makes the number of seeds in soil seed bank rare. If a large number of S. europaea in some area are harvested after July, the number of this species will dramatically decrease in that area the following year. In Seokmo Island, we carried out reintroduction experiment by sowing S. europaea seeds. On the first year, a small number of S. europaea settled and they produced seeds successfully. On April 2010, the second year, we observed many S. europaea in seed sowing sites. And we found out that plowing is more efficient than treatment sea water for settlement of S. europaea.

Soil factors determining the distribution of Phragmites australis and Phacelurus latifolius in upper tidal zone

  • To assess the environmental factors determining the zonation between Phacelurus latifolius and Phragmites australis, vegetation survey and soil analysis were performed at a tidal marsh. The vegetation of the tidal marsh was classified into P. latifolius and Suaeda japonica dominated quadrats, P. latifolius and P. australis dominated quadrats, P. australis dominated quadrats, and P. australis and other land plants dominated quadrats. The density of P. latifolius ($83.7{\pm}5.5\;shoots\;m^{-2}$) was higher than that of P. australis ($79.3{\pm}12.1\;shoots\;m^{-2}$) in each dominated quadrat but height of two species were similar. Soil environmental characteristics of P. latifolius dominated quadrats appeared to be affected by tide based on higher soil electric conductivity ($EC_{PL}=1530{\pm}152{\mu}Scm^{-1}$ ; $EC_{PA+PL}=689{\pm}578{\mu}Scm^{-1}$ ; $EC_{PA}=689{\pm}578{\mu}Scm^{-1$) and lower pH ($pH_{PL}=5.96{\pm}0.16$ ; $pH_{PA+PL}=6.28{\pm}0.31$ ; $pH_{PA}=6.38{\pm}0.22$). In redundancy analysis, environmental characteristics of P. latifolius dominated quadrats and P. australis dominated quadrats were clearly separated and those of P. latifolius and P. australis co-dominated quadrats were similar to P. australis dominated quadrats. From our investigation, P. latifolius showed relatively high competitiveness when compared to P. australis in lower tidal zone rather than upper tidal zone. Zonation of P. latifolius and P. australis seems to be a transitional zone between halophytes and land plant species.

Creation of an Environmental Forest as an Ecological Restoration

  • We created an environmental forest on the basis of ecological design around the incineration plant of Jindo Engineering and Construction Co., Ltd., which is located in Jeongwang-dong, Siheung-si, Kyunggi-do. To get ecological information of this site, physico-chemical properties of soil on salt marsh, which is located close to the syudy site and of forest soil transported from other sites for ecological restoration were analyzed. Texture of salt marsh and transported soils were loam and sandy loam, respectively. pH, organic matter, T-N, available P, and exchangeable K and Na contents of salt marsh and transported forest soils were 6.7 and 5.4, 4.1 and 0.4%, 1.0 and 0.3mg/g, 46.7 and 6.8ppm, 521 and 207ppm, and 3.8 and 0.5mg/g, respectively. Introduced plants were selected among the dominant species of forests and the species composing the potential natural vegetation around the present study site. Those plants were selected again by considering the tolerances to air pollution and to salt, and their availability. Selected trees were Pinus thunbergii, Sophora japonica, Celtis sinensis, Quercus aliena, Q. serrata, Q. dentata, and Q. acutissima. Selected sub-trees were Albizzia julibrissin, Koelreuteria poniculata, and Styrax japonica and shrubs were Rhododendron yedoense var. poukhanense, R. mucronulatum, Callicarpa japonica, Euonymus alatus, E. japonica, and R. schlippenbachii. On the other hand, introduction of herbs was not considered except for Liriope platyphylla, which was ornamentally planted in one site. Planting bed of mound type was adopted to provide the fine drainage system. Mound was designed to furnish litter, A, B, and C layers simuating the profile of forest soil. Slope of mound was mulched by rice straw of 2cm in thickness to prevent for sliding of litter and soil in cases of strong wind or heavy rain. Height of mound was designed to secure more than 1 m by combining A and B layers. Narrow zones, in which mound with stable slope degree cannot be prepared, was designed to equip the standard soil depth with the introduction of stone for supporting. On the other hand, plants with shallow root system were arranged in some zones, in which satisfactory soil depth cannot be ensured. Plants were arranged in the order of tree, sub-tree, and shrub from center to edge on the mound to make a mature forest of a dome shape in the future. Dispersion of plants was designed to be random pattern rather than clumped one. Problems on creation of the environmental forest by such ecological design were found to be management or inspection by non-specialized project operators and inspecting officers, and regulations for construction without ecological background. Alternative plans to solve such problems were suggested.

연안생태환경공간 조성지의 생물-화학적 복원지수 평가 (Evaluation of Bio-Chemical Restoration Index at the Creation Site of Ecological Environmental Zone in Coastal Area)

  • 대규모 연안개발에 따른 매립사업으로 연안역의 갯벌 또는 염습지 등의 연안생태환경공간은 해마다 감소하고 있다. 이에 따라 연안역 자연환경의 완화조치(Mitigation)의 일환으로 인공갯벌 및 염습지 개발의 필요성이 대두되어 활발한 연구가 진행되고 있으나, 인공 연안공간 조성시 복원정도의 정량적 평가에 관한 연구는 미흡한 실정이다. 따라서 본 연구에서는 연안생태환경공간 조성지의 복원정도 평가에 대한 기초적인 연구로서, 준설토 배합비에 따라 구분되는 인공갯벌(Zone. P1) 및 염습지(Zone. P2) 현장시험구를 조성하여 생물-화학적 환경인자인 시험구 내 대형저서동물 군집의 생물다양성과 개체수 및 생체량/식생(갈대)의 정착률/종속영양세균수/이화학적 특성(COD, IL, TN, TP, pH)에 대한 환경성 모니터링을 수행하였다. 이 결과를 바탕으로 각 환경인자들의 복원정도를 나타내는 복원지수를 제안하여 연안생태환경공간 조성에 따른 복원정도를 정량적으로 평가하였다.

도시자연공원의 식생구조에 따른 관리방안 - 인천광역시 월미공원의 사례 - (Structure and Management Devices of Vegetation at Weolmi Urban Nature Park, Incheon)

  • The purpose of this study is to propose vegetation management devices through analyzing the actual vegetation, flora, plant community structure and soil chemical concentrations in Weolmi urban nature park, Incheon. The actual vegetation of Weolmi Park in various areas is composed of urbanization area(2 types, 25.9%), landscape planting area(4 types, 16.1%), grass and marsh area(5 types, 7.6%) and mountain forest areal(14 types, 50.4%). The flora is composed of 295 taxa with 80 families, 253 species, 35 varieties and 7 formas, and among them there are 16 naturalized plant families, 39 species, 3 varieties. In reflection of size, the number of the species seems high but most of the them are under influence of human disturbance. Nine survey plots of plant community structure are classified into two groups. One is the semi-natural plant community(Prunus sargentii-Acer palmatum, Quercus accutissma-Prunus sargentii, Quercus serrata-Quercus accutissma-Prunus sargentii, Prunus sargentii, and Zelkova serrata-Prunus sargentii) that migrated finn the planting forest to the natural forest and the other is planting forest(Pinus koraienssis-Pinus thunbergii-Abies holophylla-Chamaectparis obtusa, Prunus sargentii, Pinus thunbergii-Alnus firma, Zelkova serrata). The average pH is 4.65 which means the soil acidity is quite high. The concentration of K, Ca, Mg and base saturation is very low. It seems that the environmental pollutants from Incheon Port and industrial plants near by survey site and long-distance transport of air pollutants from China made the soil condition worse. On the basis of the results above, six vegetation management devices are suggested: 1) removing the hazard plants(Pueraia thunbergiana and Humulus japonica), 2) natural landscape management of the middle and long term, 3) increasing species diversity, 4) Robinia pseudoacacia management, 5) keeping the naturalized plants from being distributed any further inside the mountain forest, 6) improving soil acidification.

금조산 분지의 왕미꾸리광이 ( Glyceria leptolepis Ohwi ) 개체군의 실소 및 인의 순환 (Nitrogen and Phosphorus Cycles in the Glyceria leptolepis Ohwi Population at the Mt. Geumoh Basin)

  • The seasonal pattern of pool size and withdrawal from senescing or stroage organ, and the annual magintude of internal-and plant-soil cycles for nitrogen and phosphorus in a Glyceria leptolepis Ohwi population in a marsh of the Mt. Geumoh were investigared. The population pool changed from initial size of 6.8 to the maximum of 16.1gN$m^{-2}$ for N and from 1.7 to 3.9g Pm$m^{-2}$ for P, maintaining far higher relative pool size during the first half of the growth period as compared with that for biomass. A sharp increase in N and P pool was noticed in early spring before the biomass growth was recognized, The major process supplying the demand for N and P changed as the growth progressed showing the order; absorption-withdrwal-absorption-with-drawal. The annual magnitude of plant-soil cycle for N and P was 18.0-19.1 and 2.9-3.3gm$m^{-2}$, accounting for 3 and 5% of each nutrient pool in 0-20 cm humus layer, respectively. The higher exent of internal cycle and the lower rate of annual turnover for P(1.08) as compared with those for N may suggest that this population conserves and reuses P more efficiently than N.

Endophytic Fungi of Salt-Tolerant Plants: Diversity and Ability to Promote Plant Growth

  • Suaeda australis, Phragmites australis, Suaeda maritima, Suaeda glauca Bunge, and Limonium tetragonum in the Seocheon salt marsh on the west coast of the Korean Penincula were sampled in order to identify the endophytes inhabiting the roots. A total of 128 endophytic fungal isolates belonging to 31 different genera were identified using the fungal internal transcribed spacer (ITS) regions and the 5.8S ribosomal RNA gene. Fusarium, Paraconiothyrium and Alternaria were the most commonly isolated genera in the plant root samples. Various diversity indicators were used to assess the diversity of the isolated fungi. Pure cultures containing each of the 128 endophytic fungi, respectively, were tested for the plant growth-promoting abilities of the fungus on Waito-C rice germinals. The culture filtrate of the isolate Lt-1-3-3 significantly increased the growth of shoots compared to the shoots treated with the control. Lt-1-3-3 culture filtrate was analyzed and showed the presence of gibberellins (GA1 2.487 ng/ml, GA3 2.592 ng/ml, GA9 3.998, and GA24 6.191 ng/ml). The culture filtrate from the Lt-1-3-3 fungal isolate produced greater amounts of GA9 and GA24 than the wild-type Gibberella fujikuroi, a fungus known to produce large amounts of gibberellins. By the molecular analysis, fungal isolate Lt-1-3-3 was identified as Gibberella intermedia, with 100% similarity.

분뇨처리장 방류수정화 갈대습지셀의 초기운영단계 질산성질소 제거 (NO3-N Removal of A Reed Wetland Cell Constructed for Purifying Effluent from A Night Soil Treatment Plant During Its Initial Operating Stage)

  • $NO^3$-N removal was examined from July 2002 to December 2002 of a surface-flow constructed treatment wetland cell, which was a part of a treatment wetland system composed of four wetland cells and one distribution pond. The system was established on rice paddy near the Kohung Estuarine Lake located at the southern part of the Korean Peninsula. The lake and the paddy were formed by a salt marsh reclamation project. Effluent from a secondary-level treatment plant was funneled into the system. The investigated cell was created in June 2002. Its dimensions were 87 m in length and 14 m in width. It had an open water zone at its center, which was equivalent to 10 percent of its total area. Reeds(Phragmites australis) were transplanted from natural wetlands into the cell and their stems were cut at about 40 cm height from their bottom ends. Average 25 $m^3$/day of effluent from the plant was funneled into the cell by gravity flow and average 24.2$m^3$/day of its treated effluent was discharged into the Sinyang Stream flowing into the lake. Its water depth was maintained about 0.2 m and its hydraulic detention time averaged 5.2 days. The average height of the reed stems was 45.2 cm in July 2002 and 80.5 cm in September 2002. The number of stems averaged 40.3 stems/$m^2$ in July 2002 and 74.5 stems/$m^2$ in September 2002. The reeds were established initially well. $NO_3$-N loading rate of influent and effluent averaged 173.7 and $93.5mg/m2{\cdot}day$, respectively. Removal of $NO_3$-N averaged $80.2mg/m2{\cdot}day$ and its removal rate by mass was about 50 %. Considering the initial operation of the cell and the inclusion of the cold months of November and December in the analysis period, the $NO_3$-N removal rate was good.

Changes in vegetation and flora of abandoned paddy terraces in responses to drawdown

  • In order to assess the impacts of drawdown for land-use change on a Sphagnum-marsh, we compared the vegetation and flora of the wetland before and after the drawdown with focusing on the population of Sphagnum palustre L. Remarkable changes in the coverage of S. palustre and the major vegetational components of the wetland were observed. The coverage of S. palustre markedly decreased by about 75% (from approx. 247 ㎥ in 2011 to approx. 62 ㎥ in 2015) after the drawdown. Tree species such as Salix spp. extended (from about 70% to about 83% in the total coverage of the wetland), whereas herbaceous species shrunk after the drawdown. Upland-inhabiting species such as obligate plants for uplands (OBU) increased, whereas wetland-inhabiting species such as facultative plants for wetlands (FACW) and OBW decreased in terms of vegetational coverage. The total number of plant species decreased from 70 species to 62 species after the drawdown, including the disappearance of some wetland-inhabiting species from the wetland. We suggest that the attention for further studies on the abandoned paddy terraces (APTs) and effort for the management and conservation of APTs and APT-inhabiting species that are vulnerable to human-induced disturbances have to be paid more.
