Lipid and Fatty Acid Composition in Nannochloropsis oculata Cultured in Varying Salinities (염분농도에 따른 해양미세조류(Nannochloropsis oculata)의 지질 및 지방산의 변화)

  • The quality and quantity of food organisms in fish seed production are important. The marine microalgae Nannochloropsis oculata are used as initial food organisms in the field. We investigated the effects of salinity (0, 10, 20, 30, 40 and 50 psu) on the lipid and fatty acid composition of N. oculata. Cultivation of N. oculata at varying salinities showed the highest growth rate at 20 psu. Total lipid content ranged from 17.26 to 18.63% at salinities from 0 to 50 psu). The nonpolar lipid content increased markedly at 30 psu and was highest at 15.55%. The polar lipid content was lowest at 30 psu, by 84.45%. It was also found that the omega-3 and EPA contents were inversely proportional to salt concentration. For the polar and nonpolar lipid compositions, there was no significant effect of salinity. Omega-3 polyunsaturated fatty acid content especially the content of EPA in the seawater larvae is the essential fatty acid in this food organism. It is thus advantageous to culture N. oculata at 20 psu.

Food Quality of Rainbow Trout Oncorhynchus mykiss Domesticated in Seawater (해수 순치 무지개 송어(Oncorhynchus mykiss)의 식품학적 품질 특성)

  • This study compared the food quality of domesticated(RT-DS) and freshwater (RT-F) rainbow trouts Oncorhynchus mykiss. The proximate composition of RT-DS was 73.8% moisture, 20.6% crude protein, 4.2% crude lipid, and 1.1% ash and was similar to RT-F. No differences were found in the red color, odor and taste intensities, calcium, and potassium between RT-DS and RT-F based on the Hunter a values, volatile basic nitrogen content, odor intensity and using an electronic nose, and taste intensity using an electronic tongue. The crude protein, total amino acid contents, and taurine, glutamic acid, glycine, alanine and anserine contents in free amino acids were higher in RT-DS than in RT-F. There was no difference in hardness between RT-DS and RT-F.

The Physiochemical Characteristics of Seawater and Sediment of Marine Shellfish Farm in Jindong Bay (진동만 패류양식해역의 환경특성)

  • Seawater and sediment quality analysed was calculated to examinate the present environmental characteristics and pollution load was also calculated to evaluate the effect of farming area on the coastal environment. The measurements for seawater quality demonstrate the coastal environment has relatively eutrophicated with significantly decreased DO (0.2-8.5 mg/l) and elevated COD (9.6-31.2 mg/l) in summer. It was also evident that the water quality in Jindong Bay has been influenced by residues tide from Masan Bay with high metal concentration in August of 2002. Annual total pollution load (land and farm-driven) was estimated at 37,316 ton (SS) /yr: 9,809 ton/yr (26.3%) of land-driven load, 23,576 ton/yr (63.2%) of coastal sedimentation and 3,932 ton/yr (10.5%) of feces of cultural organisms. When all ark shell seedling farms are permitted species conversion to ascidian farm, the pollution load would increase by 196%, which may be another source for accelerating the eutrophication of the environment in Jindong Bay.

Bacteriological and Physiochemical Water Quality of Seawater in Tongyeong Harbor, Korea (통영항 해수의 세균학적 및 이화학적 수질)

  • The bacteriological and physiochemical analysis of seawater in Tongyeong harbor was conducted to evaluate sanitary conditions, The samples were collected at 6 stations established once a month from January to December, 2000. During the study period, the ranges of temperature, transparency, chemical oxygen demand, dissolved oxygen, dissolved nitrogen, phosphate and chlorophyll-a were $6.8\sim25.2^{\circ}C,\;1.0\sim2.5\;m,\;1.79\sim2.41\;mg/L,\;5.7\sim10.1\;mg/L,\;6.59\sim10.53{\mu}g-at/L,\;0.56\sim1.01{\mu}g-at/L\;and\;1.21\sim9.54\;mg/m^3$, respectively, The viable cell counts of seawater in Tongyeong harbor ranged from $3.0\times10^4CFU/mL\;to\;6.9\times10^6CFU/mL$. The coliform group and fecal coliform MPN's of the samples were ranged $23\~4,600\;MPN/100\;mL$ (means 540 MPN/100 mL) and $11\~1,600\;MPN/100\;mL$ (means 210 MPN/100 mL), respectively, The coliform group was classified with IMViC reactions and pathogenic vibrios were analyzed. Two hundred eighteen strains that were obtained from seawater samples in Tongyeong harbor represented Escherichia coli group, $66.1\%$; Citrobacter freundii group, $11.0\%$; Enterobacter aerogenes, $9.6\%$; and unknown, $13.3\%$, respectively. During the study period, infectious bacteria such as Vibrio cholerae O1, Salmonella sp. and Shigella sp. were not detected from the samples, but detection ratios of V. parahaemolyticus, V cholerae non-O1 and V. vulnificus were $10.0\sim30.1\%$.

  • Discharged materials from the point or non-point source are released into the sea, and as the results, marine environment is directly affected. We must estimate the impacts of contaminants to marine pollution rapidly and accurately. Therefore, it is needed on early warning system for appreciating marine environmental impacts, and required a bioassay to evaluate abnormal changes. A bioassay test was developed to examine the effects of heavy metal contaminants on the early life stages of the marine annimals. We have studied the effects of metals on early development of a sea urchin species, Strongylocentrotus intermedius. S. intermedius embryos were tested with six metals (Cu, Ag, Zn, Cd, Cr, Ni) and showed the highest sensitivity to Cu as well as the lowest sensitivity to Cd. The order of biological impact for metals was Cu>Ag>Ni>Zn>Cr>Cd. In accordance with the results, sea urchins embryos can provide biological criteria for seawater quality assessment. The sensitivity of developmental bioassay whith S. intermedius is at intermediate level among marine organisms commonly used in aquatic bioassays. And this sea urchin can be routinely employed as a test organism for ecotoxicity assays.

Spatial Distributions and Monthly Variations of Water Quality in Coastal Seawater of Tongyeong, Korea (통영 주변 해역 수질의 공간분포 및 월 변화 특성)

  • Seawater quality was investigated each month at 30 stations near Tongyeong, South Korea, to provide data for the effective use of coastal fisheries and the reduction of economic damage to marine products. Water temperature was lowest in January and highest at the end of August. Neither extremely low water temperature below $4^{\circ}C$ nor fish damage caused by low water temperature was observed. Salinity ranged from 24.04 to 34.39 psu in the surface layer and from 29.92 to 34.39 psu in the bottom layer. The minimum salinity, attributable to rainfall events, was observed in July; salinity increased to high of about 34 psu in November. Low dissolved oxygen (DO), below 4 mg/L, was observed at Wenmun and Buksin Bays during May to October. Concentrations of $NO_2$-N, $NO_3$-N, and $PO_4$-P were low from March to September and high from October to February. Transparency was 6 m on average and was high in Wenmun Bay. Chemical oxygen demand (COD) and chlorophyll a (Chl. a) were high during summer, when the water temperature was high. With cluster analysis based on environment factors related to water quality, the study area could be divided into three main sea areas: Buksin Bay, coastal seawater, and offshore seawater. Buksin Bay was characterized by low salinity, high DO and Chl. a, and high transparency in the surface layer and by low DO and high $NH_4$-N in the bottom layer. Offshore seawater had high salinity and $NO_3$-N and low Chl. a concentration. In summer season that oyster need lots of phytoplankton, $NO_3$-N and Chl. a concentrations at this study area were low compare to Gwangy-ang and Gamak Bays. In winter, a sea squirt swallow much more than other season, the Chl. a concentrations were also low than Gwangyang and Gamak Bays.

Removal of Suspended Solids Using a Flexible Fiber Filter in a Recirculating Aquaculture System (유연성 섬유사 여과기를 이용한 순환여과식 양식장의 부유고형물 제거)

  • The suitability of a flexible fiber filter for removing suspended solid (SS) in a recirculating aquaculture system was evaluated. This study focused on variation in the performance with a change in filtering time, influent water quality, and filtering mode duration. The particle distribution diagram of the filter effluent showed that the number of particles bigger than $5-8{\mu}m$ decreased dramatically, and the removal efficiency exceeded 80%. Although the removal efficiencies of SS and chemical oxygen demand (COD) were dependent on the quality of the influent, the SS and COD concentrations of the effluent were not affected by the influent concentrations. This was despite the deterioration if water quality after feeding in the rearing tank. The performance of the filter was not affected by the filtering mode duration, feeding conditions, or filtering time. The SS concentration and turbidity of the recirculating-type rearing tank were 30% and 50% lower, respectively, than of the a non-recirculating-type rearing tank under the same operating conditions. The flexible fiber filter was applicable to a recirculating aquaculture system that uses plenty of seawater, based on its low filtering resistance $(2kg_f/cm^2)$, high flux $(330m^3/m^2/hr)$, and high fine particle removal efficiency (80%, $5-8{\mu}m$).

Salinity Changes and Bottom Water Particle Exchange Simulations in Response to Sluice Gate Operations at Saemangeum Lake (새만금 배수갑문 운영에 따른 염분 변화와 저층수의 입자교환 모의)

  • In an effort to improve water quality, the South Korean government has implemented measures to increase seawater circulation in Saemangeum Lake. We analyzed the effect of increasing the frequency of seawater circulation based on salinity levels and bottom water exchange in the lake, using an environmental fluid dynamics code model. When the sluice gate opening and shutting frequency increased from once to twice per day, the internal water level of Saemangeum Lake increased by up to ~0.7 m. The salinity increased by 2.12 psu near the western breakwater and decreased by 1.18 psu near the freshwater inlet. We analyzed the extent of bottom water exchange using a particle tracing method and observed that the residual rate of particles shallower than 5 m in water depth decreased by 2.52% in Case 2 (opening and shutting twice per day) compared to Case 1 (opening and shutting once per day). This indicates that increasing the frequency of sluice gate opening and shutting would promote enhanced bottom water exchange. Consequently, the increased salinity and bottom water exchange associated with increased seawater circulation are expected to improve water quality in Saemangeum Lake.

  • Protection against salt attack in seawater is obtained by using a dense, quality concrete with a low water-cement ratio, and a components appropriate for producing concrete having the needed salt resistance. The objective of this study is to evaluate the feature of corrosion with using the various concrete materials under marine exposure environment. According to the test results, slag powder and anti -corrosion inhibitor showed high chloride resistance effect. Also concre crack have an influence on corrosion of steel in spite of mixed design for salt resistance concrete. The requirement for low permeability is essential not only to delay the effect of salt attack, but also to afford adquate protection to reinforcement with admixtures.

Geochemical Indicators for the Recovery of Sediment Quality after the Abandonment of Oyster Crassostrea gigas Farming in South Korea (굴(Crassostrea gigas)양식 중단 이후 퇴적물 질 회복에 관한 지화학적 지표 탐색)

  • In order to evaluate the recovery of fishing ground environment after the cessation of aquaculture farming, we examined the variation in sediment quality over time using different geochemical factors and investigated whether these factors are indicators of sediment quality recovery. The study area was an oyster Crassostrea gigas, farm in Tongyeong, Geyongsangnam-do, South Korea, where aquaculture activities had been carried out for 20 years, and the changes in water and sediment environment were monitored for 14 months after the abandonment of the farm. The mean water depth was 14 m, with a tidal range of 3 m, and seawater current velocity ranged from 4.7 to 7.0 cm/s. After the abandonment of the farm, total organic carbon and total nitrogen concentrations in the surface sediments decreased significantly over time compared to those in the control site; in particular, immediately after farm abandonment, acid-volatile sulfide concentrations decreased relatively rapidly. Carbohydrate and protein concentrations in the sediments showed no significant changes; however, lipid concentrations markedly decreased. Further studies on other aquaculture taxa and various aquaculture areas are needed to establish new policies for environmental management of fishing grounds, such as relocation of fishing grounds and determination of optimal fallowing periods.
