Quantification of Vitamin $D_3$ and Its Derivatives in Marine Animal Foods by HPLC (HPLC에 의한 동물성 수산식품 중의 Vitamin $D_3$와 그 관련물질 정량에 관한 연구)

  • Contents of vitamin $D_3$ and 25-OH-vitamin $D_3$ in marine animal products(20 species) were determined by HPLC. The isomers of vitamin D, $D_2$ and $D_3$, were not clearly separated on a reversed phase, ${\mu}$ Bonda Pak, with 20% methanol-acetonitrile, and on a normal phase, Zorbax SIL. with 0.4% isopropanol-hexane, but 25-OH-vitamin $D_2$ and-$D_3$ were separated on either ${\mu}$ Bonda Pak with 10% methanol-acetonitrile, or on Zorbax SIL with 2.2% isopropanol-hexane, respectively. Although levels of vitamin $D_3$ and 25-OH-vitamin $D_3$ varied remarkably according to species, their average value(fish : $l,l87{sim}36,007$ I.U/sample 100g, mussel : $58{\sim}1,706$ I.U/sample 100g, pickle: $1,208{\sim}79,358$ I.U/sample 100g) was greatly higher than that of meat($80{\sim}100$ I.U/sample 100g) and dairy products($400{\sim}800$ I.U/sample 100g). Fatty tissues of fish and pickled fish intestines contained high level of vitamin $D_3$ and 25-OH-vitamin $D_3$, while the clam and mussel known to have various kinds of sterol including ${\Delta}^7$-sterol showed very low levels of vitamin $D_3$ and its derivative.

The Influence of Water Temperature on Filtration Rates and Ingestion Rates of the Blue Mussel, Mytilus galloprovincialis (Bivalvia) (수온에 따른 지중해담치 (Mytilus galloprovincialis; Bivalvia) 의 여과율과 섭식율 변동)

  • This study was performed to describe the influence of temperature on the clearance rate and ingestion rate of the blue mussel, Mytilus galloprovincialis with three food organisms and habitat location (shell size) of mussel. Food organisms used in this experiments were Isochrysis galbana, Chaetoceros didymus and Prorocentrum dentatum. The size of mussels inhabiting higher midlittoral zone was smaller than those of lower midlittoral zone. Regardless of the kind of food organisms, filtration rates and ingestion rates of higher midlittoral mussels were higher than those of lower midlittoral mussels in experiment temperature conditions. The variation of filtration rate and ingestion rate showed same tendency with temperature. Filtration rates and ingestion rates increased with temperature, and recorded maximum values at $20-25^{\circ}C$ of temperature, and thereafter decreased gradually. Theoretical optimum temperatures showing maximum filtration rates and ingestion rates estimated from polynomial regression curves were also in the range of $20-25^{\circ}C$. Blue mussels showed different variation of filtration rate and ingestion rate with the kind of food organisms. Filtration rates and ingestion rate based on cell number were similar regardless of habitat location(tidal elevation) and food organisms. Ingestion rates based on carbon content showed very high values in case of P. dentatum beside I. galbana and C. didymus as food organism.

Spat culture of the hard shelled mussel Mytilus coruscus in seawater pond (축제식 양식어장을 활용한 홍합 치패의 중간육성)

  • Growth and survival of the hard shelled mussel spat were investigated to confirm the possibe spat culture in seawater pond from September 2014 to April 2015. Also, we measured simultaneously environmental factors including water temperature, salinity, dissolved oxygen, pH, DIN, DIP, chlorophyll a, and abundance and dominant species of phytoplankton in seawater pond every month. Ranges of water temperature and salinity were $4.0-23.4^{\circ}C$ and 18.8-25.2 psu, respectively, which were rather lower than the reported optimal level. Monthly measured survival rates in all the spat cages were over 90%. Concentration of chlorophyll-a and abundance of phytoplankton were very high, and dominant species phytoplankton were cryptomonads and nanoflagellates. These dominant species were considered to be good food organisms for the bivalve spat. The experimental cage stocked 100 individuals per basket ($2,700ind.\;m^{-2}$) hanging in bottom of seawater pond revealed the highest growth in shell height ($7.63{\pm}4.65mm$), but all experimental cages stocked below 200 individuals per basket ($5,400ind.\;m^{-2}$) did not show statistically significant difference. We may expect that seawater pond would be one of the best culture ground for bivalve spats when appropriate measures are available.

Heavy Metal Contents of Canned Seafoods Packed in Oil (수산물 기름 담금 통조림 식품의 중금속 함량)

  • Specification and heavy metal contents of canned seafoods packed in oil were investigated. 30 species of canned tuna were classified by 4 groups as follow; group (sample codes, 1-10) composed of tuna and oil only, group (11-18) composed of tuna, vegetable and oil, group (19-27) composed of tuna, sauce and oil, and group (28-30) composed of tuna, vegetable, sauce and oil. Commercial canned shellfish packed in oil were mainly produced from sea mussel (sample codes, 31 and 32) and oyster (33-35). Can bodies of canned tuna were made by tin-plate, and used c-enamel or aluminium-paste as coating materials. In pH values of canned tuna, sample codes 1-10 (pH 5.55-5.69) and 19-27 (pH 5.17-5.85) were higher than sample codes 11-18 (pH 4.95-5.43) and 28-30 (pH 5.20-5.38). There was no difference in salinity (1.3-1.9%) and vacuum degree (15-18 mmHg) among canned samples. Heavy metal contents of canned seafoods ranged from 1.04-9.03 ppm for Sn, and 0.17-0.68 ppm for Pb. Those values are below the permitted range (less than 150 ppm for Sn and 2 ppm for Pb).

Effect of Heavy Metals on Embryonic Development in the Mussel, Mytilus galloprovincialis

  • The embryos of marine bivalves have been commonly used in bioassays for quality assessments of marine environments. Although several standard protocols for the developmental bioassay of bivalves have been proposed, only a few trials for application of these protocols in environmental assessments or for the development of a new protocol with Korean species have been conducted. As such, there is a strong need to establish standard bioassay protocols with bivalves commonly found in Korean waters. To determine the sensitivity of Mytilus galloprovincialis to establish a standard bioassay, their fertilized eggs were exposed to six metals (Ag, Cd, Cr, Cu, Ni, and Zn). The order of biological impact was Ag > Cu > Ni > Zn > Cr > Cd and their lowest observed effective concentration were 5, 16.4, 25.4, 142, 187 and 1,500${\mu}g/l$, respectively. The proportion of normal larvae appeared to decrease linearly with the logarithm of each toxicant concentration within the tested range. The average values of median effective concentrations $(EC_{50})$ from the triplicate experiments for Ag, Cd, Cr, Cu, Ni, and Zn were 6.8, 1,797, 786, 16.6, 68.1, and 139.2${\mu}g/l$, respectively. There was a more than 100-fold difference in $EC_{50}$ values of Cu and Cd. The value of $EC_{50}$ or median lethal concentration of Cu was within the range observed for other bivalve developmental bioassays. The overall sensitivity of M. galloprovincialis in the present developmental bioassay was also similar to that of other marine organisms commonly used in aquatic bioassays (e.g. sea urchins, oysters). Hence, the bioassay using the embryo of M. galloprovincialis is considered to be a useful tool to monitor and evaluate the quality of marine aquatic environments.

Sulfites Contents Survey on Dried Marine Products in Market (시판 건 해산물 중 아황산염류 함량 조사)

  • This study was performed to investigate the contents of sulfites in 2, 770 dried marine products in Seoul Chung-Bu market from March to October in 1999. Sulfites of the samples were determined by Zn powder reduction method and Monnier-Williams's modified method. Eight samples of 2, 770 (0.29%) were detected over 30ppm in SO2 contents by Monnier-Williams's modified method. In samples detected over 30ppm, six dried filefishes(130.2~573.4ppm) were imported from Vietnam, same kind of other one(114.2ppm) was from China and the other one Alaska pollack(196.3ppm) was domestic. By Zn powder reduction method, 259 dried marine products (259/2, 770=9.35%) were positive reaction (1-30ppm). These were 93 dried shrimps, 53 filefish meats, 34 dried squids, 33 slices of dried pollacks and 17 sea mussel meats. So sulfites must be use commonly to prevent black spot and preserve for many dried shrimps, filefishes meats, dried squids etc. According to results, the quality test for the imported dried marine products must be reinforced to supply safety food for the citizens.

Monitoring of Paralytic Shellfish Poison by Highly Sensitive HPLC from Commercial Shellfishes and Sea Squirts (고감도 HPLC에 의한 시판 중인 패류 및 멍게의 마비성 패류독 모니터링)

  • We monitored paralytic shellfish poison (PSP) by HPLC method in shellfishes including a sea squirt, and its products produced in domestic and imported, total 35 species, 850 samples, collected at 9 cities such as Seoul, Busan, Daegu, Ulsan, Pohang, Masan, Tongyeong, Geoje and Sacheon, 10 times by month (2 times in April and May) from March to October, 2004. PSPs were detected in a few raw samples on March, and it showed highest toxic ratio $(72.9\%)$ on third week of April (51 samples toxic in total 70 samples) and decreased suddenly to $2.4\%$ on third week of May. After then, PSPs were detected in a few samples, sporadically, and disappeared after August. Most raw shellfishes in the market were safe from PSP, showing the toxicity below 1 MU/g (quarantine level 4.0 MU/g), except oyster collected on April at Masan (1.9 MU/g), blue mussel on April (1.8 MU/g) and purplish washington clam on May at Sacheon (2.1 MU/g). However, it was strongly suggested to survey and under control for the imported scallops and ark shells which showed highly toxic in the quarantine level on May, July, even September. PSP were not detectable in the all shellfish products collected on May, July and September, except 2 boiled and dried mussels contained trace amount (0.01 MU/g), and all those products were safe as below quarantine toxin level (4.0 MU/g).

Reproductive Ecology of the Hard Shelled Mussel, Mytilus coruscus in Western Korea (한국 서해산 홍합, Mytilus coruscus의 번식생태)

  • The gonad index, gonadosomatic index, the condition index, reproductive cycle, first sexual maturity, and sex ratio of the hard shelled mussel, Mytilus coruscus were investigated by using histological analysis and morphometric data. Specimens were collected monthly in Kyeokpo, western Korea from January to December, 2006. The gonad index reached a maximum in February, and minimum in September. Both the monthly variations of the gonad index (GI) and the gonadosomatic index (GSI) reached a maximum in February. After that, their values continually decreased between March and April because of spawning. The monthly variation of the condition index coincides with the GI and the GSI. The spawning period was from February to April, with the main spawning occurring between February and March. The reproductive cycle of this species could be classified into six stages: early active stage (November to January), late active stage (November to February), ripe stage (December to April), partially spawned stage (February to April), degenerative stage (April to October), and resting stage (June to December ). Percentages of sexual maturity was over 50% for the female and male hard shelled mussels that ranges from 40.1 to 50.0 mm in shell length. The percentage was 100% for those that are over 50.1 mm. The sex ratios of females to males over 40.1 mm in shell length were not significantly different from a 1:1 sex ratio $(x^2\;=\;0.63,\;p\;>\;0.05)$. No evidence of hermaphroditism was found in histological sections of any hard shelled mussel examined.

Study on the control of marine biofouling developed on the surface of porous ceramics (세라믹 다공체 표면에 발생하는 해양 생물 오손 억제에 관한 연구)

  • Recently, removing methods of red tide has been attempted by filtering the organisms using the ceramic porous bodies. However, the marine biofouling could be developed on the surfaces of porous ceramic body after use for more than one month, and it might decrease the function of the specimen. In this paper, a method of inhibiting marine biofouling by changing the physical properties or surface-modification of ceramic porous body was studied. After experiment with six different ceramic porous bodies, it was found that the specimen of lower porosity and water absorption showed the least amount of biofouling. In addition, by increasing the surface roughness with silica particles bonded to the surface of specimen, the amount of biofouling caused by large marine life such as barnacle and mussel could be decreased. On the other hand, when the surface of specimen was coated and fused by glass powder, the amount of biofouling was rather increased. This might be due to eluted inorganic ions from the glass which can promote the growth of the microorganism. In conclusion, the environmental-friendly methods to reduce the amount of marine biofouling, such as controlling the physical properties and the surface roughness of the porous ceramics, can be possible without the use of dangerous substances. So it is expected for the results obtained to be applicable to a marine structure.

Monitoring of Polycyclic Aromatic Hydrocarbons in Sediments and Organisms from Korean Coast

  • Surface sediments (0- 5 cm), mussels (Mytilus coruscus and M. edulis) and oysters (Crassostrea gigas) were sampled at 20 stations in Korean coast during a period of February to July 2000. Samples were analyzed for polycyclic aromatic hydrocarbons (PAHs) content using gas chromatography coupled to mass spectrometer detector (GC/MSD). The sediment $\sum PAH$ concentration varied from 7 to 1,214 ng/g dry weight and the level of carcinogenic PAHs of six species ranged from 1 to 563 ng/g dry weight in surface sediments. In organisms, the levels of $\sum PAH$ were in the range of 63-876 ng/g dry weight and the concentrations of carcinogenic PAHs of six species were in the range of 4-582 ng/g dry weight. The highest PAH concentrations in sediments and organisms were in samples from Station 5 in Pohang coast and Station 8 in Jinhae coast, respectively. $\sum PAH$ concentrations in sediments and organisms collected from Korean coast were slightly low or comparable to those in other countries. The contribution of ring aromatic groups to sum of 16 PAHs in sediments and organisms showed a similar pattern for most stations. In sediments, the predominant contributions were four and five ring aromatics like fluoranthene, pyrene, benzo[a]anthracene, chrysene, benzo[b]fluoranthene, benzo[k]fluoranthene and benzo[a]pyrene. In the organisms, four ring aromatics like fluoranthene, pyrene, benzo[a]anthracene and chrysene were predominant PAH species. Several molecular indices such as phenanthrene/anthracene, fluoranthene/pyrene and LMW/HMW ratio were used to identify the origin of PAH contamination in sediments. The results indicate that PAHs of Korean coast were mainly of pyro1ytic contaminants with slight petrogenic input. A high correlation within individual PAH compound in marine sediments was observed, while correlation in organisms showed a highly significant relationship for heavier molecular weight PAHs.
