Biodegradation of marine microplastics by the whole-cell catalyst overexpressing recombinant PETase (PET분해효소(PETase) 과발현 전세포 촉매의 해양미세플라스틱 생분해 활성 연구)

  • The increased production and consumption of polyethylene terephthalate (PET)-based products over the past several decades has resulted in the discharge of countless tons of PET waste into the marine environment. PET microparticles resulting from the physical erosion of general PET wastes end up in the ocean and pose a threat to the marine biosphere and human health, necessitating the development of new technologies for recycling and upcycling. Notably, enzyme-mediated PET degradation is an appealing option due to its eco-friendly and energy-saving characteristics. PETase, a PET-hydrolyzing enzyme originating from Ideonella sakaiensis, is one of the most thoroughly researched biological catalysts. However, the industrial application of PETase-mediated PET recycling is limited due to the low stability and poor reusability of the enzyme. Here we developed the whole-cell catalyst (WCC) in which functional PETase is attached to the outer membrane of Escherichia coli. Immunoassays are used to identify the surface-expressed PETase, and we demonstrated that the WCC degraded PET microparticles most efficiently at 30℃ and pH 9 without agitation. Furthermore, the WCC increased the PET-degrading activity in a concentration-dependent manner, surpassing the limited activity of soluble PETase above 100 nM. Finally, we demonstrated that the WCC could be recycled up to three times.

Characteristics of Marine Debris collected from the Coastline of Sandbar in the Nakdong River Estuary (낙동강 하구 사주 해안에서 수거한 쓰레기 종류 및 분포량 추정)

  • This study investigated the spatial distribution of the marine debris in Jinudo in the Nakdong river estuary. Types of marine debris in Jinudo are analyzed through sampling around some portion of its sandbar shoreline. The present study and its main results are summarized as follow: 1) Monitoring system, for evaluating the volume of marine debris, was established in the coastal region of $50\;m\;{\times}\;50\;m\;(=0.002\;km^2)$ over the southern part of Jinudo. All the marine debris of conceivable form are collected and their amount was also estimated. 2) During the $1^{st}$ collection in May 2007, the total weight of the marine debris was 1,110 kg in this site, which means the density of marine debris around the shoreline is $444\;ton/km^2$. After one month later, the collected marine debris was 23.75 kg and so we have the average density of $9.5\;ton/km^2$. From these results, it is suggested that the marine debris of 316.67kg was being accumulated per day over unit area($1\;km^2$). 3) The most frequent marine debris collected during monitoring campaigns was wooden material amounting to 85.86% of the whole collection; the rest were fishing gears such as nets and buoys(5.13%), household waste(2.34%), glass(0.94%) and metals(0.27%).

  • This paper is performed to develop a tripple-tube exchanger which can improve the system efficiency. Three different tube diameters are compacted by one body(tripple-tube) to recover waste heat from heat exchanging among the fluids. With this, the tripple-tube shows higher cooling capacity than the double-tube after comparing between those two systems. The results of this paper are basic data to design the optimum tripple-tube heat exchanger.

  • Intrinsic safety is generally considered the safest method of operating electrical instrumention in potentially explosive atmospheres. The method of intrinsic safety limits the energy passing into the hazardus area. The energy limitation is provided by the use of safety barriers which are mounted in the safe area. Because of the energy limitation, regardless of the fault in the hazardous area, sufficient energy cannot be released to ignite the explosive atmosphere. The following industries are known to have hazardous locations: chemical. munitions, petrochemical, auto(paint spray booths), grain, waste water, printing, distillers, pharmaceutical. breweries, cosmetics, and utilities. In this paper, a isolator type barrier for ship(LNG, LPG, etc,) and test equipment confidence are proposed. The test equipments are designed for mechanical electrical life time test and vibration. All of test results satisfy the goal and the studied barrier shows the improved confidence.

  • Measuring of the dissolved oxygen is widely used for the environmental control of natural waters, sewage waste treatment, medical and biochemical studies, soil husbandry, food and drug process control, and prevention of corrosion in boilers. Especially, a power plants need special management for preventing accidents from corrosion, therefore, it is essential to measure the concentration of dissolved oxygen in real-time. In this paper we present a method of measuring dissolved oxygen very accurately up to pub units. This method, called polarograph method, is based on the measures of the electric current generated by the oxidation process in cathode and de-oxidation process in anode, assuming that the amount of the current is proportional to the density of dissolved oxygen.

  • The water ejector is a low pressure high flow rate volumetric pump. It utilize the energy of a low mass flow, high velocity stream to induce a large mass flow, low velocity stream. In addition, it has a very good resistances to cavitation compared to the other type of pumps, and the maintenance cost is practically nil. There has been enormous energy loss to supply the upper part water of dam which has large potential energy as mere irrigation water in domestic multipurpose dam. The new type of energy saving system which developed through the present study can economizes over 950,000 kWh per year by mixing the upper part water of dam with the waste water by the large water ejector. This paper estimates the economical efficiency of the new type of irrigation water pumping system, and further more, represents the change of performance characteristics of large water ejector, which was adapted to this system, according to the fluctuation of upper water level that seasonally changes.

  • The eco-hydrodynamic model was used to simulation water quality of Kwangyang Bay according to the environmental variation for appropriate water quality management. The mean concentration of COD was 3.3㎎/L, this exceeded the third class of water quality criteria. Waste water discharging loads showed approximately 90% of total pollutant loads. For satisfaction to below 10㎍/L of Chl. a and 2㎎/L of COD, above 35% reduction of present pollutant loads of point sources are needed.

  • Heat recovery steam generator(HRSG) is a principal component of the combined cycle power plant (CCPP) which utilizes the waste energy of the gas turbine exhaust gas. A design of the HRSG is a keypoint to achieve high cycle efficiency with competitive cost. This paper presents a brief review on the design of a HRSG which covers the basic design parameters and their effects on the performance and the investment cost. Finally the concept of the optimum design point is presented according to the selection of a pinch point temperature difference and a steam pressure as an illustrated case.

  • The fishing boat propulsion system employing the modified stern shape and the tunnel to protect a propeller is developed to increase the cruise speed and reduce he problem resulting from the open propeller accidentally catching the waste net and able on the sea. Using 3 different tunnel types, the model test was performed in the circular water channel and the panel method based on the potential theory is applied to analyze the open water performance of the propeller. In the numerical analysis using he potential-based panel method, it calculates the hydrodynamic interaction between the propeller and the tunnel and evaluates the effect of the tunnel geometry. From the numerical and experimental results differing tunnel geometries, the propulsion efficiency is increased by the larger diameter of the inlet than the outlet of the tunnel and the smaller gap between the propeller tip and the tunnel internal surface. These results provide the information of the propeller system with the tunnel and the hydrodynamic interaction between the propeller and the tunnel.
