Anti-Inflammatory and Anti-Fibrotic Activities of Nocardiopsis sp. 13G027 in Lipopolysaccharides-Induced RAW 264.7 Macrophages and Transforming Growth Factor Beta-1-Stimulated Nasal Polyp-Derived Fibroblasts

  • Nocardiopsis species produce bioactive compounds, such as antimicrobial and anti-cancer agents and toxins. However, no reports have described their anti-inflammatory and anti-fibrotic effects during nasal polyp (NP) formation. In this study, we investigated whether marine-derived bacterial Nocardiopsis sp. 13G027 exerts anti-inflammatory and anti-fibrotic effects on lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages and transforming growth factor (TGF)-β1-induced NP-derived fibroblasts (NPDFs). Nitric oxide (NO) and prostaglandin E2 (PGE2) levels were analyzed. Extract from Nocardiopsis sp. 13G027 significantly inhibited the upregulation of NO and PGE2 in LPS-activated RAW 264.7 macrophages. The expression of mitogen-activated protein kinases (MAPKs) and protein kinase B (Akt/PKB) in LPS-induced RAW 264.7 macrophages was evaluated; smooth muscle alpha-actin (α-SMA), collagen type I (Col-1), and fibronectin also phosphorylated small mothers against decapentaplegic (SMAD) 2 and 3 in TGF-β1-stimulated NPDFs. The Nocardiopsis sp. 13G027 extract suppressed the phosphorylation of MAPKs and Akt and the DNA-binding activity of activator protein 1 (AP-1). The expression of pro-fibrotic components such as α-SMA, Col-1, fibronectin, and SMAD2/3 was inhibited in TGF-β1-exposed NPDFs. These findings suggest that Nocardiopsis sp. 13G027 has the potential to treat inflammatory disorders, such as NP formation.

  • Serratia marcescens biovar A2/A6, isolated from an Indonesian freshwater source, was identified based on extensive morphological, biochemical and genetic characterization. Formation of pigment was found to be strongly influenced by environmental conditions. Placket-Burman design was used to analyze the effect of carbon and nitrogen sources. Based on results of physiological and biochemical studies, the optimum conditions for growth and pigment formation were incubation 30$^{\circ}C$ in a neutral to slightly alkaline medium containing lactic acid and beef extract.

Genetic Variability and Population Structure of Olive Flounder Paralichthys olivaceus from Stocked Areas Using Microsatellite DNA Markers (종묘방류에 따른 넙치, Paralichthys olivaceus 지역집단의 유전학적 구조)

  • Five microsatellite DNA markers were used to investigate genetic diversity and population structure of olive flounder Paralichthys olivaceus collected from four locations (YD, SC, GJ, WD) where hatchery-based seeds of the flounder have been released. The average of observed (Ho) and expected heterozygosity (He) ranged from 0.833 to 0.871, and from 0.842 to 0.876, respectively. The average number of alleles per locus ranged from 12.4 to 17.8. The proportion of stocked flounder ranged from 20.0% to 95.8% for wild-caught populations with a decreasing tendency of alleles per locus following a higher proportion of stocked flounder. There is need to implement a more careful stock-enhancement program of hatchery-based seeds and to monitor its genetic effects on wild populations to ensure conservation of natural flounder resources.

Population genetic structure and regional fragmentation of rare CO1 haplotypes of Black-tailed Gulls (Larus crassirostris: Laridae) in Korean Peninsula (한반도 괭이갈매기의 개체군 유전 구조와 희귀 CO1 유전자형의 지리적 파편화)

  • The Black-tailed gull (Larus crassirostris) is one of the most common gull species in the Northeast Asia and classified as LC (least concern) in the IUCN Red List. The bird has become a useful indicator for marine environmental pollution and habitat disruption, since the human activities impacted marine environments. Here we show that the black-tailed Gull has 6 haplotypes and some rare haplotypes are locally limited implying that the bird may not disperse far. This study may be the first report on population genetic study of Black-tailed Gull in Korea and could be used as the baseline data for monitoring and genetic study on the bird as an environmental indicator.

New Record of Scolecenchelys fuscogularis (Anguilliformes: Ophichthidae) Leptocephali from Korea, as Revealed by Morphological and Molecular Analyses

  • Three leptocephali (22.2, 22.7, 56.0 mm in total length) collected from the East/Japan Sea were identified by morphological and genetic analyses as belonging to the genus Scolecenchelys (Anguilliformes, Ophichthidae). Morphologically, the specimens were characterized by 148-158 myomeres, 10 gut swellings, dorsal fin origin above middle of the body, and 6 postanal melanophores between the anus and the caudal margin. An analysis of an 849-base pair 12S rRNA sequence of mitochondrial DNA showed that sequences are concordant with those of adult Scolecenchelys fuscogularis (genetic distance = 0.001). Furthermore total number of myomeres is consistent with the total number of vertebrae in adult S. fuscogularis. This study provides the first description of the morphological characteristics of S. fuscogularis leptocephali and their variations with size. The Korean name of S. fuscogularis is "Ga-neun-mul-baem", established by Ji et al. (2012).

  • We presented detailed morphological descriptions of the eggs, larvae and juvenile of Acanthaphritis unoorum based on specimens collected with bongo nets from Korean waters during the period May 2017-July 2018. We collected 18 individuals including eggs (n= 4, 0.77-0.85 mm in egg diameter), preflexion larvae (n= 6, 4.11-6.31 mm in standard length, SL), flexion larvae (n= 4, 6.60-7.82 mm SL), postflexion larvae (n= 3, 8.94-13.46 mm SL), and one juvenile (n= 1, 14.67 mm SL). The mitochondrial (mt) DNA 16S rRNA sequences of the eggs, and the cytochrome c oxidase subunit I (COI) sequences of the larvae were identical to those of A. unoorum adults (genetic distances <0.01). The A. unoorum larvae and the juvenile that we collected were morphologically similar to those of Dactylopsaron dimorphicum, but the A. unoorum specimens were readily distinguishable by the presence of lateral melanophores. This is the first record of A. unoorum in Korean waters. We propose a new Korean name for A. unoorum: "O-ri-bu-ri-nuntung-i".

Genetic diversity, relationships and demographic history of the small yellow croaker, Larimichthys polyactis (Pisces: Sciaenidae) from Korea and China inferred from mitochondrial control region sequence data

  • Genetic variation was surveyed at the mitochondrial control region (766bp) to test for the presence of genetic stock structure in the small yellow croaker, Larimichthys polyactis from the Yellow and East China Seas. Individuals of the small yellow croaker could not be distinguished on the basis of its location, as demonstrated using the neighbor-joining (NJ) method, unweighted pair-group method, arithmetic average (UPGMA) and the minimum spanning network (MSN). Analysis of molecular variance revealed no significant differences among collections of the small yellow croaker taken from the four locations (two locations each in Korea and China). Neutrality tests and a mismatch distribution analysis indicated that this species has recently expanded. Our findings suggest either that the small yellow croaker has a high migration capability that enables it to overcome the effects of genetic drift, or that this species expanded relatively recently and has not yet had sufficient time to differentiate.

Intraspecific diversity and phylogeography of bony lip barb, Osteochilus vittatus, in Sundaland, as revealed by mitochondrial cytochrome oxidase I (mtCOI)

  • Life history characteristics, habitat landscape, and historical events are believed to have shaped the patterns of genetic variation in many taxa. The bony lip barb, Osteohilus vittatus, represent a potamodromous fish that complete all life cycle in freshwater and is widely distributed in Southeast Asia. It usually lives in small rivers and other freshwater habitats, and movement between habitats for either food or reproduction has been typical. These life history characteristics may promote gene flow, leading to less structured populations. However, many freshwater habitats are fragmented, which restricts gene flow. We investigate how this interplay has shaped patterns of genetic variation and phylogeographic structure within this species in the Sundaland, a biodiversity hotspot with a complex geological history, using mitochondrial cytochrome oxidase I (mtCOI) as a genetic marker. Forty-six mtCOI sequences of 506 bp long were collected from ten localities, eight geographically isolated and two connected. The sequences were used for population genetic and phylogeographic analyses. Our results showed a low genetic diversity within populations but high between populations. There was a deep phylogeographic structure among geographically isolated populations but a lack of such structure in the connected habitats. Among geographically isolated populations, sequence divergence was revealed, ranging from 1.8% between Java and Sumatra populations to 12.2% between Malaysia and Vietnam. An indication of structuring was also observed among localities that are geographically closer but without connectivity. We conclude that despite high dispersal capacity, the joint effects of historical events, long-term geographic isolation associated with sea level oscillation during the Pleistocene, and restricted gene flow related to lack of habitat connectivity have shaped the phylogeographic structure within the O. vittatus over the Sundaland.

Stratigraphy and Petroleum Geochemical Characteristics of Jiaolai Basin in Shandong Province of China (중국 교래분지의 층서와 석유지화학적 특성)

  • Jiaolai Basin is the Cretaceous continental sedimentary basin developed in Shandong Province of China. It is interpreted as a pull-apart basin which is filled with fluvio-lacustrine sediments and volcanic rocks. The sedimentary strata are divided into three formations: Laiyang Formation, Qingshan Formation and Wangshi Formation in ascending order. Laiyang Formation of the early Cretaceous consists of conglomerate, sandstone and shale, which are grey, black or red in color, respectively. Qingshan Formation of early Cretaceous includes various kinds of volcanic rocks. Late Cretaceous Wangshi Formation consists of red conglomerate, sandstone and shale. Various types of oil shows are observed on many outcrops in the basin such as asphalt filing fissures, oil smelling, rocks wetted with oil. However, commercial oil discovery was not made. Laiyang Formation is the richest in terms of organic matter contents. Some grey or black shales of Laiyang Formation contain more than 1% of organic matter. Kerogens of some layers mainly consist of amorphous organic matter or pollen. Thermal maturity of the organic matter reached main oil generation zone and hydrocarbon genetic potential is fairly good. According to such geochemical data, some layers of Laiyang Formation can act as hydrocarbon source rocks.

  • 지금까지의 선형 최적화에 대한 연구는 고전적인 최적화 기법인 비선형계획법과 유동해석법을 중심으로 생물의 진화 알고리즘을 바탕으로 한 유전자 알고리즘과 인공지능에 기초를 둔 신경망이론 등이 이용되어 왔다. 또한 최근 컴퓨터의 성능이 급속도로 향상됨에 따라 전산유체역학에 기초한 시뮬레이션 평가기법도 사용되고 있다. 본 논문에서는 유전자 알고리즘을 이용한 선형 최적화 방법을 제시하였다. 그리고 광역 최적해의 효과적인 검색과 빠른 접근을 위한 방법으로 네트워크 시스템을 기반으로 한 병렬분산 유전자 알고리즘 시스템(PDGAS)을 개발하였으며 그 성능을 기존의 진화 알고리즘과 비교${\cdot}$분석함으로써 선형 최적화의 가능성을 확인하였다.
