New Algicidal Compounds from a Marine Algicidal Bacterium against Cochlodinium polykrikoides

  • In screening of algicidal bacteria, we isolated a marine bacterium which had potent algicidal effects on harmful algal bloom (HAB) species. This organism was identified as a strain very close to Bacillus subtilisby 16S rRNA gene sequencing. This bacterium, Bacillus sp. SY-1, produces very active algicidal compounds against the harmful dinoflagellate Cochlodinium polykrikoides. We isolated three algicidal compounds (MS 1056, 1070, 1084) and identified them by amino acid analyses, fast atom bombardment mass spectrometry (FAB-MS), infrared spectroscopy (IR), $^1H$, $^{13}C$, and extensive two-dimensional nuclear magnetic resonance (2D NMR) techniques including $^1H-^{15}N$ HMBC analysis. One of them, MS 1056, contains a b-amino acid residue with an alkyl side chain of $C_{15}$. MS 1056, 1070, and 1084 showed algicidal activities against C. polykrikoides with an $LC_{50}$ (6 hrs) of 2.3, 0.8, $0.6\;{\mu}g/ml$, respectively. These compounds also showed significant algicidal activities against other harmful dinoflagellates and raphidophytes. In contrast, MS 1084 showed no significant growth inhibition against various organisms coexisting with HAB species in natural environments, including bacteria, eukaryotic microalgae, and cyanobacteria, although it inhibited growth of some fungi and yeasts. These observations imply that algicidal bacterium Bacillus sp. SY-1 and its algicidal compounds could play an important role in regulating the onset and development of HABs in the natural environments.

해양세균 Cytophaga sp. AYK301의 분리.동정 및 한천분해효소 생산을 위한 최적배양조건 (Isolation and Identification of Marine Bacterium Cytophaga sp. AYK301 and Optimal Culture Conditions for the Production of Agarase)

  • 해양으로부터 한천분해능이 뛰어난 균주를 한국의 서해(전남, 여천)으로부터 분리하여 Cytophaga sp.인 것으로 동정되었으며, 이 균주를 Cytophaga sp. AYK301이라 명명하였다. 균체성장과 효소 생산성 향상을 위한 탄소원의 영향을 단당 및 복합당류를 이용하여 살펴본 결과 본 균주는 agar기질을 사용하여 배양하였을 때 효소생산량이 가장 높은 것으로 확인되었으며, agar를 0.0~0.6%(w/v) 농도별로 첨가하여 효소생산능의 최적조건을 살펴보았다. 그 결과 0.2%(w/v) agar에서 최대효소활성을 나타내었다. 질소원의 영향은 유기질소원과 무기질소원으로 나누어 실험을 행하였는데 beef extract 0.3% (w/v), $NH_4NO_3$ 0.05%(w/v)에서 각각 최고의 효소활성을 나타내었다. 또한, 초기 pH의 영향에서 pH 7.5에서 효소활성이 가장 높게 나타났고, NaCl 농도의 영향에서는 7%(w/v)에서 가장 높은 효소활성을 나타내었으며, 각종 금속이온에 대한 효소활성은 거의 영향이 없는 것으로 확인되었다. 따라서, 본 연구에서는 최적배양조건하에서 플라스크로 배양한 결과, 231 units/L를 생한하여 기본배지를 이용하였을 때보다 약 4배 정도 효소활성이 증가하였음을 확인하였다.

Physicochemical and Rheological Properties of a Novel Emulsifier, EPS-R, Produced by the Marine Bacterium Hahella chejuensis

  • The rheological properties of an exopolysaccharide, EPS-R, produced by the marine bacterium Hahella chejuensis strain 96CJ 10356 were investigated. The $E_{24}$ of $0.5\%$ EPS-R was $89.2\%$, which was higher than that observed in commercial polysaccharides such as xanthan gum ($67.8\%$), gellan gum ($2.01\%$) or sodium alginate ($1.02\%$). Glucose and galactose are the main Sugars in EPS-R, with a molar ratio of ${\~}1:6.8$, xylose and ribose are minor sugar components. The average molecular mass, as determined by gel filtration chromatography, was $2.2{\times}10^3$ KDa, The intrinsic viscosities of EPS-R were calculated to be 16.5 and 15.9 dL/g using the Huggins and Kraemer equations, respectively, with a 2.3 dL/g overlap. In terms of rigidity, the conformation of EPS-R was similar to that of caboxymethyl cellulose ($5.0{\times}10^{-2}$). The rheological behavior of EPS-R dispersion indicated that the formation of a structure intermediate between that of a random-coil polysaccharide and a weak gel. The aqueous dispersion of EPS-R at concentrations ranging from 0.25 to $1.0\%$ (w/w) showed a marked shear-thinning property in accordance with Power-law behavior. In aqueous dispersions of $1.0\%$ EPS-R, the consistency index (K) and flow behavior index (n) were 1,410 and 0.73, respectively. EPS-R was Stable to pH and salts.

한천분해효소를 생산하는 해양세균 Thalassomonas sp. SL-5의 분리 및 특성 (Isolation and characterization of a marine bacterium Thalassomonas sp. SL-5 producing β-agarase)

  • 포항 호미곶 해수에서 한천분해활성을 보이는 해양성 세균 SL-5을 분리하였으며 16S rDNA 염기서열분석으로 해양기원의 Thalassomonas 속과 가장 유사한 균주임을 확인하였다. SL-5 균주가 생성하는 한천분해효소(agarase)는 성장의존성인 것으로 확인되었고 효소활성을 위한 최적 pH는 pH 7.0(20 mM sodium phosphate 완충용액)이고 최적 온도는 $40^{\circ}C$로 나타났다. 한천분해효소는 $80^{\circ}C$까지 65%의 효소활성을 보이는 호열성 효소이지만, 내열성은 그리 높지 않았다. 한천 분해효소의 분해산물에 대한 TLC 분석결과, 본 효소가 $\beta-agarase$라는 사실을 확인할 수 있었다. 분리한 Thalassomonas sp. SL-5 균주가 생산하는 한천분해효소를 이용하여 한천으로부터 다양한 기능성 한천올리고당을 생산하는데 있어 유용하게 사용될 것으로 기대된다.

Purification and Characterization of Chitinase from a Marine Bacterium, Vibrio sp. 98CJ11027

  • Chitin-degrading marine bacterial strain 98CJ11027 was isolated from bryozoa from the coastal area of Cheju Island, Korea, and identified as a member of the genus Vibrio. The molecular mass of the main extracellular chitinase (chitinase I), purified from strain 98CJ11027, was estimated to be 98 kDa. The optimal condition for chitinase I activity is pH 6.0 and 45℃. The activity was inhibited by $Fe^+2$ and$Cu^+2$. Chitinase I displayed the hydrolysis type of chitobiosidase and catalyzed reversed hydrolysis leading to the synthesis of tetraacetylchitotetraose.

Antibacterial Activity of an Ethyl Acetate Extract of Pseudomonas sp. UJ-6 against Methicillin-Resistant Staphylococcus aureus

  • In an effort to discover an alternative antibiotic for treating infections with methicillin-resistant Staphylococcus aureus (MRSA), Pseudomonas sp. UJ-6, a marine bacterium that exhibited antibacterial activity against MRSA, was isolated. The culture broth and its ethyl acetate extract exhibited bactericidal activity against MRSA. The extract also exhibited antibacterial activity against gram-negative bacteria, which were not susceptible to vancomycin. The treatment of MRSA with the extract resulted in abnormal cell lysis. The extract retained >95% of its anti-MRSA activity after heat treatment for 15 min at $121^{\circ}C$. Thus, although most antibiotics are unstable under conditions of thermal stress, Pseudomonas sp. UJ-6 produces a heat-stable anti-MRSA substance. The results of this study strongly suggest that Pseudomonas sp. UJ-6 can be used to develop a novel, heat-stable, broad-spectrum antibiotic for the treatment of MRSA infections.

해양으로부터 fucoidan 분해세균의 분리 및 특성 (Isolation and Characteristics of Fucoidan Degrading Bacterium from Marine)

  • Ecklonia cava로부터 얻어진 fucoidan을 분해하는 해양세균은 해수로부터 분리하였다. 이 균주의 조효소는 pH8과 $50^{\circ}C$에서 fucoidan을 효율적으로 분해하였다. Crude fucoidanase는 1% (w/v) fucoidan 반응액에서 24시간 내에 약 7.1%를 가수분해하였으며 반응산물로서 endo-type 가수분해에 의한 oligosaccharide를 생산하였다. 16S rRNA 유전자 염기서열분석과 생화학적 시험의 결과로부터 SB 1493균주는 잠정적으로 Pseudoalteromonas sp.로 동정하였다.

형광세균 Beneckea sp.에 의한 Pyruvic Acid의 생산 (Production of Pyruvic Acid by Luminescent Bacterium Beneckea sp.)

  • 신선한 생선에서 분리한 형광세균들 중에서, glucose로부터 pyruvic acid생산능이 가장 좋은 균주 BL-1980을 선별하여, Beneckea sp.로 동정하였다. Glucose로부터 pyruvic acid생산을 위한 배양 최적조건이 검토되었으며, pH 7.0, 배양온도 28$^{\circ}C$에서 60시간 진탕배양하였을때 10.6g/L의 pyruvic acid가 생산되었다. 또한. 이때 사용한 3 %의 glucose로부터 pyruvic acid로의 전환율은 35.33%이었다.

Isolation and Characterization of a Chitinolytic Enzyme Producing Marine Bacterium, Aeromonas sp. J-5003

  • A chitinolytic enzyme-producing bacterium was isolated from sea water on the coast of Busan. The bacterium was identified as Aeromonas sp. based on its morphological, cultural and biochemical characteristics and designated Aeromonas sp. J-5003. The strain produced two chitinoloytic enzymes: chitinase and chitobiase. The optimum culture conditions of the strain for production of chitinoloytic enzymes were investigated. For the production of chitinase, the major components of medium were colloidal chitin $0.5\%$, glucose $0.2\%$, yeast extract $0.25\%$ and peptone $0.25\%$ while for the production of chitobiase, they were colloidal chitin $0.5\%$, galactose and tryptone $0.2\%$. The optimum cultural temperature and initial pH for the production of chitinase and chitobiase were $30^{\circ}C$ and pH 7.0, respectively.
