한국산 고등균류기 (IX) (Notes on the Korean Higher Fungi (IX))

  • 많은 고등륜류가 1990년 9월부터 1991년 10월까지 만덕산, 덕유산, 지리산에서 채집되었다. 이 고등균류들을 동정한 결과 Strobilurus(솔버섯속)은 한국산 미기록속이었다. 그리고 Strobilurus oshimae(가지솔버섯), Mycena chlorophos(야광애주름버섯), Mmarasmius pulcerripes(예쁜낙엽버섯), Pholiota astragalina(빨강비늘버섯), Cortinarius subdelibutus(물끈적버섯아재비), Boletellus shichianus(가시노랑그물버섯)은 한국산 미기록종으로 확인되었다.

Three new records of mushroom species from Hangyeong-Andeok Gotjawal, South Korea

  • This study introduces one specimen each of the genera Gymnopilus, Marasmius, and Mycena as an unrecorded species found in the Hwasun Gotjawal and Jeju Gotjawal Provincial Park of the Hangyeong-Andeok terrain, South Korea. Morphological analyses of macroscopic and microscopic features, and molecular analysis of the nuclear ribosomal internal transcribed spacer region were conducted to identify the specimens. Our results indicated that the newly identified specimens were Gymnopilus dilepis, Marasmius tenuissimus, and Mycena seminau. Here, we describe these newly identified mushroom species and provide baseline data on their diversity in Gotjawal, South Korea.

Purification and Characterization of the Laccase Involved in Dye Decolorization by the White-Rot Fungus Marasmius scorodonius

  • Marasmius scorodonius secretes an extracellular laccase in potato dextrose broth, and this enzyme was purified up to 206-fold using $(NH_4)_2SO_4$ precipitation and a Hi-trap Q Sepharose column. The molecular mass of the purified laccase was estimated to be ~67 kDa by SDS-PAGE. The UV/vis spectrum of the enzyme was nontypical for laccases, and metal content analysis revealed that the enzyme contains 1 mole of Fe and Zn and 2 moles of Cu per mole of protein. The optimal pH for the enzymatic activity was 3.4, 4.0, and 4.6 with 2,2'-azino-bis(3-ethylbenzothazoline-6-sulfonate) (ABTS), guaiacol, and 2,6-dimethoxy phenol as the substrate, respectively. The optimal temperature of the enzyme was $75^{\circ}C$ with ABTS as the substrate. The enzyme was stable in the presence of some metal ions such as $Ca^{2+}$, $Cu^{2+}$, $Ni^{2+}$, $Mg^{2+}$, $Mn^{2+}$, $Ba^{2+}$, $Co^{2+}$, and $Zn^{2+}$ at a low concentration (1 mM), whereas $Fe^{2+}$ completely inhibited the enzymatic activity. The enzymatic reaction was strongly inhibited by metal chelators and thiol compounds except for EDTA. This enzyme directly decolorized Congo red, Malachite green, Crystal violet, and Methylene green dyes at various decolorization rates of 63-90%. In the presence of 1-hydroxybenzotriazole as a redox mediator, the decolorization of Reactive orange 16 and Remazol brilliant blue R was also achieved.

치악산버섯의 분포상 (Fungal Flora of Mt. Chiak (1) -Agaric Fungi-)

  • 치악산지역에 발생하는 버섯류를 조사하기 위하여 1988년 9월부터 1990년 8월까지 6회 채집하였다. 주름버섯류를 수집하여, 최근 분류체계에 준하여 종을 분류 동정한 결과, 15과 43속 93종을 분류동정하였으며, 그중 한국미기록 6종, 남빛밤버섯 Calocybe ionides(Bull.: Fr.) Donk.; 치악송이 Tricholoma psammopus(Kalchbr.) Quel.; 흰무리애기버섯 Collybia cirrhata(Per.) Quel.; 황소낙엽버섯 Marasmius aurantioferrugineus Hongo; 양파광대버섯 Amanita abrupta Peck in Bull.; 흰오뚜기광대버섯 Amanita castanopsidis Hongo이 확인되었다. 수목과 공생하며 외생균근을 형성하는 버섯류는 9과 18속 54종이었으며, 그중 광대버섯속, 젖버섯속, 무당버섯속이 우점종으로서의 가능성을 보였다. 한편, 못버섯속, 마개버섯속, 땀버섯속, 껄껄이그물버섯속, 분말그물버섯속, 외대버섯속, 귀신그물버섯속, 쓴맛그물버섯속과 산그물버섯속은 드물게 발생하고 있었다. 치악송이는 일본잎갈나무에서만 발견된 기주특이성 종이었고, 남빛밤버섯도 같은 장소에서만 발견되었고, 못버섯속과 마개버섯속은 소나무림내에서 발견되었는데 이는 Miller(1972)의 관찰과 일치하였으며, Trappe(1962)는 그의 연구에서 마개버섯속은 소나무과에서만 발견된다고 보고한 바 있다. 사물기생균은 8과 25속 39종이 발견되었으며, 외생균근 버섯류에 비해 속은 다양하나 종은 대단히 적었다. 이 지역의 버섯류에 대하여 체계적이고 지속적인 조사를 통하여 종다양성, 분포상, 희귀촌, 우접종등에 의한 기초자료가 확보될 것으로 기대된다.

  • 1974년(年) 9월(月) 5일(日)부터 1975(年) 7월(月) 30일(日)까지 소백산(小白山) 및 안동지방(安東地方)에서 50여종(餘種)의 고등균류(高等菌類)를 채집(採集)하여 동정(同定)하였다. 그중(中) 한국(韓國) 미기록종(未記錄種)은 다음과 같다. Sing. 밤색비단그물버섯, Calodon zonatus (Fr.) $Qu{\acute{e}}l$. 갈색깔대기버섯 Patella scutellata (St. Amans) Morgan 주홍접시버섯, Peziza badia Pers. ex Merat. 자주주발버섯.

Screening Test of Wild Mushroom Extracts for Fibrinolytic Activity

  • To investigate the fibrinolytic activities of the Korean basidiomycetes, the Extracts of 50 wild mushrooms were tested for their fibrinolytic activites. Extract from Tricholoma sejunctum showed 175% increased activity to that of plasmin 1.0 U/ml. Marasmius siccus showed 54% of activity, and Laetiporus sulphureus var. miniatus and Macrolepiota procera, 43% and 26% activities, respectively to that of plasmin. But, Cystoderma amianthinum, Lepiota sp., Coprinus sp., Lycoperdon sp. were less than 10% of plasmin activity.

Thermostable Xylanase from Marasmius sp.: Purification and Characterization

  • We have screened 766 strains of fungi from the BIOTEC Culture Collection (BCC) for xylanases working in extreme pH and/or high temperature conditions, the so-called extreme xylanases. From a total number of 32 strains producing extreme xylanases, the strain BCC7928, identified by using the internal transcribed spacer (ITS) sequence of rRNA to be a Marasmius sp., was chosen for further characterization because of its high xylanolytic activity at temperature as high as $90^{\circ}C$. The crude enzyme possessed high thermostability and pH stability. Purification of this xylanase was carried out using an anion exchanger followed by hydrophobic interaction chromatography, yielding the enzyme with >90% homogeneity. The molecular mass of the enzyme was approximately 40 kDa. The purified enzyme retained broad working pH range of 4-8 and optimal temperature of $90^{\circ}C$. When using xylan from birchwood as substrate, it exhibits $K_m$ and $V_{max}$ values of $2.6{\pm}0.6\;mg/ml$ and $428{\pm}26\;U/mg$, respectively. The enzyme rapidly hydrolysed xylans from birchwood, beechwood, and exhibited lower activity on xylan from wheatbran, or celluloses from carboxymethylcellulose and Avicel. The purified enzyme was highly stable at temperature ranges from 50 to $70^{\circ}C$. It retained 84% of its maximal activity after incubation in standard buffer containing 1% xylan substrate at $70^{\circ}C$ for 3 h. This thermostable xylanase should therefore be useful for several industrial applications, such as agricultural, food and biofuel.

계룡산(鷄龍山) 지역(地域)의 균류상(菌類相)에 관한 연구(硏究)(I) (The Fungal Flora of Mt. Gyeryong National Park(I))

  • 본연구(本硏究)는 국립공원(國立公園) 계룡산지역(鷄龍山地域)의 자연보존(自然保存) 및 자연학습원조성(自然學習園造成)의 기초확립(基礎確立)을 위(爲)한 사업(事業)의 일환(一環)으로서 동지역(同地域)에 발생(發生)하는 야생균류(野生菌類)의 분포상(分布狀)을 구명(究明)하고자 실시(實施)하였다. 1981年 7月부터 10月의 고온다습기(高溫多濕期)에 계룡산지역(鷄龍山地域)에 발생(發生)하는 야생고등균류(野生高等菌類)를 조사(調査)한 결과(結果) 담자균(擔子菌) 19과(科) 53속(屬) 93종(種)과 자양균 5과(科) 7속(屬) 8종(種)이 확인(確認)되었다. 그중(中) 우점종(優點種)은 Laccaria, Marasmius, Amanita, Suillus, Boletus, Russula, Lactarius, Lycoperdon등(等)에 큰 군낙(群落)도 발견(發見)되었다. 본보고(本報告)에는 국내기록종(國內記錄種)만을 수록(收錄)하였다.

Twelve Undescribed Species of Macrofungi from Korea

  • A survey of the indigenous fungal resources of Korea was undertaken during 2014-2015. All specimens collected in this study were identified at the species level, based on their morphological characteristics and rDNA sequence data. Among them, 12 macrofungal species, viz. Agaricus guizhouensis, Amanita orientifulva, Armillaria cepistipes, Crepidotus inhonestus, Daldinia childiae, Elmerina cladophora, Lycoperdon scabrum, Marasmius brunneoaurantiacus, Otidea bufonia, Pluteus hongoi, Pluteus variabilicolor, and Russula grisea have not been previously reported in Korea.

Eight Previously Unreported Species of Fungi Identified in Mt. Manggyeong, Korea

  • A total of 118 fungal specimens were collected from Mt. Manggyeong, Chungnam province, Korea between July and October 2014. All specimens were identified to the species level using a combination of morphological characteristics and DNA sequencing. Out of 118 specimens, eight species, Amanita manginiana, A. pilosella, Calvatia holothurioides, Cantharellus tabernensis, Dicephalospora rufocornea, Gymnopus menehune, Marasmius brunneospermus, and Russula cerolens, have not been previously reported in Korea.
