Syntheses and Arene Transport Studies of Water Soluble Aminomethylcalixarene (수용성 아미노메틸칼릭스아렌의 합성 및 방향족 물질의 통과실험 연구)

  • Calix[6]arene react with formaldehyde and secondary amines to yields water soluble Mannich bases which can be converted to the corresponding quaternary salts. Treatment of the quaternary salts with a nucleophile such as cyano, ethoxy, and hydride yields p-substituted calix[6]arenes. Calix[8]arene too react with formaldehyde and diallyl amine to yield a water soluble Mannich base. The transport of neutral arenes through an aqueous phase along a concentration gradient mediated by those of water soluble calixarenes as molecular carrier was studied in a U-type cell. Naphthalene, anthracene, pyrene, and fluoranthene are tested as a neutral solid guest compounds for the transport experiment.

  • The triketones 3-6 were obtained by alkylation of cyclopentanone or cyclohexanone with the appropriate cycloalkanone bis-mannich base (1) or (2). Schmidt reaction of the tricketones 3-6 afforded the corresponding trcyclic lactames 7-10 respectively. reduction of 7 and 9 gave compounds 11 and 12 respectively.

  • ${\alpha}$-terthienyl the first isolated from natural products shows potent antitumor activity, which encouraged us to study terpyridine and its biological properties. Terpyridine has also been reported as having carcinogenicity, and it's erivatives showed high cytotoxic activities against several human cancer cell lines and topoisomerase I inhibitory ctivity. Mannich free base from condensation reaction was allowed to react with pyridinum salts to give activity. (omitted)

Synthesis of Diazacrown Ethers Containing Phenolic Side Arms and Their Complex with Divalent Metal Ions

  • The aminomethylation of phenols with para-substituents by the Mannich reaction has successfully been accomplished to produce the Mannich bases 2-6. The compounds 7-8 have also been synthesized in order to identify the effect of the side arms and t he macrocycle in the complex formation. Protonation constants and stability constants of the double armed diaza-18-crown-6 ethers 2-7 with metal ions have been determined by potentiometric method at 25 $^{\circ}C$ in 95 % methanol solution. Under a basic condition (pH > 8.0), the double-armed crown ethers 2-6 revealed stronger interaction with divalent metal ions than the simple diazacrown ether 1. The stability constants with these metal ions were Co 2+ < Ni2+ < Cu2+ > Zn 2+ in increasing order, which are in accordance with the order of the Williams-Irving series. The stability constants with alkali earth metal ions were Ca 2+ < Sr 2+ < Ba 2+ in increasing order, which may be explained by the concept of size effect. It is noteworthy that the hosts 2-6, which have phenolic side arms and a macrocycle, bind stronger with metal ions than the hosts 1 and 7. On the other hand, the host 8, which has phenolic side arms with a pyperazine ring,provided comparable stability constants to those with the host 3. These facts demonstrate that phenolic side arms play a more important role than the azacrown ether ring in the process of making a complex with metal ions especially in a basic condition. In particular, the log KML values for complexation of divalent metal ions with the hosts 2-6 had the sequence, i.e., 2 (R=OCH3) < 3 (R=CH3) < 4 (R=H) < 5 (R=Cl) < 6 (R=CF3). The stability constants of the hosts 5 and 6 containing an electron-withdrawing group are larger than those of the hosts 2 and 3 containing an electron-donating group. This substituent effect is attributed to the solvent effect in which the aryl oxide with an electron-donating group has a tendency to be tied strongly with protic solvents.

Bioluminescence immunoassay for neurotransmitter, serotonin using aequorin as a Label (Aequorin을 표지물질로 사용한 신경전달물질, 세로토닌에 대한 생물발광면역분석법)

  • A sensitive competitive heterogeneous bioluminescence immunoassay for serotonin was developed using photoprotein, aequorin as a label for the first time with the optimal assay conditions; especially, serotoninavidin conjugate was prepared by Mannich reaction and the synthetic process of serotonin-avidin conjugate was optimized by controlling the initial molar ratios of serotonin, formaldehyde and avidin (1:12,000:25). The developed bioluminescence immunoassay for serotonin showed good sensitivity (LOD of 0.68 ng/mL) with wide area of dynamic range ($5.0{\times}10^{-10}\;M\sim5.0{\times}10^{-7}\;M$). (cf. the range for serotonin in human blood serum is $151{\pm}45\;ng$/mL). In addition, cross-reactivity studies demonstrated that 5-methoxytryptamine showed some cross-reactivity (28.0%), whereas 3-methylindole, melatonin and 5-hydroxylindole-3-acetic acid showed no crossreactivity, and good recoveries were obtained in serum. Thus, this developed method provides a good tool to monitor serotonin in serum.

An Efficient Synthesis of 3,4-Dihydro-3-substituted-2H-naphtho[2,1-e][1,3]oxazine Derivatives Catalyzed by Zirconyl(IV) Chloride and Evaluation of its Biological Activities

  • An efficient and novel one-pot synthesis of new 3,4-dihydro-3-substituted-2H-naphtho[2,1-e][1,3]oxazine derivatives from 1-naphthol, various anilines and formalin at room temperature grinding is presented. The six-membered N,O-heterocyclic skeleton was constructed via zirconyl(IV) chloride promoted Mannich type reaction. In vitro antimicrobial activities of synthesized compounds have been investigated against Gram-positive Bacillus subtilis, Gram negative Escherichia coli and two fungi Candida albicans and Aspergillus niger in comparison with standard drugs. The results of preliminary bioassay indicate that some of title compounds possess significant antibacterial and antifungal activity.

Synthesis and Pharmacological Studies for New Benzotriazole and Dibenzodiazepine Derivatives as Antipsychotic Agents

  • New benzotriazoles (5-8) or dibenzodiazepine derivatives (11-18) were synthesized starting from 3-[(2-amino- 4,5-disubstitutedphenyl)amino]-5,5-disubstitutedcyclohex-2-enones (1-4) through internal coupling of their diazonium salts or internal Mannich reaction in the presence of aromatic aldehydes. Pharmacological evaluation of benzotriazole and dibenzodiazepine derivatives for their clozapine-like properties revealed that dibenzodiazepine 11 bearing 4-bromophenyl group exhibited the same antipsychotic activity as the reference drug clozapine while the activity of benzotriazole 7 was 25% lesser than that of clozapine. Moreover, compounds 7 and 11 did not show significant CNS depressant activity as well as no or slight neurotoxicity on contrast to clozapine when tested in mice using forced swim, rotarod and horizontal screen tests.

  • Chloroacylation of 3-amino-2-phenylpyrazole-4-carboxamide (2) using chloroacetyl-(propionyl) chloride affording 6-chloromethyl(ethyl)-1-phenylpyrazolo[3,4-d]pyrimidin-4[5H]-one (3) or (6). Chlorine atom in compound (3) or (6) underwent nucleophilic substitution reaction with primary or secondary amines to give 6-alkyl(aryl)aminomethyl(ethyl)-1-phenylpyrazolo[3,4-d]pyrimidin-4[5H]-one (4a-g,7a-f). When arylaminomethyl( ethyl)pyrazolopyrimidine was treated with formaline (30%) solution in ethanol, underwent Mannich reaction to afford imidazopyrazolopyrimidines (5a-e) and pyrazolopyrimidopyrimidines (8a-e). Chloromethylpyrimidine derivative 3 was converted into the corresponding mercaptomethylpyrazolopyrimidene 9, Which cyclized using bromomalononitrile or phenacyl bromide into pyrazolopyrimidothiazine 11,12.
