유충 Manduca sexta 리포포린에 의한 지방체로의 디아실글리세리드 운반 (The transfer of diacylglycerol from lipophor in to fat body in larval Manduca sexta)

  • 본 논문은 리포포린에 의해 디아실글리세리드(DAG)가 Manduca sexta 유충 지방체로 운반되는 과정을 조사하였다. $[^3H]$-DAG 표지 리포포린($[^3H]$-DAG-Lp)을 시간별로 유충 지방체와 배양하여 지방체로 운반되는 DAG의 방사능을 결정하였다. $[^3H]$-DAG-Lp와 지방체를 배양하면 지방체에 DAG가 축적되며 이의 일부는 지방체에서 트리아실글리세리드(TAG)로 전환되는 것을 알 수 있었다. Suramin과 표지되지 않은 리포포린(unlabeled Lp)의 존재 하에서는 지방체로 운반되는 DAG가 억제되는 데, 이는 DAG를 운반하는 데 리포포린 수용체가 관여한다는 사실을 알 수 있었다. Suramin의 효과는 다소 복잡하지만 리포포린 수용체에 결합하여 막의 특성을 변화시켜 DAG 운반속도에 영향을 주는 것 같다. 지질 운반과정이 수용체-매개 내포작용이라는 사실을 조사하기 위하여 내포작용 억제자인 ammonium chloride와 chloroquine을 처리하였다. 그 결과 리포포린과 지방체사이에서의 지질 운반 기작이 수용체-매개 과정이라는 사실을 보여준다.

Control Effectiveness Analysis of the hawkmoth Manduca sexta: a Multibody Dynamics Approach

  • This paper presents a control effectiveness analysis of the hawkmoth Manduca sexta. A multibody dynamic model of the insect that considers the time-varying inertia of two flapping wings is established, based on measurement data from the real hawkmoth. A six-degree-of-freedom (6-DOF) multibody flight dynamics simulation environment is used to analyze the effectiveness of the control variables defined in a wing kinematics function. The aerodynamics from complex wing flapping motions is estimated by a blade element approach, including translational and rotational force coefficients derived from relevant experimental studies. Control characteristics of flight dynamics with respect to the changes of three angular degrees of freedom (stroke positional, feathering, and deviation angle) of the wing kinematics are investigated. Results show that the symmetric (asymmetric) wing kinematics change of each wing only affects the longitudinal (lateral) flight forces and moments, which implies that the longitudinal and lateral flight controls are decoupled. However, there are coupling effects within each plane of motion. In the longitudinal plane, pitch and forward/backward motion controls are coupled; in the lateral plane, roll and side-translation motion controls are coupled.

Immunolocalization of Allatotropin Neuropeptide in the Developing Brain of the Silk Moth Bombyx mori

  • Polyclonal antiserum against Manduca sexta allatotropin has been utilized to investigate the localization of allatotropin-immunoreactivity in the brain of the si1k moth Bombyx mori. Manduca sexta allatotropin-immunoreactive (Mas-AT-IR) neurons were found in all larval brains investigated, but not in prepupal, pupal and adult brains. In the larval stages, first appearance of Mas-AT-immunoreactivity w8s shown in the brain of first instar larvae, which contains four pairs of bilateral Mas-AT-IR cell bodies. Labeled neurons increased to six pairs in the second instar larval brain, including two pairs of median neurosecretory cells in the pars intercerebralis. In the third and fourth instar larvae, five pairs of labeled cell bodies were distributed throughout each brain. In the fifth instar, there were about ten pairs of bilateral cell bodies in the day-1 brain, about seven pairs in the day-3 brains, and five pairs in the day-5 brains, respectively. Mas-AT-labeling was observed in both axons within nervi corpora cavdiaci (NCC) 1+11 and corpora allata. This suggests that the Mas-AT produced from the brain neurons is transported via some axons of the NCC 1+11 and nervi corpora allati I to the corpora allata, which appears to be a main accumulation site for the Mas-AT neuropeptide in some brain neurons produced in B. mori.

Localization of Manduca sexta Allatotropin Neuropeptides in Developing Ventral Nerve Cord of the Silk Moth Bombyx mori

  • This study has been carried out to determine localization of Manduca sexta allatotropin (Mas-AT) neuropeptide in developing ventral nerve cord of the silk moth Bombyx mori with polyclonal antisera against Mas-AT. Suboesophageal ganglion (SOG) of the second to fifth instar larvae and 3-day-old pupae showed two to ten Mas-AT-immunoreactive (Mas-AT-IR) cell bodies. There were two to three pairs of labeled cell bodies in each thoracic ganglion (TG) from third instar larvae to adults, with the exception of TG from prepupae. One pair of labeled cell bodies was localized in each abdominal ganglion (AG) 1 to 6 from third instar larvae to 3-day-old pupae, whereas in 5-day-old pupae to adults there was one pair in a similar location of AG 1 to 5. The seventh neuromeres of terminal abdominal ganglia (TAG) from third instar Iarvae to 3- day-old pupae contained four labeled large cell bodies. In each of AG 1 to 7, these cell bodies showed similar allatotropin-immunoreactivity in appearance. Some labeled axons, projected from Mas-AT-lR cells in each of those AG, were extended to the nerves N 1 and N 2.

  • Allatotropin is a 13-residue amidated neuropeptide isolated from pharate adult heads of the tobacco hornworm, Manduca serta and strongly stimulates biosynthesis of juvenile hormones in adults, but not larval, lepidopteran corpora allata. From a Bombyx mori midgut cDNA library, a cDNA that encodes a 130-amino-acid polypeptide containing M. sexta allatotropin sequence was isolated. The B. mori allatotropin cDNA consists of 1196 nucleotides. (omitted)
