GM 유채에 도입된 β-glucosidase 1 (AtBG1)의 단회투여독성시험 (Single-dose Oral Toxicity Study of β-glucosidase 1 (AtBG1) Protein Introduced into Genetically Modified Rapeseed (Brassica napus L.))

  • 유채는 십자화과로 분류되는 유지작물로써 전 세계적으로 재배되는 작물이다. 이전 연구결과에서 가뭄저항성을 증대시키는 것으로 보고된 애기장대의 ${\beta}$-glucosidase 1 (AtBG1) 유전자를 도입함으로써 가뭄저항성 형질전환 유채를 개발하였다. 새로 개발된 형질전환 작물들은 그 이용에 대한 안전성에 대하여 어떠한 정보도 제공되지 않기 때문에 작물의 안정성에 대한 증명이 필수적이다. 이러한 안정성 평가는 작물의 역사적 사용과 과학적 증거를 기초로 한다. 이번 연구에서는 AtBG1이 도입된 형질전환 유채의 급성투여독성시험을 통하여 암수 마우스 각각에서 한계투여 용량을 확인하는 것을 목적으로 하였다. 이를 위해 OECD의 급성독성의 권고안에 따라 AtBG1 단백질을 암수 각각 5마리의 ICR 마우스에 몸무게 1 kg 당 2,000 mg을 부형제에 녹여 강제구강투여 하였다. 투여일부터 14일 동안 운동성, 병리학적 이상, 몸무게를 측정하였고 마지막 14일에는 안락사 후에 부검을 통해 AtBG1 투여군과 비투여군의 차이를 확인하였다. 이상의 실험을 통하여 AtBG1 단백질이 운동성, 병리학적 이상, 몸무게 및 부검 결과에서 대조구와 실험구, 두 그룹간의 특이적인 차이를 보이지 않음을 확인하였다. 또한 AtBG1 단백질은 급성독성의 측면에서 안전한 물질로 판단되며, 한계허용치 용량이 2,000 mg을 상회함을 확인하였다. 따라서, 이는 차후 AtBG1 도입 유채의 인체위해성 평가 자료로 사용될 수 있을 것으로 사료된다.

Variation of Ginkgolides and Bilobalide Contents in Leaves and Cell Cultures of Ginkgo biloba L.

  • Ginkgolides (GK) and bilobalide are valuable compounds that belong to the lactone terpene. The contents of these metabolites were determined by HPLC from female and male tree of Ginkgo biloba L. The productivity of G. biloba cells was also compared with the corresponding individual trees. High variations in the ginkgolides and bilobalide were observed from different individuals, plant parts, and cultured cells. The ginkgolides and bilobalide contents were different depending on the plant parts. Callus was obtained from various plant tissues, and NAA was better at callogenesis than 2,4-D in both the female and male trees. The plants and their corresponding cells showed considerable variation in their ginkgolides and bilobalide concentrations. The ginkgolides and bilobalide contents were not correlated with the production between dominant trees and their corresponding cells. Light irradiation enhanced the production of GK-A and GK-B, however, the concentration of bilobalide decreased under dark conditions.

2n 배우자 형성 2배체 감자의 육성연구 (Studies on 2n gametophyte Producing Diploid Potato Clones)

  • 우수 이위체 clone을 선발하고 이를 FDR clone으로 육성하고자 본 실험 이 수행되었다. 1) 도입된 2X종자를 포장 및 온실에서 재배하여17 clone이 선발되었으며 이들의 특성이 조사되 고, 이들 Clone을 인공교배, 개방수분, 집단수분하여 그후대에서 18clone 이 선발되었다. 2) 선발된 clone들은 2n웅성과 2n자성배우자 형성에 대하여 조사되었다. 그중 D6-21만이 27∼30%의 2n웅성 배우자를 FDR에 의해 형성하였고. 다른 clone들은 2n웅성 배우자 형성율이 7% 미만이었다. 3) 자성 2n배우자를 생성하는 clone은 발견되지 않았다.

작은 생물을 연구하는 과학자에 대한 초등학생들의 인식 및 이미지 분석 (Perceptions and Image Analysis of Elementary Students on Scientists studying Small Organisms)

  • We investigated perceptions and image analysis on scientists studying small organisms reflected in elementary student's drawing using a modified version of the Drawing-A-Scientist-Test. The participants were 530 of fifth and sixth graders consisted of 449 ordinary students and 81 science gifted students. The data were collected from associated words, images and explanatory notes depicted by students engaged in questionnaires. The results indicated that a larger number of students reminded small sized animals and/or plants as words associated with small organisms. In addition, some students depicted anthropomorphic or abstract microorganisms. In this study, more stereotypes of scientists' appearance were exhibited at sixth graders and city region group. Most of the students depicted indicators such as lab coat, glasses, scientific instruments for observing, indoor, male and young, whereas only a few students depicted collaborative work. There was statistically significant difference between girls and boys, because boys perceived male scientists only, while half of girls depicted female. More frequent research instruments and scientific captions were used when science gifted students depicted scientists studying small organisms. These results could be contributed to education on microorganisms in elementary science.

The Stable Production of Organic Seed to Distribute Certified Seed of Waxy Corn Hybrid

  • In order to produce the organic foods in accord with international standard, organic seeds should be used in organic farming. This study was conducted to establish the stable production of organic seed of waxy com by examining the growing characteristics, seed yield, and the economy for seed production by organic farming. The optimal sowing timing for organic seed production of waxy com hybrid was within 10 days of the $1^{st}$ of May with yield of 88~90% of conventional seed production. The optimal planting density was 41,600 plants/ha ($80{\times}30$ cm) for organic seed production of waxy com. The weight of 100 seeds and seed productivity increased at the planting ratio of 2:1 mother plant:male plant. Growth and seed production were improved by removing male plant at 7~10 days after silking. Organic fertilizer (mixed oil cake) was applied at a rate of 4~6 Mg/ha before sowing. Black plastic mulching was used for weed control. In addition, sex pheromone trap and bio-control agents were used for safe pest control and low labour cost.

참나리와 Asiatic hybrid "Dreamland"의 교잡 후대 특성 (The Characteristics of Progenies derived from Lilium lancifolium and Asiatic hybrid "Dreamland")

  • 참나리(Lilium lancifolium)는 우리나라 자생나리로 생육이 왕성하고 후자리움에 내성이 있으며 주아를 형성하는 등 많은 장점을 가진다. 이를 자방친으로 하여 Asiatic hybrid "Dreamland"의 상향이고 흑갈색의 반점이 없는 형질을 이입시키기 위해 교배를 수행하였다. 참나리와 "Dreamland"의 화분발아율은 각각 30과 60%이었다. 이들 후대의 개화일자별 화분의 평균 발아율은 25-57%로 양친의 중간에 분포하였다. 개화는 정식 후 124일째 가장 많았다. 엽형지수는 5 이상인 식물체가 약 99%로 나타나 자방친인 참나리의 영향이 우세하였다. 화색은 4가지로 분리되었다. Orange와 light orange가 각각 51.5와 44.2%로 대부분을 차지하여 자방친의 영향이 우세하였다. 그리고 양친의 중간색인 orange yellow와 light yellow orange는 각각 3.1과 1.2%로 분리되었다. 꽃잎의 반점은 1개체를 제외한 나머지 모든 개체에서 관찰되었다. 개화 방향은 상향, 측향 및 하향 3가지로 분리되었다. 측향 즉, 자방친과 화분친의 중간형이 73% 이었다. 주아는 49.4% 형성되었으며 그 중 최대 25개까지 착생되었다. 평균 주아수는 5개이었다.

Biology of a predatory bug Eocanthecona furcellata Wolff (Hemiptera : Pentatomidae) on Vapourer tussock moth larvae: a major pest of tasar silkworm food plants

  • Tropical Tasar culture is forest and agro-based activity that covers agricultural and industrial activity. Tasar silk is produced by the silkworm, Antheraea mylitta which is polyphagous in nature. A large number of pests are reported to attack both the host plant as well as silkworm. As the rearing is conducted outdoor silkworms are exposed to attack of various pests during their life cycle. Eocanthecona furcellata (Hemiptera: Pentatomidae) is the major predator of tasar silkworm during young age rearing. Both nymphs and adults of the stink bug suck the heamolymph of silkworm leading to death. The present work was undertaken to study the life cycle of E. furcellata on larvae of vapourer tussock moth which is a major pest of tasar host plants. The incubation period, nymphal duration and total developmental period from egg to adult was $8.0{\pm}0.45d$, $16.0{\pm}3.24$ and $22.0{\pm}4.20d$, respectively. Pre-oviposition, oviposition and longevity of male and female stink bug was $3.20{\pm}0.38$, $17.60{\pm}1.86$, $42.40{\pm}1.94$, $37.00{\pm}3.18d$, respectively. Analysis of the data revealed no significant difference in life cycle of the predator when provided silkworm and vapourer tussock moth larvae as hosts. Which clearly indicates that E. furcellata can multiply on vapourer tussock moth larvae during non rearing periods. Adoption of recommended pest control measures for defoliators of tasar host plants during non-rearing periods will help in reducing the incidence of host plant pests and also in reducing the incidence of stink bug during rearing season. During the study it was also observed that larvae semilooper, boll headed caterpillar and nymphs of leaf hopper serve as alternate/secondary hosts to E. furcellata during non rearing seasons.

가정내 식물 가꾸기 활동이 초등학생들의 정서지능에 미치는 영향 (The Effects of Plant Raising Activities in Home Upon School Student' Emotion Intelligence)

  • 가정내 식물 가꾸기 활동이 초등학생들의 정서지능에 미치는 영향을 검증하고자 하였다. 실험집단의 사후검사시 통제집단에 비하여 정서인식, 정서표현, 정서이입, 정서조절, 정서활용모두에서 유의차가 나타나 가정내 식물 가꾸기 활동이 초등학교 고학년 학생들의 정서지능에 긍정적으로 영향을 미치는 것으로 나타났다. 성별에 따른 정서지능 변화에서도 실험집단의 남학생은 통제집단의 남학생보다 높게 나타났고, 정서인식, 정서표현, 정서이입, 정서조절, 정서활용 모두에서 유의차가 나타났으며, 실험집단의 여학생도 같은 경향이었다. 따라서 가정내 식물 가꾸기 활동은 초등학교 고학년 학생들의 정서지능 향상에 긍정적인 영향을 미치는 것으로 볼 수 있다.

Development of a CMS-specific marker based on chloroplast-derived mitochondrial sequence in pepper

  • Molecular markers developed from the flanking sequences of two cytoplasmic male sterility (CMS)-associated genes, orf456 and ${\Psi}atp6-2$, have been used for marker-assisted selection of CMS in pepper. However, in practice, the presence of orf456 and ${\Psi}atp6-2$ at substoichiometric levels even in maintainer lines hampers reliable selection of plants containing the CMS gene. In this study, we developed a novel CMS-specific molecular marker, accD-U, for reliable determination of CMS lines in pepper, and used the newly and previously developed markers to determine the cytoplasm types of pepper breeding lines and germplasms. This marker was developed from a deletion in a chloroplast-derived sequence in the mitochondrial genome of a CMS pepper line. CMS pepper lines could be unambiguously determined by presence or absence of the accD-U marker band. Application of orf456, ${\Psi}atp6-2$and accD-U to various pepper breeding lines and germplasms revealed that accD-U is the most reliable CMS selection marker. A wide distribution of orf456, but not ${\Psi}atp6-2$, in germplasms suggests that the pepper cytoplasm containing both orf456 and ${\Psi}atp6-2$ has been selected as CMS cytoplasm from cytoplasm containing only orf456. Furthermore, factors other than orf456 may be required for the regulation of male sterility in pepper.

초등교사의 과학과 환경 교육에 대한 수업실태 분석 및 인식도 (A Study on the Elementary School Teachers Perception of the Subjects of Environment-related Education in Science and of Their Instructional Methods)

  • The purpose of this study was to analyze the elementary school teachers'perception toward the environment-related subjects in science education and to investigate their instructional methods in delivering the subjects in the classroom. The participants of this study were 300 elementary school teachers sampled from elementary schools in Kyung Nam Province. The findings of this study were as follows; First, more than half of the teachers surveyed showed that the environment-related units and contents in the science textbook were insufficient to the existing issues. Second, the large portion of teachers responded that the teaching materials in the textbook were insufficient to deliver the contents. Third, the largest portion of respondents replied that their preferred instructional method was a teacher-directed instruction followed by a case study approach. Fourth, in terms of the effectiveness of environmental education, both male and female teachers showed negative responses. Furthermore, male teachers were more negative than female teachers. Fifth, total means of their preference of the environment-related units was 3.13. The unit for sixth graders titled`Environmental Pollution and Protecting Nature'showed the highest preference, whereas the unit for fifth graders titled`Structure and Functions of Plants'showed the lowest preference. Sixth, there was no statistically significance in gender and their working areas. Seventh, there was statistically significant differences between male and female teachers in terms of teaching experience. In detail, the teachers with 5 to 9 teaching experience showed the lowest scores, while the teacher with more than 15 years teaching experience showed the highest scores in their perception of the units. Eighth, there was a significant difference between two groups in terms of their educational background. For example, two-year college graduates showed higher preference than four-year college graduates.
