Association between consumption of milk and dairy products, calcium and riboflavin, and periodontitis in Korean adults: Using the 2007-2010 Korea National Health and Nutrition Examination Surveys (한국 성인의 우유 및 유제품과 칼슘 및 리보플라빈 섭취량과 치주염 간의 연관성: 2007~2010년 국민건강영양조사 자료를 이용하여)

  • Purpose: The current study was designed to investigate the relationship of dietary calcium and riboflavin and their main dietary source (milk and dairy products) with the risk of periodontitis using data from 2007 to 2010 Korea National Health and Nutrition Examination Surveys. Methods: A total of 1,690 adults aged ${\geq}40$ years were included. We used results of dental examination regarding all sextant information on probing depth of at least two index teeth, nutritional assessment by a single 24-hour dietary recall, and demographic and medical information. The periodontitis group was defined as those who had 3-4 points, and the normal group was defined as those who had 0 points of Community Periodontal Index at all locations of six examination sites using a probe. Results: Using multiple logistic regression analysis, after adjustment for age, body mass index, energy intake, income, smoking, and alcohol drinking, we found an inverse relationship between consumption of dairy products and risk for periodontitis (OR: 0.465, 95% CI: 0.224-0.964) and between dietary riboflavin intake more than the estimated average requirements and risk for periodontitis (OR: 0.535, 95% CI: 0.300-0.954) in males. Conclusion: Adequate intake of milk dairy products and riboflavin may be recommended for prevention of periodontitis in the Korean male population.

  • Sexed sperm can contribute to increase the profitability of the cow industry through the production of offspring of the craved sex, such as males for meat or females for dairy production. Therefore, the utilization of sexed sperms plays a very important role in the production of offspring of superior cattle. In this study, we examined the pregnancy rates and calves sexing proportion of male and female calves produced using AI, both performed using sexed and conventional sperm. In the result, the conception rates after ET were 73.3% (33/45) sexed semen and 52% (55/104) conventional semen. Thus, the sex ratio for sexed-semen inseminations was 70% (21/30) females for singleton births within a 272 to 292 day gestation interval. The sex ratio for conventional semen was 61% (34/56) females for births. As a result, it is suggested that the use of sex classification sperm will play a very important role in the offspring production of Korean bovine.

Use of the Non-electrophoretic Method to Detect Testis Specific Protein Gene for Sexing in Preimplantation Bovine Embryos

  • Testis-specific protein (TSPY) is a Y-specific gene, with up to 200 copy numbers in bulls. In order to make bovine embryo sexing under farm condition more feasible, the possibility of using a non-electrophoretic method to detect the TSPY gene for sexing bovine early embryos was examined. Primers were designed to amplify a portion of the TSPY gene and a common gene as an internal control primer. PCR optimization was carried out using a DNA template from bovine whole blood. Furthermore, embryo samples were diagnosed by this method and the sexing results were contrasted with those of the Loop-Mediated Isothermal Amplification (LAMP) method. The results showed that TSPY was as reliable a sexing method as LAMP. Forty-three morula and blastocyst embryos collected from superovulated donor dairy cattle were sexed by this method, and twenty-one embryos judged to be female embryos were transferred non-surgically to recipients 6 to 8 days after natural estrus. Out of 21 recipients, 9 were pregnant (42.86%) and all delivered female calves. The results showed that the sex predicted by this protocol was 100% accurate. In conclusion, the TSPY gene was a good male specific marker and indicated that a non-electrophoretic method was feasible and accurate to detect the TSPY gene for sexing preimplantation bovine embryos.

A Survey of Food and Nutrient Intakes of Korean Men by Age Groups (한국 남성의 연령별 식품 및 영양소 섭취량 조사)

  • A dietary survey was conducted using 24-hour recall method in 80 male elementary school children, 83 high school students, 87 adults and 98 elderly people over 65 years of age to discuss food and nutrient intakes among different age groups. In observing food intake by age groups, the percentage of plant foods to total food intake was significantly higher in the elderly than other groups and that of animal foods was the highest in the children. The children consumed significantly higher amounts of milk and dairy products. Carbohydrate energy percent in the elderly people was 68.0%, significantly higher than other age groups. All age groups consumed Ca below Korean RDA and especially, 65.1% of the adolescents and 68.4% of the elderly consumed below 75% of RDA. Nutrient adequacy ratio (NAR) of most nutrients was lowest in the elderly. Mean adequacy ratio (MAR), an index of overall dietary quality, was 0.94 for the children, 0.84 for the adolescents, 0.84 for the adults, and 0.73 for the elderly. The above results suggest that food and nutrient intakes have been differently affected with advancing age, and overall dietary quality of the elderly people is lower than that of other age groups.   • This survey was carried out to investigate the correlationship between the nutrition knowledge scores and food habit of the mothers and food preference and other factors of the 272 elementary school children in Masan. The results were summarized as follows : 1. Children's most favorite food was fruits, but they dislike spicy vegetables and some fermented foods. Preferences of the male children to food groups such as protein, calcium and carbohydrate were significantly higher than those of the female children. 2. The rate of the children who take between-meal snacks 2 o 3 times a day was high and those children who skip breakfast take snacks more often. They prefer to eat fruits, dairy products and cakes as their snacks. The body weight of the children who take snacks more frequently was rather lower. 3. As the mother's nutrition knowledge scores rise, so do those of food habit scores. The nutrition knowledge scores go up according to the education degree of the mothers, but the food habit scores seem to have little to do with their education degree. The nutrition knowledge scores of the working mothers were lower than those of the non-working mothers. 4. There was strong positive correlation between the food preference on the food group and those on the other food groups of the children. As the food habit scores of the mothers rise, the children prefer to take calcium and carbohydrate group. Those who take more fat showed higher body weight.

Effects of Fetal Calf Serum and Gonadotropins Supplemented to the Medium on Maturation and Fertilization In Vitro of Porcine Follicular Oociytes (성선(性腺)자극호르몬과 우태아혈청(牛胎兒血淸)첨가가 돼지난포란(卵胞卵)의 체외성숙(體外成熟) 및 체외수정(體外受精)에 미치는 영향)

  • This study was carried out ot investigate the effects of fetal calf serum (FCS) and gonadotropins supplemented to the medium on maturation and fertilization in vitro of porcine follcular oocytes. Ovaries were obtained from gilts at local slaughter-house. Oocyte-cumulus complexes were recovered by puncturing the ovarian follicles(3~5 mm in diameter). The complexes from individual ovaries were pooled in a $0.4m{\ell}$ droplet of medium covered with paraffin oil, then washed twice in fresh droplet and cultured for 36hrs in culture media according to experimental conditions. Boar epididymal spermatozoa were capacitated by preincubation for 4hrs in m-KRB medium and the preincubated spermatozoa were insemenated in the fertilization medium containing the cultured oocytes. The results obtained in this study are summarized as follows: 1. The maturation rates of oocytes cultured in m-KRB and m-KRB supplemented to 10% FCS were 82 and 37%, respectively. When PMSG, hCG. and PMSGt hcG($10Iu/m{\ell}$) were added to the media supplemented to 10% FCS, the maturation rates were 66, 58 and 68%, respectively. 2. Expansion of cumulus cells was not occured in m-KRB and m-KRB supplemented to 10% FCS. However, when PMSG, hCG and PMSG+hCG($10Iu/m{\ell}$) were added to m-KRB supplemented to 10% FCS, the expansion rates of cumulus cell layers were 92, 13 and 91%, respectively. 3. When oocytes were mltured in m-KRB, the rates of penetration and formation of male pronucle: were 93 and 7%, respectively. By adding FCS and gonadotropin to m-KRB, the penetration and formation of male pronuclei were 100 80%, respectively.

Evaluation of Nutrient Intake and Food Variety in Korean Male Adults according to Framingham Risk Score (Framingham Risk Score에 의한 한국 성인 남성의 영양소 및 식품 섭취의 다양성 평가)

  • The purpose of this study was to evaluate dietary intake according to the risk of coronary heart disease (less than 10% = low-risk group; 10~20% = middle-risk group) based on Framingham risk score (FRS), on 122 male adult subjects. The body weight and body mass index were not significantly different between the groups, while height of the low-risk group was shown to be significantly high compared to that of the middle-risk group. The daily energy intake was shown to be significantly high in the low-risk group with 1,910.88 kcal, compared to 1,606.63 kcal of the middle-risk group. As a result of analyzing nutrient intake per 1,000 kcal of energy, while the low-risk group had significantly high intake of animal protein, fat, and animal fat compared to the middle-risk group, the intake of plant protein, carbohydrate, and plant iron was found to be significantly low. The daily food intake was shown to be significantly high in the low-risk group (1,445.16 g), compared to the middle-risk group (1,075.12 g). The low-risk group was found to have significantly high intake of sugars, eggs, and beverages compared to the middle-risk group, while mushrooms intake was significantly high in the middle-risk group. Dietary variety score (DVS) was significantly high in the low-risk group with 26.42, compared to 22.66 of the middle-risk group. Dietary diversity score (DDS) was indicated to be significantly high in the low-risk group with 3.70, compared to 3.27 of the middle-risk group. The low-risk group was indicated to have significantly high score in DDS of dairy products and fruit group, compared to the middle-risk group. In the correlation between diversity index of food intake (DVS and DDS) and FRS, DDS was shown to have significantly negatively correlation with FRS after adjusting for confounding factors. To sum up these results, the adult males with low-risk of coronary heart disease had more various consumptions of fruits and milk, compared to the subjects with the middle-risk. The proportion of consuming major food groups such as cereals, meat group, milk, fruits, and vegetables more than a fixed quantity was indicated to be high. Accordingly, dietary habit for intake of various food seems to be necessary, to prevent coronary heart disease.

Yearly trend of milk intake in Korean children and adolescents and their nutritional status by the milk intake level using 2007-2015 Korea National Health and Nutrition Examination Survey (아동과 청소년의 연도별 우유 섭취량 변화와 우유 섭취량에 따른 영양상태 평가: 2007-2015 국민건강영양조사를 바탕으로)

  • Purpose: This study examined the yearly trend of milk consumption and the nutritional status of subjects aged 6-18 years using the 2007-2015 Korea National Health and Nutrition Examination Survey data. Methods: Milk and dairy products were classified into plain milk, flavored milk, and dairy products (ice cream, milkshakes, cheese, and yogurt, etc.). This study compared the milk and dairy products intakes, some nutrients intakes and percent of dietary reference intakes for Koreans in the milk intake and non-milk intake groups. Results: Plain milk intake decreased with year (male, p = 0.0199; female, p < 0.0001; elementary school, p = 0.0013; high school, p = 0.0061), whereas flavored milk and dairy products intake in these subjects increased with year. In all subjects, 49.9% of subjects did not drink milk at all. The intakes of energy, protein, fat, calcium, phosphorus, and riboflavin in the milk intake group were significantly higher than those in the non-milk intake group, even after adjusting for covariates (p < 0.05). The odds ratio of the prevalence of nutritional deficiency in the non-milk intake group was 3.2 times higher than that of the milk intake group (p < 0.001), even after adjusting for covariates. The odds ratio for the prevalence of excess intake of the energy/fat was not significant with milk intake. Conclusion: The prevalence of milk intake decreased every year in the subjects. Calcium deficiency and nutritional deficiency were very high in the non-milk intake group. Efforts should be made to improve the calcium status in children and adolescents by strengthening nutrition education about the importance of milk intake.

Embryo Gender Ratio and Developmental Potential after Biopsy of In Vivo and In Vitro Produced Hanwoo Embryos

  • The present study was to assess the in vitro viability and sexing rate of bovine embryos. Blastocysts were harvested on day 7~9 day after insemination(in vitro and in vivo), and the sex of the embryos was examined using the LAMP method. Embryo cell biopsy was carried out in a $80{\mu}l$ drop $Ca^{2+}$, $Mg^{2+}$ free D-PBS and, biopsied embryos viability were evaluated after more 12 h culture in IVMD culture medium. The formation of recovered embryo to expanded and hatching stages had ensued in higher of sexed embryo in vivo than in vitro (100% vs. 89%, p<0.05), and in vitro, the rates of degeneration after sexing were significantly (p<0.05) higher in vitro than in vivo(11% vs. 0.0%). The rates of the predicted sex were female 61% vs. 56%, and male 39% vs. 44% in vivo and in vitro, respectively. The rates of survival following different biopsy methods were seen between punching and bisection method in vivo and in vitro (100% vs. 89% and 100% vs, 78% respectively). Biopsy method by punching was significantly (p<0.05) higher than bisection between produced embryos in vivo and in vitro. The present data indicate that with microblade after punching for embryo sexing results in high incidence of survivability on development after embryo biopsy. It is also suggested that LAMP-based embryo sexing suitable for field applications.

QuEChERS-based determination of tissue residues and acute toxicity of pyraclofos in rat (QuEChERS 법을 이용한 Rat 조직내 Pyraclofos 잔류 분석 및 급성독성 평가)

  • Environmental pesticides used for insect control can be transferred from plants to animals even to livestock animals through food chain. Human beings also can be exposed to pesticides by consuming polluted dairy products, including meats, eggs and other milk products. Therefore, the Ministry of Food and Drug Safety (MFDS) established Standard for Pesticide Residue Limits in dairy products. The QuEChERS (quick, easy, cheap, effective, rugged and safe) methods for detecting residual pesticides are relatively well established for fruits and vegetables, however, the methods for meat have not been appropriately studied yet. In the present work, pyraclofos was used as an organophosphate pesticide to examine its tissue residue in experimental animals by QuEChERS methods. For this, pyraclofos (150 mg/kg body weight) was orally administered to male rats once a day for 2 days. After 6, 12, and 24 hr of the treatment, the tissue residues in liver and femoral muscle of the rats were determined using QuEChERS methods followed by HPLC analyses. In preliminary studies, the recovery rates of spiking samples of pyraclofos demonstrated approximately 109~110% from the tissues. In previous study, pyraclofos tissue residues were observed with significantly high levels in livers and muscles at 6 hr of oral treatment. Then, they were almost completely disappeared after 24 hr of the administration, indicating the orally exposed pyraclofos is rapidly absorbed and distributed to body organs, then quickly excreted from the body with a negligible level of tissue residue. The alterations in blood chemistry as well as the histopathology of heart, lung, liver, spleen and kidney have also been investigated in the experimental animals for assessing acute toxic effects of pyraclofos. The obtained blood chemistry indexes (ALT and AST) showed maximum peak values at 12 hr after the oral administration and decreased to the normal levels at 24 hr of the treatment. Histopathologic observation exhibited acute hepatic damages at 24 hr of the treatment. In conclusion, we suggest that QuEChERS method can be adequately optimized for the analysis of pyraclofos residues in animal tissues.
