  • 1. Formalin $50\~60ppm$, malachite green 0.2ppm, dipterex 0.6ppm 되도록 혼합하여 순환수조내의 어류기생성원충류인 Chilodonella, Costia, Trichodine, Ichthyophthirius등을 근절할 수 있었으며 이때 어체의 아가미 조직에 변화는 관찰되지 않았다. 이때에 기생충은 6기간 후부터 구제되었다. $2.130\~140ppm$ formalin에 $24\~48$시간 약욕시킨 잉어의 아가미 상피에 병리학적 변성을 볼 수 있었다. 이 경우 치료약에 장시간 약욕시킴으로서 새엽상피가 변성 괴사되고 새엽지주세포가 떨어졌다.

  • A simple, rapid, reproducible and selective UV-Visible spectrophotometric method has been developed for determination of germanium in a week acid medium (pH = 3.0) in the presence of mandelic acid (MA) and malachite green (MG). A single extraction in chlorobenzene was used for the analysis. The germanium-MA ion association complex exhibits an absorption maximum at ${\lambda}_{max}=618nm$ with MG as counter ion and has an apparent molar absorptivtity of $1.313{\times}10^5L\;mol^{-1}\;cm^{-1}$. The values obtained by this UV-Visible spectrophotometric method are in good agreement with the values from inductively coupled plasma-mass spectrometer analysis.

포도 저장을 위한 아황산가스 발생제의 제조 및 처리 방법 (Preparation and Treatment of Sulfur Dioxide Gas Generating Agent for Storage of Grape Fruits)

  • 아황산가스 발생제를 자가 제조하여 포도의 저장 중 병해발생 방지에 이용하는 방법을 확립하기 위하여 sulfite 화합물의 아황산가스 발생 정도와 지속 기간을 측정하고 아황산가스 발생 조절 및 간이 검출 방법을 개발하였다. 포도의 저장 중 잿빛곰팡이병을 일으키는 B. cinerea의 포자 발아와 균사생장은 각각 400 ppm과 3200 ppm의 아황산가스 농도 조건에서 억제되었다. 5종류의 sulfite 및 bisulfite 화합물 중 sodium hydrosulfite는 아황산가스 발생량이 가장 높았으며 이를 비닐 포장하여 포장의 pinhole수를 조절하거나 산 또는 알카리의 pH 조절제를 혼합하면 가스 발생량과 발생 지속 기간을 조절할 수 있었다. Malachite green 용액은 아황산가스에 의해 정량적으로 탈색되어 아황산가스 검출에 이용할 수 있었으며 여과지에 흡수하여 건조할 경우 아황산가스 검출지로 활용할 수 있었다. MBA 포도를 속효성과 지효성의 아황산가스 발생제를 동시에 처리하여 저온 저장할 경우 병해 발생을 감소시킬 수 있었다.

Isotherm, kinetic and thermodynamic studies of dye removal from wastewater solution using leach waste materials

  • In this study, Malachite Green (MG) dye removal from synthetic wastewaters by adsorption process using raw boron enrichment waste (BEW) and it's modifications (with acid and ultrasound) were aimed. 81% MG removal was obtained by BEW at optimum equilibrium conditions (time: 40 min., dosage: 500 mg/dm3, pH: 5-6, speed: 200 rpm, 298 K). MG removal from wastewaters using acid modified boron enrichment waste (HBEW) was determined as 82% at optimum conditions (time: 20 min., dosage: 200 mg/dm3, pH: 10, speed: 200 rpm, 298 K). For ultrasound modified BEW (UBEW), the highest MG removal percent was achieved as 84% at optimum conditions (time: 30 min, dosage: 375 mg/ dm3, pH: 8, speed: 200 rpm, 298 K). The equilibrium data of Malachite Green was evaluated for BEW, HBEW and UBEW adsorbents by using sorption isotherms such as Langmuir, Freundlich and Temkin models, out of which Langmuir model (R2 = 0.971, 0.987 and 0.984) gave better correlation and maximum adsorption capacity was found to be 147.05, 434.78 and 192.30 mg/g, respectively. The adsorption kinetics followed the pseudo-second-order kinetic equation for sorption of MG onto wastes. A look at thermodynamic data reveals that natural sorption is spontaneous and endothermic because of free negative energy exchange and positive change in enthalpy, respectively. The results indicated that boron enrichment waste, and HCl and ultrasound-modified boron enrichment waste served as good alternative adsorbents in dye removal from wastewater.

Application of Three-phase Hollow Fiber LPME using an Ionic Liquid as Supported Phase for Preconcentration of Malachite Green from Water Samples with HPLC Detection

  • A novel three-phase hollow fiber liquid phase microextraction was developed for the determination of malachite green (MG) in environmental waters, which selected [BMIM][$PF_6$] mixed with 1% trioctylphosphine oxide (TOPO) as supported phase. Several parameters (accepter phase pH, sample pH, supported phase membrane, volume of accepter phase, salinity, extraction time) that could affect extraction performance were investigated. Under the optimal extraction conditions, the established approach showed excellent characters as: high enrichment factor (212), wide linear range ($0.20-100{\mu}gL^{-1}$), low detection limit ($0.01{\mu}gL^{-1}$), good reproducibility (RSD, 8.9%, n=5) and satisfactory recovery (84.0-106.2%). The method was applied to detect MG at Yangtze River and pond waters in Zhenjiang, Jiangsu province, and 4 sites among 15 sampling sites were found MG with the concentration of $1.73-11.06{\mu}gL^{-1}$, which confirmed that the proposed environmentally friendly method was simple and effective for monitoring MG in aquatic system.

사찰에 사용된 적색 및 녹색안료의 종류와 입자특성 분석 (Species and Characteristics of Particles for Traditional Red and Green Pigments used in Temples)

  • 사찰의 단청 등 회화에 사용된 적색과 녹색안료에 대해 그 종류를 밝혀내고, 이들의 광물입자 특성을 관찰하여 서로 비교분석하고자 하였다. 성분분석 결과, 적색안료는 진사(Cinnabar)와 석간주(Hematite), 연단(Minium) 등 3가지 종류가 사용된 것으로 확인되었고, 녹색안료는 녹염동광(Atacamite)과 뇌록석(Celadonite) 그리고, 일부 석록(Malachite)이 확인되었다. 이는 사찰별로 독특한 안료를 사용하였다기 보다는 다양한 종류의 안료를 회화의 특성에 맞게 모두 이용하였음을 알 수 있다. 안료의 입자특성에서는 적색안료 중에서 같은 진사라 할지라도 매끄럽거나 길쭉하고 각진모양, 판상 등 자형(hedral) 또는 타형(anhedral)의 다양한 형태를 가지고 있으며, 이는 녹색안료에서도 유사한 패턴을 보였다. 특히 지금까지 안료의 성분과 결정구조분석을 통해 광물을 확인하는 방법이 주로 이용되어왔으나 이에 비해 입자의 형태적 특성을 이용하는 것이 색상과 종류가 유사한 안료 그룹내에서는 이들을 보다 세밀하게 구분할 수 있고, 인자화 하는데 유리한 방법일 수 있을 것으로 판단된다.

  • "Kimchi", which is a main side-dish on Korean table is the pickled vegitables with seasoning substances : such as onion red pepper, salt, water and etc. In this paper, the variation of pH, acidity (calculated as lactic acid) and sugar contents (calculated as dextrose) are investigated during the fermentation period of Kimchi and the effects of antiseptics (Brilliant green, malachite green, sodium benzate, salicylic acid, P-dichlor amino sulfonyl-benzoic acid) and hydrochlonic acid to prevent spoiling are studied on the Kimchi which is prepared by a simple method in this laboratory.

Simultaneous Kinetic Spectrophotometric Determination of Sulfite and Sulfide Using Partial Least Squares (PLS) Regression

  • The partial least squares (PLS-1) calibration model based on spectrophotometric measurement, for the simultaneous determination of sulfite and sulfide is described. This method is based on the difference between the rate of the reaction of sulfide and sulfite with Malachite Green in pH 7.0 buffer solution and at 25 ${^{\circ}C}$. The absorption kinetic profiles of the solutions were monitored by measuring the decrease in the absorbance of Malachite Green at 617 nm in the time range 10-180 s after initiation of the reactions with 2 s intervals. The experimental calibration matrix for partial least squares (PLS-1) calibration was designed with 24 samples. The cross-validation method was used for selecting the number of factors. The results showed that simultaneous determination could be performed in the range 0.030-1.5 and 0.030-1.2 $\mu$g m$L ^{-1}$ for sulfite and sulfide, respectively. The proposed method was successfully applied to simultaneous determination of sulfite and sulfide in water samples and whole human blood.

Bioprocess of Triphenylmethane Dyes Decolorization by Pleurotus ostreatus BP Under Solid-State Cultivation

  • With an aim to evaluate dye decolorization by white rot fungus on natural living conditions, reproducing by solid-state fermentation, the process of triphenylmethane dyes decolorization using the white rot fungus P. ostreatus BP, cultivated on rice straw solid-state medium, has been demonstrated. Three typical dyes, including malachite green, bromophenol blue, and crystal violet, were almost completely decolorized by the fungus after 9 days of incubation. During the process of dye decolorization, the activities of enzyme secreted by the fungus, and the contents of soluble components, such as phenolic compounds, protein, and sugar, changed regularly. The fungus could produce ligninolytic, cellulolytic, and hemicellulolytic enzymes and laccase was the most dominant enzyme in solid-state medium. Laccase, laccase isoenzyme, and the laccase mediator could explain the decolorization of malachite green, bromophenol blue, and crystal violet by the fungus in solid-state medium, respectively. It is worth noting that the presence of the water-soluble phenolic compounds could stimulate the growth of fungus, enhance the production of laccase, and accelerate dye decolorization.
