Great success has been being achieved in identification and utilization of candidate genes affecting economically important as well as disease related traits in animals for the past two decades. This article reviews the current status of candidate genes in animals, with particular emphasis on the disease causing genes in livestock. Two different types of phenotypes, monogenic and polygenic, are also described. Finally, commercialized animal DNA markers, which can be used for the molecular breeding in near future, are discussed.
Food production is a major role of agriculture. It has been projected that the world population continues to increase by the middle of the 21st century, and the population growth results in raising a serious problem of food shortage. Thus we have to increase food as possible. A considerable amount of crops have been abandoned due to short-life after postharvest. Ethylene is a factor responsible for the postharvest loss in crops, especially horticultural crops. If we can reduce ethylene production or sensitivity by genetic engineering, we can develop, so called,“long-life crop”conferring long postharvest lives. During last two decades, intensive research for molecular dissection of ethylene biosynthesis has been carried out, and the researchers have succeeded in engineering ethylene productivity in some crops. On the other hand, after the successful isolation of Arabidopsis ethylene receptor gene ETR1, the homolog genes have been isolated in various plant species. Currently the characterization of these genes and alteration of ethylene sensitivity using the genes are in progress. This review summarizes current progress in the analysis of these genes, and discusses genetic engineering of ethylene sensitivity using these genes.
Avermectin and its analogs are major commercial antiparasitic agents in the fields of animal health, agriculture, and human infections. To increase our understanding about the genetic mechanism underlying avermectin overproduction, comparative transcriptomes were analyzed between the low producer S. avermitilis ATCC31267 and the high producer S. avermitilis ATCC31780 via a S. avermitilis whole genome chip. The comparative transcriptome analysis revealed that fifty S. avermitilis genes were expressed at least two-fold higher in S. avermitilis ATCC31780. In particular, all the avermectin biosynthetic genes, including polyketide synthase (PKS) genes and an avermectin pathway-specific regulatory gene, were less expressed in the low producer S. avermitilis ATCC31267. The present results imply that avermectin overproduction in S. avermitilis ATCC31780 could be attributed to the previously unidentified fifty genes reported here and increased transcription levels of avermectin PKS genes.
The root hair provides a major entering spot for the symbiotic legume rhizobia. It is obvious that dynamic cell wall modification occurs in the plant root hair during the early microbe invasion. Expansins are nondestructive cell wall-modifying proteins that are involved in cell growth and differentiation. Among about 40 expansin genes in Arabidopsis, two expansin genes are expressed specifically in the root hair cell. Orthologous genes of this Arabidopsis root hair expansins have been found in other Brassica members, rice, and Medicago truncatula (a legume). In this review, I discuss the probable function of expansins during the early symbiotic process between the root hair and microbes and the regulation of root hair expansin genes in a comparative approach.
One hundred and three random complementary DNA clones representing rainbow trout intestine were par1i811y sequenced as an approach to analyze the transcribed sequences of its genome. Of the sequences generated, 60.0% of the ESTs were represented by 40 known genes. Thirty-five clones of unknown gene products potentially represented 34 novel genes. The most Bbundantly represented messages were the 28S ribosomal protein (6.5%) and beta actin (5.8%). The genes involved in ribosome formation (18%) accounted for the major gene expression. Development of EST panels representing the genes expressed in a particular tissue will be useful in determining the role of these genes in normal function and in response to developmental, hormonal, environmental and physiological changes.

The purpose of this study is to expression pattern of melanogenesis associate genes on cultured melanocyte layer cells in Korean Brindle Cattle(Dark, Brindle and Yellow) were analyzed to evaluate the effects of sex hormones on the control of melanogenesis pathways. Korean Brindle Cattle(Dark, Brindle and Yellow) melanocyte in the skin cells was collected. after the addition of estrogen and testosterone, the culture was analyzed for expression of cell activity and melanin genes for 72 hours. For the analysis of estrogen in different coat color other than the melanogenesis-related genes it is increasingly yellow showed low expression. in particular, the cells of the brindle coat color is low active and expression of genes. However, the testosterone was low, the expression of cell activity inhibiting MMP-2. the expression of melanin genes actually showed a tendency to increase gradually, which is testosterone compared with the estrogen to be considered that affect the skin cell layer brindle coat color. In this study, stimulation with estrogen triggered the inhibition of MC1R of the melanocyte in brindle coat color, but testosterone is induced MC1R in melanocyte. Therefore, considered the eumelanin or phaeomelanin activation are controlled caused by differential expression of sex hormones on melanocyte in Korean Brindle Cattle.

Rice blast caused by Magnaporthe oryzae is one of the most serious diseases that affect the quantity and quality of rice production. The use of resistant rice varieties would be the most effective way to control the rice blast. However R gene incorporation into the rice variety takes time and pathogen could overcome the R gene effects after for a while. For monitoring the rice blast resistance gene distribution in Korean varieties, the four major blast resistance genes against M. oryzae were screened in a number of Korean rice varieties using molecular markers. Of the 120 rice varieties tested, 40 were found to contain the Pi-5 gene, 25 for the Pi-9 gene, 79 for Pi-b and 40 for the Pi-ta gene. None of these rice varieties includes tested 4 R genes. 3 R genes combination, Pi-5/Pi-9/Pi-b, Pi-5, Pi-9.Pi-ta, or Pi-9/Pi-b/Pi-ta were found in 12 varieties, the rice blast disease severity were showed as resistant in the rice verities containing Pi-9/Pi-b/Pi-ta R genes combination, respectively. Also pathogenic diversity of M. oryzae isolates collected in the rice field from 2004 to 2015 in rice field in Korea were analyzed using rice blast monogenic lines, each harboring a single blast resistance gene. Compatibility of blast isolates against rice blast monogenic lines carrying the resistance genes Pi5, Pi9, Pib, and Piz showed dynamic changes by year. It indicates that pathogen has high evolutionary potential adapted host resistances to increase fitness and would lead to rice blast resistance bred into the cultivar becoming ineffective eventually.
MspTL is the major surface protein of Treponema lecithinolyticum associated with periodontitis and endodontic infections. Our recent investigation revealed that MspTL induces proinflammatory cytokines and intercellular adhesion molecule 1 in THP-1 cells and periodontal ligament cells. In this study we conducted oligonucleotide microarray analysis to investigate the global transcriptional regulation in THP-1 cells stimulated with purified recombinant MspTL. MspTL upregulated the expression of 90 genes in THP-1 cells at least four fold, and the functions of these genes were categorized into adhesion, apoptosis/antiapoptosis, cell cycle/growth/differentiation, chemotaxis, cytoskeleton organization, immune response, molecular metabolism, proteolysis, signaling, and transcription. The majority of the modified genes are known to be NF-${\kappa}B$-responsive and interferon-stimulated genes (ISGs). The expression of 12 selected genes was confirmed by real-time RT-PCR. Because prostaglandin $E_2(PGE_2)$ is an important inflammatory mediator and Cox-2 was found to be induced by MspTL in the microarray analysis, we determined the level of $PGE_2$ in the culture supernatants of MspTL-treated cells and found that MspTL significantly increased $PGE_2$. Our results provide insight into the gene regulation of host cells in response to MspTL, and may contribute to the understanding of the molecular mechanism in periodontitis.
Blast disease caused by the fungal pathogen, Magnaporthe oryzae, is one of the most damaging diseases in rice. The use of resistant varieties is an effective measure to control the disease, however, many resistant varieties were broken down to their resistance effects by the differentiating of new virulent isolates. This study was done to analyze the haplotypes of 31 microsatellite markers linked to five major R genes and two QTLs and to identify the alleles for the putatively novel genes related to durable resistance to blast in 56 Korean japonica and four indica varieties. The 31 microsatellite markers produced 2 to 13 alleles(mean = 5.4) and had PICi values ranging from 0.065 to 0.860(mean=0.563) among the 60 rice accessions. Cluster analysis based on allele diversities of 31 microsatellite markers grouped into 60 haplotypes and ten major clusters in 0.810 genetic similarity. A subcluster IV-1 grouped of early flowering varieties harboring Piz and/or Pi9(t) on chromosome 6 and Pita/Pita-2 gene on chromosome 12. The other subcluster V-1 consisted of four stable resistance varieties Donghae, Seomjin, Palgong and Milyang20. The analysis of putative QTLs associated with seven blast resistance genes using ANOVA and linear regression showed high significance to blast resistance across regions and isolates in the markers of two genes Piz and/or Pi9(t) and Pita/Pita-2. These results illustrate the utility of microsatellite markers to identify rice varieties is likely carrying the same R genes and QTLs and rice lines with potentially novel resistant gene.

Many of plant defense responses are consequence of transcriptional activation of related genes. We have developed a modified differential screening procedure to isolate tobacco genes that are involved in the defense responses against TMV infection. A cDNA library was constructed from Nicotiana glutinosa leaves infected by TMV under temperature shift conditions. Each of plasmid DNA in the library was hybridized on a set of slot blots to a pool of cDNA probes prepared from either TMV-infected or mock-treated tobacco leaves. Among 900 plasmid DNAs, 81 clones exhibiting significantly enhanced or reduced level of hybridization to either probe were selected for nucleotide sequencing. The clones were listed into 61 genes considering redundancy between the sequences. The genes were identified to be defense-related genes including PR-genes and genes involved in primary or secondary metabolisms. This results supports the implication that plant defense process entails a major shift in total cellular metabolisms rather than activation of a limited number of defense-related genes. Expression patterns of a number of defense-related genes. Expression patterns of a number of selected genes were examined in northern blot analyses. It is notable that the clone 630 of unknown function exhibits expression pattern similar to those of previously known PR-genes. Experiments to elucidate the roles in defense mechanism of a couple of genes newly identified in this study are in progress.
