Detection of Eight Different Events of Genetically Modified Maize by Multiplex PCR Method

  • Multiplex PCR was performed to simultaneously detect eight different events of genetically modified (GM) maize. Specific primers were constructed from GA21, T25, TC1507, Mon810, Mon863, Event176, Bt11, and NK603 events of GM maize. Using this PCR method, specific GM maize was monitored in commercialized foods and feed.

  • It has been hypothesized that efficient exclusion of methylated retrotransposons and repeated DNA region is one of the rapid and cost-effective approaches for comprehensive gene discovery in large genome size of maize. Three kinds of methylation-sensitive restriction enzymes, HapII, MspI and McrBC, were used to identify the restriction frequency of cytosine methylation sites in maize genome. Roughly 60% of total maize genomic DNA was restricted less than 500bp by McrBC, and the most of restricted small size fraction was composed retrotransposon. In order to validate the efficient construction of gene-rich shotgun library, we compare two gene-rich methyl-filtration shotgun libraries using in vivo and in vitro methyl-filtration system. The size selected DNA fraction by Sau3A-McrBC enzyme treated was very stable and has not appeared modification in E. coli, but most insert DNA size of partially digested with Sau3A were decrease less than 500bp by bacterial methylation-modification system. In compare of retroelements portion, A 44.6% of the sequences were retroelement in unmethyl-filtered library, and the most of them was Copia type, such as Prem, Opie and Ji. The portion of retroelement was drastically decreased to 25% and 20% by in vivo and in vitro filtration system, respectively.

Studies on the Fibrinolytic Effect of Germinated Grain Seeds

  • In this study, seven grain seeds(sorghum maize, buckwheat, soy bean, mung bean, red bean, and barngrass) and germinated seven grain seeds were examined the fibrinolytic activity through fibrin plate assay and SDS-PAGE. The results obtained were as follows : 1. In the fibrin plate assay, the extracts of maize, barngrass, sorghum and buckwheat showed fibrinolytic activity. Especially, Maize of them showed fibrinolytic activity that was almost similar to plasmin, fibrinolytic enzyme used as a positive control. 2. In the SDS-PAGE of seven grain seeds, fibrinolytic activity was remarkably shown in mung bean and red bean. 3. In the fibrin plate assay of germinated grain seeds, buckwheat(5 mm), buckwheat(10 mm) and soy bean(10 mm) showed a level of fibrinolytic activity that was about 0.3 fold than 1.0 unit of plasmin, Also maize(10 mm) of them showed a level of fibrinolytic activity that was about 0.5 fold than 1.0 unit of plasmin. As a result, maize of grain seeds was found that it has a strong fibrinolytic activity.

Roles of Azospirillum spp. Inoculation in Two Consecutive Growth of Maize Plants

  • Two consecutive green house experiments were carried out to examine the effect of Azospirillum spp. inoculation on growth, nitrogen and phosphorus accumulation in maize plants grown in pots. There were eight treatments including an uninoculated control and Azospirillum strains OAD-3, OAD-9, AZ-22, AZ-8, AZ-9, Azospirillum brasilense BR-11001 and Azospirillum lipoferum BR-11080. The inoculated plants showed higher values in each of the following measurements; plant height, total dry mass and nitrogen and phosphorus accumulation in shoot when compared to the uninoculated control plants in two consecutive experiments conducted in the same soil. Among Azospirillum strains, Azospirillum sp. OAD-3 inoculated plants showed higher nitrogen accumulation by 44.5% and 45.1%, total dry mass by 48.6% and 66.9% in two consecutive experiments respectively. The nitrogen concentration in the maize plants was not changed significantly in the first experiment, however it increased significantly in the second experiment due to Azospirillum inoculation. In addition, Azospirillum sp. OAD-9 and A. brasilense BR-11001 also proved to be effective with respect to total dry mass, total nitrogen accumulation and total phosphorus accumulation. The nitrogen concentrations in maize plants were increased in the second experiment due to Azospirillum inoculation.

Morphological Properties of Lintnerized Maize Starches with Different Amylose Content (아밀로오스 함량이 다른 산처리 옥수수전분의 형태학적 특성)

  • Morphological properties on lintnerized maize starches with different amylose content were investigated. With increasing the lintnerization periods and decreasing the amylose content, hydrolysis rate was increased. As amylose content of starch was increased, the degree of damage with acid treatment was decreased by SEM. With increasing hydrolysis, iodine affinity, apparent amylose content and ${\lambda}_{max}$ of lintnerized starches were decreased. Water binding capacities of lintnerized starches were higher than those of native starches.

THE USE OF CASSAVA IN BROILER FEEDING IN THE SOUTH PACIFIC

  • An experiment was conducted to investigate the nutritional and economic effects of complete replacement of maize with sun-dried cassava (SDC) in the diet of broiler chickens raised from one day of age to seven weeks. The complete replacement of maize by SDC resulted in a 10 percent reduction (p<0.05) in final bodyweights (1.91 vs 1.72 kg); and a 5 percent reduction (p>0.05) in average feed intake (4.01 vs 3.81 kg). Feed efficiency was not affected (p > 0.05). Cost per kilogramme of feed was reduced by nearly 30 percent and cost per kilogramme of bodyweight gain lowered by about 26 percent by using SDC diet. Relative profit return after accounting for cost of feed and cost of day old chick was higher by 11 percent on the SDC diet. It was concluded that despite the reduction in final bodyweight, the attractive economic return obtained from using SDC, a locally produced ingredient, may be justified in place of maize which is imported.

Genetic Diversity and Population Structure of maize, Zea mays, in Both Landraces and Cultivar Lines

  • Enzyme electrophoresis was used to estimate genetic diversity and population structure of maize, Zea mays L. (Graminales) in Korea. In nine populations, fourteen of the 24 loci (58.3 %) showed detectable polymorphism. Genetic diversity (0.205) was higher than average values for species with similar life history traits. Although our data are relatively small and the landraces not direct ancestors of cultivar, apparently the domestication process has eroded the levels of genetic variation of maize. The recent cultivars were found to have fewer alleles per locus (1.42 vs. 1.56), fewer alleles per polymorphic locus (2.27 vs. 2.33), lower percent polymorphic locus (33.3% vs. 41.7%), and lower diversity (0.159 vs. 0.185) than landraces. These genetic diversity parameters indicated that the cultivar populations were genetically depauperate relative to landlaces. The GST value of nine populations was 0.239. Nearly 76% of the total genetic diversity in Zea mays was apportioned within populations. The indirect estimate of gene new based on mean GST was moderate (Nm=0.80).
