Application and Validation of an Optimal Analytical Method using QuEChERS for the determination of Tolpyralate in Agricultural Products (QuEChERS법을 활용한 농산물 중 제초제 Tolpyralate의 최적 분석법 선발 및 검증)

  • BACKGROUND: Pesticides are broadly used to control weeds and pests, and the residues remaining in crops are managed in accordance with the MRLs (maximum residue limits). Therefore, an analytical method is required to quantify the residues, and we conducted a series of analyses to select and validate the quick and simple analytical method for tolpyralate in five agricultural products using QuEChERS (quick, easy, cheap, effective, rugged and safe) method and LC-MS/MS (liquid chromatography-tandem mass spectrometry). METHODS AND RESULTS: The agricultural samples were extracted with acetonitrile followed by addition of anhydrous magnesium sulfate, sodium chloride, disodium hydrogencitrate sesquihydrate and trisodium citrate dihydrate. After shaking and centrifugation, purification was performed with d-SPE (dispersive-solid phase extraction) sorbents. To validate the optimized method, its selectivity, linearity, LOD (limit of detection), LOQ (limit of quantitation), accuracy, repeatability, and reproducibility from the inter-laboratory analyses were considered. LOQ of the analytical method was 0.01 mg/kg at five agricultural products and the linearity of matrix-matched calibration were good at seven concentration levels, from 0.0025 to 0.25 mg/L (R2≥0.9980). Mean recoveries at three spiking levels (n=5) were in the range of 85.2~112.4% with associated relative standard deviation values less than 6.2%, and the coefficient of variation between the two laboratories was also below 13%. All optimized results were validated according to the criteria ranges requested in the Codex Alimentarius Commission (CAC) and Ministry of Food and Drug Safety (MFDS) guidelines. CONCLUSION: In conclusion, we suggest that the selected and validated method could serve as a basic data for detecting tolpyralate residue in imported and domestic agricultural products.

Effect of Combined Application of Lime and Organic Matter , and of Calcium Silicate on the Growth and Cadmium Content of Chinese Cabbage (석회(石灰), 유기물(有機物)의 병용(竝用) 및 규산(硅酸)칼슘의 시용(施用)이 배추의 생육(生育) 및 카드뮴함량(含量)에 준 영향(影響))

  • A small pot experiment, filled with one hundred fifty gram of Cd amended soil, was conducted in order to learn the effect of combined application of lime and organic matter and of calcium silicate on Chinese cabbage(Brassica pekinensis, Var. Seoul, Heungnong, Miho 70 days). Results obtained are as follows: 1. The application of lime without organic matter depressed the growth of cabbage at the first and second harvests, but, at the later part of third harvest the growth was facilitated so as to harvest very good yield. 2. The combined application of lime and organic matter gave not only a good growth of cabbage from the first crop, but also lowered the cadmium content of dry cabbage bellow that of cabbage harvested from the plot applied with slaked lime alone. Those effect of combined application of both materials were lasted untill the third harvest. 3. Although gave little effect on soil pH, calcium silicate raised a normal good cabbage, and depressed the cadmium content of dry cabbage, by 0.21 me per 100g, which is higher than that of slaked lime plot cabbage, but within the range of no harm to cabbage growth. 4. The control and organic matter plots resulted in a remarkable soil pH drop and high cadmium content in dry cabbage, which lead the crop to a death from the second crop. 5. A negative correlation was observed between the contents of cadmium and calcium in dry cabbage crop, but positive correlations between those of cadmium and magnesium or cadmium and potassium. These relations were grown up as the harvesting were proceeding.

Production of Sedum Extract Adding Jelly and Assessment of Its Physicochemical Properties (돌나물 즙을 첨가한 젤라틴 젤리의 제조 및 품질특성)

  • Sedum sarmentosum, also blown as stonecrop (dolnamul), is a widely consumed herb, and is used as an ingredient in salads in Korea. Unfortunately, sedum is perishable and vulnerable to tissue damage during preservation. Therefore, this feasibility study was performed in order to increase the availability of sedum and increase its value. Various concentrations of sedum extract (0.5-3%) were added to gelatin jelly, and their physicochemical properties were determined. The ascorbic acid content of the sedum jelly increased in proportion to the sedum extract concentration. Calcium content of the sedum jelly was 4 to 28 times higher than that of the control. Contrary to the control, iron was detected in the sedum jelly (0.023-1.031 mg/100 g dry weight). Furthermore, magnesium and potassium levels were higher in the sedum extract groups. There were significant differences (p < 0.05) in greenness (a value) and yellowness (b value) between the control and the sedum extract groups. However, significant differences between the 2% and 3% sedum extract groups were not detected. As sedum extract concentration increased, the pH level of soft jellies (solid state) decreased. Therefore, hardness and gumminess were decreased significantly. These results are in agreement with the sensory evaluation. According to sensory tests, the values for palatability, appearance, and color in the 2% sedum extract group were higher than those of the 0.5-1% and 3% sedum extract groups.

  • 부식은 재료와 사용 환경과의 상호작용에 의한 결과로서 일반적으로 두께의 감소와 균열의 발생 및 파손 등의 문제로 나타난다. 특히 사용환경 중에서 해수 분위기는 금속의 부식에 가장 유리한 조건이다. 따라서 해양환경 중 항만이나 조선 및 해양 산업 등에 많이 이용되는 강 구조물은 이에 대응하기 위하여 도장방식이나 음극방식을 사용하고 있다. 여기서 음극방식은 피방식체를 일정전위로 음극 분극하는 원리로써 외부전원을 인가하거나 비전위의 금속을 전기적으로 연결하여 방식하는 방법이다[1]. 한편, 해수 중에서 이와 같은 원리로 음극방식 할 경우에는 피방식체인 강재표면에 부분적으로 칼슘 또는 마그네슘 화합물 등의 생성물이 부착하는 현상을 볼 수 있게 된다. 이와 같이 수산화마그네슘($Mg(OH)_2$)및 탄산칼슘($CaCO_3$)을 주성분으로 하여 석출되는 석회질 피막(calcareous deposits)은 피방식체에 유입되는 음극방식 전류밀도를 감소시켜 주거나 물리적 장벽의 역할을 함으로써 외부의 산소와 물 등 부식환경으로부터 소지금속을 보호한다[2]. 그러나 석회질 피막은 소지금속과의 결합력, 막의 균일한 분포, 내식성 및 제작시간의 단축 등 해결해야 할 과제가 있다. 또한 여러 가지 환경 조건 등의 영향을 받아 그 피막의 형성 정도도 가늠하기 어렵기 때문에 음극방식 설계 시 그 정도에 따른 영향을 고려-반영하기가 곤란하다. 따라서 본 연구에서는 석출속도, 밀착성 및 내식특성을 향상시키기 위해 전착프로세스를 통해 해수 중 기체를 용해시켜 석회질 피막을 제작하고 막의 결정구조 제어 및 특성을 분석-평가하였다. 본 연구에 사용된 강 기판(Steel Substrate)은 일반구조용강(KS D 3503, SS400)을 사용하였으며, 외부전원은 정류기(Rectifier, xantrex, XDL 35-5T)를 사용하여 3 및 $5A/m^2$의 조건으로 인가하였다. 양극의 경우에는 해수에 녹아있는 이온 이외에 다른 성분들이 환원되는 것을 방지하기 위해 불용성 양극인 탄소봉(Carbon Rod)을 사용하였다. 이때 석출속도, 밀착성 및 내식특성 향상을 위해 해수에 주입한 기체의 양은 0.5 NL/min였으며, 기판 근처에 고정하여 음극 부근에서의 반응을 유도하였다. 각 조건별로 제작된 막의 표면 모폴로지, 조성원소 및 결정구조 분석을 실시하였으며, 석회질 피막의 밀착성과 내식특성을 평가하기 위해 규격에 따른 테이핑 테스트(Taping Test, ISO 2409)와 3 % NaCl 용액에서 전기화학적 양극 분극 시험을 진행하여 제작된 막의 내구성과 내식성을 분석-평가하였다. 시간에 따른 전착막의 외관관찰 결과 전류밀도의 증가와 함께 상대적으로 많은 피막이 형성되었고, 용해시킨 기체에 의해 더 치밀하고 두터운 피막이 형성됨을 확인할 수 있었다. 성분 및 결정구조 분석 결과 $Mg(OH)_2$ 성분의 Brucite 및 $CaCO_3$ 성분의 Calcite 및 Aragonite 구조를 확인하였으며, 용해시킨 기체의 영향으로 $CaCO_3$ 성분의 Aragonite 구조가 상대적으로 많이 검출되었다. 밀착성 및 내식성 평가를 실시한 결과 해수 중 용해시킨 기체에 의해 제작한 시편의 경우 견고하고 화학적 친화력이 높은 Aragonite 결정이 표면을 치밀하게 덮어 전해질로부터 산소와 물의 침입을 차단하는 역할을 하여 기체를 용해시키지 않은 3 및 $5A/m^2$ 보다 비교적 우수한 밀착성 및 내식 특성을 보이는 것으로 사료된다.

Instant Tea Preparation from Cassia tora Seeds (결명자로부터 인스턴트차 제조)

  • In order to develop a type of instant tea cassia tora seed, changes of constiuent amounts before and after roasting $(3{\pm}0.5min\;at\;170{\pm}5^{\circ}C)$ effective extracting solvent and it's optimum concentration, dehydration method and sensory scores were investigated. Moisture, lipid, reducing sugar, emodin and rhein were decreased by roasting, whereas protein, and total sugar of cassia tora increased. Color intensity and extracting efficacy were also increased. Of three kinds of solvent (water, methanol and ethanol), ethanol was highest in redness and yellowness of exeracts, and filtrate yield. Effective extracting concentration of ethanol for cassia tora was 50%(V/V). Freeze dry product(FDP) and spary dry product(SDP) showed coffee and aloe-like smell, coffee like brown color, rapid soluble in cold and hot water, and fast caking in air during storage. Free sugar contents were high in order of raffinose>fructose >glucose>maltose>sucrose Metallic contents were high in order of sodium>calcium>potassium>magnesium>iron. In sensory score for color, taste and aroma, FDP and SDP were both above 8.0 point, however, of them FDP more or less higher than SDP In addion, score in developing value as an instant tea was above 8.0 point (valuable).

Echinostoma aegyptica (Trematoda: Echinostomatidae) Infection in Five Riparian People in Savannakhet Province, Lao PDR

  • Human infection with Echinostoma aegyptica Khalil and Abaza, 1924 (Trematoda: Echinostomatidae) is extremely rare. In this study, we confirmed E. aegyptica infection in 5 riparian residents living along the Mekong River in Savannakhet Province, Lao PDR. The patients revealed eggs of Opisthorchis viverrini/minute intestinal flukes, echinostomes, and other parasites in fecal examinations using the Kato-Katz technique. Following treatment with praziquantel 30-40 mg/kg and pyrantel pamoate 10-15 mg/kg in a single dose and purging with magnesium salts, adult specimens of various helminth species were collected. Among the trematodes, echinostome flukes of 4.5-7.6 mm in length (n = 134; av. 22.3 specimens per case) were of taxonomic interest and subjected in this study. The flukes were morphologically characterized by having total 43-45 collar spines arranged in 2 alternating rows (corner spines usually 5 on each side) and compatible with previous descriptions of E. aegyptica. The patients were mixed-infected with other helminths, so specific clinical manifestations due to this echinostome fluke were difficult to determine. The present paper describes for the first time human E. aegyptica infections in Lao PDR. This is the second report of human infection (2nd-6th cases) with E. aegyptica in the world following the first one from China.

Determination of Mineral Nutrient Concentrations in Fish Growing Water and Lettuce Leaf for Hydroball Aquaphonics (하이드로볼 배지경 아쿠아포닉스에서 사육수 및 상추 잎의 무기이온 농도 구명)

  • This experiment was aimed to identify concentrations of mineral nutrients in leaf lettuce (Lactuca sativa) grown on hydroball aquaponics and in the water for growing fish by conducting two experiments. The experiment I (Expt. I) was conducted with 12 fishes (F12) with and without filter, hydroball and plants (H12 (12 fishes, hydroball), FHP12 (12 fishes, filter, hydroball, 6 plants) and HP12 (12 fishes, hydroball, 6 plants)), and the experiment II (Expt. II) was with and without plants (FH15 (15 fishes, filter, hydroball), FHP15 (15 fishes, filter, hydroball, 6 plants)). The pH level in the water of all the treatments was decreased during the growing period, and the pH and EC of the water were lower in all the treatment with plants than those without plants in both Expt. I and Expt. II. When compared with adequate nutrient concentrations for hydroponics, nitrate nitrogen (NO3-N) and phosphorus (P) concentrations in the fish growing water were higher under the FHP15 treatment in Expt. II; however, potassium (K), calcium (Ca), and magnesium (Mg) were only 16, 49, and 82% of hydroponics, representatively, and iron (Fe) was not detected. The fresh weight of lettuce harvested from the FHP15 treatment was 38 g, only a 30% of marketable lettuce yield. The T-N and P contents of the leaf tissue grown under the FHP15 treatment were close to the optimal level; however, the K, Ca, and iron (Fe) contents were less than the optimal with no deficiency symptom.

Changes on the Components of Yam Snack by Processing Methods (제조방법에 따른 마 스낵의 성분 변화)

  • The study was intended to develop a yam snack to increase the consumption and the added value of the yam which has many kinds of medicinal effects and functions. Moisture content of a freeze-drying yam snack was higher than those of hot wind drying and vacuum drying yam snack. However, carbohydrate content was opposed to it. The content of soluble proteins in freeze drying yam snack was 6.43 mg/100g, and lower than that those in hot wind drying and vacuum drying yam snack. The contents of total sugar and reducing sugar were not noticeably different by processing methods. The major organic acids of yam snack were malic, acetic, acids, and acetic acid. Citric acid were rich contented in hot wind and vacuum dried yam snack, but the content of malic acid in freeze dried yam snack was higher than those in hot wind and vacuum dried yam snack. The contents of the amino acids and total amino acids showed the highest contents in vacuum dried snack and the next came in hot wind dried snack and freeze dried snack order. The content of free amino acids were not different by processing methods. Total content of vitamin C were not different by processing methods, but a ascorbic acid was higher in freeze dried yam snack than those in other. The dehydroascorbic acid contents of the hot-air and vacuum dried snack was higher than those of the freeze dried snack, Potassium, sodium and magnesium were the main minerals of the yam snacks.

Anaerobic Treatment of Leachate Solubilized from Thermal Hydrolysis of Sludge Cake (하수슬러지 케이크 열수분해 탈리액의 혐기성 분해 특성)

  • This study was performed to evaluate the feasibility of anaerobic pretreatment for the leachate solubilized from thermal hydrolysis of sewage sludge cake. Overall process for the treatment of sludge cake consists of thermal hydrolysis, crystallization of magnesium, ammonium, and phosphate (MAP) for the leachate and anaerobic digestion of supernatant from MAP crystallization. The experimental evidence showed that the optimum ratio of Mg : P for the struvite crystallization of leachate solubilized from thermal hydrolysis of sludge cake was 1.5 to 1.0 as weight basis at the pH of 9.5. With this operational condition, the removal efficiencies of ammonia nitrogen and phosphorous achieved 50% and 97%, respectively. The mesophilic batch test showed that the ultimate biodegradability of the supernatant from MAP crystallization reached 63% at S/I ratio of 0.5. The readily biodegradable fraction of 90% ($S_1$) of the MAP supernatant BVS (Biodegradable Volatile Solids, $S_0$) degraded with $k_1$ of $0.207day^{-1}$ for the initial 17 days where as the rest slowly biodegradable fraction ($S_2$) of 10% of BVS degraded with $k_2$ of $0.02day^{-1}$ for the rest of the operational period. Semi-Continuously Fed and Mixed Reactor (SCFMR) was chosen as one of the best candidates to treat the MAP supernatant because of its total solids content over 6%. Maximum average biogas production rates reached 0.45 v/v-d and TVS removal efficiency of 37~41% was achieved at an hydraulic retention time (HRT) of 20 days and its corresponding organic loading rate (OLR) of 1.43 g VS/L-d.

  • 철강은 기본적으로 강도가 우수하고 그 매장량이 풍부할 뿐만 아니라 대량생산이 가능하다 또한 다른 금속과 합금을 구성하여 또 다른 특성을 부여할 수 있기 때문에 현재 전 세계 금속 생산량의 95%를 차지할 정도로 많이 사용되며, 각종 산업과 기술이 발달함에 따라 그 중요도는 점점 더 커져가고 있다. 하지만 철강은 사용 환경 중 부식에 의해 그 수명과 성능이 급격히 저하되기 때문에 내식성을 향상시키기 위하여 도장이나 도금 등의 표면처리를 포함한 다양한 방법이 적용되고 있다. 그 중 철강재의 도금 표면처리방법은 주로 아연을 이용한 용융도금이나 전기도금 등과 같은 습식 프로세스가 널리 사용되고 있다. 여기서 아연은 철보다 이온화 경향은 크나 대기 환경 중 산소와 물과 반응하여 Zn(OH)2와 같은 화합물을 형성함으로써 철강재 표면상 부식인자를 차단(Barrier)함은 물론 사용 중 철 모재가 노출되는 결함이 발생하는 경우에는 철을 대신하여 희생양극(Sacrificial Anode) 역할을 하기 때문에 철의 부식방식용 금속으로 가장 많이 사용되고 있다. 한편 최근에는 철강의 사용 환경이 다양해짐은 물론 가혹해지고 있어서 이에 따른 내식성 향상이 계속해서 요구되고 있는 추세이다. 따라서 본 연구에서는 철강재의 내식성을 향상시키기 위한 일환으로 현재 많이 사용되고 있는 용융아연도금 강판 상에 아연보다 활성이 높은 마그네슘(Mg)을 건식 프로세스 방법 중에 하나인 PVD(Physical Vapour Deposition)법에 의해 코팅하는 것을 시도하였다. 일반적으로 PVD법에 의해 진공증착하는 경우에는 그 도입가스로써 불활성가스인 아르곤(Ar)을 사용하는 경우가 대부분이나 여기서는 상대적으로 비활성이면서 그 크기가 작은 질소(N2)가스를 도입하여 그 증착 막의 몰포로지는 물론 결정구조도 제어하여 그 내식특성을 향상시키고자 하였다. 본 연구에서는 철강재의 내식성을 향상시키기 위한 방법으로 마그네슘(Mg)를 PVD(Physical Vapor Deposition)법 중 진공증착법(Vacuum Deposition)을 사용하여 용융아연도금 강판 상에 마그네슘 증착 막을 형성하였다. 즉, 여기서는 진공증착 중 질소(Nitrogen, N2)가스를 도입하여 진공챔버(Vacuum Chamber)내의 진공도를 $1{\times}10^{-1}$, $1{\times}10^{-2}$, $1{\times}10^{-3}$, $1{\times}10^{-4}$로 조절하며 제작하였다. 또한 제작된 시편에 대해서는 SEM(Scanning Electron Microscope) 및 XRD(X-Ray Diffraction)을 사용하여 형성된 아연도금상 마그네슘 막의 표면 몰포로지 및 결정구조의 변화를 분석함은 물론 침지시험, 염수분무시험, 분극시험을 통해 이 막들에 대한 내식특성을 분석 평가하였다. 상기 실험결과에 의하면, 진공 가스압이 증가됨에 따라 마그네슘 막의 두께는 감소하였으며, 그 몰포로지의 단면은 주상정(Columnar)에서 입상정(Granular) 구조로 변화하며 표면의 결정립은 점점 미세화 되는 경향을 나타냈다. 이때의 표면의 결정배향성(Crystal orientation)은 표면에너지가 상대적으로 큰 면이 우세하게 나타나는 경향이 있었다. 또한 본 실험에서 형성한 진공증착 막은 비교재인 용융아연도금강판보다 우수한 내식성을 나타냈고, 본 형성 막 중에는 마그네슘 막 두께가 작음에도 불구하고 질소 가스압이 가장 큰 조건일수록 내식성이 우수한 경향을 나타냈다. 이상의 결과는 철강재의 내식성 향상을 위한 응용표면처리설계에 기초적인 지침을 제공할 수 있을 것으로 기대된다.
