Anti-inflammaory effects of the Gamroeum in vivo and in vitro (감로음(甘露飮)의 항염증 효과에 대한 실험적 연구)

  • Objectives : The present study was conducted to evaluate the anti-inflammatory effects of the Gamroeum water extracts (GRE) in vivo and in vitro. Methods : The effects of GRE on anti-inflammation were measured by production of NO, $PGE_2$ (Prostaglandin $E_2$), iNOS (inducible Nitric Oxide Synthase), COX-2, $NF{\kappa}B$ (Nuclear Factor kappa B), TNF-$\alpha$ (Tumor Necrosis Factor-alpha) and IL-$1{\beta}$ (Interleukin-$1{\beta}$), IL-6 in Raw 264.7 macrophage cells stimulated with LPS. Results : 1. In machrophage cells, LPS displayed significant stimulatory effects on the production of NO and $PGE_2$. However, GRE showed significant inhibitory effects on NO and $PGE_2$ release. The level of NO and $PGE_2$ was decreased by GRE in a concentration dependent manner as compared with LPS only group. 2. Immunoblot analysis verified that LPS stimulation significantly increased the iNOS and COX-2 protein level, but GRE suppressed the induction of iNOS and COX-2 protein at a concentration dependent manner. 3. GRE reduced the elevated production of TNF-$\alpha$, IL-$1{\beta}$ and IL-6 by LPS. Moreover, the inhibitory effects of GRE was occurred in a dose-dependent manner. 4. GRE significantly reduced the expression of NF-${\kappa}B$ protein in nuclear fraction. 5. GRE effectively inhibited the increases of hind paw skin thicknesses and inflammatory cell infiltrations induced by carrageenan treatment. It, therefore, considered that GRE will be favorably inhibited the acute edematous inflammations. Conclusions : These results indicated that GRE could have anti-inflammatory capacity by inhibiting the production of NO, $PGE_2$ and cytokines in vitro and by reducing the formation of carrageenan-induced paw edema in vivo. Moreover, inhibitory effects of GRE on the macrophage activation were attributable to the reduction of some of inflammatory factors by inhibiting iNOS and COX-2 through the suppression of NF-${\kappa}B$.

Immuno-enhancing Effect of Protaetia brevitarsis seulensis (white grub) Extracts on RAW 264.7 Cell Line. (굼벵이 추출물의 RAW 264.7 세포에 대한 면역증강 효과)

  • Protaetia Brevitarsis Seulensis(white grub) has been traditionally used as medicinal stuff to treat blood stasis, occlusion of menstruation, tetanus and liver cancer in Asian countries (Korea, Japan, China, Taiwan, India and Myanmar). Especially, Donguibogam, which is traditional korean medicinal book, described the white grub as traditional medicine to treat hepatic diseases and vascular disorders. The white grub has been considered as highly nutritional food. The major constituents of white grub are rich in protein, healthy fats, iron, calcium. Recent studies announced that white grub has hepatoprotective effect and anti-microbacterial effect. However, the immuno-enhancing effect of white grub extracts in RAW 264.7 macrophage cells has not been studied yet. In this study, the various concentrations of white grub extract were examined to find immuno-enhancing effects on RAW 264.7 cells. Cytotoxicity was determined by MTT assay and immuno-enhancing effect of white grub extract was investigated by measuring nitric oxide (NO) production compared with only lipopolysaccharide (LPS) treatment. White grub extracts (0.001 - 10 mg/ml) did not show cytotoxicity. Additionally, white grub extracts (0.001 - 1mg/ml) had Immuno-enhancing effect on RAW 264.7 cells compared with only LPS treated group. These results might be provided proof to develop beneficial immuno-enhancing material for human health.

Immune System-Stimulating Activities of Mucilage Polysaccharides Isolated from Opuntia humifusa (천년초에서 분리한 점질다당의 면역자극 활성)

  • To examine the new practical utilization of mucilages in Opuntia humifusa, the Korean pear cactus, the polysaccharides were isolated from O. humifusa, and immuno-stimulating activities were assayed. The main polysaccharide, CNC-E, was prepared by a commercial enzyme treatment, water extraction, and ethanol precipitation. The molecular mass of CNC-E was estimated to be about 700 kDa, and it consisted mainly of arabinose, galactose and xylose in addition to two minor sugars such as rhamnose and fucose. On the other hand, CNC-E showed considerably high splenocyte proliferation activity in a dose-dependent manner. Peritoneal macrophages stimulated with CNC-E produced cytokines such as IL-6, IL-12, IL-10, and TNF-${\alpha}$. The intravenous administration of CNC-E significantly augmented the cytotoxicity of natural killer (NK) cells against Yac-1 tumor cells. Especially, NK cells obtained from the mice treated with $100{\mu}g$ of CNC-E showed threefold higher cytolytic activity than those of untreated mice. CNC-E also showed potent anti-complementary activity in a dose-dependent fashion. Identification of C3 activation products by the crossed immunoelectrophoresis using anti-human C3 and the anti-complementary activity of CNC-E in a $Ca^{2+}$-free condition suggested complement activations by CNC-E that occur via both alternative and classical pathways. These results indicate that Korean pear cactus contains selected polysaccharides that provide immuno-stimulating activities beneficial to human health.

Suppressive Effects of Chrysanthemum zawadskii var. latilobum Flower Extracts on Nitric Oxide Production and Inducible Nitric Oxide Synthase Expression (구절초 꽃 추출물의 Nitric Oxide 생성과 Inducible Nitric Oxide Synthase 발현 억제 효과)

  • In this study, we investigated the effect of C. zawadskii extract on nitric oxide (NO) production, prostaglandin E2 (PGE2) production, protein and mRNA expression of inducible nitric oxide synthase (iNOS) in LPS-induced RAW 264.7 macrophage cells. C. zawadskii extract (5~50 μg/mL) significantly inhibited LPS-induced NO production in a concentration-dependent manner ranging from 23.3% to 100%. Consistent with the inhibitory effect on NO production, C. zawadskii extract inhibited the protein expression and mRNA expression of iNOS. Although flower extracts of C. zawadskii was not effective on the expression of PGE2 and COX-2, flower extracts of C. zawadskii, however, showed a strong anti-inflammatory activity through inhibition of NO production and iNOS expression. The present results suggest that C. zawadskii extract has an inhibitory effect on NO production, and thus can be used as an anti-inflammatory agent.

Taxifolin Inhibited the Nitric Oxide Production and Expression of Pro-inflammatory Cytokine mRNA in Lipopolysaccharide-stimulated RAW264.7 Cells

  • In previous works, we found that solvent extract of Opuntia humifusa Raf., a member of the lactaceae family, displayed potent anti-oxidative and anti-inflammatory activities. Thus, all solvent fractions, except for the water layer, showed potent scavenging effects. According to activity-guided fractionation, one of active radical scavenging principles in the ethyl acetate fraction was found to be taxifolin. In this study, we investigated whether taxifolin showed anti-oxidative activity. In addition, taxifolin modulated nitric oxide (NO) release and the expression of pro-inflammatory cytokine mRNA such as interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6, granulocyte-macrophage colony-stimulating factor (GM-CSF), and TNF-${\alpha}$. Taxifolin showed potent anti-oxidant activity with the $IC_{50}\;of\;8.5{\pm}1.4\;and\;9.3{\pm}1.0{\mu}M$ using xanthine/xanthine oxidase (XO) assay and 2,2-Diphenyl-lpicrylhydrazyl radical (DPPH) assay, respectively. We next determined the role of taxifolin on the immunomodulating activity using murine macrophage cell line RAW264.7 cells. Taxifolin dose-dependently inhibited NO production in lipopolysaccharide (LPS)-activated RAW264.7. It also significantly blocked the expression of inducible NO synthase (iNOS) mRNA in the LPS-stimulated RAW264.7 cells. In addition, taxifolin potently suppressed the expression of IL-$1{\beta}$, IL-6 and GM-CSF mRNA in LPS-activated RAW264.7 cells, but not that of TNF-${\alpha}$ Moreover, taxifolin significantly inhibited the transcriptional activity of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) and activator protein -1 (AP-1). These results suggest that taxifolin may downregulate inflammatory iNOS, IL-$1{\beta}$, IL-6 and GM-CSF gene expressions through inhibition of NF-K and AP-1 activation in LPS-stimulated RAW264.7 cells.

Screening for Anti-inflammatory Activities in Extracts from Korean Herb Medicines (국산 생약 추출물의 항염증 활성 스크리닝)

  • Cosmetics are products used over long periods by the public, and their safety is very important. Contact dermatitis induced by cosmetics is the result of an inflammatory response of the skin to direct irritancy. The initial event that this inflammatory response is observed is the release of pro-inflammatory cytokines. In this study, the anti-inflammatory activities of extracts from Korean herb medicines were investigated using RAW264.7 macrophage. Among the fifty one extracts tested, the ethanol extracts from Biotae Orientalis Folium, Biotae Orientalis Folium (roasted), Cyperi Rhizoma, Nepetae Spica, Benincasae Semen, Dioscoreae Rhizoma, Dioscoreae Rhizoma (roasted), Mori Ramulus, Pini Ramulus and Alismatis Rhizoma reduced the cytotoxicity and inhibited the productions of Nitric oxide (NO) and cytokines such as interleukin (IL)-1${\beta}$, IL-6 and tumor necrosis factor (TNF)-${\alpha}$n lipopolysaccharide (LPS)-induced RAW264.7 macrophage. Additionally, they didn't induce the skin irritation when tested the human patch test. Overall, the result of this study suggests that the extracts of the ten Korean herb medicines are useful cosmetic agents for preventing the skin irritation.

Effects of Red-Koji Fermented Scutellariae Radix Extracts on Lipopolysaccharide-induced Rat Acute Lung Injury (홍국발효 황금이 Lipopolysaccharide 유발 급성 폐손상에 미치는 영향)

  • In the present study, the possibility of whether the pharmacological effects of Scutellariae Radix Aqueous Extracts(SR) were favorably changed by report that lipopolysaccharide(LPS)-induced rat acute lung injury was treated with Red-Koji(Monascus purpureus 12002) fermentation. Three different dosages of Red-Koji fermented SR extract(fSR), 125, 250 and 500 mg/kg were orally administered once a day for 28 days before LPS(Escherichia coli 0111:B4) treatments, and then 5 hours after LPS treatment(500 ${\mu}g$/head, intra trachea instillation), all rats were sacrificed. Changes in the body weights, lung weights, pulmonary transcapillary albumin transit, arterial gas parameters(pH, $PaO_2$ and $PaCO_2$) bronchoalveolar lavage fluid(BALF) protein, lactate dehydrogenase(LDH) and proinflammatory cytokine tumor necrosis factor-${\alpha}$(TNF-${\alpha}$), interleukin-$1{\beta}$(IL-$1{\beta}$) contents, total cell numbers, neutrophil and alveolar macrophage ratios, lung malondialdehyde(MDA), myeloperoxidase(MPO), proinflammatory cytokine TNF-${\alpha}$ and IL-$1{\beta}$ contents were observed with histopathology of the lung, changes on luminal surface of alveolus(LSA), thickness of alveolar septum, number of polymorphonuclear neutrophils(PMNs). As results of LPS-injection, dramatical increases in lung weights, pulmonary transcapillary albumin transit increases in $PaCO_2$, decreases in pH of arterial blood and $PaO_2$, increases of BALF protein, LDH, TNF-${\alpha}$ and IL-$1{\beta}$ contents, total cells, neutrophil and alveolar macrophage ratios, lung MDA, MPO, TNF-${\alpha}$ and IL-$1{\beta}$ contents increases were detected with decreases in LSA and increases of alveolar septum and PMNs numbers, respectively as compared with intact control. Especially fSR 125 mg/kg showed quite similar favorable effects on the LPS-induced acute lung injuries as compared with 60 mg/kg of ${\alpha}$-lipoic acid and 250 mg/kg of SR. The results suggest that over 125 mg/kg of fSR extracts showed favorable effects on the LPS-induced acute lung injury mediated by their antioxidant and anti-inflammatory effects. Moreover, increases of the pharmacological effects of SR on LPS-induced acute lung injury were observed by Red-Koji fermentation in this study, at least 2-fold higher.

  • Antitumor components were obtained from the cultured mycelia of Pleurotus pulmonarius by ethanol precipitation. The protein-bound polysaccharide was subjected to DEAE-­Sephadex column chromatography and Sephadex G-200 gel filtration. The antitumor fraction $C_1$ was isolated. The inhibition ratio of fraction $C_1$ was 81.8 % in the doses of 10 mg/kg/day for 10 days. The antitumor fraction $C_1$ consisted of a polysaccharide and a protein. The protein-moiety was composed of 14 amino acids. From the peritoneal cell populations in the mice given antitumor fraction $C_1$, the injection of the fraction caused the influx of peritoneal macrophages at two days when compared with those of soluble starch. This was named pulmonaran after its species name.

Comparison of Chemical Composition and Immune-enhancing Activity of the Four Lines of Korean Native Chickens (4계통 재래종 닭고기의 화학적 특성 및 면역활성)

  • This study was conducted to compare the general composition and immunomodulatory activity of breast and thigh meats from four lines of Korean native chickens: Yeonsan Ogye, Hyunin Black, Hwangbong, and Hoengseong Yakdak. White Leghorn was used as a control. Fifteen male chickens (three chickens in each line) were grown under the same conditions and slaughtered at 13 weeks old. The four lines of Korean native chickens, regardless of the part, had higher contents of crude fat (p<0.05) than White Leghorn. The cholesterol contents were significantly higher in Hyunin Black and significantly lower in Hoengseong Yakdak than those of other chickens (p<0.05). The immunomodulatory effect, assessed by macrophage cell proliferation and nitric oxide production, was only observed in the breast meat of the four lines of Korean native chickens. The phagocytic activity of macrophage cells was significantly augmented by the breast meat of Hyunin Black and Hoengseong Yakdak. Furthermore, the mRNA expressions of pro-inflammatory cytokines, such as IL-4, IL-10 and $IFN-{\gamma}$, was significantly suppressed by Korean native chickens compared with White Leghorn. These results suggested that the four lines of Korean native chickens exhibited greater immune-enhancing activity than White Leghorn.

The role of nitric oxide as an effector of macrophage-mediated cytotoxicity against Trichomonas vaginalis (질편모충에 대한 대식세포의 세포독성에 있어서 NO의 역할)

  • The purpose of this study is to determine whether nitric oxide is involved in the extracellular killing of Trichomoncs uasinalis by mouse (BALB/c) peritoneal macrophages and RAW264.7 cells activated with LPS or rIFN-γ and also to observe the effects of various chemicals which affect the production of reactive nitrogen intermediates (RNl) in the cytotoxicity against T. vnginnlis. The cytotoxicity was measured by counting the release of (3H)-thymidine from labelled protozoa and NOa was assayed by Griess reaction. Nemonomethyl-L-arginine (L-NMHA), Nenitro-L-arginine methyl ester (NAME) and arginase inhibited cytotoxicity to T. vaginnlis and nitrite production by activated mouse perioneal macrophagrs and RAW 264.7 cells. The addition of excess L-arginine competitively restored trichomonacidal activity of macrophages. Exogenous addition of FeSO4 inhibited cytotoxicity to T. vaginaLis and nitric products of macrophages. From above results, it is assumed that nitric oxide plays an important role in the host defense mechanism of macrophages against T ucfinalis.
