Ethanol의 면역독성에 대한 인삼엑기스의 영향 (Effect of Panax ginseng Extracts on the Immunotoxicity of Ethanol)

  • Experiments were performed on mice to investigate the effect of panax ginseng extracts on the immunotoxicity of ethanol. Immune response were evaluated by antibody production, Arthus reaction, delayed type hypersensitivity (DTH), Rosette froming cell (RFC) and macrophage activity in mice, sensitized and challenged with sheep red blood cells. The weight of liver, spleen and thymus were measured. Following results obtained in this experiment. The exposure of ethanol decreased humoral and cellular immune response, the body weight and macrophage activity. Ginseng extracts such as ethanol extract, petroleum ether extract and n-butanol fraction were significantly increased the body weight. The administration of ginseng ethanol extract and ginseng petroleum ether extract were restored or increased humoral and cellular immune response. Macrophage activity was decreased by ethanol, but restored by the ginseng extracts.

  • The purpose of this research was to investigate effect of water extract of DangKwi-Eum-Ja ka Sumsoo(DESE) on the cytotoxicity of human epidemloid cell, A431 cells. The effects of DESE on the proliferation of A431 cells, Balb/c 3T3 cells, mouse thymocytes and splenocnes were estimated by MTT colorimetric assay, and nitric oxide production from mouse peritoneal macrophage was estimated by Griess method. DESE inhibited the proliferation of A431 cells at $10{\mu}g/ml$, and did not affect the proliferation of Balb/c 3T3 cells. DESE decreased the cytotoxicity of mitomycin C or cisplatin on A431 cells, increased the cytotoxicity of mitomycin C or cisplatin on Balb/c 3T3 cells. DESE inhibited the proliferation of mouse thymocytes and splenocytes at $100{\mu}g/ml$. DESE did not affect the nitric oxide production from mouse peritoneal macrophage in vitro, but decreased the nitric oxide production from DESE-treated mouse peritoneal macrophage.

  • The interaction between fibronectin (FN) and its receptors controls cell attachment and migration, two crucial events during monocyte development and differentiation. To investigate the functional role of FN and its receptor, we have studied adhesion of monocyte to two different regions of FN (38- and 85-kDa domain), as well as the expression of the integrin during monocle differentiation. Anti-integrin Rl subunit antibody completely blocked the attachment of FN-coated latex beads to macrophage, but the effect of anti-integrin u4 antibody was much less significant. Rat monocyte expressed integrin $u4\beta1$ predominantly, while macrophage expressed $\alpha5\beta1$ as analyzed tv flow cvtometer and western blot. From these results, it can be suggested that these two integrins plan different roles during monocyte differentiation.

Immunomodulatory Response Induced by Ginseng

  • There has been continuing interest in the development of synthetic and natural compounds that modify the immune response particularly for the treatment of AIDS and cancer. During the past fifty years, numerous scientific studies have been published on ginseng. Modem human studies have investigated preventive effect of ginseng on several kinds of cancer, its long term immunological effect on HIV patients, its effect on cell mediated immune functions in healthy volunteers. Similarly non clinical studies on animal model system have studied the chemopreventive action of ginseng on cancer and immunological properties of ginseng. The precise mechanism of action of ginseng, however, not clearly understood. Considering its wide-ranging therapeutic effects, this study is being undertaken to elucidate the general mode of action of ginseng, especially to test our hypothesis that its biological action may be mediated by the immune system.

  • Alginates are polysaccharides with gel-forming properties composed of 1,4-linked $\beta$-D-mannuronic acid (M), $\alpha$-L-guluronic acid (G), and alternating (MG) blocks. The M-and the MG-blocks, but not the G-blocks, have been known to be the active components of the alginates in experimental models. In this study, we have examined the ability of high M-alginate to activate immune cells. Alginate induced the macrophage anti-viral activity and the lymphocyte blastogenesis, and enhanced cytotoxicity of natural killer cell. In addition, alginates stimulated the macrophages to induce the production of $H_{2}O_{2}$, whereas alginates had no effect on NO production and suppressed the production of TNF-$\alpha$. These findings suggest that high M-alginate may be modulating various elements of the host immune response.

  • Objectives: This experimental study was carried out to prove the effect of Saenghyuldan(SHD; shengxiedan) on bone marrow failure induced by cyclophosphamide(CY) and irradiation in mice. Methods: The following were performed; immunopathology, histopathlogical findings of bone marrow and in the smear of myelocyte. hematopoietic cytokine(IL-3, GM-CSF, TPO), hematopoietic stem cell colony assay, humoral immunity(LPS mitogen response), cell-mediated immunity (Con A mitogen response) and nonspecific immunity(macrophage adherence & phagocytosis) in vitro or vivo. Results: SHD showed a protective effect on bone marrow failure induced by cyclophosphamide(CY) and irradiation in mice. SHD increased lymphoproliferative responses to LPS and Con A, and activated macrophage adherence and phagocytosis to SRBC. Conclusions: We expect that SHD can be used to treat bone marrow failure and immune suppression induced by the chemotherapy or radiation.

마우스에 있어서 식이들기름이 면역반응에 미치는 영향 (Effect of Perilla Oil Diet on the Immune Response in Mice)

  • The effects of perilla oil diet on the immune response in mice have been studied. ICR male mice were divided into 4 groups and were fed on the experimental diet for 4 weeks. Mice were sensitized and challenged with sheep red blood cell (S-RBC). Immune response were evaluated by antibody production, Arthus reaction, delayed type hypersensitivity (DTH), Rosette forming cell (RFC) and macrophage activity. The weight of body, liver, thymus and spleen were measured. The body weight was increased but thymus weight was not altered by them. The perilla oil diet decreased the weight of liver and spleen in mice. It reduced antibody production, Arthus reaction, DTH and RFC, macrophage activity. These results showed that the high perilla oil diet decresed more humoral and celluar immune response than the low perilla oil diet. It decreased the phagocytic activity on the reticuloendothelial system in mice.

  • Nitric oxide (NO) and prostagladins(PGs) produced by inducible nitric oxide synthase(iNOS) and cyclooxygenase(COX-2) are known as inflammatory mediator. Modulation of these enzymes, induced by many stimuli(LPS, IFN-gamma, TNF-alpha, phorbol ester, etc), is a potent strategy as treatment of inflammatory diseases. Treatment of murine macrophage RAW 264.7 cell line with indole compound(IND-6) markedly reduced lipopolysacchride(LPS) stimulated NO production in a concentration-related manner. (omitted)

  • B cell-activating factor belonging to the TNF family (BAFF) is primarily expressed by macrophages and stimulates B cell proliferation, differentiation, survival, and Ig production. In this study, we explored the effect of lactoferrin (LF) on BAFF expression by murine macrophages. We determined the level of BAFF expression at the transcriptional and protein levels using RT-PCR and ELISA, respectively. LF markedly enhanced BAFF expression in mouse macrophages at both the transcriptional and protein levels. Overexpression of Smad3/4 further increased LF-induced BAFF transcription while DN-Smad3 abolished the LF-induced BAFF expression. These results demonstrate that LF can enhance BAFF expression through Smad3/4 pathway.
