A biotransformation approach using microbes as biocatalysts can be an efficient tool for the targeted modification of existing antibiotic chemical scaffolds to create previously uncharacterized therapeutic agents. By employing a recombinant Streptomyces venezuelae strain as a microbial catalyst, a reduced macrolide, 10,11-dihydrorosamicin, was created from rosamicin macrolide. Its chemical structure was spectroscopically elucidated, and the new rosamicin analog showed 2-4-fold higher antibacterial activity against two strains of methicillin-resistant Staphylococcus aureus compared with its parent rosamicin. This kind of biocatalytic approach is able to expand existing antibiotic entities and can also provide more diverse therapeutic resources.

The purpose of this study is to investigate the prevalence of resistance to macrolide, lincosamide, streptogramin and ketolide antibiotics in Korea. The antibiotic susceptibility test was performed to the macrolide erythromycin, clarithromycin, azithromycin, josamycin, the lincosamide clindamycin, the streptogramin synercid and the ketolide ABT -773 against 337 clinical Staphylococcus aureus(SAU). Coagulase-negative Staphylococci (CNS) and Enterococci isolates exhibited an average percentage of 64%, 56%, and 81 % of resistance to erythromycin, respectively.
About two thirds of the naturally occurring antibiotics have been discovered from actinomycetes. Therefore, the probability of discovering further new antibiotics from actinomycetes is declining as many known metabolites are isolated repeatedly. However, various efforts leave been made in order to enhance the probability of discovering novel compounds. In the present study, we have developed new screening strategies based on the antibiotic biosynthetic pathway, and the genetic information, utilizing polymerase chain reaction. We have selected macrolide type polyketides. In order to divide the ansamycin group antibotic of macrolide type polyketides, we have selected 3-amino-5-hydroxybenzoic acid (AHBA) moiety which contains a biosynthetically unique structural element in the group as a target molecules. Oligonucleotide primers were designed to amplify DNA fragments of macrolide type polyketide synthase and AHBA synthase genes from fourteen actinomycetes species. This method was successfully applied to all three of the known macrolide type polyketide produccing actinomycetes tested. In addition, it also identified the presence of potential macrolide type polyketide producing genes from seven actinomycetes that were known to produce none of macrolide type polyketides, and AHBA biosynthetic genes in one actinomycetes. This technique is potentially useful for the screening of new antibiotices and cloning of their biosynthetic genes.
Purpose: Macrolide resistance rate of Mycoplasma pneumoniae has rapidly increased in children. Studies on the clinical features between macrolide susceptible-M. pneumoniae (MSMP) and macrolide resistant-M. pneumoniae (MRMP) are lacking. The aim of this study was to identify the macrolide resistance rate of M. pneumoniae in Korean children with M. pneumoniae penupmonia in 2015 and compare manifestations between MSMP and MRMP. Methods: Among 122 children (0-18 years old) diagnosed with M. pneumoniae pneumonia, 95 children with the results of macrolide sensitivity test were included in this study. Clinical manifestations were acquired using retrospective medical records. Results: The macrolide resistant rate of M. pneumoniae was 87.2% (82 of 94 patients) in children with M. pneumoniae pneumonia. One patient showed a mixed type of wild type and A2063G mutation in 23S rRNA of M. pneumoniae. There were no significant differences in clinical, laboratory, and radiologic findings between the MSMP and MRMP groups at the first visit to our hospital. The time interval between initiation of macrolide and defervescence was significantly longer in the MRMP group ($4.9{\pm}3.3$ vs. $2.8{\pm}3.1days$, P=0.039). Conclusion: The macrolide resistant rate of M. pneumoniae is very high in children with M. pneumoniae pneumonia in Korea. The clinical manifestations of MRMP are similar to MSMP except for the defervescence period after administration of macrolide. Continuous monitoring of the occurrence and antimicrobial susceptibility of MRMP is required to control its spread and establish strategies for treating second-line antibiotic resistant M. pneumoniae infection.
Dihydrochalcomycin (GERI-155), produced by Streptomyces sp. KCTC-0041BP isolated from Korean soil, is a 16-membered macrolide antibiotic consisting of two deoxysugar moieties at C-5 and C-20 positions of a branched lactone ring. The cloning and sequencing of a gene cluster for dihydrochalcomycin biosynthesis revealed a 63-kb nucleotide region containing 25 open reading frames (ORFs). The products of all of these 25 ORFs playa role in dihydrochalcomycin biosynthesis and self-resistance against the compounds synthesized. At the core of this cluster lies a 39.6-kb polyketide synthase (PKS) region encoding eight modules in five giant multifunctional protein-coding genes (gerSI-SV). The genes responsible for the biosynthesis of deoxysugar moieties, D-chalcose and D-mycinose, and their modification and attachment were found on either side of this PKS region. The involvement of this gene cluster in dihydrochalcomycin biosynthesis was confirmed by disruption of the dehydratase (DH) domain in module 3 of the PKS gene and by metabolite analysis.
ErmSF는, 235 rRNA의 A2058에 methylation 시켜서 macrolide-lincosamide-streptogramin B ($MLS_B$)계 항생제의 부착을 저해함으로써 항생제의 활성을 억제하는 내성인자 단백질인 ERM 단백질의 하나로, 다른 ERM 단백질과는 달리 긴 N-terminal end region을 가지고 있고 이 부위의 25%를 arginine이 차지하고 있다. 특히 $^{58}RARR^{61}$ 부위에 arginine이 모여 있어서 여기에 존재하는 R의 역할을 알아보기 위해 1-57, 1-59 그리고 1-60과 1-61이 제거된 결손 변이 단백질을 대장균에 발현하고 그 활성을 성장곡선을 작성하여 알아보았다. 그 결과 R60과 R6l이 활성에 중요한 것으로 관찰되었다. 1-59의 아미노산이 결손 된 유전자를 사용하여 R60A, R61A와RR60, 61AA의 위치선정 치환 변이 단배질의 세포내 활성을 측정하여 본 결과 R60의 역할이 R6l보다 큰 것으로 관찰되었으며 이들의 활성에서의 역할은 상호보완적인 것으로 나타났다. 그리고 이 아미노산들이 2차 구조인 $\alpha$-helix의 일부일 것으로 추정되었다.
지속성 및 유발성 발한의 두 macrolide-lincosamide-streptogramin B 저항성 인자가 한 Staphylococcus aureus DHI 균주의 염색체 DNA 및 plasmid pDE1(7.4kb)로부터 각각 분리되었다. pDE1상의 유발성 Em 저항성 인자의 염기서열은 이미 보고 된 바 있는 pE194상의 ermC와 동일하였으며 지속성 Em 저항성 인자의 경우는 그 제한효소 인식부위의 mapping 결과로 보아 ermCdb전자에서 유발성 기구에 관여하는 leader peptide 부위가 결여된 인자인 것으로 밝혀졌다.
Streptomyces padanus PMS-702 strain produces a polyene macrolide antibiotic fungichromin and displays antagonistic activities against many phytopathogenic fungi. In the present study, experimental formulations were assessed to improve the production of fungichromin, the efficacy of PMS-702 on the suppression of sporangial germination, and the reduction of cucumber downy mildew caused by Pseudoperonospora cubensis. PMS-702 strain cultured in a soybean meal-glucose (SMG) medium led to low levels of fungichromin accumulation and sporangial germination suppression. Increasing medium compositions and adding plant oils (noticeably coconut oil) in SMG significantly increased fungichromin production from 68 to $1,999.6{\mu}g/ml$. Microscopic examination reveals that the resultant suspensions significantly reduced sporangial germination and caused cytoplasmic aggregation. Greenhouse trials reveal that the application of PMS-702 cultural suspensions reduced downy mildew severity considerably. The addition of Tween 80 into the synthetic medium while culturing PMS-702 further increased the suppressive efficacy of downy mildew severity, particularly when applied at 24 h before inoculation or co-applied with inoculum. Fungichromin at $50{\mu}g/ml$ induced phytotoxicity showing minor necrosis surrounded with light yellowish halos on cucumber leaves. The concentration that leads to 90% inhibition (IC90) of sporangial germination was estimated to be around $10{\mu}g/ml$. The results provide a strong possibility of using the S. padanus PMS-702 strain as a biocontrol agent to control other plant pathogens.
ERM 단백질은 23S rRNA의 A2058에 methylation시킴으로써 macrolide-lincosamide- streptogramin B $(MLS_B)$계 항생제의 부착을 저해하여 항생제의 활성을 억제하는 내성 인자 단백질로 monomethylase와 dimethylase로 나누어진다. Dimethylase와 비교되는 monomethylase의 특성을 밝히기 위해 dimethylase (ErmSF)와 monomethylase (TlrD)를 동시에 보유한 Streptomyces fradiae에서 tlrD를 클론하고 대장균에서 최초로 대략생산을 시도하여 $37^{\circ}C$에서 세포내 전체 단백질의 55%를 차지할 정도로 대량생산된 불용성 단백질을 얻어내었다. 그러나 ErmSF와는 달리 낮은 온도에서 대량생산된 단백질이 용해성 단백질로 전환되지 않고 불용성 단백질로 남아있었다. Thioredoxin과 샤페론인 GroESL은 모두 ErmSF의 경우와 마찬가지로 용해성 단백질로의 전환에 도움을 주지 않았다. 이러한 차이점은 천재까지 전혀 밝혀지지 않은 단백질내의 구조적 특성에 의한 monomethylase와 dimethylase의 차이점을 밝힐 수 있다는 가능성을 말해주는 것으로 추정된다. 그러나 ErmSF의 경우와 동일하게 SDS-PAGE에서 검색되지 않은 미량의 발현된 용해성 단백질이 TlrD를 함유한 세포에 항생제에 대한 내성을 나타내게 하였고 이렇게 발현된 내성은 monomethylase에 의한 내성에서 기대되는 내성과 일치하였다.
