A Study on Comparison of Changes in Ecological Characteristics for Bulgwangcheon(stream) Close-to Nature Section (불광천 자연형 하천 정비구간의 생태적 특성 변화 비교 연구)

  • The aim of this study was to provide basic data in managing the project that was carried out on Bulgwangcheon in a nature-friendly way to improve the conditions around the areas, which was brought to completion in 2002, based on changes in ecological characteristics. For this propose, this study examined documents related to the project, compared physical and enviromnental changes before and after the project was conducted and analyzed changes in the stream ecosystem. The result showed that in areas that effluent water was often observed, especially when it rained, the river wall was washed away and vegetation was found damaged. As for actual vegetation, this study compared planting coverage of each section of the research area and actual vegetation charts. The results indicated that Lespedeza spp., Aster koraiensis among mixed seeds that were planted in the reservoir path were almost swept away while Festuca arundinacea dominated the areas. Phragmites communis, Miscanthus sacchariflorns and Salix gracilistyla which had been planted in a small number were also almost washed out though a small number of them were left to form a colony. After examining the topography and structure of the plant community, this study found that areas where mixed seed were planted had changed into two types of vegetation: First type of area is dominated by P and R which are usually raised in apron with abundant floating particles. The second type of area is dominated by dry gramineous plant such as F and A. Areas around low flow channel where Phragmites communis, Miscanthus sacchariflorus and Salix gracilistyla planting construction method is applied was washed away with the width of low flow channel reduced. Though P, M and S formed a small community in some areas around the low flow area, they were in small number and in composition of simple plant species. Two ways were suggested in this study to manage the stream in an ecological way. First, adequate revetment construction methods should be applied by monitoring the flow of the stream as well as considering the flood control of urban streams. Second, target vegetation communities that are suitable for the environment of the stream should be chosen and be plantedconstantly with high density. At the same time, ornamental native plants shouldn't be planted as they have been and disturbing vegetation should be removed.

Development of Economic Culture System Using Wastewater for Microalgae in Winter Season (폐수를 이용한 겨울철 경제적 미세조류 배양 시스템의 개발)

  • The outdoor mass cultivation is not possible for microalgae in Korea all year round, due to cold winter season. It is not easy to maintain proper level of productivity of microalgae even in winter. To prevent a drastic decrease of temperature in a greenhouse, two layers were covered additionally, inside the original plastic layer of the greenhouse. The middle layer was made up of plastic and the inner layer, of non-woven fabric. Acrylic transparent bioreactors were constructed to get more sunlight, not only from the upper side but also from the lateral and bottom directions. In winter at freezing temperatures, six different culture conditions were compared in the triply covered, insulated greenhouse. Wastewater after anaerobic digestion was used for the cultivation of microalgae to minimize the production cost. Water temperature in the bioreactors remained above $10^{\circ}C$ on average, even without any external heating system, proving that the triple-layered greenhouse is effective in keeping heat. Algal biomass reached to 0.37g $L^{-1}$ with the highest temperature, in the experimental group of light-reflection board at the bottom, with nitrogen and phosphorus removal rate of 92% and 99%, respectively. When fatty acid composition was analyzed using gas-chromatography, linoleate (C18 : 3n3) occupied the highest proportion up to 61%, in the all experiment groups. Chemical oxygen demand (COD), however, did not decrease during the cultivation, but rather increased. Although the algal biomass productivity was not comparable to warm seasons, it was possible to maintain water temperature for algae cultivation even in the coldest season, at the minimum cost.

Effect of Silicate Mineral Filtered Water and Silicate Mineral Additive on Growth Performance and Pork Quality (규산염 광물질로 정수된 급이수 및 사료내 규산엽광물질 첨가제의 급여가 돼지의 성장능력 및 돈육의 품질에 미치는 영향)

  • This experiment was conducted to investigate the effect of filtered water with silicate minerals on growth performance and pork quality. A total of 64 pigs were allotted at 37.95 kg body weight to 4 treatments in a randomized complete block(RCB) design. Each treatment had 4 replicates and treatments were I) Con(basaI diet). 2) Si-I(consumed silicate mineral filtered water and silicate mineral additive during 4 weeks before market weight), 3) Si-Il( consumed silicate mineral filtered water and silicate mineral additive during 8 weeks before market weight) and 4) Si-I1I(consumed silicate mineral filtered water and silicate mineral additive during 12 weeks before market weight). Silicate complex was added at 0.8% to basal diet. All pigs were allowed to feed and water ad libitum for 12 weeks. During last 4 weeks, average daily gain(ADG) in Si-Il and Si-III group was higher than that in Si-I group(P < 0.05). However overall experimental period, there were no significant differences in growth performance among all treatments. Acceptability of cooked pork at 7th day after slaughter in Si-lII group was higher than that in Con group(P < 0.05). The value of pH of fresh pork at 6 hours after slaughter was lower in Si-I1I group than that in Con group(P < 0.05). TBA values of fresh pork at 14th day after slaughter were lower in Si-II and Si-III groups than Con group(P < 0.05). These results demonstrated that pork quality could be improved when pigs consumed both silicate mineral and silicate mineral filtered water. However, growth performance was not improved by the consumption of silicate mineral filtered water and silicate mineral.

Effects of a Soil-Born Paenibacillus spp. Strain KPB3 on Suppression of Bacterial Wilt Disease Caused by Ralstonia solanacearum (토양에서 분리한 Paenibacillus spp. KPB3의 Ralstonia solanacearum에 의한 세균성 풋마름병 억제 효과)

  • Two hundred bacterial strains were isolated from the soil around healthy tomato plants in a polyvinyl house, where most of the other plants showed bacterial wilt symptoms. The strains were screened in vitro for their antibacterial activity. Among them, a strain, KPB3 showed strong bactericidal activity against bacterial wilt pathogen, Ralstonia solanacearum. The strain KPB3 was identified using physiological and biochemical tests, and 16S rRNA analyses. Based on these tests, the strain was found to be closer to genus Paenibacillus. To control the bacterial wilt caused by R. solanacearum, greenhouse experiments were conducted to determine the effectiveness of the Paenibacillus strain KPB3. Drench application of this strain ($4{\times}10^8$ CFU $mL^{-1}$) into the pots containing tomato plants, post-inoculated with the pathogen, R. solanacearum could drastically reduce the disease severity, compared to the non-treated plants. To evaluate effectiveness of this strain under field conditions, experiments were carried out in polyvinyl houses infested with R. solanacearum, during spring and autumn of the year 2006. It was observed that, during spring, bacterial wilt was more prevalent compared to the autumn. During spring, 50.9% disease incidences occurred in non-treated controls, while, Paenibacillus strain KPB3 treated plants showed 24.6% disease incidences. Similarly, during autumn, around 17.2% plants were infected with bacterial wilt in non- treated polyvinyl houses, compared to the Paenibacillus strain KPB3 treated plants, which showed 7.0% disease incidences. These results demonstrated that, Paenibacillus strain KPB3 is a potential biological control agent against bacterial wilt caused by R. solanacearum, effective under greenhouse as well as field conditions. This is the first report showing biocontrol of R. solanacearum using a Paenibacillus spp. under field conditions.

EFFECT OF ACID-TREATMENT ON DENTIN BONDING (산 처리가 상아질 접착에 미치는 영향)

  • The purpose of this study was to evaluate the effect of various acid treatments on dentin bonding. Freshly extracted human teeth were uprightly embedded in self curing acrylic resin, and their occlusal surfaces were grinded to expose flat dentin surfaces. The specimens were divided into 4 groups. Specimens of one group were not treated so as to be a control and those of the other three groups were threated with 10% polyacrylic acid, 10% phosphoric acid, and 10-3 solution(10% citric acid/3% ferric chloride) respectively. Primer, bonding resin and composite resin were applied over the treated dentin surfaces sequentially. All specimens were stored in $37^{\circ}C$ distilled water for 24 hours, then the tensile bond strength was measured and the treated dentin surfaces and fracured dentin surfaces were examined under a scanning electron microscope. The results were as follows: Bond strengths of acid-treated groups were higher than those of the untreated group. In the acid-treated groups, bond strength was found to be the highest in the 10-3 solution group followed by the 10% phosphoric acid group and the 10% polyacrylic acid group(P<0.01). On SEM examination of dentin surfaces, the untreated dentin surface showed a remaining smear layer and closed dentinal tubules. Dentin surfaces treated with 10 % polyacrylic acid showed a clean dentin surface without the smear layer, but showed remaining smear plugs in dentinal tubules. A dentin surface treated with 10% phosphoric acid or 10-3 solution showed open dentinal tubules without the smear layer or smear plugs. On SEM observation of the fractured dentin-resin interface, the untreated group showed that failure occurred in the smear layer. The group treated with 10% polyacrylic acid showed no resin tag remained in the dentinal tubules, but resin tags in the dentinal tubules were observed in the group treated with the 10% phosphoric acid or the 10-3 solution. On the failure mode examination, the higher the bond strength of the group, the higher the frequency of cohesive failure. The coefficient between bond strength and cohesive failure rate was 0.71.

Chemical and Spectroscopic Characterization of Peat Moss and Its Different Humic Fractions (Humin, Humic Acid and Fulvic Acid) (피트모스에서 추출한 휴믹물질(휴믹산, 풀빅산, 휴민)의 화학적 및 분광학적 물질특성 규명)

  • Peat humin(p-Humin), humic acid(p-HA) and fulvic acid(p-FA) were isolated from Canadian Sphagnum peat moss by dissolution in 0.1M NaOH followed by acid precipitation. After purification cycles, they are characterized for their elemental compositions and, acid/base properties. Functionalities and carbon structures of the humic fractions were also characterized using FT-IR and solid state $^{13}C$-NMR spectroscopy. Those results are compared with one another and with soil humic substances from literatures. Main purpose of this study was to present a chemical and spectroscopic characterization data of humic substance from peat moss needed to evaluate its environmental applicability. The relative proportions of the p-Humin, p-HA and p-FA in the peat moss was $76\%,\;18\%,\;and\;3\%$, respectively, based on the total organic matter content ($957{\pm}32\;g/kg$). Elemental composition of p-Humin were found to be $C_{1.00}H_{1.52}O_{0.79}N_{0.01}$ and had higher H/C and (N+O)/C ratio compared to those of p-HA($C_{1.00}H_{1.09}O_{0.51}N_{0.02}$) and p-FA($C_{1.00}H_{1.08}O_{0.65}N_{0.01}$). Based on the analysis of pH titration data, there are two different types of acidic functional groups in the peat moss and its humic fractions and their proton exchange capacities(PEC, meq/g) were in the order p-FA(4.91) >p-HA(4.09) >p-Humin(2.38). IR spectroscopic results showed that the functionalities of the peat moss humic molecules are similar to those of soil humic substances, and carboxylic acid(-COOH) is main function group providing metal binding sites for Cd(II) sorption. Spectral features obtained from $^{13}C$-NMR indicated that peat moss humic molecules have rather lower degree of humification, and that important structural differences exist between p-Humin and soluble humic fractions(p-HA and p-FA).

Changes in Physico-Chemical Properties of Deer Meat Packed Different ways During Storage at 4°C (포장방법에 따른 사슴고기의 냉장저장 중 이화화적 성질의 변화)

  • A total of 5 female elk deer (220 $kg{\pm}10$ kg) were included in a study on the changes in physico-chemical properties of deer meat during storage at $4^{\circ}C$. The deers were exposed to normal pre-slaughter handling and put under anesthesia before slaughtered. The loin and leg cuts were deboned from the carcass after 24hrs slaughter. The samples weighing approximately 300 g were packaged using three packaging methods : Atmospheric packaging(A), Vacuum packaging (V) and Modified atmospheric at packaging (M: 80% $CO_2$ +20% $N_2$)and stored for 3, 7, 11, 15, 20 and 25days at $4^{\circ}$. The ability of moisture maintenance tended to be decreased with the passage of storage time, and that of the meats treated with the vacuum wrapping was lower than that with other wrapping methods. TBARS values of the meats with the regardless of treatments was increased with the passage of storage time, although the values with vacuum wrapping was lowest. VBN values of the meats with the regardless of treatments was also increased with the passage of storage time, although the values with vacuum wrapping was also lowest. The pH values of the meats with the regardless of treatments was also increased with the passage of storage time, although the values with vacuum wrapping was also lowest. For the meat softness, the value of the meats treated with atmospheric packaging was increased with the passage of storage time, but the values with vacuum wrapping and modified atmospheric packaging were respectively decreased. There were no significant differences in luminosity of leg, although loin treated with vacuum wrapping was significantly brighter than that with atmospheric packaging and modified atmospheric packaging. In general, the index of red color of the meats treated with vacuum wrapping was higher than that with atmospheric packaging and modified atmospheric packaging. The index of yellow color tended to be decreased after increment for a time with the passage of storage time, although the value with vacuum wrapping was generally higher than that with other wrapping methods.

Analysis of Respiratory Motion Artifacts in PET Imaging Using Respiratory Gated PET Combined with 4D-CT (4D-CT와 결합한 호흡게이트 PET을 이용한 PET영상의 호흡 인공산물 분석)

  • Purpose: Reduction of respiratory motion artifacts in PET images was studied using respiratory-gated PET (RGPET) with moving phantom. Especially a method of generating simulated helical CT images from 4D-CT datasets was developed and applied to a respiratory specific RGPET images for more accurate attenuation correction. Materials and Methods: Using a motion phantom with periodicity of 6 seconds and linear motion amplitude of 26 mm, PET/CT (Discovery ST: GEMS) scans with and without respiratory gating were obtained for one syringe and two vials with each volume of 3, 10, and 30 ml respectively. RPM (Real-Time Position Management, Varian) was used for tracking motion during PET/CT scanning. Ten datasets of RGPET and 4D-CT corresponding to every 10% phase intervals were acquired. from the positions, sizes, and uptake values of each subject on the resultant phase specific PET and CT datasets, the correlations between motion artifacts in PET and CT images and the size of motion relative to the size of subject were analyzed. Results: The center positions of three vials in RGPET and 4D-CT agree well with the actual position within the estimated error. However, volumes of subjects in non-gated PET images increase proportional to relative motion size and were overestimated as much as 250% when the motion amplitude was increased two times larger than the size of the subject. On the contrary, the corresponding maximal uptake value was reduced to about 50%. Conclusion: RGPET is demonstrated to remove respiratory motion artifacts in PET imaging, and moreover, more precise image fusion and more accurate attenuation correction is possible by combining with 4D-CT.

Effect of Flower and Fruit Thinner on Fruit Set and Fruit Quality of 'Gamhong' Apples (적화제 및 적과제가 '감홍' 사과의 착과와 과실 품질에 미치는 영향)

  • This study investigated the effects of flower and fruit thinning agents on fruit set and harvested fruit quality attributes in 'Gamhong' apples. Lime sulfur, MaxCel (1.9% BA), and Fruitone (3.5% NAA) were applied either at post-bloom or fruitlet stages to mature 'Gamhong/M.9' trees. In 2011, the numbers of fruits per cluster in terminal flowers were 1.74, 0.82, and 1.15 for the control, lime sulfur, and Maxcel (applied at 10-mm fruit stage) treatments, respectively. The percentages of single fruit per cluster were 36.0, 47.9, and 48.7% for the control, lime sulfur, and Maxcel (10 mm) treatments, respectively, while the percentages of clusters with three fruits per cluster were 22.9, 1.4, and 5.8%. In lateral flowers, fruit numbers per cluster were 1.20, 0.36, and 0.50 for the control, lime sulfur, and Maxcel (10 mm) treatments, respectively. In 2012, all the thinning treatments showed a positive effect on flower and fruit thinning, compared with the control. Moreover, the treatment with thinning agents did not affect fruit quality. Overall, the results suggest that a single application of flower or fruit thinning agents would be sufficient, rather than the mixed application of thinning agents, based on the observed decrease in fruit setting.

Characteristics of Salt Tolerance in Tree Species(I) - Relationship between Tree Species Distribution and Soil Salt Concentration in East Coastal Forest - (수종간(樹種間)의 내염력(耐塩力) 특성(特性)에 관(關)한 연구(硏究)(I) - 동해안림(東海岸林)의 수종분포(樹種分布)와 염분농도(塩分濃度)와의 관계(關係) -)

  • Relationship between distribution of tree species and salt concentration in soil was studied in order to understand the salt tolerance of tree species in the middle part of Korean east coast and its results were as follows; 1) The tree species in the area mostly consist of Pinus thunbergii, Pinus densiflora, Rosa rugosa, Lespedeza bicolor, Amorpha fruticosa, Quercus dentata, Rhododendron mucronulatum, Rohinia pseudoacacia and others. 2) Pinus thunbergii was dominant species and Rosa rugosa gradually disappeared at the distance from the beach line to 200 meters toward inland. Pinus thunbergii tended to disappear gradually and Pinus densiflora was dominant at the distance from 200 meters to 300 meters inland. 3) Pinus thunbergii was dominant below 50 meters in altitude while Pinus densiflora was dominant above 50 meters. 4) NaCl content tended to decrease as proceeding to inland, higher altitude and to shallow depth of soil. 5) Change in pH in terms of distance toward inland and altitude was not significant, but change in pH along soil depth was conspicuously decreasing from pH 7.0 of top soil to pH 6.5 50cm below. 6) Pinus thunbergii was densely distributed to the area where soil NaCl content was higher than 100 vpm, while Pinus densiflora dominated the area of less than 100 vpm. Rosa rugosa was shown to dominate the area of over 100 vpm soil NaCl content. 7) NaCl content in tree tended to decrease, as proceeding to inland, rapidly within 150 meters distance from the beach line and gradually at further distances. NaCl content in leaf was about 600 ppm, branch 350ppm and root 250 ppm.
