Localization and characteristics of lactate and palate dehydrogenase in the sparganum and adult worm of Spirometra erinacei (만손열두조충에서 젖산탈수소효소와 말산탈수소효소의 분포와 특징)

  • This study investigated the enzyme histochemical localization and characteristics of lactate (LDH) and malate dehydrogenase (MDH) related with the oxidation-reduction metabolism in the sparganum and adult of 5. erinacei. By enzyme histochemical assay, activity of LDH was strong in the tegument and subtegumental muscle layers of the adult and sparganum. Activity of MDH was strong in the tegument of the sparganum and subtegumental muscle layers of the adult. However it was weak in the tegument of the adult. By electrophoresis, 45 kDa band was major and common in LDH of adults and spargana. The 150 kDa molecule was the major and common band in MDH of adults and r -spargana (from experimentally infected rats) . By isoelectrofocusing, isoelectric points (Pl) or 4 MDH isogyme from adult worm were 6.0.6.5, 6.7 and 7.1, respectively. Pl 6.0 was the major band. The active range of pH for MDH was about pH 6-8 and the optimum pH was pH 7 The effective temperature on the MDH was about $30^{\circ}C$$50^{\circ}C$ and the optimum temperature was about 40℃ in spargana md adult worm. In the stability against heat, when MDH was heated at 85℃ for 10 seconds, the activity was denatured perfectly. Maximum activity or MDH was 19.4 unit in the s-sparganum (from snakes), 24.5 unit in the r-sparganum (from rats) and 108.0 unit in the adult worm. The maximum activity was higher in adults than in spargana. The present result showed us that the nutrients absorbed through the tegument were transferred into inner tissues and were utilized as the source of metabolism. According to the habitat of the parasite, the isozymes of LDH and MDH are activated differently, and by this different activation the sparganum and adult can adapt themselves to parasitic circumstances.

Effect of Package Size and Pasteurization Temperature on the Quality of Sous Vide Processed Spinach (Sous Vide 가공 시금치의 품질에 미치는 포장단위 및 살균온도의 영향)

  • Microbial lethal value and nutrient retention of sous vide processed spinach were evaluated with mathematical model prediction and experimental trial for different package sizes and pasteurization temperatures. The package size covers 500 g, 1 kg and 2 kg, while the pasteurization temperature includes 80, 90 and 97$^{\circ}C$. The basic process scheme consists of filling blanched spinach into barrier plastic film pouch, sealing under vacuum, pasteurization in hot water with over pressure and final cooling to 3$^{\circ}C$. Pasteurization condition was designed based on attainment of 6 decimal inactivation of Listeria monocytogenes at geometric center of the pouch package by heating cycle, which was determined by general method. Heat penetration property of the package and thermal destruction kinetics were combined to estimate the retention of ascorbic acid and chlorophyll. Smaller packages with shorter pasteurization time gave better nutrient retention, physical and chemical qualities. Larger package size was estimated and confirmed experimentally to give higher pasteurization value at center, lower ascorbic acid and chlorophyll contents caused by longer heat process time. Lower pasteurization temperature with longer process time was predicted to give lower pasteurization value at center and lower ascorbic acid, while chlorophyll content was affected little by the temperature. Experimental trial showed better retention of ascorbic acid and chlorophyll for smaller package and higher pasteurization temperature with shorter heating time. The beneficial effect of smaller package and higher pasteurization temperature was also observed in texture, color retention and drip production.

Antioxidative Activity of Rooibos Tea(Aspalathus linearis) Extracts (용매에 따른 Rooibos Tea(Aspalathus lineais) 추출물의 항산화 효과)

  • Total phenolic compounds and antioxidative activities of rooibos tea(Aspalathus linearis) fractions were studied. Three extracts, using hexane, ethyl acetate, and ethanol, were prepared. Total phenolic compounds were 3069.3 mg/100 g extract in the hexane fraction, 18604.4 mg/100 g extract in the ethyl acetate fraction, and 13458.8 mg/100 g extract in the ethanol fraction. Levels of vanillic acid, caffeic acid, syringic acid, 4-coumaric acid, and ferulic acid were analyzed by RP-HPLC, and totals of 3452.6 and 3156.1 mg/100 g of extract were found in the ethanol and ethyl acetate fraction, respectively. In the DPPH assay, the ethanol fraction(82.2% of contol) and the ethyl acetate fraction(78.9%) showed the highest free radical scavenging capacities. The induction period of each tea fraction in the fish oil rancimat assay was measured. When 500 ppm of the ethanol fraction was applied, a 1.19 h induction period was observed. This was 2-fold greater than the induction period of the control.

Improvement of Physical Properties for Edible Films from Alaska Pollack Protein (명태 단백질로 제조한 가식성 필름의 물성 개선)

  • The edible films were prepared from the protein of alaska pollack, Theragra chalcogrmma. Effects of plasticizer, cross linker and laminated film on physical properties such as tensile strength (TS), elongation (E) and water vapor permeability (WVP) of films were investigated. In adding various kinds of plasticizers, TS of the films prepared with propylene glycol (PG) was the highest, and followed sorbitol, polyethylene glycol 200 (PEG 200) and glycerol. Elongation of the films prepared with glycerol was the highest, then sorbitol, PEG 200 and PG. WVP of films showed lower in order of PG, sorbitol, glycerol and PEG 200.75 decreased with the increment of plasticizer concentration, but elongation increased, The addition of both PG and PEG 200 effected weakly on elongation, so they were inadequate as plasticizer for the film. Mixtures of glycerol and sorbitol, which showed opposing both TS and elongation in the films, could control the physical properties of the films. With increasing relative humidity, TS decreased, while elongation and equilibrium moisture content increased. By adding the cross linkers such as ascorbic acid, citric acid and succinic acid, TS and m of films increased, while elongation decreased. Ascorbic acid, citric acid, succinic acid were most effective for TS at 0.2, 0.1 and $0.1\%, respectively. Laminated film with alaska pollack protein and corn zein improved TS above two times, reduced WVP about $20\~30\%$, as compared with the Elm from alaska pollack protein. Two films did not show the difference to oxygen permeability, but they showed about tenfold greater oxygen resistance than polyethylene film. Laminated film showed higher b and $\Delta$E value of color difference, lower a and L value than the film from alaska pollack protein.

MEASUREMENT OF PULPAL BLOOD FLOW USING A LASER DOPPLER FLOWMETER (Laser Doppler flowmeter를 이용한 치수혈류 측정)

  • Blood supply rather than nerve supply implies pulp vitality. To evaluate pulp vitality clinically, electric pulp test and thermal test which are based on sensory nerve response have been used in addition to many auxiliary data such as past dental history, visual inspection, radiographic examination, percussion, palpation and transillumination test. However, reactivity of the nerves to the stimulation is not synonymous with normalcy. Therefore measurement of pulpal blood flow using a laser Doppler flowmeter became a new trial to test the pulp vitality. The purpose of the present study was to evaluate normal pulpal blood flow level of maxillary teeth in adult to provide a guideline in determining the vitality of dental pulp. Pulpal blood flow was measured in maxillary central and lateral incisors, canines, first and second premolars and first molars of seventy nine adults of 22 - 30 years old using a laser Doppler flowmeter (PeriFlux 4001, Perimed Co., Stockholm, Sweden, 780 nm infrared laser, 1mW). For directly-made splints, silicone rubber impressions were taken directly from the mouth. For indirectly-made splints, alginate impressions were taken from the mouth and stone cast were made. After making depressions on the buccal surfaces of the cast teeth to indicate the hole positions, second impressions with vinyl polysyloxane putty were taken from the cast. Holes for the laser probes were made at the putty impressions 4mm above the gingival level. Laser probe (PF416 dental probe, 1.5mm) was inserted in the prepared hole and the splint was set in the mouth. After 10 minutes of patient relaxing, pulpal blood flow was recorded for 5 minutes on each tooth. The recorded flow was saved in the computer and calculated with a software 'Perisoft' version 5.1. Pulpal blood flow was also recorded in six teeth of five individuals with no response to electric pulp test and cold test, with periapical radiolucency, or with history of root canal treatment to compare with nonvital teeth. The difference between the mean flow values of each group of teeth were analyzed using one-way ANOVA and Duncan's Multiple Range test. The results were as follows: 1. The average pulpal blood flow values of all the tested teeth of each location were between 9 - 16 Perfusion Unit. Pulpal blood flow value was highest in maxillary lateral incisors, followed by first premolars, second premolars, canines, central incisors, and then first molars (p<0.01). 2. In six anterior teeth, indirectly-made splint group showed higher pulpal blood flow values than directly-made splint group (p<0.01). In posterior teeth, however, there was no significant flow value difference between directly-made splint group and indirectly-made splint one (p>0.05). 3. Teeth with vital pulps showed higher signal values than teeth with nonvital pulps (p<0.01), and the flow photographs showed heartbeat-synchronous fluctuations and vasomotions, while those were absent in non vital tooth.

Optimizing Ingredients Mixing Ratio of Mungbean Pancake (빈대떡의 재료혼합비율의 최적화)

  • The sensory acceptability, texture profile analysis and nutritional evaluation were peformed in Korean traditional mungbean pancake (MPC) and modified MPC containing squid meat and soybean to standardize the recipes for healthy fast food market potentiality. Optimal ingredient formulations were revealed as mung-bean 55$\%$, pork 13$\%$ and vegetables 32$ \%$ for traditional MPC, and pork 3$\%$, squid 42$\%$ and soybean 55$\%$ for modified MPC using response surface methodology. Flavor and hardness correlated highly with overall accept-ability rather than appearance and color of traditional MPC. Higher squid levels raised adhesiveness, springi-ness and resiliences of modified MPC, but soybean decreased these textural attributes. Protein, lipid and total calorie of modified MPC were lower than those of traditional MPC. Computed protein efficiency ratio (C-PER) and degree of gelatinization of modified MPC were superior than traditional MPC.

Anti-Inflammatory Effect of Ethanol Extract from Onion (Allium cepa L.) Peel on Lipopolysaccharide-Induced Inflammatory Responses in RAW 264.7 Cells and Mice Ears (LPS로 유도된 RAW 264.7 세포와 마우스 귀 조직에 대한 양파(Allium cepa L.) 껍질 에탄올 추출물의 항염증 효과)

  • Inflammation is a complex process involving a variety of immune cells, which defend the body from harmful stimuli. However, pro-inflammatory cytokines and inflammatory mediators can also exacerbate diseases such as cancer. Onion peel contains several phenolic compounds, including quercetin at an amount 20 times greater in peel than edible flesh. Therefore, in this study, the anti-inflammatory effects of onion peel ethanol extract (OPEE) were investigated lipopolysaccharide-induced inflammatory response. In our results, NO production decreased in a dose-dependent manner. Secretion of IL-6, $TNF-{\alpha}$, and $IL-1{\beta}$ was suppressed by 44%, 53%, and 60% respectively, at $100{\mu}g/mL$. Moreover, OPEE also suppressed expression of COX-2, iNOS, $NF-{\kappa}B$, and MAPKs in a dose-dependent manner. Formation of mice ear edema was reduced at the highest dose tested compared to the control, and reduction of ear thickness was observed in the histological analysis as well. In the acute toxicity test, no morality was observed in mice administered 5,000 mg/kg body weight of OPEE over a 2-week observation period. These results suggest that OPEE may have significant effects on inflammatory factors and be a potential anti-inflammatory material.

Relation of Health Status, Nutrient Intake, and Dietary Self-Efficacy to the Obesity Levels of Male Workers (남성 근로자의 비만정도에 따른 건강상태와 영양상태 및 식이 자기효능감과의 관계)

  • This study was intended to collect the baseline information on health status, nutrient intakes and dietary self-efficacy according to the obesity levels of male workers. Nutrient intakes of 224 male workers were assessed by 24 hr recall method; also, dietary attitude and dietary self-efficacy were investigated by self-administerd questionnaires. Biochemical characteristics such as blood pressure (SBP, DBP), total cholesterol(TC), hemoglobin (Hb), blood glucose (BG), SGOT, SGPT and ${\gamma}-GTP$ were assessed. Subjects were divided into normal, overweight and obese groups by body mass index (BMI). The health status was analyzed as normal group, health concerned group and disease suspected group. Weight, percent ideal body weight (PIBW), BMI and body circumference (waist, hip) and waist-hip ratio were high in the order of obese group, overweight group and normal group. Body fat mass was high also in the order of obese, overweight and normal group. Blood pressure (SBP, DBP) was the highest in obese group. SGOT, SGPT, ${\gamma}-GTP$ were higher in the obese group than normal and overweight groups. It was Proven that the percentage of those evaluated as low nutrient intakes were 71% for Ca, 50.9% for Zn, 70.5% for vitamin B2, 56.3% for vitamin C and 81.3% for folic acid. The dietary attitude scores were high according to the dietary self-efficacy. The age was positively correlated with dietary attitude, and the dietary attitude was positively correlated with dietary self-efficacy and total energy intakes. Dietary attitude showed the greatest total causal effect in relation to dietary self-efficacy. The results of this study showed some health problems and nutritional problems, indicating the need for nutritional management for male workers.

Tensile Bond Strength of Composite Resin Treated with Er:YAG Laser (Er:YAG 레이저를 활용한 와동형성시 컴포짓 결합강도)

  • This in vitro study evaluated the influence of a flowable composite resin on the tensile bond strength of resin to enamel and dentin treated with Er:YAG laser and diamond bur. 96 Buccal enamel and mid-coronal dentin were laser-irradiated using an Er:YAG laser and treated with diamond bur. Each groups(48) were divided two small groups depends on acid-etching procedure. Light-cure flowable resin(Metafil Flo) and self-cure resin(Clearfil FII New Bond) were used in this study. After surface etching with 37% phosphoric acid and the application of an adhesive system, specimens were prepared with a hybrid composite resin. After 24hours storage in distilled water at 37$^{\circ}C$, all samples were submitted to the tensile bond strength evaluation, using a universal testing machine(Z020, Zwick, Germany). The obtained results were as follows: 1. TBS of acid-etching group were higher than those of non-etching group in both enamel and dentin treated with Er:YAG laser and diamond bur. Laser 'conditioning' was clearly less effective than acid-etching. Moreover, acid etching lased enamel and dentin significantly improved the microTBS of M-Flo. 2. In enamel, TBS of laser-irradiated group were lower than those of bur-prepared group. However, in flowable resin subgroup, there were not differed those between two groups in dentin. 3. In laser-treated group, TBS of flowable composite resin were higher than those of self-curing resin in dentin, however, there was no difference in enamel. From this study, we can conclude that the self- and light-cure composite resin bonded significantly less effective to lased than to bur-cut enamel and dentin, and that acid-etch procedure remains mandatory even after laser ablation. We suggest that Er:YAG laser was useful for preparing dentin cavity with flowable resin filling.

Comparison of the Results in Gastric Carcinoma Patients undergoing Billroth I and Billroth II Gastrectomiesy (원위부 위암에서 위절제술 후 위재건술식에 따른 잔위기능 및 영양 증상학적 삶의 질 비교)

  • Purpose: The proper reconstruction technique to use after a distal subtotal gastrectomy for a gastric carcinoma, there has been a subject for debated what is the proper reconstruction technique. The aim of this study was to compare the gastricemptying time and the quality of life following both B-I and B-II reconstructions after a distal gastrectomy for a gastric adenocarcinoma. Materials and Methods: We studied 122 patients who had undergone a distal gastrectomy for a gastric adenocarcinoma between June 1999 and July 2002 at our hospital. 51 patients underwent B-I group, and 71 patients underwent B-II group. To evaluate the gastric-emptying time, we analyzed the T1/2 time by means of radionuclide scintigraphy using a gamma camera after ingestion of an $^{99m}Tc$-tin-colloid steamed egg. The nutritional status was measured by the weight change. Postgastrectomy syndrome was evaluated using an abdominal symptoms survey. Dumping syndrome was measured using the Sigstad dumping score. Results: The gastric-emptying time was somewhat delayed in the B-I group after a 6 month period, but there was no difference after 12 months between the two groups. There was less weight loss in the B-I group than in the B-II group (P=0.023). Fewer abdominal symptoms were occurred in the B-I group than in the B-II group. Dumping syndrome occurred less frequently in the B-I group than in the B-II group (P=0.013). Conclusion: In our study, the Billroth I reconstruction led to less weight loss, a better nutritional status, and a better quality of life than the Billroth II reconstruction. We concluded that after a distal subtotal gastrectomy, the Billroth I reconstruction would be considered when the procedure is oncologically suitable.
