It were reported that antifungal mechanism of Enterobacter cloacae is a volatile ammonia that produced by the strain in soil, and the production of ammonia is related to the bacterial urease activity. A powerful bacterium SH14 against soil-borne pathogen Fusarium solani, which cause root rot of many important crops, was selected from a ginseng pathogen suppressive soil. The strain SH14 was identified as Bacillus subtilis by cultural, biochemical, morphological method, and $API^{circledR}$ test. From several in vitro tests, the antifungal substance that is produced from B. subtilis SH14 was revealed as heat-stable and low-molecular weight antibiotic substance. In order to construct the multifunctional biocontrol agent, the urease gene of Bacillus pasteurii which can produce pathogenes-suppressive ammonia transferred into antifungal bacterium. First, a partial BamH I digestion fragment of plasmid pBU11 containing the alkalophilic B. pasteurii l1859 urease gene was inserted into the BamH I site of pEB203 and expressed in Escherichia coli JM109. The recombinant plasmid was designated as pGU366. The plasmid pGU366 containing urease gene was introduced into the B. subtilis SH14 with PEG-induced protoplast transformation (PIP) method. The urease gene was very stably expressed in the transformant of B. subtilis SH14. Also, the optimal conditions for transformation were established and the highest transformation frequency was obtained by treatment of lysozyme for 90 min, and then addition of 1.5 ${mu}g$/ml DNA and 40% PEG4000. From the in vitro antifungal test against F. solani, antifungal activity of B. subtilis SH14(pGu366) containing urease gene was much higher than that of the host strain. Genetical development of B. subtilis SH14 by transfer of urease gene can be responsible for enhanced biocontrol efficacy with its antibiotic action.
The purpose of this study was to verify the usefulness of MTT analysis as a tool of measurement of the periodontal ligament cell viability from the extracted rat molar. A total of 80 Sprague-Dawley white female rat of 4 week-old with a body weight of 100 grams were used. The maxillary left and right, first and second molars were extracted under Ketamine anesthesia. Twenty-four teeth of each group (divided as five groups depending upon the time-lapse after extraction such as Immediate, 10, 20, 40 and 60 minutes) were immersed in $200{\mu}l$ of MTT solution (0.5 mg/ml) and processed for optical density measurements. Another 10 teeth of each group were treated as same as above and sectioned at $10{\mu}m$ for microscopic examination. All measurements values were divided by the value of hematoxylin-eosin staining which represented the volume of each corresponding samples. Immediate and 10 minute groups showed highest MTT values followed by 20, 40, and 60 minutes consecutively. Statistical significance (p<0.05) existed between all groups except in immediate versus 10 minute groups and 40 versus 60 minutes. Histological findings also showed similar findings with MTT results in crystal shape and crystal numbers between the experimental groups. These data indicate that in vivo MTT analysis nay be of value for evaluation of the periodontal ligament cell viability without time- consuming cell culturing processes.

This study aimed to reconsider the theory that the renowned Amisan(峨眉山) terraced garden at north of Gyotaejeon(交泰殿) was artificially made, by reviewing the historical records and drawings. It has been widely accepted that Amisan was made of the digged soil from Gyeonghoeji(慶會池). But several arguments about artificial mountain theory of Amisan that completely not be found in historical records have been raised in this study. The results were summarized as follows; the inherent contradiction in existing opinion, the discordance between the time of building Gyeonghoeji and Gyotaejeon, the existence of the mountain range which connect Baekaksan and Amisan appeared in Dohyeong(圖形), historical documents written in the years of kingdoms of Youngjo(英祖) and Gojong(高宗), a high position seen from Heungbogjeon(興復殿) in the north Amisan through the wall in the east but impassable, an opinion about realization Amisan as geomantic term of Amisa(蛾眉砂) at the time of Gyeongbok Palace reconstruction, and preservation of the mountain range in Gyeongbok Palace that comes from the result of the arguments in main mountain of Gyeongbok Palace in the year of Sejong(世宗). In addition, it was investigated why the slop in the north of Gyotaejeon was named as Aminsan and why the artificial mountain theory is appeared and made a conclusion that the Amisan comes from the change of the pronunciation of the geomantic term "Amisa", and modeling the yijing[意景] of Amisan which is a sacred place of Taoism and Buddhism in Sichuan[四川] of Chinaand the view of construction to mean defeating a spirit of smallpox which had to be cured. And it seems to be a result which retroactively applied the artificial mountain theory of Amisanis the technique of 'constructing mountain with digged pond dirt' to the relationship between Gyeonghoeji and Amisan. The greater part of mountain range which was connecting with Baekaksan and Amisan was seriously disconnected with large scale of exposition by the Japanese colonial period in 1915. But low slope is kept about 70 meters along the trail northeast of Gyotaejeon. Accordingly, it is judged that the range has not been entirely destroyed. And according to the result of elevation analysis, discontinuous slope form certain axis is found, so the mountain range of Amisan is approximately estimated. This basic research about the mountain range of Amisan might provide a critical clue about restoration of topography in Gyeongbok Palace.

Recently in Korea, in the case of metropolis, the urban signalized intersections are controlled by traffic information center or ITS center. Cheon-an City also established traffic information center through the 1st.-$\sim$3rd. ITS public construction and has managed this center that includes bus information service, traffic information collection and providing service, parking information service, and traffic responsive control system. In the Cheon-an metropolitan traffic signal operation, traffic signal controllers were grouped by the each main traffic flow axes and performed with coordinated signal timing for the signalized arterials, and also cycle and split changed by realtime traffic demands. Cheon-an urban traffic responsive control system was evaluated by intersection delay and speed, then it was verified that the delay decreased and vehicle speed improved. However, the rural signal control system to connect adjacency town was evaluated to have lower status than urban area due to the unimproved TOD (Time of day) plan. Therefore actuated signal control was examined for substitutive control system in isolated signal intersection. The aim of this article is to compare actuated signal control with TOD mode in the rural intersection of Cheon-an and to fine superiority of these two control mode, with evaluation of vehicle delay by using HCM(2000) method and by micro-simulation CORSlM. The result of field test show that actuated signal control gave better performance in delay comparison than the existing TOD signal control. And simulation outcome verified that non-optimized TOD has higher delay than optimized TOD mode, non-optimal actuated mode, and optimal actuated signal control mode. Particularly, these three modes delays had not different values according to the paired sample t-test. This is because small traffic demands were loaded in each links. This suggested actuated signal control is expected to be more effective than TOD mode in some rural isolated intersections which frequently need to survey for traffic volume.
This study was designed to determine the optimal ratio of dropwort powder in castella by adding the powder at levels of 0, 3, 6, 9, and 12% respectively. The properties of the castella were analyzed by specific gravity, specific volume, color determinations, texture properties and sensory evaluation. The Specific gravity increased with increasing amount of dropwort powder. However, the specific volume decreased with increasing dropwort powder. For the color values, as more dropwort powder was added, the L-value decreased. The castella with 9% dropwort powder had a higher hardness, gumminess, and chewiness. A sensory panel perceived that the external and internal color of the castella become darker with the dropwort powder substitution and the grain size decreased with increasing amount dropwort powder, while sweet taste showed no significant difference. The order of overall preference was DP 9>DP 6>DP 12>CON>DP 3. Therefore, the substitution of 9% of wheat flour with dropwort powder was recommended in the production of castella.

Along with recent improvement of recycling technique, the quality of the recycled concrete aggregate have become very competitive to the natural concrete aggregate. Therefore, a practical use of the recycled concrete aggregate may be possible for structural members. Majority studies about the recycled concrete aggregate was emphasized a limitation of fundamental study concerned with a strength characteristics and durability of the recycled aggregate concrete, there is use for the structural members. Therefore, for the extension of application of recycled concrete aggregate, this investigation verifies the strength characteristics recycled concrete aggregate of the spun-concrete products with various coarse and fine recycled aggregate replacement ratio(coarse recycled aggregate: 0%, 20%, 40%, 60%, 100%; fine recycled aggregate: 0%, 30%, 60%, 100%) and with addition of cellulose fibers(0%, 0.01%, 0.03%, 0.05%, 0.08%). From the test results, The strength of spun concrete used with recycled aggregate [NR specimen], was measured as 72MPa, was found to be very approximately to the strength of spun concrete used with the natural aggregate(NN specimen), was measured as 74MPa, when only fine aggregate was replaced with the recycled. Therefore, the fine recycled concrete aggregate can be successfully used in the spun high strength concrete product. The compressive strength of all specimens used the specialty cellulose fiber were measured as about 70M Pa, however, the increasement of the specialty cellulose fiber content is showed to decrease compressive strength of spun concrete. Therefore, it is anticipated that the specialty cellulose fiber can be applied to the various spun concrete products.

We investigated whether or not Schisandra chinensis (SC), a traditional herbal medicine, has protective effects against alcohol-induced fatty liver and blood alcohol clearance. Two tests focused on acute intoxication and chronic ethanol treatment were carried out. For the chronic ethanol treatment test, rats were fed ethanol by intragastric administration everyday for 8 weeks to induce alcoholic fatty liver. Ethanol treatment significantly increased blood alcohol concentration at 90 min after acute ethanol intoxication. Compared with the two ethanol-treated groups, rats administered ethanol along with SC extracts showed an approximately 13% increased blood alcohol clearance rate at 360 min. Chronic ethanol treatment significantly increased serum and hepatic triglyceride (TG) levels, and caused fatty degeneration of liver. Ethanol treatment also elevated the serum total-cholesterol (TC) level. However, after feeding of ethanol plus SC extracts, ethanol-induced elevation of hepatic TG levels reversed, whereas elevation of serum TG and TC levels was not observed after treatment with SC extracts. Ethanol treatment significantly increased ${\gamma}$-GT, aspartate aminotransferase, and alanine aminotransferase activities after 8 weeks. Compared with the ethanol-fed group, rats administered ethanol plus SC extracts for 4 weeks showed attenuated fatty degeneration as well as decreased hepatic function test values. SC administration also significantly increased intracellular lipid accumulation in hepatocytes and reduced steatosis score and hepatic TG levels, as measured by biochemical and histolopathological analyses. Our results indicate that the protective effects of SC are accompanied by a significant decrease in hepatic TG levels, thereby suggesting SC has the ability to prevent ethanol-induced fatty liver, by reducing hepatic TG and enzyme levels in alcoholic rats.

This study was conducted to examine the relationships between milk and milk product intake and bone health. The data from the 2008~2011 Korean National Health and Nutrition Examination Survey (KNHANES) were used for data analysis. Subjects included 4,626 men and 6,144 women aged 19 to 64 years. Daily intake frequency of milk and milk products was obtained using a food frequency questionnaire and divided into two categories: less than one serving per day and more than one serving per day. Bone mineral density (BMD) values of total femur, femoral neck, and lumbar spine were compared based on daily intake frequency, and relationships between milk and milk product daily intake frequency and osteoporosis risk were evaluated based on logistic regression. In men aged 30~39, BMDs of total femur and femoral neck were significantly higher in the group that consumed milk more than one serving per day (P<0.05). Intake frequency of milk and milk products was also significantly related to both BMDs of total femur and femoral neck. The odds ratio (OR) for milk intake frequency (more than one serving per day) compared to intake frequency less than one serving per day was 0.36 [95% confidence interval (CI) 0.21~0.62], and the OR for milk and milk products intake frequency (more than one serving per day) was 0.49 (95% CI 0.28~0.86) in women aged 50~64. These results indicate that increased consumption of milk and its products is associated with reduced risk of bone health disease, and adequate intakes of milk and milk products might play an important role in maintaining optimum bone health. Further research on the causal relationship and dose-response association between milk intake and bone heath using prospective cohort data is required prior to applying the observed results to programs that prevent bone health problems.

Objectives: The purpose of this study is to investigate the effects of 38-hour sleep deprivation on fine motor performance. The Motor Performance Series (MPS) in the Vienna Test System (computerized neurocognitive function tests) was used in this study. Methods: Twenty four subjects participated in this study. Subjects had no past history of psychiatric disorders and physical illness. Subjects had normal sleep-waking cycle without current sleep disturbances and were all right-handed (Annett's Hand Preference Questionnaire: above +9 points). To minimize the learning effects, familiarization with the Vienna Test System was performed one day before the study. Subjects were to get up at 6:00 in the morning after getting enough sleep according to his or her usual sleep-wake cycle. After awakening, subjects remained awake for 38 hours under continuous surveillance. During two consecutive study days, the subjects tested MPS at 7 AM and 7 PM each day, which means the MPS was done four times in total. During the experiment, anything that could affect the subjects' sleep such as coffee, tea, alcohol, a nap, tiring sports, and all medications were prohibited. Results: In MPS, the fine motor functions of both hands decreased after 38 hours of sleep deprivation. The decrement in motor performance was prominent in the dominant right hand. In the right hand, the total number of tapping was reduced (p<.005), and the number of misses (p<.05) and the length of misses (p<.05) of line tracking, the total length of inserting a short pin (p<.01), the total length of inserting a long pin (p<.05), and the number of misses in aiming (p<.05) increased. Such performance decrement was distinct in the morning sessions. Conclusions: These results suggest that fine motor performance decrement during sleep deprivation is predominant in the right hand, which exerts maximal motor function. The finding of decrement in motor function in tapping during sleep deprivation also suggested that the time required for exhaustion of muscles is shortened during sleep deprivation. More deterioration of motor performance was shown in the morning, which could be explained as circadian rhythm effects.
Background : Following several decades of decline, the incidence of tuberculosis has recent1y begun to increase in many countries of this the control of this disease has been impeded by the emergence of multi-drug resistant tuberculosis (MDR-TB). The development of rapid diagnostic methods and effective new drugs are needed to control MDR-TB. One of the new drugs for MDR-TB is rifabutin (RBU) which has been known to be effective in some patients with MDR-TB. A few reports showed that some types of mutations of the rpoB gene, which were known to be present in 96-98% of rifampicin-resistant M. tuberculosis, were associated with the rifampicin-resistant but RBU-susceptible phenotype. This study was performed to investigate the correlation between RBU susceptibility and the patterns of rpoB gene mutations in Korean MDR-TB. Methods : Sixty-five clinical isolates of multi-drug resistant Mycobacterium tuberculosis, gathered from patients who visited the Asan Medical Center from July 1997 to June 1999, were investigated. Clinical responses to rifabutin-containing regimen were evaluated. An RBU susceptibility test and sequencing analysis of rpoB gene were performed, and the results were analyzed to confirm which mutations correlated with RBU-susceptible MDR-TB. Results : Fifty-three of 56 (95%) clinical isolates of MDR-TB had 60 mutations of the rpoB gene. The most frequent mutations were found at codon 531 (43%), and two mutations were combined in seven clinical isolates. Five of 53 (10%) clinical isolates showed the RBU-susceptible phenotype, and in them the characteristic patterns of point mutations were found at codon 509, 516, and 526. Conclusion : The frequency and pattern of mutations of the rpoB gene of Korean MDR-TB isolates were similar to those in western countries, where the prevalence of tuberculosis is low, but some show RBU-susceptible phenotypes. RBU-susceptible MDR-TB isolates showed the characteristic pattern of mutations of the rpoB gene which could be used to rapidly diagnose RBU susceptibility.
