Human METCAM/MUC18, a cell adhesion molecule (CAM) in the immunoglobulin-like gene super family, plays a dual role in the progression of several epithelium cancers; however, its role in the nasopharyngeal carcinoma (NPC) remains unclear. To initiate the study we determined human METCAM/MUC18 expression in tissue samples of normal nasopharynx (NP), NPCs, and metastatic lesions, and in two established NPC cell lines. Immunoblotting analysis was used for the determination in lysates of frozen tissues, and immunohistochemistry (IHC) for expression in formalin-fixed, paraffin-embedded tissue sections of 7 normal nasopharynx specimens, 94 NPC tissue specimens, and 3 metastatic lesions. Human METCAM/MUC18 was expressed in 100% of the normal NP, not expressed in 73% of NPC specimens (or expressed at very low levels in only about 27% of NPC specimens), and expressed again in all of the metastatic lesions. The level of human METCAM/MUC18 expression in NPC tissues was about one fifth of that in the normal NP and metastatic lesions. The low level of human METCAM/MUC18 expression in NPC specimens was confirmed by a weak signal of RT-PCR amplification of the mRNA. Low expression levels of human METCAM/MUC18 in NPC tissues were also reflected in the seven established NPC cell lines. These findings provided the first evidence that diminished expression of human METCAM/MUC18 is an indicator for the emergence of NPC, but increased expression then occurs with metastatic progression, suggesting that huMETCAM/MUC18, perhaps similar to TGF-${\beta}$, may be a tumor suppressor, but a metastasis promoter for NPC.

Background: In this study, we investigated whether lobetyolin, lobetyol, and methyl linoleate derived from Codonopsis pilosula affect MUC5AC mucin secretion, production, and gene expression from airway epithelial cells. Methods: Confluent NCI-H292 cells were pretreated with lobetyolin, lobetyol, or methyl linoleate for 30 minutes and then stimulated with phorbol 12-myristate 13-acetate (PMA) for 24 hours. The MUC5AC mucin gene expression, and mucin protein production and secretion were measured by reverse transcription polymerase chain reaction and enzyme-linked immunosorbent assay, respectively. Results: Lobetyolin, lobetyol, and methyl linoleate inhibited the gene expression of MUC5AC mucin induced by PMA; lobetyolin did not affect PMA-induced MUC5AC mucin production. However, lobetyol and methyl linoleate inhibited the production of MUC5AC mucin; lobetyolin and lobetyol did not significantly affect PMA-induced MUC5AC mucin secretion from NCI-H292 cells. However, methyl linoleate decreased the MUC5AC mucin secretion. Conclusion: These results suggest that among the three compounds, methyl linoleate can regulate gene expression, production, and secretion of MUC5AC mucin by directly acting on the airway epithelial cells.
Background: The mucin components of the gastric gel layer function as a protective and lubricating factor against luminal acid and proteolytic enzymes. Alteration of mucin expression in gastric preneoplastic and neoplastic lesions has suggested potential roles in neoplastic processes. This study aimed to assess the clinicopathological and prognostic significance of MUC-2, MUC-4 and MUC-5AC in Japanese gastric cancer. Methods: Expression of MUC-2, -4 and -5AC was evaluated on tissue microarrays of gastric carcinomas and adjacent non-cancerous mucosa specimens by immunohistochemistry and compared with clinicopathological parameters and survival time of the patients. Results: The three mucins were found to be expressed to a lesser extent in gastric carcinomas in comparison with non-cancerous mucosa (p<0.05). MUC-2 expression was negatively correlated with tumor size, depth of invasion, and TNM staging of gastric cancer (p<0.05), while that of MUC-5AC was negatively associated with the depth of invasion, venous invasion, lymph node metastasis and TNM staging (p<0.05), but positively with MUC-4 and MUC-2 expression (p<0.05). There was higher MUC-2 expression in intestinal- than diffuse-type carcinomas (p<0.05). Kaplan-Meier analysis indicated no relationship between expression of the three mucins and the cumulative survival rate of patients, even stratified according to the depth of invasion (p>0.05). Conclusion: Down-regulated expression of MUC-2, -4 and -5AC may be involved in pathogenesis, invasion, metastasis or differentiation of gastric carcinoma. Their altered expression might therefore be employed as an indicator of pathobiological behavior.

$CLB_{2.0}$, a constituent of PM, induces secretion of multiple cytokines and chemokines that regulate airway inflammation. Specifically, IL-6 upregulates $CLB_{2.0}$-induced MUC5AC and MUC1 expression. Interestingly, of the tight junction proteins examined, claudin-1 expression was inhibited by $CLB_{2.0}$. While the overexpression of claudin-1 decreased $CLB_{2.0}$-induced MUC5AC expression, it increased the expression of the anti-inflammatory mucin, MUC1. $CLB_{2.0}$-induced IL-6 secretion was mediated by ROS. The ROS scavenger N-acetyl-cysteine inhibited $CLB_{2.0}$-induced IL-6 secretion, thereby decreasing the $CLB_{2.0}$-induced MUC5AC expression, whereas $CLB_{2.0}$-induced MUC1 expression increased. $CLB_{2.0}$ activated the ERK1/2 MAPK via a ROS-dependent pathway. ERK1/2 downregulated the claudin-1 and MUC1 expressions, whereas it dramatically increased $CLB_{2.0}$-induced MUC5AC expression. These findings suggest that $CLB_{2.0}$-induced ERK1/2 activation acts as a switch for regulating inflammatory conditions though a ROS-dependent pathway. Our data also suggest that secreted IL-6 regulates $CLB_{2.0}$-induced MUC5AC and MUC1 expression via ROS-mediated downregulation of claudin-1 expression to maintain mucus homeostasis in the airway.
연구배경: 기관지 점액의 과분비는 천식의 중요한 기전중의 하나이며, 특히천식 환자에서는 MUC5AC가 중요한 역할을 하는 것으로 알려져 있다. MUC5AC는 다양한 유전자 다형성을 갖는 것으로 알려져 있으나 MUC5AC 유전자 다형성과 천식과의 관계를 본 연구는 거의 없는 실정이다. 따라서 본 연구는 MUC5AC 프로모터 부위의 유전자 다형성과 천식과의 관계를 조사하였다. 방법: 강원대학교 병원에서 78명의 천식환자와 이들과 성별, 나이가 일치하는 78명의 대조군을 선정하였다. 이들로부터 전혈을 채취하여 DNA를 분리하여 PCR과 RFLP를 이용하여 MUC5AC 프로모터의 -1274G>A 유전자 다형성을 분석하였다. 모든 대상환자는 의무기록지를 검토하여 주된 증상과 투여 약제를 확인하였으며, 이들에서의 폐기능, 총 호산구수, 혈청 IgE, 피부반응검사 결과를 조사하였다. 결과: 천식환자의 평균 나이는 $47.7{\pm}16.1$세, 남자가 38.5%이었으며, 평균 $FEV_1$은 $84.4{\pm}22.3%$이었다. MUC5AC 프로모터의 -1274G>A 유전자 다형성은 두 군간에 차이가 없었다(p=0.752, Cod). 천식 증상의 심한 정도, $FEV_1$, 총 호산구수, 혈청 IgE, $PC_{20}$, 아토피의 유무에 따라서도 MUC5AC 프로모터의 -1274G>A 유전자 다형성은 차이가 없었다. 결론: MUC5AC 프로모터의 -1274G>A 유전자 다형성은 천식과는 무관하였으며, 여러 가지 임상적인 지표들과도 상관관계를 보이지 않았다.
배 경 : MUG1은 고분자량의 당화 당단백으로 정상적으로 선상피의 선단부에서 발현되나, 종양세포에서는 비정상적인 발현 양상을 보인다. CD44는 세포-세포간 부착, 세포-기질간 부착에 관여하는 당단백이다. 일부 종양에서 MUC1 과 CD44는 종양의 침습과 전이에 관계한다고 알려져 있다. 본 연구에서는 비소세포폐암에서의 MUC1과 CD44의 standard form (CD44s)의 발현과 조직형, TNM-병기와의 상관성을 조사하였다. 대상 및 방법 : 수술적 방법으로 얻은 비소세포폐암 80예의 파라핀포매 조직절편을 대상으로 하여 MUC1과 CD44s에 대한 면역조직화학염색을 시행하였다. 결 과 : MUC1 은 편평상피암종의 12/43예 (27.9%), 선암종의 12/37예 (32.4%)에서 양성으로 조직형에 따른 차이는 보이지 않았다(p=0.660). CD44s는 편평상 피암종의 36/43예(83.7%), 선암종의 14/37예 (37.8%)에서 양성으로 선암종에 비해 편평상피암종에서 발현이 유의하게 높았다(p<0.001). 편평상피암종에서 TNM-병기에 따른 MUC1의 양성율은 통계적으로 유의한 차이를 보이지 않았다. 선암종에서 MUC1 양성은 N0에서 4/22예(18.2%), N1-2에서 8/15예 (53.3%)로 림프절 침범과 관련되었다(p=0.036). 편평상피암종에서 CD44s의 발현은 T-병기에 따른 의미있는 차이는 없었으나, N0기에서 16/21예 (72.7%), N1-2기에서 9/22예 (38.1%)로 CD44s의 발현의 감소는 림프절 침범과 관련되었다(p=0.031). 편평상피암종의 TNM-병기 I기에서 15/18예(83.3%), II-III기에서 10/25예 (40.0%)에서 CD44s 양성으로 CD44s 발현의 감소는 TNM-병기의 진행과 관련되었다(p=0.006). 결 론 : 비소세포폐암에서 MUC1과 CD44s는 종양의 조직형에 따라 다른 발현 양상을 보이고, 각각은 종양의 침습과 전이에 관여하는 것으로 보인다.
Phenotypic changes after plasmid curing experiment suggested that the bacteriocin production phenotype ($Bac^{+}$) might be linked to a chromosomal DNA and the mucin production phenotype ($Muc^{+}$) might be linked to a 62.5 kilobase (kb) plasmid (pMUC62) in Lactobacillus plantarum 27 isolated from meat starter culture. The non-mucoid ($Muc^{-}$) variants were missing pMUC62 but they produced bacteriocin as the wild strain ($Bac^{+}$). There was no difference in antibiotic resistance and sugar fermentation patterns between the wild strain ($Bac^{+}$$Muc^{+}$) and the nonmucoid ($Bac^{+}$$Muc^{-}$) variants. Antimicrobial spectrum of bacteriocin produced by both wild strain and $Muc^{-}$ variant of Lb. plantarum 27 included strains of Pediococcus acidilactici (A, M, H), Pediococcus sp. isolated from meat, Lactobacillus sp. isolated from meat, Lb. plantarum NCDO 955 and Staphylococcus aureus 485. Neither of the tested Gram negative bacteria were inhibited by bacteriocin. Antimicrobial activity of crude bacteriocin was retained after autoclaving, DNase or catalase treatment and exposure from pHs 4 to 9 but was lost after treating with several proteolytic enzymes and exposure at pH 10.
In this study, we investigated whether adenosine, adenine, uridine and homogentisic acid derived from Pinellia ternata affect the secretion, production and gene expression of MUC5AC mucin from airway epithelial cells. Confluent NCI-H292 cells were pretreated with adenosine, adenine, uridine or homogentisic acid for 30 min and then stimulated with PMA (phorbol 12-myristate 13-acetate) for 24 h. The MUC5AC mucin gene expression, mucin protein production and secretion were measured by RT-PCR and ELISA, respectively. The results were as follows: (1) Adenine and homogentisic acid decreased PMA-induced MUC5AC mucin gene expression, although adenosine and uridine did not affect the mucin gene expression; (2) Adenosine, adenine, uridine and homogentisic acid inhibited PMA-induced MUC5AC mucin production; (3) Homogentisic acid inhibited the secretion of MUC5AC mucin from NCI-H292 cells. These results suggest that, among the four compounds examined, homogentisic acid showed the regulatory effect on the steps of gene expression, production and secretion of mucin, by directly acting on airway epithelial cells.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) induces coronavirus disease 2019 (COVID-19) and may increase the risk of adverse outcomes in lung cancer patients. In this study, we investigated the expression and function of mucin 1 (MUC1) after SARS-CoV-2 infection in the lung epithelial cancer cell line Calu-3. MUC1 is a major constituent of the mucus layer in the respiratory tract and contributes to pathogen defense. SARS-CoV-2 infection induced MUC1 C-terminal subunit (MUC1-C) expression in a STAT3 activation-dependent manner. Inhibition of MUC1-C signaling increased apoptosis-related protein levels and reduced proliferation-related protein levels; however, SARS-CoV-2 replication was not affected. Together, these results suggest that increased MUC1-C expression in response to SARS-CoV-2 infection may trigger the growth of lung cancer cells, and COVID-19 may be a risk factor for lung cancer patients.
연구배경 : 만성폐쇄성폐질환에서 나타나는 기도점액의 과다분비는 이 질환의 중요한 병리학적 소견이며 호흡곤란 등 환자의 증상을 악화시키는 요인 중의 하나이다. 기도 점액을 구성하는 여러 성분 중 Muc 유전자에 의해 만들어지는 당 단백이 흡연에 의해 생성이 증가하는데 이에 관여하는 세포 내 신호전달 과정에 대하여 확실히 밝혀진 바가 없다. 저자는 Muc 유전자 중 인체의 기도에 가장 많이 분비되는 Muc5ac 점액 생성을 담당하는 Muc5ac 유전자의 발현이 흡연에 의하여 증가하는데 관여하는 세포 내 신호전달 과정을 알아보고자 하였다. 재료 및 방법 : 사람 폐선암 세포주인 A549 세포를 배양하여 Muc5ac 유전자의 promotor를 luciferase reporter plasmid를 사용하여 세포 내에 transfection시키고 5% 담배연기 추출물로 자극하여 배양하였다. 또 세포 내 신호전달에 관여하는 표피성장인자 수용체 kinase의 억제제인 AG1478, mitogen-activated protein kinase kinase(MAPKK) 억제제인 PD98059, p38 mitogen-activated protein kinase 억제제인 SB203580으로 각각 전 처치 후 역시 5% 담배연기 추출물로 자극 배양하였다. 배양된 세포에서 단백질을 추출하여 luciferase 분석을 통하여 Muc5ac promoter 활성도를 측정하고 Western blot을 이용하여 표피성장인자 수용체와 mitogen-activated protein kinase (MAPK)인 extracellular signalrelated kinase (ERK)1/2, p38 MAPK, c-Jun N-terminal kinase (JNK)의 발현을 확인하였다. 또 세포에서 RNA를 추출한 후 Muc5ac primer를 이용하여 역전사효소 중합연쇄반응을 수행하여 Muc5ac mRNA 발현을 관찰하였다. 결 과 : 1. Muc5ac promoter를 삽입한 A549 세포를 5% 담배연기 추출물로 자극하였을 때 의의 있게 luciferase 활성도가 증가하였고(P<0.001) 자극하는 시간이 3시간이었을 때 luciferase 활성도가 최고치를 보였다(P<0.01). 또 담배연기 추출물 자극은 표피성장인자 수용체를 인산화시켰으며 인산화는 AG1478과 PD98059에 의하여 억제되었다. 2. AG1478 혹은 PD98059로 전 처치 후 5% 담배연기 추출물로 자극한 경우 5% 담배연기 추출물 단독으로 자극한 것에 비하여 유의하게 lucifearse 활성도가 억제되었고 (P<0.01) 세 가지 종류의 MAPK 중 ERK1/2와 p38 MAPK의 인산화는 관찰되었으나 JNK의 인산화는 관찰되지 않았다. 역전사효소 중합연쇄반응을 이용하여 관찰한 Muc5ac mRNA 발현은 담배연기 추출물에 의해 증가되었고 PD98059와 AG1478에 의하여 역시 억제되었다. 3. 담배연기 추출물에 의하여 인산화 된 ERK1/2는 PD98059에 의하여 인산화가 감소하였고 p38 MAPK의 인산화는 PD98059와 SB203580에 의하여 감소하였으며 이 두 가지 억제제는 모두 luciferase 활성도를 유의하게 억제시켰다(P<0.0001). 결 론 : 담배연기 추출물은 Muc5ac 유전자의 발현을 증가시켜 기도 내 점액 분비를 증가시키며 이는 표피성장인자 수용체를 매개로 ERK1/2와 p38 MAPK를 경유하여 세포 내 신호전달이 이루어진다고 생각된다. 따라서 점액 유전자 활성화를 매개하는 신호전달 과정을 차단하는 약제나 방법이 개발된다면 과도한 점액분비를 치료할 수 있을 것으로 생각한다.

제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
