Expression of Membrane-Type Matrix Metalloproteinase 1 and 2 in Mouse Oocytes, Embryos, Ovary and Oviduct (생쥐 난자와 배아 및 난소와 수란관의 Membrane-Type Matrix Metalloproteinase 1 및 2의 유전자 발현)

  • Membrane type matrix metalloproteinases(MT-MMPs) have been suggested to play an important role during structural remodeling of various tissue. Expression patterns of MT1-MMP and MT2-MMP mRNAs were investigated in oocytes, embryos, ovary and oviduct of mouse during their differentiation or periovulatory period using RT-PCR technique. Both cDNA products of MT1- and MT2-MMP of immature oocytes were barely discernable with a minimum amount but the expressions were distinct in mature oocytes regardless that they were matured in vivo or in vitro. MT2-MMP was not expressed by 2-cell embryos but was expressed by 4-cell stage embryos. From the morula stage untill hatched blastocyst stage, embyos showed intesnse expression of MT2-MMP with a sudden increase at blastocyst stage. While mouse ovarian tissues showed both expression of MT1- and MT2-MMP, there was no stage-specific difference throughout the estrous cycle. Mouse oviducts also exhibit constant amount of both MT1- and MT2-MMP expressions throughout periovulatory period, i.e., before or after ovulation. These observations lead to suggest that the differential expressions of maternal MT1- and MT2-MMP during meiotic resumption of mouse oocytes and embryonic expression of MT2-MMP particularly at blastocyst stage might play a role in the differentiation of mouse oocytes and/or embryos. The precise function of MT1- and MT2-MMP with regards to their participation in the remodeling of ovarian and oviductal tissues remains in a question.

pT7MT, a Metallothionein 2A-Tagged Novel Prokaryotic Fusion Expression Vector

  • In the present article, a novel fusion expression vector for Escherichia coli was developed based on the pTORG plasmid, a derivative of pET32a. This vector, named pT7MT(GenBank Accession No DQ504436), carries a T7 promoter and it drives the downstream gene encoding Metallothionein 2A(MT2A). There are in-framed multiple cloning sites(MCS) downstream of the MT2A gene. A target gene can be cloned into the MCS and fused to the C-terminal of the MT2A gene in a compatible open reading frame(ORF) to achieve fusion expression. The metal-binding capability of MT2A allows the purification of fusion proteins by metal chelating affinity chromatography, known as $Ni^{2+}$-affinity chromatography. Using this expression vector, we successfully got the stable and high-yield expression of MT2A-GST and MT2A-Troponin I fusion proteins. These two proteins were easily purified from the supernatant of cell lysates by one-step $Ni^{2+}$-affinity chromatography. The final yields of MT2A-GST and MT2A-Troponin I were 30mg/l and 28mg/l in LB culture, respectively. Taken together, our data suggest that pT7MT can be applied as a useful expression vector for stable and high-yield production of fusion proteins.

  • Thirty nine samples of striped field mice (Apodemus agrarius coreae Thomas) from eight localities in the Korean peninsula were used for the analyses of mitochondria1 DNA (mtDNA) fragment patterns resulted from the digestion with eight restriction enzymes. A total of 31 fragments were recognized and seven mtDNA clones were revealed: one clone consisted of 32 among 39 samples from eight localities (1 of 1 from Sogcho, 4 of 5 from Mt. Chiak, 3 of 3 from Mt. Weolak, 2 of 2 from Mt. Sogri, 2 of 2 from Mt. Deokyoo, 3 of 4 from Mt. Jiri, 2 of 4 from Haenam, and 15 of 18 from Cheongju). The nucleotide-sequence divergences (p) among seven mtDNA clones ranged from 0.2% to 2.3% and distinct subgroups were not resulted from the grouping of these clones. It is confirmed that striped field mcie from the Korean peninsula is a single subspecies of Apodemus agrarius (A. agrarius coreae) because they were not divided into separate subgroups in their mtDNA genotypes.

Isolation and mRNA Expression of Metallothionein Isoforms from Rockbream Oplegnathus fasciatus (돌돔(Oplegnathus fasciatus)으로부터 Metallothionein 유전자 Isoform들의 분리 및 발현 특징 분석)

  • Genetic determinants of two metallothionein isoforms (MT-A and MT-B) were isolated and characterized from the perciform species, rockbream (Oplegnathus fasciatus). Rockbream MT-A and MT-B shared a high degree of homology at amino acid levels with representative orthologs from other perciform species, especially with respect to the conserved cysteine residues. At the genomic level, both MT-A and MT-B genes represent a tripartite structure typical of vertebrate MT genes. However, rockbream MT-B showed unusually large introns (1.2 kb and 0.8 kb for intron I and II, respectively), a phenomenon that has rarely been seen in other vertebrate MT genes. MT-A and MT-B transcripts were ubiquitously detected in a wide array of tissues, wherein brain and eye showed the highest basal expression levels, and the fin exhibited the lowest expression of both isoforms. The basal expression of MT-A in most tissues was significantly higher (ranging from 4- to 10-fold) than that of MT-B. Upon heavy metal exposures to Cd, Cu or Zn at 25 ppb for 48 h, MT-A and MT-B transcripts in the liver were significantly activated by Cd and moderately by Zn. On the other hand, exposure to Cu did not result in alterations of MT-A, nor in the significant suppression of MT-B. Following bacterial challenges with Escherichia coli, Edwardsiella tarda or Streptococcus iniae, MT isoforms in the liver, kidney and spleen were highly modulated and exhibited a pattern that was dependent on the bacterial species, tissues and isoforms. These results suggest that the two MT isoforms could be taken into account as potential indicators of metal toxicity and immune perturbations of this aquaculture-relevant species.

Solubility and Stability of Melatonin in Propylene glycol and 2-hydroxypropyl-${\beta}$-cyclodextrin vehicles

  • The physicochemical properties of melatonin (MT) in propylene glycol (PG) and 2-hydroxypropyl-.betha.-cyclodextrin $(2-HP{\beta}CD)$ vehicles were characterized. MT was endothermally decomposed as determined by differential scanning calorimetry (DSC). Melting point and heat of fusion obtained were $116.9{\pm}0.24^{\circ}C $.and $7249{\pm}217 cal/mol$., respectively. MT as received from a manufacture was very pure, at least 99.9%. The solubility of MT in PG solution increased slowly until reaching 40% PG and then steeply increased. Solubility of MT increased linearly as concentration of $2-HP{\beta}CD$ without PG INCREASED$(R^2=0.993)$. MT solubility in the mixtures of pg and $2-HP{\beta}CD$ also increased linearly but was less than the sum of its solubility in $2-HP{\beta}CD$ and PG individually. The MT solubility was low in water, simulated gastric or intestinal fluid but the highest in the mixture of PG(40v/v%) and $2-HP{\beta}CD$ (30w/v%) although efficiency of MT solubilization in $2-HP{\beta}CD$ decreased as the concentration of PG increased. MT was degraded in a fashion of the first order kinetics $(r^2>0.90)$. MT was unstable in strong acidic solution (HCl-NaCl buffer, pH 1.4) but relatively stable in other pH values of 4-10 at $70^{\circ}C$. In HCl-NaCl buffer, MT in 10% PG was more quickly degraded and then slowed dpwm at a higher concentration. However, the degradation rate constant of MT in 2-HP.betha.CD was not changed significantly when compared to the water. The current studies can be applied to the dosage formulations for the purpose of enhancing percutaneous absorption or bioavailability of MT.

Effects of several herbs on the expression of MT1 and MT2 melatonin receptors in C6 glial cells (수종의 한약재 열수추출물이 멜라토닌 수용체 발현에 미치는 영향)

  • Objective : This study was to investigate the effects of several herbs on the levels of MT1 and MT2 melatonin receptors Methods: It was investigated the effects of several herbs such as WEDL, WEZV, WEFO, WEOC on the levels of MT1 and MT2 melatonin receptors using C6 glial cell model. ${\beta}-estradiol$ treatment, as a positive control group, under non-cytotoxic condition. Results : 1. The water extracts of Dimocarpus long (WEDL) induced the levels of MT2 melatonin receptor expression in a concentration-dependent manner without altering the levels of MT1 melatonin receptor expression. 2. The treatment with the water extract of Zizyphus vulgaris (WEZV) induced the levels of MT1 melatonin receptor expression and the levels of MT2 melatonin receptor expression was not affected. 3. The levels of MT1 as well as MT2 melatonin receptor expression were markedly up-regulated in the water extract of Fossilia ossis (WEFO) and the water extract of Ostreae caro (WEOC)-treated C6 cells. 4. The combination treatment with WEDL and WEZV induced not only the levels of MT1 melatonin receptor expression but also MT2 melatonin receptor expression, but the synergic effects of the combination treatment with WEFO and WEOC were not detected in C6 cells. Conclusion : The study provides important new insights into the possible mechanisms on the regulation of melatonin receptor synthesis by WEDL, WEZV, WEFO and WEOC.

Investigation of the efficacy of mycotoxin-detoxifying additive on health and growth of newly-weaned pigs under deoxynivalenol challenges

  • Objective: This study evaluated the effects of feeding diets naturally contaminated with deoxynivalenol (supplemental 2 mg/kg) on health, growth, and the effects of a mycotoxin-detoxifying additive in newly-weaned pigs. Methods: Thirty-six pigs (27 day-old) were housed individually and assigned to 3 treatments for 5 weeks: CON (diet containing minimal deoxynivalenol), MT (diet with supplemental 1.9 mg/kg of deoxynivalenol), and MT+D (MT + mycotoxin-detoxifying additive, 0.2%, MegaFix, ICC, São Paulo, Brazil). The mycotoxin-detoxifying additive included bentonite, algae, enzymes, and yeast. Blood was taken at week 2 and 5. Jejunal tissue were taken at week 5. Data were analyzed using the MIXED procedure of SAS. Results: Pigs fed MT+D tended to have decreased (p = 0.056) averaged daily feed intake during week 1 than MT. At week 2, serum aspartate aminotransferase/alanine aminotransferase in MT tended to be lower (p = 0.059) than CON, whereas it was increased (p<0.05) for MT+D than MT, indicating hepatic damages in MT and recovery in MT+D. Pigs fed MT had lower (p<0.05) blood urea nitrogen/creatinine than CON, supporting hepatic damage. At week 5, pigs fed MT tended to have reduced (p = 0.079) glucose than CON, whereas it was increased (p<0.05) for MT+D than MT, indicating impaired intestinal glucose absorption in MT, which was improved in MT+D. Pigs fed CON tended to have increased (p = 0.057) total glutathione in jejunum than MT, indicating oxidative stress in MT. Pigs fed MT+D had a reduced (p<0.05) proportion of Ki-67-positive cells in jejunum than MT, indicating lower enterocyte proliferation in MT+D. Conclusion: Feeding supplemental 1.9 mg/kg of deoxynivalenol reduced growth and debilitated hepatic health of pigs, as seen in leakage of hepatic enzymes, impaired nitrogen metabolism, and increase in oxidative stress. The mycotoxin-detoxifying enhanced hepatic health and glucose levels, and attenuated gut damage in pigs fed deoxynivalenol contaminated diets.

Ecological Characteristic of Abies koreana Stand Structure of Mt. Jirisan and Mt. Hallasan (지리산과 한라산의 구상나무 임분 구조의 생태적 특성)

  • This study compared the stand structure of Abies koreana forests between Mt. Jirisan and Mt. Hallasan by analyzing DBH distribution, stem vitality, crown structure, importance value, species diversity, and DCA. The data were acquired through the national long-term ecological research project conducted by the Ministry of Environment. In the comparison of DBH class distribution, the stem density of A. koreana with DBH < 20 cm was higher in Mt. Hallasan than that in Mt. Jirisan; however, the stem density of A. koreana with DBH > 20 cm was higher in Mt. Jirisan than that in Mt. Hallasan. For A. koreana stem vitality, the ratio of alive standing was higher in Mt. Jirisan (72.0%) than in Mt. Hallasan (60.7%), whereas the ratio of alive leaning was higher in Mt. Hallasan (10.2%) than in Mt. Jirisan (1.1%). A. koreana stand in Mt. Jirisan was a mature stand with four evenly developed and distinct layers, whereas the tree layer in Mt. Hallasan was less than 10 m and the stand had three layers without a subtree layer. Mt. Hallasan's importance value of A. koreana was 39.4% higher than Mt. Jirisan's importance value of 26.6%. The species diversity of the A. koreana community of Mt. Jirisan was 2.52 times higher than that of Mt. Hallasan, which was 1.58. DCA results revealed that the average distance between Mt. Jirisan and Mt. Hallasan was relatively distinguished, and the degree of scattering of species composition of Mt. Jirisan was relatively dense compared with that of Mt. Hallasan.

Modification of cell wall structural carbohydrate in the hybrid poplar expressing Medicago R2R3-MYB transcription factor MtMYB70

  • The isolation, cloning, and characterization of an R2R3-MYB transcription factor gene (MtMYB70) from the model legume Medicago truncatula is reported. MtMYB70 consists of a 768-bp coding sequence corresponding to 255 amino acids. Sequence alignment revealed that MtMYB70 cDNA contains conserved R2R3-type MYB domains with highly divergent C terminal regions. MtMYB70 was found to have relatively low sequence homology with known R2R3-MYB genes. Phylogenetic analysis placed the R2R3-MYB domain of MtMYB70 closest to PtMYB1, a known activator of lignin biosynthesis. Overexpression of MtMYB70 under the control of the 35S promoter in transgenic poplar did not cause a significant difference in total lignin content relative to the control, but glucan content was significantly increased in transgenic poplar. Therefore, MtMYB70 might have regulatory role in the biosynthesis of cell wall structural carbohydrates.

Transcriptional Alteration of Two Metallothionein Isoforms in Mud Loach (Misgurnus mizolepis) Fry during Acute Heavy Metal Exposure

  • Altered mRNA expression of two metallothionein isoforms (MT-IA and MT-IB) in response to acute heavy metal exposure was examined in mud loach, Misgurnus mizolepis, fry using a real-time RTPCR assay. Sublethal exposure (1 or 5 ${\mu}M$) to Cd, Cr, Fe, Mn, Ni, and Zn resulted in highly variable transcriptional responses of the two MT isoforms to the heavy metal ions, including upregulation, a steady state, and downregulation. Overall, the most potent inducer of both MT isoforms was Cd (up to 6-fold). Another exposure experiment using a series of doses of Cu revealed that the stimulation patterns of the two MT isoforms differed: MT-IA transcription was soon saturated at higher concentrations (about 2-fold at 1-4 ${\mu}M$ of Cu), whereas the activation of MT-IB was more dependent on the treatment dose (increased up to 5-fold at 3 ${\mu}M$). The isoform-specific allotment of constitutive and inducible functions was not as clear in fry as in adult tissues. Coordinated interaction between the MT-IA and MT-IB isoforms was hypothesized based on the finding that MT-IA represented a primary action under 'less stressful' or 'sublethal' conditions, whereas the activation of MT-IB became important under 'more stressful' or 'lethal' circumstances in this species.
