The effects of salt are studied on control of weeds and growth of Korean turfgrass(Zoysia japon-ica Steud.). And the mobility of salt were determined in a lawn field. At post-emergence stage, the growth of the 13 weeds such as Trifolium repens, Erigeron canadensis, Artemisia prieceps, Equi-setum arvense, etc were controlled by treatment of salt and salt water, Pea-annua and Equisetum arvense were tolerant to salt compared with the other 11 weeds. At pre-emergence stage, the salt controlled the germination of the weeds in soil. 300~500 kg /10a salt and 20~30 % salt waterwere enough for the control of weeds, However, salt water injuried the lawngrass except the dormancy stage. The mobility of salt in soil was increased by rainfall. After 160mm of rainfall, the salinity in the soil treated with salt 500kg /l0a was below 0.3 ms/cm in surface soil and about 0.1 ms /cm in soil 30 cm depth. 식물 세포나 조직의 배양에서 배지의 salt strength, sucrose 농도, 질소 함량과 질소원의 종류 및 그 비율 등이 세포 또는 조직의 생장에 영향을 미친다. 본 연구에서는 Agar를 함유한 MS 기본배지와 BA 1 mg/L를 첨가한 MS 기본 액체배지에서 계대배양을 통하여 생산한 다신초를 액아와 본엽이 $2{\sim}3$매 되도록 분할한 신초를 이용하여 MS 액체배지의 salt strength와 sucrose 농도 및 $NH_4/NO_3$ 비율이 고추냉이 신초의 생육에 미치는 영향을 조사하여 고추냉이 유묘의 생산에 필요한 최적 배지조건을 구명하였다. 고추냉이 신초의 증식에는 MS 액체배지의 salt strength는 기본조성이 가장 적합한 것으로 나타났으며, $1/4{\sim}1/2x$ 또는 2x MS 배지에서는 고추냉이 유묘의 생장이 크게 억제 되었다. 기본조성의 MS 액체배지에 sucrose 3%를 첨가하였을 때 고추냉이 신초의 생장이 가장 우수한 것으로 나타났으나 sucrose 농도 $1{\sim}6%$ 범위에서 신초의 수와 길이는 유의성 있는 차이를 보이지 않았다. 기본조성의 MS 액체배지에서 질소원으로 ${NH_4}^+$과 ${NO_3}^-$의 비율을 10:50으로 조절하였을 때 고추냉이 신초의 생장이 가장 우수한 것으로 나타났으며, ${NH_4}^+$의 비율이 증가할수록 생장은 억제되었다.
Deionized water, methanol, and ethanol were investigated for their effectiveness at dissolving LiCl-KCl-UCl3 at 25, 35, and 50℃ using inductively coupled plasma mass spectrometry (ICP-MS) to study the concentration evolution of uranium and mass ratio evolutions of lithium and potassium in these solvents. A visualization experiment of the dissolution of the ternary salt in solvents was performed at 25℃ for 2 min to gain further understanding of the reactions. Aforementioned solvents were evaluated for their performance on removing the adhered ternary salt from uranium dendrites that were electrochemically separated in a molten LiCl-KCl-UCl3 electrolyte (500℃) using scanning electron microscopy with energy dispersive spectroscopy (SEM-EDS). Findings indicate that deionized water is best suited for dissolving the ternary salt and removing adhered salt from electrodeposits. The maximum uranium concentrations detected in deionized water, methanol, and ethanol for the different temperature conditions were 8.33, 5.67, 2.79 μg·L-1 for 25℃, 10.62, 5.73, 2.50 μg·L-1 for 35℃, and 11.55, 6.75, and 4.73 μg·L-1 for 50℃. ICP-MS analysis indicates that ethanol did not take up any KCl during dissolutions investigated. SEM-EDS analysis of ethanol washed uranium dendrites confirmed that KCl was still adhered to the surface. Saturation criteria is also proposed and utilized to approximate the state of saturation of the solvents used in the dissolution trials.
소금의 이산화황 발생 원인을 구명하기 위하여 갯벌천일염 (MSS)과 구운 소금(RS)의 시간 경과(RS1, RS2, RS3 및 RS4)에 따른 일반성분, 중금속, 무기질 함량을 분석하고 이산화황 발생 및 각 소금의 환원력을 비교 분석하였다. 이산화황과 아황산은 모두 검출되지 않았으며, 황산이온은 MSS 및 RS에서 각각 35,601.65 ppm과 29,878.15~36,097.45 ppm으로 시간이 지날수록 감소되는 경향을 나타내었다. ORP는 MSS(181.15 mV)에서 가장 낮았고 RS1(58.55 mV)에서 가장 높았다. 수분은 MSS가 9.34%였으나 RS에서 크게 감소되었으며, RS의 NaCl은 94.77~95.77%로 증가되었다. RS 시간 경과에 따라 수분과 염도에 차이는 없었다. 불용분과 사분은 MSS에 비해 RS에서 더 높은 함량을 나타내었고 Ca, K, Mg는 유의한 차이를 나타내지 않았다. MSS에 비해 RS의 Cl(556,487.5~612,305.0 ppm) 함량이 높았으며, Br은 MSS(628.1 ppm)에서 더 높은 함량을 나타내었으나 $NO_3$는 모두 유의한 차이를 보이지 않았다. MSS에 비해 RS에서 Pb, As, Hg가 높게 나타났으나 Pb, As, Cd, Hg에서 기준치 이상의 검출은 없었다. MSS와 RS의 Co, Cu, Se, U는 차이가 없었으나 Li, Al, Mn, Fe, Sr은 RS에서 더 높은 함량을 나타내었으며, Mn은 굽는 시간 경과에 따라 감소하는 경향을 나타내었다. 이상의 결과에서 볼 때 MSS와 RS에서 인체에 유해한 영향을 미치는 이산화황 발생은 없었으며, RS 시간 경과에 따른 이산화황 발생도 없는 것으로 나타났다. 중금속의 위해성은 안전 수준이며, 이외 발생될 수 있는 잠재적인 위해성에 대한 관심이 필요할 것으로 판단된다.
In order to improve physico-chemical properties and to enhance stability of drugs, amino acid salt has been widely adoptd in pharmaceutical synthesis. Acetylsalicylic acid lysinate is one of the widely used analgesics and it is a good example of t5his synthesis. In the case of bacetylsalicylic acid lysinate synthesis, racemization of natrually occurred lysine is esential because the racemic lysine salt of the drug shows better yield, crystallinity and dryness than that of the L-lysine salt. To esatablish a simple, practical and economical process for L-lysine racemization, L-lysine treatments with phosphoric acid and with acetic acid were compared and the optimum conditions for its process and derivatization were investigated by chiral separation methods using GC_MS spectroscopy.
Salt stress is one of the major abiotic stresses affecting the yield of ginseng (Panax ginseng C. A. Meyer). The objective of this study was to identify bio-marker, which is early responsive in salt stress in ginseng, using proteomics approach. Ginseng plants were exposed to 5 ds/m salt concentration and samples were harvested at 0, 6, 12 and 18 hours after exposure. Total proteins were extracted from ginseng leaves treated with salt stress using Mg/NP-40 buffer and were separated on high resolution 2-DE. Approximately $1003{\pm}240$ (0 h), $992{\pm}166$ (6 h), $1051{\pm}51$ (12 h) and $990{\pm}160$ (18 h) spots were detected in colloidal CBB stained 2D maps. Among these, 8 spots were differentially expressed and were identified by using MALDI-TOF/TOF MS or/and LC-MS/MS. Ethylene response sensor-1 (spot GL 1), nucleotide binding protein (spot GL 2), carbonic anhydrase-1 (spot GL 3), thylakoid lumenal 17.9 kDa protein (spot GL 4) and Chlorophyll a/b binding protein (spot GL 5, GL 6) were up-regulated at the 12 and 18 hour, while RuBisCO activase B (spot GL 7) and DNA helicase (spot GL 8) were down-regulated. Thus, we suggest that these proteins might participate in the early response to salt stress in ginseng leaves.
The influence of carrageenan addition on the gelatinization of salt-based surimi gels was investigated by measuring compressive properties and Hunter color scale values. Hydrocolloid kappa (${\kappa}$)-carrageenan at 0%, 0.2%, and 1.0% and NaCl (2% w/w), KCl (1.5% w/w), and a mixture of NaCl (2% w/w) and KCl (1.5% w/w), were added to Alaska pollock surimi. Gel compressive properties were assessed by measuring the breaking force and gel strength. The gelling property of ${\kappa}$-carrageenan-induced surimi gel was significantly increased by the incorporation of KCl rather than NaCl. The addition of ${\kappa}$-carrageenan increased the breaking force and gel strength of surimi gels. Gels with 1% ${\kappa}$-carrageenan and KCl had the highest breaking force and gel strength. The addition of ${\kappa}$-carrageenan caused an increase in the whiteness values of the surimi gels.
Lilium oriental hybrid 'Casa Blanca'의 기내인편에서 자구를 대량생산하기 위하여 MS배지의 염류농도 및 당, 광조건 등이 자구의 형성 및 비대에 미치는 영향을 조사하였다. MS배지의 1/2~2배 염류농도에서 자구의 형성은 차이를 나타내지 않았으나 3배 염류농도에서 자구의 형성이 현저히 억제되었다 자구의 비대는 MS기본배지에서 가장 촉진되었다. 배지의 NH$_4$$^{+}$ : NO$_3$의 비율이 1 : 2와 1 : 3일 때, 또는 MS기본배지 (NH$_4$$^{+}$ : NO$_3$= 약 1 : 2)가 인편에서 자구의 형성 및 비대에 적절하였다. 따라서 MS 기본배지가 자구의 형성 및 비대에 양호한 것으로 나타났다. 당의 농도가 높아지면 자구형성은 억제되고 자구비대는 촉진되었으며, 당 9~12% 첨가배지에서 자구비대가 양호하였다. 배지내 활성탄의 첨가는 인편에서 자구의 형성을 억제하였으나 형성된 자구의 비대는 촉진하였다. 특히 활성탄 1 g/L를 첨가한 배지가 자구의 비대에 가장 효과적이었다. 자구의 형성 및 형성된 자구의 비대는 명, 암간에 차이를 나타내지 않았다. 따라서 L. Oriental Hybrid 'Casa Blanca'의 기내 대량번식은 당 9%와 활성탄 1 g/L가 첨가된 MS기본배지에서 기내인편을 배양하여 자구를 형성 및 비대시키고 형성된 자구의 인편을 계대배양하는 것이 효율적이었다.
An efficient transformation system for high-throughput functional genomic studies of kiwifruit has been developed to overcome the problem of necrosis in Actinidia arguta explants. The system uses Agrobacterium tumefaciens strain EHA105 harbouring the binary vector pART27-10 to inoculate leaf strips. The vector contains neomycin phosphotransferase (nptII) and ${\beta}$-glucuronidase (GUS) (uidA) genes. A range of light intensities and different strengths of Murashige and Skoog (MS) basal salt media was used to overcome the problem of browning and/or necrosis of explants and calli. Callus browning was significantly reduced, resulting in regenerated adventitious shoots when the MS basal salt concentration in the culture medium was reduced to half-strength at low light intensity ($3.4\;{\mu}mol\;m^{-2}\;s^{-1}$) conditions. Inoculated leaf strips produced putative transformed shoots of Actinidia arguta on half-MS basal salt medium supplemented with 3.0 $mg\;l^{-1}$ zeatin, 0.5 $mg\;l^{-1}$ 6-benzyladenine, 0.05 $mg\;l^{-1}$ naphthalene acetic acid, 150 $mg\;l^{-1}$ kanamycin and 300 $mg\;l^{-1}$$Timentin^{(R)}$. All regenerated plantlets were deemed putativ transgenic by histochemical GUS assay and polymerase chain-reaction analysis.
헤드스페이스/가스크로마토그래프/질량분석계(HS/GC/MS)을 이용하여 환경 시료 중 폐기물 시료인 슬러지 중의 휘발성 유기화합물을 분석하였다. 본 연구에서는 헤드스페이스를 이용한 휘발성 물질의 분석에서 염(salt)의 첨가 유무, 평형온도 및 평형시간 등이 미치는 영향을 파악하기 위해서 수행되었다. NaCl을 첨가한 경우와 첨가하시 않은 경우 이들 분석물질의 회수율 변화는 NaCl을 넣었을 때 1,2-dibromo-3-chloropropane이 2.61배로 가장 큰 회수율의 변화를 보였으며, trlchlorofluoromethane이 1.07배로 가장 적은 변화를 나타내었다. 또한, 평형온도는 $80^{\circ}C$에서 $90^{\circ}C$(30분)로 증가됨에 따라 피이크의 면적이 크게 증가한 것으로 나타났으며, 평형도달 시간은 30분($85^{\circ}C$)이 최적 조건으로 조사되었다. 또한, 55종의 분석대상물질의 재현성도 매우 높게 나타나, 이 방법을 이용한 폐기물 시료중의 휘발성 유기화합물의 분석이 가능하리라 판단되어진다.
