Bacterial lysates have become a common ingredient for natural health care. Lactic acid bacteria (LAB) could serve as potential candidates for lysate production: the lactic acids produced by LAB have been utilized for their moisturizing, antimicrobial, and rejuvenating effects, while other substances provide topical benefits and health effects for the skin. Our study aimed to obtain lysate from a LAB S. macedonicus MBF 10-2 through an optimized fermentation using the Response Surface Methodology. Strain MBF10-2 was cultivated in a 2L fermenter tank in de Man Rogosa and Sharpe (MRS) medium and in plant-based peptone modified MRS, i.e. Soy-peptone and Vegitone. The duration and the medium composition (dextrose and soy peptone or proteose peptone) were adjusted to obtain an optimum production of cell lysate. Central Composite Design was employed for Design Expert 7.0.0 by adjusting 3 factors: dextrose (1%, 1.5%, 2%, 2.5%, 3%), soy or proteose peptone (0.5%, 0.75%, 1%, 1.25% and 1.5%), and duration of fermentation (8, 10, 12 14, and 16 h for MRS-Soy peptone and 15, 17, 19, 21, and 23 h for MRS Vegitone). Bacteriocin-Like Inhibitor Substance activity of lysate and pH were used as indicators. The optimum condition for lysate production using MRS Soy Peptone and Vegitone are as follows: dextrose concentration 2.5%, plant-based peptone 1.25%, while optimum fermentation duration were 11.18 h (MRS Soy Peptone) and 17 h (MRS Vegitone) with a starter concentration of 10% at $OD_{600nm}$$0.2{\pm}0.05$. However, the standard MRS medium produced better quality lysate compared to MRS plant-based peptones.
The superoxide dismutase (SOD) activity of seven Bifidobacterium spp. strains was examined by an indirect SOD assay method. Some Bifidobacterium spp. showed significant levels of SOD activity. However, we could not observe any significant differences between anaerobic and aerobic cultures. Furthermore, although several Bifidobacterium spp. exhibited some degree of tolerance to paraquat which produces superoxide radicals, the apparent SOD activity of these strains was not correlated with their resistance to paraquat. In addition, when we added increasing amounts of manganese or iron to MRS medium which had been prepared without either of the metal ions, the apparent SOD activity of cell free extracts (CFEs) was increased with increasing concentration of both metal ions. To our surprise, the heat-denatured CFEs also showed nearly identical correlative patterns. Based on these results, the apparent SOD activity was likely due to a nonenzymatic dismutation. These results strongly suggest that high concentration of divalent metal ions ($Mn^{2+}$, $Fe^{2+}$) in MRS medium result in nonenzymatic dismutation which can lead to false positive SOD activities in Bifidobacerium spp.
The MRS medium is widely used as an optimized medium for the growth of Lactobacillus spp. and also used for the growth of Leuconostoc spp. Leuconostoc mesenteroides shows quite different physicochemical properties compared to Lactobacilli spp. and it is one of the major strain of kimchi fermenting microorganisms with its usefulness in our traditional foods and availability in biotechnology in the future, specifically tailor-made medium is necessary for the growth of Leuconostoc mesenteroides. Sequential experimental designs (Plackett-Burman, fractional factorial, steepest ascent, central composite design and response surface methodology) were introduced to optimize and improve the Leuconostoc medium. Fifteen medium ingredients were investigated and fructose, sodium acetate and ammonium citrate were determined to give a critical and positive effect for cell-growth. The yield of biomass using the optimal medium was improved more than that of the MRS medium and the result of fed-batch culture showed the capability of the improvement in cell mass similar to the E.coli system.
폐콘크리트 파쇄시 발생하는 재생잔골재의 품질을 검토하였으며, 재생잔골재를 활용한 모르타르의 압축강도, 휨강도 및 흡수율을 검토하였다. 또한, 프리캐스트 콘크리트 제품용 골재로서의 적용성에 대하여 평가하였다. 재생잔골재의 밀도 및 흡수율은 각각 $2.31\;g/cm^3$ 및 8.07%로 KS F 2573의 2종에 해당되는 품질이었다. 혼합시멘트 MRS1, MRS2 및 MRS3 사용 모르타르의 재령 28일 압축강도는 물-시멘트비 35%, 양생온도 $40^{\circ}C$의 조건에서 각각 15.8, 27.4 및 48.7MPa로 최대값을 발현하였다. 모르타르의 최대휨강도는 혼합시멘트 MRS1 및 MRS2 사용시 물-시멘트비 35.0%, 양생온도 $40^{\circ}C$, 혼합시멘트 MRS3사용시 물-시멘트비 37.5%, 양생온도 $40^{\circ}C$의 조건에서 발현하였다. 혼합시멘트 MRS1, MRS2 및 MRS3를 사용한 모르타르의 흡수율의 범위는 각각 $8.3{\sim}7.3%,\;6.5{\sim}8.5%$ 및 $3.5{\sim}6%$의 범위로 나타났다. 그러므로 혼합시멘트와 재생잔골재의 비율을 적절히 조절함으로써 MRS1는 저강도용, MRS2는 보통강도용, MRS3는 고강도용으로 다양한 프리캐스트 콘크리트 제품에 적용될 수 있을 것으로 기대된다.
For the utilization of animal blood produced in slaughter for the cultivation of lactic acid bacteria, the nitrogen sources in a complex(MRS) medium were replaced by blood plasma proteins. Focusing the purpose on the industrial production of a probiotics, the hydrolytic activities of three industrially applicable proteases were compared for the effective digestion of the proteins, and Alcalase(the product of Novo Nordisk) was selected with comparatively high activity. The growth of Streptococcus thermophilus KCCM12020 was best among the four strains of lactic acid bacteria tested. With Alcalase-digested proteins in the medium, the growth rates and the final cell concentrations were higher than those with non-digested proteins. The cell mass produced in the medium containing blood proteins as nitrogen sources, $2.5{\times}10^9$ CFU/ml, was significantly high and about 70% of that in MRS medium, showing a great possibility for the utilization of animal blood proteins as economic nitrogen sources in the production of cell mass of lactic acid bacteria.
Saccharide utilizations for the growth by Bifidobacterium longum and Bifidobacterium breve were compared. B. longum fermented glucose more rapidly than lactose as a carbon source whereas B. breve fermented lactose at a rate higher than that of glucose. The highest cell concentration, in the case of B. longum, was obtained when cultivated in a jar fermentor that contained modified MRS medium that half the beef extract was replaced by the same amount of tuna extract, and that pH was controlled at 6.0. B. breve showed the best growth when grown in a jar fermentor containing the MRS medium with lactose instead of glucose, controlled at pH 6.0. The optimal concentration of peptone in MRS medium for the growth of B. breve was 5 g/l.
The bacteriocin-producing lactic acid bacteria Enterococcus faecium DB1 was isolated from Korean traditional gajami sikhae. Culture conditions were optimized by response surface methodology (RSM) to maximize bacteriocin DB1 production. E. faecium DB1 displayed the highest bacteriocin activity when grown in modified MRS medium containing sucrose, rather than glucose, as a carbon source. The effects of temperature, initial pH, and sucrose concentration were tested to determine the optimum conditions for maximum bacteriocin production by E. faecium DB1. A central composite design was used to control the three variables in the experiment. RSM revealed that the optimum values for bacteriocin production were 27.66 g/L sucrose, temperature of $34.37^{\circ}C$, and an initial pH of 6.54. A 2.08-fold increase in bacteriocin production was obtained with sucrose-containing MRS medium compared to production in standard MRS medium.
Soybean curd whey(SCW) containing plenty of nutrients is the discarded by-product in soybean curd processing. To test the potential utilization of SCW as a medium for the cultivation of Lactobacilus acidophilus, the chemical composition of SCW, as well as the growth, acid production, acid-tolerance, and bile-tolerance of L. acidophilus in SCW-based media were investigated. Sucrose and stachyose, the main free sugars of SCW, were 0.42% and 0.41%, respectively. SCW contained 36.1mg/L of total free amino acids. L. acidophilus KFRI 150 showed lower cell growth and acid production in SCW than those in MRS broth. In optimized SCW-based medium supplemented with 1.0% glucose, 0.5% yeast extract, and 0.2% $K_{2}HPO_{4}$, the growth and acid production of L. acidophilus KFRI 150 increased by twice against those in SCW. In optimized SCW-based medium, the viable counts of four L. acidophilus strains were mostly at the level of $10^{9}$/ml, which is similar to those in MRS broth. Each acid-tolerance and biletolerance of four L. acidophilus strains cultured in optimized SCW-based medium and MRS broth showed no dist-inguishable difference.

B. longum, B. bifidum, B. adolescentis, B. animalis, B. auglutum, B. breve의 생육활성에 대한 중합도 4와 6의 영향을 검토하기 위하여 modified-MRS배지 상에 탄소원으로 중합도 4와 6를 대체하여 생육활성을 비교한 결과 B. longum에서는 D.P 6 galactomannooligosaccharide를 탄소원으로 대체한 경우 표준 MRS배지와 비교하여 10배의, D.P. 4을 처리한 경우에도 7.6배의 상대 활성을 나타내어 가장 우수한 생육활성을 나타냈었으며, B. bifidum의 경우에서도 D.P 6에서 6.6배, D.P 4에서 4.8배의 활성을 나타내었으며 B. animalis에 있어서는 D.P. 6의 경우 3.8배의 상대활성을 나타내었다. Bifidobacterium 7균주 모두에 대해서 중합도 6의 올리고당이 중합도 4의 올리고당보다 생육활성에 크게 기여하는 것으로 나타났다. 신규 MRS 배지의 제품화와 관련하여 가장 유리한 탄소원 대체소재는 Bacillus sp. 유래 galactosyl mannooligosaccharide 인 ${Gal^3}{M_4}$가 가장 유리한 소재로 결정되어 신규 배지의 scale-up 공정을 구축중에 있다.
