Inhibition of Enzymatic Browning of Crown Daisy by Maillard Reaction Products (마이얄반응생성물(Maillard reaction product)에 의한 쑥갓의 효소적갈면 억제)

  • The study was conducted to investigate the effect of Maillard reaction products (MRPs) on enzymatic browning of crown daisy (Chrysanthmum coronarium var. spatiosum). The MRPs prepared by heating various amino acid and sugar at $90^{\circ}C$ caused a strong inhibitory effect on crown daisy polyphenol oxidase (PPO, ${\sigma}$-diphenol oxygen oxidoreductase, EC 1.10.3.1). As the reaction time of the solution containing glycine and glucose increased at $90^{\circ}C$, the production of MRPs was increased, whereas the amounts of glycine and glucose were decreased. Accordingly, the inhibitory effect of crown daisy PPO activity by MRPs was increased as the amounts of synthesized MRPs were increased. The MRPs synthesized from the various amino acids and sugars significantly reduced the PPO activity, particularly MRPs prepared by glutamine and xylose. The Michealis-Menten constant value ($K_m$) of crown daisy PPO with catechol as a substrate was 22.0 mM, and MRPs were a noncompetitive inhibitor against crown daisy PPO.

  • Resource procurement is an important management area because cost of resource covers 40% of total construction project cost and resource delivery has direct relationship with project performance. Integration of cost provides various potentials for effective and efficient project control. This study investigates the usefulness of time in resource procurement management focused on materials. These days, construction projects have characterized manufacture because of industrialization and component. Therefore, application of systematic resource planning has been requested in the construction. There are many companies conducting procurement of resource on the web by applying MRP, ERP etc. in the construction. However, in applying them in the construction yet, there is obstruction. MRP has the character doing its function under accurate cost prediction of resource. But prediction of resource is difficult in industry mechanism of the construction. If accurate cost prediction of resource is possible in the construction, it will be expected to reduce cost of procurement of resource substantially by applying successful resource planning model in the manufacture. On the basis of recent current, the purpose of study is to present procurement of resource system that period observance of construction and minimization of stock is possible by reflecting accurate lead-time to apply proactive thought to be able to cope with alteration of construction schedule efficiently in analyzing resource planning of the construction site.

Cytotoxicity, Toxicity, and Anticancer Activity of Zingiber Officinale Roscoe Against Cholangiocarcinoma

  • Cholangiocarcinoma (CCA) is an uncommon adenocarcinoma which arises from the epithelial cells of the bile ducts. The aim of the study was to investigate the cytotoxicity, toxicity, and anticancer activity of a crude ethanolic extract of ginger (Zingiber officinale Roscoe) against CCA. Cytotoxic activity against a CCA cell line (CL-6) was assessed by calcein-AM and Hoechst 33342 assays and anti-oxidant activity was evaluated using the DPPH assay. Investigation of apoptotic activity was performed by DNA fragmentation assay and induction of genes that may be involved in the resistance of CCA to anticancer drugs (MDR1, MRP1, MRP2, and MRP3) was examined by real-time PCR. To investigate anti-CCA activity in vivo, a total of 80 OV and nitrosamine (OV/DMN)-induced CCA hamsters were fed with the ginger extract at doses of 1000, 3000, and 5000 mg/kg body weight daily or every alternate day for 30 days. Control groups consisting of 10 hamsters for each group were fed with 5-fluorouracil (positive control) or distilled water (untreated control). Median $IC_{50}$ (concentration that inhibits cell growth by 50%) values for cytotoxicity and anti-oxidant activities of the crude ethanolic extract of ginger were 10.95, 53.15, and $27.86{\mu}g/ml$, respectively. More than ten DNA fragments were visualized and up to 7-9 fold up-regulation of MDR1 and MRP3 genes was observed following exposure to the ethanolic extract of ginger. Acute and subacute toxicity tests indicated absence of any significant toxicity at the maximum dose of 5,000 mg/kg body weight given by intragastric gavage. The survival time and survival rate of the CCA-bearing hamsters were significantly prolonged compared to the control group (median of 54 vs 17 weeks). Results from these in vitro and in vivo studies thus indicate promising anticancer activity of the crude ethanolic extract of ginger against CCA with the absence of any significant toxicity. Moreover, MDR1 and MRP3 may be involved in conferring resistance of CCA to the ginger extract.

Tyrosinase inhibitory activity and antimicrobial activity by mixtures of ultrasonicated chitosan and Maillard reaction products (초음파 처리 키토산과 메일라드 반응액 혼합물에 의한 tyrosinase 억제 활성 및 항균력 분석)

  • The aim of this study was to investigate the tyrosinase inhibitory activity and antimicrobial activity by mixtures of ultrasonicated chitosan and Maillard reaction products. Analysis of tyrosinase was purified from potato and confirmed by active staining after SDS-PAGE. Maillard reaction products (MRPs) were formed from various sugars (glucose, fructose, galactose, xylose, arabinose or ribose) and cystein. MRPs inhibited the tyrosinase purified from potato. The highest tyrosinase inhibitory activity was shown by MRP from glucose and cystein. Ultrasonicated chitosan (over 1 hr) showed antimicrobial activity at the concentration of 1% against E. coli and S. aureus. For the development of antibrowning agent with antimicrobial activity, tyrosinase inhibitory and antimicrobial activity by the mixtures of ultrasonicated chitosan and MRP were tested. 1:1 mixture of ultrasonicated chitosan and MRP from glucose and cystein was the best antibrowning agent having antimicrobial activity.

  • The Maillard reaction is a non-enzymatic reaction between amino and carbonyl groups. During milk processing, lactose reacts with milk protein through this reaction. Infant formulas (IFs) are milk-based products processed with heat-treatments, including spray-drying and sterilization. Because IFs contain higher Maillard reaction products (MRPs) than breast milk, formula-fed infants are subject to higher MRP exposure than breast milk-fed ones. In this study, we investigated the optimization of conditions for minimal MRP formation with the addition of $\small{L}$-carnitine ($\small{L}$-car), pyridoxine hydrochloride (PH), and $\small{DL}$-${\alpha}$-tocopheryl acetate (${\alpha}$-T) in an IF model system. MRP formation was monitored by response surface methodology using fluorescence intensity (FI) and 5-hydroxymethylfurfural (HMF) content. The optimal condition for minimizing the formation of MRPs was with $2.3{\mu}M$ $\small{L}$-car, $15.8{\mu}M$ PH, and $20.6{\mu}M$ ${\alpha}$-T. Under this condition, the predicted values were 77.4% FI and 248.7 ppb HMF.

Effects of Saengjihwangeum-ja Extracts on the Expression of Inflammatory Response in Human Monocyte Cells Induced by Advanced Glycation End Product (생지황음자(生地黃飮子) 추출물이 단핵세포에서 당화종말산물로 유도된 염증반응에 미치는 효과)

  • Saengjihwangeum-ja (SJHEJ) was recorded in DongEuiBoGam as being able to be used for treatment of Sogal whose concept had been applied to Diabetes Mellitus (DM). Modification of proteins by long term circulation of glucose leads to the formation of advanced glycation end product(AGE). Recent immunological studies demonstrated that ligation of AGE play an important role in the development of diabetic complications including atherosclerosis, which includes activation, adhesion, and migration of monocytes. Also, AGE and Maillard reaction product(MRP) could augment monocyte inflammatory responses via ligation of AGE receptor. In this study, the effects of SJHEJ extracts on the expression of inflammatory response-related genes such as tumor necrosis factor-$\alpha$, monocyte chemoattractant protein-1, interferon-g-inducible protein-10, and cyclooxygenase-2 in the human monocyte cell line, THP-1 cells. Reverse transcriptase-polymerase chain reaction revealed that SJHEJ had inhibitory effects on the expression of the TNF-a, MCP-1, IP-10, COX2, IL-1b genes in MRP-induced THP-1 cells. Treatment with SJHEJ had reduced reactive oxygen production in THP-1 cells stimulated by MRP. These inhibitory effects might be exerted via prevention of oxidative stress in activated monocytes. In addition, radical scavenging activity of SJHEJ was increased. These results suggest that SJHEJ has a beneficial effects for improve diabetic vascular complication.

Cyanidin and Cyanidin-3-O-β-D-glucoside Suppress the Inflammatory Responses of Obese Adipose Tissue by Inhibiting the Release of Chemokines MCP-1 and MRP-2

  • Obesity-induced inflammation plays a crucial role in obesity-related pathologies such as type II diabetes and atherosclerosis. Adipose tissue macrophages and the cell-derived proinflammatory chemokines are key components in augmenting inflammatory responses in obesity. Anthocyanins such as cyanidin and $cyanidin-3-O-{\beta}-D-glucoside$ (C3G) are known to elicit anti-inflammatory activities by suppressing the production of proinflammatory mediators such as tumor necrosis factor alpha and nitric oxide in LPS-stimulated macrophages. In the present study, we investigated whether cyanidin and C3G have the potential to suppress the inflammatory responses of adipose cells. Cyanidin and C3G not only suppressed the migration of RAW 264.7 macrophages induced by mesenteric adipose tissue-conditioned medium, but also inhibited the activation of the cells to produce inflammatory chemokines such as monocyte chemoattractant protein-1 (MCP-1) and macrophage inflammatory protein-related protein-2 (MRP-2) in a dose-dependent manner. Cyanidin and C3G also inhibited the release of MCP-1 and MRP-2 from adipocytes and/or macrophages. These findings suggest that cyanidin and C3G may suppress the inflammatory responses of adipose tissue in obesity.

  • This paper aims to review and discuss a graph-based approach for the representation and analysis of building circulation using BIM models. To propose this approach, the authors survey diverse researches and developments which are related to building circulation issues such as circulation requirements in Korea Building Act, spatial network analysis, as well as BIM applications. As the basis of this paper, UCN (Universal Circulation Network) is the main reference of the research, and the major goal of this paper is to extend the coverage of UCN with additional features we examined in the survey. In this paper we restructured two major perspectives on top of UCN: 1) finding major factors of graph-based circulation analysis based on UCN and 2) restructuring the UCN approach and others for adjusting to Korean Building Act. As a result of the further studies in this paper, two major additions have demonstrated in the article: 1) the most remote point-based circulation representation, and 2) virtual space-based circulation analysis.

  • A lot-sizing model for multi-stage MRP systems is proposed, in which known demands must be satisfied. In this model, an approach with considerations of initial inventory and limited production capacity is involved. Most of the studies on the lot-sizing techniques for multi-stage material requirement planning systems have been focused upon two basic approaches. One approach is to develope an algorithm yielding an optimal solution. Due to the computational complexity and sensitivity of the optimal solution to the problem of lot sizing, heuristic approaches are often employed. In this paper, the heuristic approach is used by sequential application of a single-stage algorithm with a set of modified cost by the concept of multi-echelon costs. The proposed method is compared with an lot-sizing method(Florian-Klein Model) to prove its effectiveness by numerical examples.

A Queueing Modeling for A ATM Performance Analysis (ATM 성능분석을 위한 대기행렬 모델링)

  • ATM 망을 효율적으로 구축하고 여러 가지 형태의 제어를 통하여 망 자원을 안정적으로 관리하기 위해서는 망의 성능에 대한 다양한 관점에서의 분석이 필수적이며, 그 기본이 되는 것이 ATM 다중화기에 대한 성능 분석이다. ATM 다중화기에 입력되는 트래픽을 분석하는데 있어서 MRP(Markov Renewal Processes) 또는 SMP(Semi-Markov Processes)는 자동 상관계수를 계산하기가 비교적 용이해서 높은 양의 상관관계를 가지는 버스티한 트래픽을 표현하기에 적절한 구조를 가지고 있으며, 입력 트래픽의 머무는 시간이 어떠한 분포이든 표현 가능한 장점이 있다. 본 연구에서는 ATM 트래픽을 분석하는데 있어서 입력되는 on/off 소스를 MRP로 모형화하고, 이를 도착과정으로 하는 이산시간 MR/D/1B 대기시스템으로 구성하여 ATM 다중화기의 셀 손실확률 등의 성능분석을 제시한다. 또한 본 연구에서 제시한 방법에 대한 타당성 검증을 위하여 시뮬레이션과 비교 검토한다.
