방송뉴스제작에 있어서 기자.PD 협업의 시너지효과 탐구 - KBS 2TV <뉴스투데이>를 중심으로 - (Study of Synergy Effects of Collaboration in News Program between TV Reporter & Producer - Focusing on KBS 2TV -)

  • 기자와 PD 협업의 본격적인 시작이라고 볼 수 있는 <뉴스투데이>의 시너지 효과 분석을 통하여 기자와 PD 협업의 바람직한 방향에 대한 함의를 얻기 위한 연구다. <뉴스투데이>는 PD시스템, 기자시스템, 더블 리포팅시스템, 공동작업 시스템 등 4가지의 대표적인 협업양식을 시도했으며, 각 양식은 현장취재, 반영, 편집, 글쓰기의 주체에 의해 구별되었다. 공동작업 시스템을 제외한 기자시스템과 PD시스템에서는 완성한 꼭지 프로그램의 단순한 합산만으로 엄밀한 의미의 시너지는 적다고 할 수 있다. 협업의 성과로는 기존의 뉴스와는 다른 영상, 길이, 관점, 사운드를 가진 새로운 뉴스가 출현했다는 것과, 새로운 프로그램을 배태시킨 토양을 제공했으며, 8시 뉴스시간대라는 새로운 장르 리터러시를 생성시켰다는 것이다. 협업의 성공 요인으로는 구성원이라는 변인이 중요하다는 것을 발견했다. 적절한 비율을 가진 시스템의 균형 역시 성공요인으로 들 수 있다. 하지만, 직종의 벽이라는 한계는 넘기 어려웠고, 협업지향적인 리더가 사라지자, 조직이 와해되었으며, 프로그램은 폐지의 수순을 밟았다.

온라인 정보의 경제적 가치 평가 모델 개발 및 적용 (Development and Application of an Economic Value Assessment Model for Online Information)

  • 지식정보 사회가 점차 발전되어 감에 따라 온라인으로 제공되고 있는 정보에 대한 가치와 그 중요성이 날로 증가되고 있다. 하지만 정당화된 방법론이나 모델의 부재로 인하여 정보의 가치에 관련된 연구는 다소 미진한 실정이다. 이에 본 연구에서는 온라인 정보의 경제적 가치 평가 모델을 제안하고, 제안된 모델을 이용하여 실제로 정보통신부의 지원사업으로 개발 운영중인 정보통신 통합정보시스템(즉, ITFIND)에서 온라인으로 제공되고 있는 정보의 경제적 가치를 산출하였다. 경제적인 가치를 측정하기 위해 첫째, 정보의 카테고리별 이용가치와 교환가치를 측정하였고, 둘째, 이용자의 연간 수요량을 통해 정보 카테고리별 연간 일인당 $CS^{UV}$$CS^{EV}$를 산출하였으며, 마지막으로 ITFIND 전체 이용자의 연간 일인당 모평균 $CS^{UV}$$CS^{EV}$ 추정을 통해 ITFIND에서 제공하고 있는 정보의 연간 경제적 가치인 $CS^{UV}$$CS^{EV}$를 산출하였다.

Poly(Ethylene Glycol)-branched Polyethylenimine-poly(L-phenylalanine) Block Copolymer Synthesized by Multi-initiation Method for Formation of More Stable Polyelectrolyte Complex with Biotherapeutic Drugs

  • An amphiphilic cationic branched methoxy poly (ethylene glycol)-branched polyethylenimine - poly(L-phenylalanine) (mPEG-bPEI-pPhe) block copolymer was successfully synthesized by ring-opening polymerization (ROP) of N-carboxyanhydride of L-phenylalanine (Phe-NCA) with mPEG-bPEI for the preparation of more stable polyelectrolyte complex (PEC) included a hydrophobic interaction. mPEG-bPEI was firstly prepared by the coupling of mPEG and bPEI using hexamethylene diisocyanate (HMDI). The structural properties of mPEG-bPEI-pPhe copolymers were confirmed by $^1H$ NMR. The copolymers exhibited a self-assemble behavior in water above critical aggregate concentration (CAC) in the range of 0.01-0.14 g/L. The CAC of copolymers obviously depended on the hydrophobic block content in the copolymers (the value decreased with the increase of the pPhe block content). The cationic copolymers have the ability to form multi-interaction complex (MIC) with bovine serum albumin (BSA) and plasmid DNA through multi-interaction (electrostatic and hydrophobic interaction). The physicochemical characterization of the complex was carried out by the measurement of zeta potential and particle size. Their zeta-potentials were positive (approximately +10 mV) and their sizes decreased with increasing pPhe contents in the copolymers (PPF/BSA wt% ratio = 2). The complex showed good stability at high ionic strength. Therefore, mPEG-bPEI-pPhe block copolymer was considered as a potential material to enhance the stability of complex including biotherapuetic drugs.

Identification and Characterization of the Acid Phosphatase HppA in Helicobacter pylori

  • An acid phosphatase (HppA) activated by $NH_4Cl$ was purified 192- and 34-fold from the periplasmic and membrane fractions of Helicobacter pylori, respectively. SDS-polyacrylamide gel electrophoresis revealed that HppA from the latter appears to be several kilodaltons larger in molecular mass than from the former by about 24 kDa. Under acidic conditions (pH${\leq}$4.5), the enzyme activity was entirely dependent on the presence of certain mono- and/or divalent metal cations (e.g., $K^+$,$ NH_4{^+}$, and/or $Ni^{2+}$). In particular, $Ni^{2+}$ appeared to lower the enzyme's $K_m$ for the substrates, without changing $V_{max}$. The purified enzyme showed differential specificity against nucleotide substrates with pH; for example, the enzyme hydrolyzed adenosine nucleotides more rapidly at pH 5.5 than at pH 6.0, and vice versa for CTP or TTP. Analyses of the enzyme's N-terminal sequence and of an $HppA^-$ H. pylori mutant revealed that the purified enzyme is identical to rHppA, a cloned H. pylori class C acid phosphatase, and shown to be the sole bacterial 5'-nucleotidase uniquely activated by $NH_4Cl$. In contrast to wild type, $HppA^-$ H. pylori cells grew more slowly. Strikingly, they imported $Mg^{2+}$ at a markedly lowered rate, but assimilated urea rapidly, with a subsequent increase in extracellular pH. Moreover, mutant cells were much more sensitive to extracellular potassium ions, as well as to metronidazole, omeprazole, or thiophenol, with considerably lowered MIC values, than wild-type cells. From these data, we suggest that the role of the acid phosphatase HppA in H. pylori may extend beyond 5'-nucleotidase function to include cation-flux as well as pH regulation on the cell envelope.

녹차추출물과 보존료의 식중독세균에 대한 항균활성 비교 (Comparison of Antibacterial Activities of Green Tea Extracts and Preservatives to the Pathogenic Bacteria)

  • Survival of pathogenic bacteris(S. aureus, L. monocytogenes, E. coli and S. typhimurium) in tryptic soy broth containing green tea water extract(GTW), green tea ethanol extract(GTE), potassium sorbate (PS) and sodium benzoate(SB) stored at various pH was evaluated. Tryptic soy broth(TSB) containing 0∼2%(w/v) of green tea extracts and preservatives adjusted to pH 5.5, 6.0, 6.5 and 7.0 was inoculated approximately 105 CFU/ml of pathogenic bacteria and incubated at 35$^{\circ}C$ for 24∼48 hours. Survival of bacteria was determined by viable cell counts of bacterial culture at each pH. Minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) of green tea extracts and preservatives against pathogenic bacteria were derived from survival curves of each bacteria. Antibacterial activities of green tea extracts increased with increasing pH but those of preservatives decreased with increrasing pH. S. aureus was the most sensitive strain to GTW and GTE but the most resistant to PS and SB. The MICs of green tea extracts to S. aureus were 0.52∼0.98% at pH 5.5∼6.0 and non inhibitory at pH 7.0. S. typhimurium was the most resistant to green tea extracts while the most sensitive to SB. The MICs of green tea extracts to S. typhimurium were 0.46∼1.62% at pH 5.5∼6.0 and 2% of PS was bactericidal at pH 5.5. 1.0∼2.0% of GTE were bactericidal to all strains tested except L. m9oncytogenes at pH 7.0. GTE was most efficient at inactivating pathogenic bacteria, generally followed by GTW, PS and SB.

Isolation and Evaluation of Protective Effect against Fusarium Wilt of Sesame Plants of Antibiotic Substance from Bacillus polymyxa KB-8

  • An antibiotic compound was isolated from the culture of an antagonist against Fusarium oxysporum f. sp. sesami, Bacillus polymyxa strain KB-8, and tested for the control of Fusarium wilt of sesame in greenhouse conditions. Optimum conditions for culturing the antagonist to obtain the maximum antibiotic activity were determined using different culture media, initial medium acidity, and incubation periods for which yeast -malt extract agar with the initial acidity of pH 5 and over 13 days culture were best. Antibiotic substances extracted by methanol had 2 main fractions, KB-8A and KB-8B, in thin layer chromatography (OLC) with Rf values of 0.35 and 0.67 in a solvent system of chloroform : methanol = 7 : 3. The fraction KB-8A wa purified further by XAD-2, silica gel and Sephadex LH-20 column chromatography, and crystalization. Its minimum inhibitory concentrations (MICs) were $12.8\mu\textrm{g}$/ml for F. oxysporum and Alternaria mali, $6.4\mu\textrm{g}$/ml for Colletotrichum gloeosporioides and Rhizoctonia solani, and $3.2\mu\textrm{g}$/ml for Phytophthora capsici. Soil drenching of antibiotic KB-8A in the concentrations of $13.0\mu\textrm{g}$/ml and $26.0\mu\textrm{g}$/ml effectively inhibited the Fusarium wilt of sesame in a greenhouse test, which appeared to be comparable to the fungicide benlate of $6.5\mu\textrm{g}$ a. i./ml.

비선형 해석법을 이용한 IMT2000 중계기용 1W 전력증폭기 제작 연구 (A Study on the Fabrication of 1W Power Amplifier for IMT2000 Repeater Using Nonlinear Analysis)

  • 본 논문에서는 IMT2000 중계기용으로 1.88-1.98 ㎓ 대역 전력증폭기를 저 가격, 소형으로 개발하였다. 이 전력증폭기는 두 단으로 구성되어 있으며 초단은 P-HEMT (ATF-34143, 800 micron gate width, Agilent Technologies)를 사용하였으며, 종단은 GaAs FET(EFA240D-SOT89, 2400 micron gate width, Excelics Semiconductor)를 사용하였다. 개발된 전력 증폭기는 전체 주파수 대역인 1880-1980 ㎒에서 이득이 29.5㏈, 1㏈ gain compression point는 29.5dBm, 3rd order intercept point(OIP3)는 42dBm 그리고 입출력 return loss는 -10㏈/-l2㏈ 이다. 본 연구에서는 이 전력 증폭기를 설계하기 위하여 각 소자의 비선형 모델을 사용하여 전력증폭기의 여러 가지 비선형 특성을 설계할 수 있었으며, 이 전력 증폭기의 크기는 42(L) x 34(W) mm으로 소형화가 가능하였다.

Effects of the Synthetic Coprisin Analog Peptide, CopA3 in Pathogenic Microorganisms and Mammalian Cancer Cells

  • A synthetic coprisin analog peptide, 9-mer dimer CopA3 (CopA3) was designed based on a defensin-like peptide, Coprisin, isolated from the bacteria-immunized dung beetle Copris tripartitus. Here, CopA3 was investigated for its antimicrobial activity and cancer cell growth inhibition. CopA3 showed antimicrobial activities against various pathogenic bacteria and yeast fungus with MIC values in 2~32 ${\mu}M$ ranges, and inhibited the cell viabilities of pancreatic and hepatocellular cancer cells, except MIA-Paca2, Hep3B, and HepG2 cells, in a dose-dependent manner. The average $IC_{50}$ values of CopA3 against pancreatic and hepatocellular cancer cells were 61.7 ${\mu}M$ and 67.8 ${\mu}M$, respectively. The results indicate that CopA3 has potential in the treatments of pancreatic and hepatocellular cancers as well as microorganism infection disease.

욕창 감염의 세균 역학과 항생제 감수성 (Bacteriology and Antibiotics Sensitivity for Pressure Sore)

  • Purpose: Pressure sore wound develops inevitably in long-term, immobilized and hospitalized patients. Sore wound infection is common problem and makes healing process difficult. We aimed to identify the pathogens of the purulent discharge in sore wound and to obtain information for appropriate antibiotics through a sensitivity test Methods: The bacteriologic study was made on 120 cases of patients who admitted or visited our hospital from 2004 January to 2005 December for sore wound treatment. Culture material was collected in BBL transport media with cotton swab and cultured by MacConkey agar plate. The method of MIC by VITEK and Microscan was used for sensitivity test. Results: Among 120 specimens, organisms were isolated from 77(64.2%) cases. Gram positive organisms were cultured in 73 specimens, Gram negative organisms in 46 specemens, and fungi in 2 specimens. Mixed infection by Gram (+) and Gram (-) bacteria were observed in 34 specimens. Among them, S. aureus was the most common isolate in 24(31.2%) patients and 10 (13.0%) S. Aureus isolates were MRSA. The most prevalent Gram-negative organism was Escherichia coli in 20 patients(25.9%). Vancomycin and teicoplanin showed highest sensitivity to Gram-positive organisms and imipenem and amikacin to Gram-negative organisms. Conclusion: Pressure sore wound demands consideration of multimodal therapeutic aspects and these findings would be useful informations to physicians, nurses and clinical assistants in understanding the nature of sore wound and selecting appropriate antibiotics.

선형성이 우수한 GaAs MESFET 저항성 혼합기 설계 (The Desing of GaAs MESFET Resistive Mixer with High Linearity)

  • 본 논문에서는 선형성이 우수한 MIC형태의 GaAs MESFET 저항성 혼합기를 설계하였다. 설계된 저항성 혼합기는 채널저항을 이용하기 위해 게이트단에만 바이어스 전압을 인가하였으며 LO 신호를 게이트단에 입 력시키고 드레인단에는 LO- RF간의 적절한 격리도를 얻기 위하여 삽입된 7 -jXlle hairpin 대역통과 여파기를 통하여 RF 신호를 가하여서 소오스단에서 단락회로와 저역통과 여파기를 통해 IF 신호를 얻는 것이다 .. LO 신호와 RF 신호에 대한 간략화된 등가회로를 추출하여 변환손실을 계산하였으며 하모닉 발란스 해석의 결과 와 비교하였다. 제작된 S-band 수신용 혼합기의 변환손실은 7 -jXlle hairpin 대역통과 여파기의 3.0-3.4 dB 정 도의 삽입손실을 고려하여 8.2 -10.5 dB로 얻을 수 있었고, 왜곡 특성에서 IP3in는 26.5 dBm의 선형적인 특 성을 Vg=-0.85~-1.0 V에서 얻을 수 있었다.
