Genomic Analysis of Dairy Starter Culture Streptococcus thermophilus MTCC 5461

  • The lactic acid bacterium Streptococcus thermophilus is widely used as a starter culture for the production of dairy products. Whole-genome sequencing is expected to utilize the genetic basis behind the metabolic functioning of lactic acid bacterium (LAB), for development of their use in biotechnological and probiotic applications. We sequenced the whole genome of Streptococcus thermophilus MTCC 5461, the strain isolated from a curd source, by 454 GS-FLX titanium and Ion Torrent PGM. We performed comparative genome analysis using the local BLAST and RDP for 16S rDNA comparison and by the RAST server for functional comparison against the published genome sequence of Streptococcus thermophilus CNRZ 1066. The whole genome size of S. thermophilus MTCC 5461 is of 1.73Mb size with a GC content of 39.3%. Streptococcal virulence-related genes are either inactivated or absent in the strain. The genome possesses coding sequences for features important for a probiotic organism such as adhesion, acid tolerance, bacteriocin production, and lactose utilization, which was found to be conserved among the strains MTCC 5461 and CNRZ 1066. Biochemical analysis revealed the utilization of 17 sugars by the bacterium, where the presence of genes encoding enzymes involved in metabolism for 16 of these 17 sugars were confirmed in the genome. This study supports the facts that the strain MTCC 5461 is nonpathogenic and harbors essential features that can be exploited for its probiotic potential.

Production of the Rare Ginsenoside Rh2-MIX (20(S)-Rh2, 20(R)-Rh2, Rk2, and Rh3) by Enzymatic Conversion Combined with Acid Treatment and Evaluation of Its Anti-Cancer Activity

  • The ginsenoside Rh2 has strong anti-cancer, anti-inflammatory, and anti-diabetic effects. However, the application of ginsenoside Rh2 is restricted because of the small amounts found in Korean white and red ginsengs. To enhance the production of ginsenoside Rh2-MIX (comprising 20(S)-Rh2, 20(R)-Rh2, Rk2, and Rh3 as a 10-g unit) with high specificity, yield, and purity, a new combination of enzymatic conversion using the commercial enzyme Viscozyme L followed by acid treatment was developed. Viscozyme L treatment at pH 5.0 and $50^{\circ}C$ was used initially to transform the major ginsenosides Rb1, Rb2, Rc, and Rd into ginsenoside F2, followed by acid-heat treatment using citric acid 2% (w/v) at pH 2.0 and $121^{\circ}C$ for 15 min. Scale-up production in a 10-L jar fermenter, using 60 g of the protopanaxadiol-type ginsenoside mixture from ginseng roots, produced 24 g of ginsenoside Rh2-MIX. Using 2 g of Rh2-MIX, 131 mg of 20(S)-Rh2, 58 mg of 20(R)-Rh2, 47 mg of Rk2, and 26 mg of Rh3 were obtained at over 98% chromatographic purity. Then, the anti-cancer effect of the four purified ginsenosides was investigated on B16F10, MDA-MB-231, and HuH-7 cell lines. As a result, these four rare ginsenosides markedly inhibited the growth of the cancer cell lines. These results suggested that rare ginsenoside Rh2-MIX could be exploited to prepare an anti-cancer supplement in the functional food and pharmaceutical industries.

A 60-GHz LTCC SiP with Low-Power CMOS OOK Modulator and Demodulator

  • In this paper, a 60 GHz LTCC SiP with low-power CMOS OOK modulator and demodulator is presented. The 60 GHz modulator is designed in a 90-nm CMOS process. The modulator uses a current reuse technique and only consumes 14.4-mW of DC power in the on-state. The measured data rate is up to 2 Gb/s. The 60 GHz OOK demodulator is designed in a 130nm CMOS process. The demodulator consists of a gain boosting detector and a baseband amplifier, and it recovers up to 5 Gb/s while consuming low DC power of 14.7 mW. The fabricated 60 GHz modulator and demodulator are fully integrated in an LTCC SiP with 1 by 2 patch antenna. With the LTCC SiP, 648 Mb/s wireless video transmission was successfully demonstrated at wireless distance of 20-cm.

Studies on the Composition, Productivity, Regrowth and Soil Properties of Grasslands in Kimhwa and Chuiwon Areas (금화 철원지역 자연초지의 군락구조, 생산력, 재생력 및 토양특성에 관한 연구)

  • The investigation was made on the relationship among vegetation, net productivity, regrowth and soils of Moonheri-Wasoori, Soodowon-Sungilkyo and Mt. Kumhak grasslands in Chulwon area. The following conclusions have been made as the results of this study. The vascular flora of the grassland in Moonheri-Wasoori was composed of 102 species, the most of which were Medicago denticulata, Miscanthus sinensis, Arundinella hirta, Festuca ovina and Cassia nomame. The flora of Soodowon-Sungilkyn grassland was composed of 74 species, the most of which were Arundinella hirta, Miscanthus sinensis, Carex nanella, Festuca ovina and Cassia nomame. It of Mt. Kumfiak grassland was composed of 78 species, the most of which were Arundinella hirta, Carex nanella, Festuca ovina, Miscanthus sinensis and Aster scaber. These five species contributed greatly to the standing crop of live material which was in excess of 60% of it. The peak standing crop of each 413.7, 307.6 and $348.7g /m^2$ in Moonheri-Wasoori, Soo- dowon-Sungilkyn and Mt. Kuruhak grasslands wars reached in August under flooded conditions as a result of the growth of dominant species, Medicago denticulata and Arundinella hirta. The net prodution of organic matter was slight in some intervals but occurred throughout the growing season After cutting of grasses, the regrowth of new leaves mainly depended on growing stage and edaphic factors. Characteristics of the natural grassland soils, especially, concerning water, total nitrogen, avail-able phosphorus and exchangeable calcium were clarified and might be depend upon the quantity of plant production. Nitrogen, phosphorus and calcium concentration of leaves of Medicago denticulata, Arundinella hirta and Miscanthus sinensis were higher than of stems. Even if diem we diffurmem in the a-mb of nutrients among Medicago denticulata hirta and Miacanthus sinensis for single species and stands. they were covered by variations within species in the present survey.   • These days, the use of alternative energy is recognized as one of countermeasures to rising oil prices and climate-change agreements. Especially, it is regarded as a main element to realize 'Low-Carbon Green Growth', one of the key policies of MB government. However, though alternative energy, renewable energy, is a main policy issue of the present administration, there are many problems to solve: the lack of recognition of its energy, the low level of its technology, its expert's shortage, and its limited application fields. Therefore, to stimulate the use of alternative energy in Korea and to change the recognition of this energy, it is needed to utilize it to urban street furniture that many people visit and use. In particular, if alternative energy is first applied to street furniture that needs energy for its management, the effect of publicity and energy saving will be so great. In this study, for the basic research to introduce and promote various alternative energy sources such as solar power, wind power, small hydro power, and human power, we analyzed the present condition of development of Korean and foreign street furniture items. Furthermore, we evaluated the level of development of these street furniture products and the design types of them and from these evaluations suggested the design direction that can solve problems in development and improve the possibility of application of these energy sources to these street furniture items. Through these analyses, this study aims to enhance the image of future environment-friendly city and make new index for the application of alternative energy.

  • Cyanobacteria (blue-green algae) are not only the first oxygenic organisms on earth but also the foremost primary producers in aquatic environment. Massive growth of cyanobacteria, in eutrophic waters, usually changes the water colour to green and is called as algal (cyanobacterial) bloom or green tide. Cyanobacterial blooms are a result of high levels of primary production by certain species such as Microcystis sp., Anabaena sp., Oscillatoria sp., Aphanizomenon sp. and Phormidium sp. These cyanobacterial species can produce hepatotoxins or neurotoxins as well as malodorous compounds like geosmin and 2-methylisoborneol (MIB). In order to solve the nationwide problem of hazardous cyanobacterial blooms in Korea, the following technically and strategically sound approaches need to be developed. 1) As a long-term strategy, reduction of the nutrients such as phosphorus and nitrogen in our water bodies to below permitted levels. 2) As a short term strategy, field application of combination of already established bloom remediation techniques. 3) Development of emerging convergence technologies based on information and communication technology (ICT), environmental technology (ET) and biotechnology (BT). 4) Finally, strengthening education and creating awareness among students, public and industry for effective reduction of pollution discharge. Considering their ecological roles, a complete elimination of cyanobacteria is not desirable. Hence a holistic approach mentioned above in combination to addressing the issue from a social perspective with cooperation from public, government, industry, academic and research institutions is more pragmatic and desirable management strategy.

Fast Intra Prediction Mode Decision Algorithm Using Directional Gradients For H.264 (방향성 기울기를 이용한 H.264를 위한 고속 화면내 예측 모드 결정 알고리즘)

  • H.264/AVC video coding standard uses the rate distortion optimization method which determines the best coding mode for macroblock(MB) to improve coding efficiency. Whereas RDO selects the best coding mode, it causes the heavy computational burden comparing with previous standards. To reduce the complexity, in this paper, a fast intra prediction mode decision algorithm using directional gradients is proposed. The proposed algorithm is composed of 2-path structure. In the first path, $16{\times}16$ intra prediction mode is determined using directional gradients. In the second path, 3 modes instead of 9 modes are chosen for RDO to decide the best mode for $4{\times}4$ block. Finally, the two modes determined in the two-path decision process are compared to decide the final block mode. Experimental results show that the computation time of the proposed method is decreased to about 77% of the exhaustive mode decision method with negligible quality loss.

A Study of the Boot ROM S/W Design and Verification for the Next Generation LEO Satellite (차세대 저궤도 위성의 Boot ROM 소프트웨어 설계 및 검증에 대한 연구)

  • The next generation LID satellite has 64KB PROM which contains the boot loader and the monitor software, and two 4MB NVMEMs which are used for flight software storage. The boot loader has two operation modes which are the flight software mode and the monitor mode. In the flight software mode, it checks CRC checksum of selected NVMEM and copies flight software image from NVMEM to RAM And then it starts VxWorks RTOS in RAM, creates flight software tasks, and starts execution of flight software. In the monitor mode, it activates monitor software which performs NVMEM reprogramming and board-level testing on the ground. This paper is to present the design of Boot ROM software and verification method using simulator.

Myocardial Protective Effect of Histidine-Tryptophan-Ketoglutarate Solution and Cold Blood Cardioplegic Solution in Pediatric Cardiac Surgery (소아 개심술에서 Histidine-Tryptophan-Ketoglutarate 용액과 냉혈심정지액의 심근보호 효과)

  • Background: There is limited data on comparisons between the effect of histidin-etryptophan-ketoglutarate (HTK) solution and cold blood cardioplegic (CBC) solution in pediatric cardiac surgery. The purpose of this study was to compare the myocardial protective effect of HTK solution and CBC solution in patients undergoing pediatric cardiac surgery. Materials and Methods: We selected 49 patients with ventricular septal defect and atrial septal defect. HTK solution was used in 21 patients and CBC solution was used in 28 patents. HTK solution was given as a single dose, whereas CBC solution was used in the usual multi-dose method. The incidence of EKG change and concentration of Troponin T and CK-MB were compared for the evaluation of myocardial damage. Results: There were no significant differences in the incidence of ST, T segment change by EKG and serial cardiac enzyme levels between two groups. Conclusion: These results suggested that the myocardial protective effect of HTK solution was similar to CBC solution in simple pediatric cardiac surgery.

Model Algorithms for Estimates of Inhalation Exposure and Comparison between Exposure Estimates from Each Model (흡입 노출 모델 알고리즘의 구성과 시나리오 노출량 비교)

  • Objectives: This study aimed to review model algorithms and input parameters applied to some exposure models and to compare the simulated estimates using an exposure scenario from each model. Methods: A total of five exposure models which can estimate inhalation exposure were selected; the Korea Ministry of Environment(KMOE) exposure model, European Centre for Ecotoxicology and Toxicology of Chemicals Targeted Risk Assessment(ECETOC TRA), SprayExpo, and ConsExpo model. Algorithms and input parameters for exposure estimation were reviewed and the exposure scenario was used for comparing the modeled estimates. Results: Algorithms in each model commonly consist of the function combining physicochemical properties, use characteristics, user exposure factors, and environmental factors. The outputs including air concentration ($mg/m^3$) and inhaled dose(mg/kg/day) are estimated applying input parameters with the common factors to the algorithm. In particular, the input parameters needed to estimate are complicated among the models and models need more individual input parameters in addition to common factors. In case of CEM, it can be obtained more detailed exposure estimates separating user's breathing zone(near-field) and those at influencing zone(far-field) by two-box model. The modeled exposure estimates using the exposure scenario were similar between the models; they were ranged from 0.82 to $1.38mg/m^3$ for concentration and from 0.015 to 0.180 mg/kg/day for inhaled dose, respectively. Conclusions: Modeling technique can be used for a useful tool in the process of exposure assessment if the exposure data are scarce, but it is necessary to consider proper input parameters and exposure scenario which can affect the real exposure conditions.
