Background and Objectives: The role of thyroid-stimulating hormone (TSH) signaling on osteoblastic differentiation is still undetermined. The aim of this study was to investigate the effects of 5-aza-2'-deoxycytidine (5-azacytidine) on TSH-mediated regulations of osteoblasts. Materials and Methods: MG63, a human osteoblastic cell-line, was treated with 5-azacytidine before inducing osteogenic differentiation using osteogenic medium (OM) containing L-ascorbic acid and ${\beta}$-glyceophosphate. Bovine TSH or monoclonal TSH receptor stimulating antibody (TSAb) was treated. Quantitative real-time PCR analyses or measurement of alkaline phosphatase activities were performed for evaluating osteoblastic differentiation. Results: Studies for osteogenic-related genes or alkaline phosphatase activity demonstrated that treatment of TSH or TSAb alone had no effects on osteoblastic differentiation in MG63 cells. However, treatment of 5-azacytidine, per se, significantly increased osteoblastic differentiation and combination treatment of 5-azacytidine and TSH or TSAb in the condition of OM showed further significant increase of osteoblastic differentiation. Conclusion: Stimulating TSH signaling has little effects on osteoblastic differentiation in vitro. However, in the condition of epigenetic modification using inhibitor of DNA methylation, TSH signaling positively affects osteoblastic differentiation in human osteoblasts.
In order to search for anti-influenza virus type A agents from Korean traditional prescriptions (herb complexes), we selected 63 traditional prescriptions, based on a review of the Korean traditional medicine books. Both methanol extracts and boiling-water extracts were tested, by means of the Haemagglutination Inhibition Test (HIT). Three of the 63 methanol extracts: CM-22, CM-26, CM-48 (see explanation of nomenclature below), showed efficacy against influenza virus type ACM-22 showed anti-influenza virus type A activity at the range of $313{\mu}g/ml$ to $9.75{\mu}g/ml$, CM-26 showed antiviral activity at the range of $156{\mu}l/ml$ to $4.87{\mu}g/ml$, CM-48 showed anti-influenza virus type A activity at the range of $625{\mu}g/ml$ to $19.5{\mu}g/ml$, respectively. Three of the water extracts: CW-14, CW-34, CW-61 were active. CW-14 showed anti-influenza virus type A activity at the range of l0mg/ml to $78{\mu}g/ml$, CW-34 showed antiviral activity at the range of 10mg/ml to $625{\mu}g/ml$ and CW-61 showed anti-influenza virus type A activity at the range of l0mg/ml to $313{\mu}g/ml$, respectively. In order to determine cytotoxicity of each extracts, chicken red blood cells were incubated with the various concentration of extracts of Korean traditional prescriptions. CW-14, CW-34 and CW-61 did not show cytotoxic effect against red blood cells whereas CM-22, CM-26 and CM-48 showed cytotoxic effect against red blood cells at the range of l0mg/ml to $625{\mu}g/ml$, 10mg/ml to $313{\mu}g/ml$ and 10mg/ml to $313{\mu}g/ml$, respectively. These results indicated that Korean traditional pres criptions may be inhibit either attachment of virus to cell surface receptor or penetration of the virus into cell during the initial stage of infection.
곰팡이 독소인 ochratoxin A (OA)는 동물과 사람에게 간 및 신장의 손상을 일으키는 것으로 알려져 있다. 본 연구는 산화적 손상과 관련이 있는 OA-유발 지질과산화에 대한 항산화 비타민의 완화작용에 대하여 조사하였다. 수컷 ICR 마우스에 비타민 C (10 mg/kg)와 비타민 E (63.8 mg/kg)를 복강 투여하고, 한 시간 후에 0.1M $NaHCO_3$에 녹인 OA를 4 mg/kg/day의 용량이 되게 복강 투여하였다. 4일간 반복투여 한 후 간과 신장의 microsome 내의 superoxide dismutase (SOD) 및 catalase 활성과 malondialdehyde (MDA) 생성을 측정하였다. 아울러 comet assay에 의해 세포의 손상과 항산화 비타민의 완화 작용에 대한 관련성을 알아보았다. 결과들은 간과 신장에서 모두 i) OA투여에 의해 SOD, catalase 활성 및 MDA 수치가 유의성(p<0.05) 있게 증가하였고, ii)항산화 비타민 혼합 투여에 의해 OA에 의한 SOD, catalase 활성 및 MDA 형성이 유의성 있게 저하되었으며. iii) OA-유발 지질과산화와 관련이 있는 혈구세포의 손상도 항산화 비타민에 의해 완화되었다. 이러한 결과들은 항산화 비타민이 OA중독에 의한 간과 신장의 손상의 예방에 사용될 수 있을 것으로 사료된다.
In this paper, the in vitro biocompatibility of graphene film (GF) with osteoblasts was evaluated through cell adhesion, viability, alkaline phosphatase activity, F-actin and vinculin expressions, versus graphite paper as a reference material. The results showed that MG-63 cells exhibited stronger cell adhesion, better proliferation and viability on GF, and osteoblasts cultured on GF exhibited vinculin expression throughout the cell body. The rougher and wrinkled surface morphology, higher elastic modulus and easy out-of-plane deformation associated with GF were considered to promote cell adhesion. Also, the biomineralization of GF was assessed by soaking in simulated body fluid, and the GF exhibited enhanced mineralization ability in terms of mineral deposition, which almost pervaded the entire GF surface. Our results suggest that graphene promotes cell adhesion, activity and the formation of bone-like apatite. This research is expected to facilitate a better understanding of graphene-cell interactions and potential applications of graphene as a promising toughening nanofiller in bioceramics used in load-bearing implants.
Objectives: Reactive oxygen species (ROS) have been implicated in the pathogenesis of a wide range of acute and longterm neurodegenerative diseases. This study was undertaken to examine whether Sunghyangchungisan(SHCS), a well-known prescription in Korean traditional medicine, might have beneficial effects on ROS-induced brain cell injury. Methods: Human neuroglioma cell line A172 and H2O2 were employed as an experimental model cell and oxidant. Results: SHCS effectively protected the cells against both the necrotic and apoptotic cell death induced by H2O2. The effect of SHCS was dose-dependent at concentrations ranging from 0.2 to 5mg/ml. SHCS significantly prevented depletion of cellular ATP and activation of poly (ADP-ribose) polymerase induced by H2O2. It also helped mitochondria to preserve its functional integrity estimated by MTT reduction ability. Furthermore, SHCS significantly prevented H202-induced release of cytochrome c into cytosol. Determination of intracellular ROS showed that SHCS might exert its role as a powerful scavenger of intracellular ROS. Conclusions: The present study provides clear evidence for the beneficial effect of SHCS on ROS-induced neuroglial cell injury. The action of SHCS as an ROS-scavenger might underlie the mechanism.
Hairy cell leukemia (HCL) is an uncommon chronic B-cell lymphoproliferative disorder that is characterized by cytopenia, splenomegaly, and mononuclear cells displaying cytoplasmic projections. We experienced a case of hairy cell leukemia that developed in a 38-year-old man. He showed marked splenomegaly without palpable lymphoadenopathy. A complete blood cell count revealed leukopenia ($3300/{\mu}{\ell}$ with 63% of lymphocyte) and the peripheral blood smear showed abnormal lymphoid cells with cytoplasmic projections. The bone marrow smear revealed abnormal lymphocytes and severe myelofibrosis. Tartrate-resistant acid phosphatase reactivity was strongly positive in the hairy cells. The immunophenotyping results of lymphoid cells were CD5(-), CD10(-), CD19(+), CD25(+), CD103(+), CD20(+), lambda(+). The patient was treated with 2-Chlorodeoxyadenosine at a daily dose of 0.1mg/Kg by a continuous intravenous infusion for 7 days. The patient achieved complete remission.
We investigated the quality of 12 commercial salted and fermented anchovy Engraulis japonicas sauces by measuring their chemical compositions, bacteria concentrations, and biogenic amine contents. The sauces had a moisture content of 67.13-69.83% (mean: 68.17%), salinity of 20.00-25.84% (mean: 22.29%), pH of 5.14-6.28 (mean: 5.63), volatile basic nitrogen content of 119.12-273.37 mg/100 g (mean: 199.19 mg/100 g), total nitrogen content of 0.82-1.50% (mean: 1.30%) and amino nitrogen content of 550.17-1,086.62 mg/100 g (mean: 774.99 mg/100 g). The viable cell counts ranged from not detected to $1.6{\times}10^3CFU/mL$, and the number of biogenic amine-forming bacteria was very low or undetectable. The tested samples contained 372.32-2,111,61 mg/kg (mean 813.48 mg/kg) histamine, 29.62-144.29 mg/kg (mean 98.14 mg/kg) cadaverine, 87.89-530.84 mg/kg (mean 329.91 mg/kg) tryptamine, 20.89-127.17 mg/kg (mean 60.49 mg/kg) putrescine, and 13.08-109.91 mg/kg (mean 57.74 mg/kg) tyramine. Whereas no spermidine or spermine was detected in any sample. These results strongly suggest the necessary of monitoring the biogenic amine contents of commercial salted and fermented anchovy sauces carefully to ensure consumer health.
In this study, crystallization was effectively suppressed in Al-based metallic glasses (Al-MGs) during pulverization by cryo-milling by applying an extremely low processing temperature and using a surfactant. Before Al-MGs can be used as an additive in Ag paste for solar cells, the particle sizes of the Al-MGs must be reduced by milling. However, during the ball milling process crystallization of the Al-MG is a problem. Once the Al-MG is crystallized, they no longer exhibit glass-like behavior, such as thermoplastic deformation, which is critical to decrease the electrical resistance of the Ag electrode. The main reason for crystallization during the ball milling process is the heat generated by collisions between the particles and the balls, or between the particles. Once the heat reaches the crystallization temperature of the Al-MGs, they start crystallization. Another reason for the crystallization is agglomeration of the particles. If the initially fed particles become severely agglomerated, they coalesce instead of being pulverized during the milling. The coalesced particles experience more collisions and finally crystallize. In this study, the heat generated during milling was suppressed by using cryo-milling with liquid-nitrogen, which was regularly fed into the milling jar. Also, the MG powders were dispersed using a surfactant before milling, so that the problem of agglomeration was resolved. Cryo-milling with the surfactant led to D50 = 10 um after 6 h milling, and we finally achieved a specific contact resistance of 0.22 mΩcm2 and electrical resistivity of 2.81 μΩcm using the milled MG particles.
소뼈를 하소하여 천연 하이드록시아파타이트 분말을 얻은 다음, $1100^{\circ}C$ 및 $1200^{\circ}C$로 상압 소결하여 기공율이 각각 35 % 및 18 %인 다공성 하이드록시아파타이트를 제조한 후 이들 시편의 생체친화성을 평가하였다. 골아세포의 성장속도과 세포독성의 유무에 따라 평가되는 생체친화성은 MG-63 세포의 부착능과 분화되어 증식되는 세포의 수로 측정하였다. in vitro에서 세포 반응을 관찰한 결과 소뼈로부터 제조된 하이드록시아파타이트 시편은 독성이 없고 생체 친화성이 좋은 시편이었다. 세포배양 1일 후부터 하이드록시아파타이트 표면에 세포가 부착되었고, 세포배양 후 3일 후에는 핵 주위로 세포질이 퍼져나갔으며, 5일 후에는 부착된 세포의 수가 크게 증가하였다. 배양 초기에는 밀도에 관계 없이 세포의 부착능이 유사하였지만, 세포 배양 시간이 증가하면서 기공율이 높은 시편에서 세포의 부착 및 성장이 더 활발하게 진행되었다.
This study was purpose to investigate that Gucheokbogol-tang(GBT), mainly used in osteoporosis, has possible regulatory effect on bone regeneration by operating in proliferation and calcification of osteoblast. So that, their cytotoxicity and proliferation was investigated with MTT by ELISA, and morphological change was observed by optical microscope and electron microscope. And activation of alkaline phosphates(ALP) which is secreted in early stage of bone formation was measured, and accumulation of $Ca^{2+}$ in the process of calcification was investigated by alizaline red S assay(ARS). The results were as follows: The MG-63 cell, originated from human cell was activated in $10^{-5}$ treated group, and the level of ALP was also increased in the treated group, highest on the third day. And from the outcome of ARS assay of calcification process in the Gucheokbogol-tang(GBT) treated group for 21days. These results suggested that Gucheokbogol-tang(GBT) is effective in proliferation and calcification of osteoblast.
