Effects of nicotine on the formation of osteocalcin and osteoprotegerin and synthesis of its mRNA in MG63 osteoblast-like cell (니코틴이MG63 조골세포주의 오스테오칼신과오스테오프로 테제린의 생성 및 mRNA 발현에 미치는 영향)

  • The purpose of this study was to evaluate the correlation between nicotine and the activity of bone forming cell. MG63 osteoblast-like cells were used for this study. Several factors were examined including the proliferation of cell, alkaline phosphatase activity, the formation of osteocalcin and osteoprotegerin. and the synthesis of its mRNA. MG63 osteoblast-like cells were incubated for 1, 2, 3 and 6 days with nicotine added to the culture medium in 1.0 ${\mu}M$, 1.0mM, 2.5mM, 5.0mM, 7.5mM, and 10.0mM concentrations. The proliferation of MG63 osteoblast-like cells was temporarily activated at the low nicotine concentrations. At high concentrations (>5.0 mM), however. it was suppressed. Alkaline phosphatase activity was suppressed in a dose-dependent manner as the concentration of nicotine increased. Osteocalcin decreased in a dose-dependent manner at high nicotine concentrations of more than 7.5mM and the same result was show when the osteoblasts were treated with low concentrations for longer than 3 days. There was a difference in the influence of nicotine on the synthesis of osteocalcin mRNA and formation of osteocalcin itself at 1 and 3 days. Generally, osteoprotegrin notably declined in all experimental groups. However, the level of its mRNA increased at high nicotine concentrations of more than 7.5mM after 3 days and more than 5.0mM after 6days.

The Effect of Osteoblast like Cell on Low Intensity Pulsed Ultrasound Used in Current Clinical Practice (임상에서 쓰이는 저 출력 초음파(Low Intensity Pulsed Ultrasound)가 조골세포에 미치는 영향)

  • In this study, we investigated the proliferative and adhesional effect of human osteoblast like MG-63 cell treated with low intensity pulsed ultrasound (LIPUS). We tested the effectiveness of LIPUS in human osteoblast like MG-63 cells. Cell proliferation was measured using a water soluble tetrazolium salts-1 assay. The mRNA expression of alkaline phosphate, vascular endothelial growth factor (VEGF), integrin alpha 2, colla 1A1 were performed by reverse transcription-polymerase chain reaction. LIPUS was no cytotoxicity in human osteoblast like MG-63 cells. In addition, the data show that treatment with 1 MHz and 3 MHz LIPUS on increased proliferation 7 days after. There were significant increased in mRNA expression of alkaline phosphatase, osteocalcin, VEGF, integrin alpha 2 and colla 1A1 (p<0.05). Therefore, the LIPUS significantly increased differential expression of mRNA levels in osteoblast like MG-63 cell and new possibilities in dental clinical practice.

Growth Inhibitory Effect and Changes in Membrane Phospholipid Fatty Acid Composition on MG-63 and AZ-521 Human Cancer Cells by Linoleic Acid (리놀레산에 의한 인체암세포의 성장 억제효과 및 암세포막 인지질 지방산 변화)

  • Linoleic acid(LA) was examined to evaluate its potential as a chemotherapeutic agent for MG-63 human osteosarcoma and AZ-521 gastric cancer cells. The treatment of LA(0.005% for 6 days) to the MG-63 and AZ-521 cancer cells inhibited growth of the cancer cells by 54% and 52%, respectively as compared to that of the controls. It also exhibited that LA with 0.01% concentration decreased the [$^3$H] thymidine incorporation by more than 90% in the both cancer cells. In additions we observed morphological changes in MG-63 and AZ-521 cells under inverted microscope, and the changes in membrane fatty acid compositions of the cancer cells when LA was added at the level of 0.005%. The treatment with LA revealed that the contents of 16:0 and 18:0 decreased significantly, but fatty acids that C numbers are more than 20 and unsaturated(20:4, 22:6, and 24:4) increased, concomitantly the morphological changes of the cells were observed.

Bee Venom induces apoptosis and inhibits COX-2 in human osteosarcoma cell line MG-63 (봉독이 골육종세포주에서 세포사멸 및 COX-2 억제에 미치는 영향)

  • 목적 : 한의학에서 관절염이나 진통치료에 사용되어 왔던 봉독약침액이 인간 골육종 세포주인 MG-63 세포에서 항종양효과가 있는지 연구하고자 한다. 특히 본 실험에서는 이러한 봉독의 종양발생 억제작용이 세포사멸과 관련이 있는지, 그리고 프로스타글란딘 합성 효소인 cyclooxygenase(COX)-2의 억제와 관련이 있는지를 연구하고자 한다. 방법 : 인간 골육종 세포주에서 세포사멸의 변화를 관찰하기 위해서 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium brimide(MTT) assay, 4,6-diamidino-2-phenylindole (DAPI), DNA fragmentation assay 및 reverse transcription-polymerase chain reaction(RT-PCR) 방법을 이용하였다. 결과 : 세포독성 검사에서 봉독은 MG-63 세포에서 농도-의존적으로 세포독성을 나타내었다. 이러한 봉독의 세포독성이 세포사멸로 인한 것인지를 여러 가지 형태로 검사한 결과 봉독에 의한 세포독성은 TUNEL 검사와 DAPI 염색시 세포사멸의 특징적인 소견들을 나타내었고, flow cytometric 분석에서도 세포사멸을 의미하는 세포주기의 변화들을 나타내었다. 봉독이 COX-2의 발현에 미치는 영향을 RT-PCR로 실험한 결과 봉독은 COX-2 mRNA의 발현을 선택적으로 억제하였다. 결론 : 본 실험의 결과 봉독은 COX-2 mRNA의 발현을 억제함으로써 골육종 세포에서 세포사멸을 유발하고 그 결과 항종양효과를 나타내는 것으로 보여진다.

저소득층 노인의 실태에 관한 연구

  • 저소득층 노인 대상의 급식서비스 영양효과를 분석하기 위한 기초연구로써 광주광역시 북구 지역 저소득층 노인 100명을 대상으로 2002년 11월 20일~12월 15일까지 실태조사를 통해 건강ㆍ영양문제를 도출하고 급식서비스 제공에 따른 영양효과를 분석한 결과는 다음과 같다. 수혜를 받은 대상자의 평균 연령은 72.44세, 신장은 158.50cm, 체중은 52.29kg 이었고 이로부터 구한 평균 체질량지수는 20.86kg/$m^2$이었다. 평균 영양소 섭취량은 열량 1,389,67㎉, 단백질 53.50g, 지질 26.20g, 탄수화물 234.41g, 섬유질 5.56g, 칼슘 481.15mg, 인 882,76mg, 철분 8.59mg, 나트륨 4,208.98mg, 칼륨 2,069.77mg, Retinol 408,69RE, Thiamin 0.84mg, Riboflavin 0,73mg, 나이아신 11.04NE, Ascorbic acid 80.63mg, 콜레스테롤 157.63mg 이었고, 연령이 증가할수록 영양제 섭취 비율이 증가하였다.

Effect of Extracts from Root Bark of Ulmus parvifolia on Inhibition of Growth and DNA Synthesis of Human Cancer Cells. (느릅나무 근피 추출물에 의한 인체 암세포 증식 및 DNA 합성 억제효과)

  • Growth and DNA synthesis inhibitory effects of extracts from root bark of Ulmus parvifolia on MG-63 human osteosarcoma cells, HT-29 human colon cancer cells and K-562 leukemia cancer cells were studied. The root bark extract of Ulmus parvifolia was extracted with methanol, hot water and juice. The methanol extract showed the highest inhibitory effect on growth of MG-63, HT-29 and K-562 cancer cells by >85%. The treatment of hot water and juice extracts from root bark of Ulmus parvifolia also inhibited growth of the above cancer cells with increasing concentration. DNA synthesis of MG-63 and HT-29 cancer cells was significantly inhibited by adding methanol, hot water and juice extracts from root bark of Ulmus parvifolia with increasing concentration, showing that the inhibitory effect of growth was more effective on HT-29 cancer cells. These results suggest that the methanol extract from root bark of Ulmus parvifolia may have specific active com-pounds on anticancer effect. The hot water extract also showed a strong inhibitory effect on growth of cancer cells, indicating that the active compounds may be stable to heat.

St. John's Wort (Hypericum perforatum) stimulates human osteoblastic MG-63 cell proliferation and attenuates trabecular bone loss induced by ovariectomy

  • BACKGROUND/OBJECFTIVES: The effect of St. John's Wort extract (SJW) on MG-63 cell proliferation and trabecular bone loss induced by ovariectomy was examined. MATERIALS/METHODS: Proliferation, expression of estrogen receptor (ER) ${\alpha}$ and ER ${\beta}$, and gene expressions of osteoprotegerin (OPG), osteocalcin (OC) and alkaline phosphatase (ALP) were examined in MG-63 cells treated with or without SJW. Ovariectomized rats were treated with SJW at the dose of 100 or 200 mg/kg/day, ${\beta}$-estradiol-3-benzoate (E2), or vehicle only (OVX-C), and sham operated rats were treated with vehicle only (Sham-C). Serum ALP and C-telopeptide (CTX), and femoral trabecular bone loss were examined. RESULTS: SJW increased MG-63 cell proliferation and expression of ER ${\alpha}$ and ER ${\beta}$, and positive effect was shown on gene expressions of ALP, OC and OPG. SJW also showed estrogen like effect on bone associated with slowing down in trabecular bone loss. Histopathology by H&E showed rats treated with SJW displayed denser structure in metaphyseal region of distal femur compared with rats in OVX-C. SJW was shown to reduce serum CTX in OVX rats. CONCLUSION: The present study provides new insight in preventing estrogen deficiency induced bone loss of SJW and possibility for its application in bone health supplement.

Antimicrobial Activity of Streptococcus mutans by Herbal Medicine Extracts (한약재 추출물의 Streptococcus mutans에 대한 항균효과)

  • This study was to investigate the antibacterial effect of 63 species herbal medicine extracts on S. mutans growth by paper disc method and then examined the effects of S. mutans growth by some selected herbal medicines. Antibacterial effect of 63 species medicinal herb extracts on S. mutans was superior to Lycopi herba, Scutellariae radix in regular order and antibacterial activity(20 mg/ml concentration) through paper disc method were measured 6.3 mm and 3.5 mm, respectively. The growth of S. mutans in control medium was the highest at 8hrs, while that of medicinal herbs extract added-medium(2 mg/ml) was at 16 hrs. The pH values of the control media and media supplemented with Lycopi herba and Scutellariae radix extract were 5.08, 5.80, and 5.74 at 8 hrs, respectively. In the change of total carbohydrate of the control media and supplemented with Lycopi herba and Scutellariae radix extract were 0.81 mg/ml, 1.70 mg/ml and 1.84 mg/ml at 8 hrs, respectively. In the change of proteins of the control media and supplemented with Lycopi herba and Scutellariae radix extract were 8.39 mg/ml, 9.52 mg/ml and 9.28 mg/ml at 8 hrs, respectively. The polysaccharide contents of the control media and supplemented with Lycopi herba and Scutellariae radix extract were 300 mg/100 ml, 200 mg/100 ml and 220 mg/100 ml at 8hrs, respectively.
