Middle East respiratory syndrome coronavirus (MERS-CoV) belongs to the genus Betacoronavirus and causes severe morbidity and mortality in humans especially when infected patients have underlying diseases such as chronic obstructive pulmonary disease (COPD). Previously, we demonstrated that MERS-CoV-encoded ORF8b strongly inhibits MDA5- and RIG-I-mediated induction of the interferon beta (IFN-β) promoter activities. Here, we report that ORF8b seemed to regulate MDA5 or RIG-I differentially as protein levels of MDA5 were significantly down-regulated while those of RIG-I were largely unperturbed. In addition, ORF8b seemed to efficiently suppress phosphorylation of IRF3 at the residues of 386 and 396 in cells transfected with RIG-I while total endogenous levels of IRF3 remained largely unchanged. Furthermore, ORF8b was able to inhibit all forms of RIG-I; full-length, RIG-I-1-734, and RIG-I-1-228, the last of which contains only the CARD domains. Taken together, it is tempting to postulate that ORF8b may interfere with the CARD-CARD interactions between RIG-I and MAVS. Further detailed analysis is required to delineate the mechanisms of how ORF8b inhibits the MDA5/RIG-I receptor signaling pathway.
Severe acute respiratory syndrome coronavirus (SARS-CoV2) is a major coronavirus that infects humans with human Coronavirus (HuCoV)-229E, HCoV-OC43, HCoV-HKU1, HCoV-NL63, Severe acute respiratory syndrome coronavirus (SARS-CoV) and Middle east respiratory syndrome coronavirus (MERS-CoV). SARS-CoV2 is currently a global pandemic pathogen. In this study, we developed conventional RT-PCR based diagnostic system for the detection of SARS-CoV2 which is relatively inexpensive but has high stability and a wide range of users. Three conventional RT-PCR primer sets capable of forming specific band sizes by targeting the ORF1ab [232 nucleotide (nt)], E (200 nt) and N (288 nt) genes of SARS-CoV2 were developed, respectively, and it were confirmed to be about 10~100 times higher detection sensitivity than the previously reported methods. In addition, a standard positive control that can generate specific amplicons by reacting with the developed RT-PCR primers and verify the false-positiv from contamination of the laboratory was produced. Therefore, the diagnostic system that uses the RT-PCR method is expected to be used to detect SARS-CoV2.
Over a span of few decades, the world has seen the emergence of new viruses that have posed serious problems to global health .COVID-19 is a major pathogenic threat to the modern world that primarily shoots the respiratory system of human beings. Wuhan which is the capital city of Hubei, China was the first place in the world where first cases of COVID-19 emerged and the scores of cases significantly increased at an immense rate leading to city isolation and establishment of new specially designed hospitals. SARS-CoV had emerged from bats in china (2002) and MERS-CoV from camels transmitted via bats in Middle East (2012) where the previous versions of COVID-19 took place. Infections with SARS-CoV-2 are now widespread, like Nuclear Chain Reaction (NRC). In this review we will discuss the COVID-19 origin, transmission, incubation, diagnosis and therapies available at the present scenario.
연구목적 본 연구는 메르스 생존자에서 감염 당시 낙인에 대한 인식, 12개월 후 이들이 사용하는 대처전략, 24개월 후 우울 및 외상후 스트레스 증상에 끼치는 영향 간의 연관성을 확인하고자 하였다. 방 법 63명의 2015년 메르스 생존자를 대상으로 코호트 연구를 진행하여 12개월 및 24개월 후에 인구학적 정보, 메르스 감염의 심각도, 우울 및 외상후 스트레스 증상, 대처전략, 낙인에 대한 인식 등을 조사했다. 결 과 감염 당시 낙인의 인식은 12개월 후 부정적 대처전략(p=0.003), 24개월 후 우울(p=0.058) 및 외상후 스트레스 증상(p=0.011)과 연관성을 보였다. 12개월 후 부정적 대처전략과 24개월 후 우울(p=0.002) 및 외상후 스트레스 증상(p<0.001) 간에도 유의한 연관성이 있었다. 결 론 감염자에 대한 낙인은 생존자들의 장기적 정신 건강에 부정적 영향을 줄 수 있다. 향후 신종감염병 유행 시 낙인 문제를 개선하고 생존자들이 적응적 대처전략을 가지도록 도와야 한다.
The Middle East respiratory syndrome coronavirus (MERS-CoV) outbreak in South Korea in 2015 has drawn public attention regarding the legal regulation of infectious disease control in Korea. This paper discusses the interpretive and legislative concerns regarding the Infectious Disease Prevention and Control Act, its ordinance and enforcement regulations, as well as public statements from the relevant administrative agency. Future improvements are also proposed.
본 논문은 2013년 5월 국제바이러스 분류 위원회에서 중동호흡기증후군을 일으키는 신종 코로나바이러스를 메르스코로나바이러스(MERS-CoV)라 명명한 후, 2015년 5월 국내에 감염자가 들어와 이후 확산되고, 중동 호흡기 증후군 환자 발생 후 국내 첫 사망자가 발생한 일개 병원 지역응급의료센터 내원 후 확산을 막기 위하여 시행하였던 경험들을 공유하고자 한 내용이다.
COVID-19(Coronavirus Disease 2019)는 RNA 형 바이러스로써 점막감염(粘膜感染)과 비말전파(飛沫傳播)로 전염되는 급성 호흡기성 질병이다. 2019 년 12 월 중국 후베이 우한에서 처음 감염이 보고된 후 빠르게 글로벌로 확산되었고, 현재 여러 국가와 지역이 Lockdown 상태에 있다. COVID-19 의 치사율은 국가별, 연령별 차이는 있으나 사스(SARS-CoV), 메르스(MERS-CoV) 등과 비교하여 높다고 할 수 없다. 그러나 COVID-19 는 신종 코로나바이러스로써 아직 백신(Vaccine)과 항바이러스제가 개발되지 않았고 다른 질병과 비교하여 빠른 감염 속도때문에 의료 공백, 사회적 혼란, 경제적 손실을 크게 일으키고 있다. 따라서 바이러스의 확산 양상을 데이터 분석을 통하여 예측할 수 있다면 사회·경제적인 폐해를 줄일 수 있어 Bass 모델과 R 패키지를 이용하여 COVID-19 확산 예측 모형을 계량적으로 제시하였다.
The direction of evolution can estimate based on the variation among nonsynonymous to synonymous substitution. The simulative study investigated the nucleotide sequence of closely related strains of respiratory syndrome viruses, codon-by-codon with maximum likelihood analysis, z selection, and the divergence time. The simulated results, dN/dS > 1 signify that an entire substitution model tends towards the hypothesis's positive evolution. The effect of transition/transversion proportion, Z-test of selection, and the evolution associated with these respiratory syndromes, are also analyzed. Z-test of selection for neutral and positive evolution indicates lower to positive values of dN-dS (0.012, 0.019) due to multiple substitutions in a short span. Modified Nei-Gojobori (P) statistical technique results also favor multiple substitutions with the transition/transversion rate from 1 to 7. The divergence time analysis also supports the result of dN/dS and imparts substantiating proof of evolution. Results conclude that a positive evolution model, higher dN-dS, and transition/transversion ratio significantly analyzes the evolution trend of severe acute respiratory syndrome coronavirus 2, severe acute respiratory syndrome coronavirus, and Middle East respiratory syndrome coronavirus.

The world is going through the most unprecedented time with the outbreak of novel Coronavirus disease (COVID-19), which has become a threat to millions. A Coronavirus is a group of viruses that cause a variety of diseases in mammals and birds leading to a range of illnesses in humans including common cold and more severe forms like severe acute respiratory syndrome Coronavirus (SARS-CoV), Middle East respiratory syndrome Coronavirus (MERS-CoV) and COVID-19, which are life-threatening. The virus gets its name from its shape which takes the form of a crown with protrusions around it. In December 2019, a pneumonia outbreak was reported in the Wuhan City of China, which was later traced to a novel strain of Coronavirus and termed as Novel COVID-19. It typically causes flu-like symptoms including fever, cough and shortness of breath and is transmitted through human-to-human and there is no cure for it till now. Thus, this bibliometric study has been carried out to analyze the research progress in Coronavirus and literature published during a period of 30 years (1989-2019). Data for the study were fetched from Web of Science(WoS) multidisciplinary database and the publication trends in terms of total articles, productive countries, institutions, journals, productive authors, most cited articles and authors, etc have been analyzed. In total, 4917 articles were retrieved; these were from 711 sources and were contributed by 14442 authors. The collaboration index was 3.11, which clearly indicates that there has been a lot of collaboration in this field. The most preferred journal for the study period was "Journal of Virology" and the maximum contribution has been from the University of Hong Kong.
