Effects of ${\alpha}$-lipoic acid and L-carnosine supplementation on antioxidant activities and lipid profiles in rats

  • ${\alpha}$-Lipoic acid and L-carnosine are powerful antioxidants and are often used as a health supplement and as an ergogenic aid. The objective of this study was to investigate the effects of ${\alpha}$-lipoic acid and/or L-carnosine supplementation on antioxidant activity in serum, skin, and liver of rats and blood lipid profiles for 6 weeks. Four treatment groups received diets containing regular rat chow diet (control, CON), 0.5% ${\alpha}$-lipoic acid (ALA), 0.25% ${\alpha}$-lipoic acid+0.25% L-carnosine (ALA+LC), or 0.5% L-carnosine (LC). Superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) and lipid peroxidation products, malondialdehyde (MDA) concentrations, were analyzed in serum, skin, and liver. Blood lipid profiles were measured, including triglycerides (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL-C), and low density lipoprotein cholesterol (LDL-C). Skin and liver SOD activities of the ALA and LC groups were higher than those of the CON group (P<0.05), but serum SOD activity was higher only in the LC group compared to that in the CON group (P<0.05). Additionally, only liver GSH-Px activity in the LC group was higher than that of the CON and the other groups. Serum and skin MDA levels in the ALA and LC groups were lower than those in the CON group (P<0.05). Serum TG and TC in the ALA and ALA+LC groups were lower than those in the CON and LC groups (P<0.05). The HDL-C level in the LC group was higher than that in any other group (P<0.05). LDL-C level was lower in the ALA+LC and LC groups than that in the CON group (P<0.05). Thus, ${\alpha}$-lipoic acid and L-carnosine supplementation increased antioxidant activity, decreased lipid peroxidation in the serum, liver, and skin of rats and positively modified blood lipid profiles.

The Effect of Exercise Therapy on Serum Lipid Level and Antioxidants of obese College Female Students (운동요법이 비만여대생의 혈청지질 및 항산화계에 미치는 영향)

  • The purpose of this study is to analyze the effects of exercise therapy on serum lipid levels and antioxidants of obese and normal college female students. The subject group composed of ten normal-weight students(below 30% body fat ratio) and ten obese students(above 30% body fat ratio). After a pilot test, the subjects were given an eight-weeks exercise program. Before and after the exercise program, the subjects were given test for serum lipid and antioxidants were analyzed. The SAS program was used in the data analysis. The statistical measurements employed here were T-test, Wilcoxon signed rank test, and Wilcoxon rank sum test. The results of this research are as follows. 1) The effects of exercise therapy on serum lipid levels : Before the exercise therapy, the levels of Total-cholesterol, Triglyceride and LDL-cholesterol of the obese group were highier than those of the normal-weight group. However, the HDL-cholesterol levels were highier in the normal-weight group than in the obese group, but these differences were not significant. With the exercise therapy, the levels of Total-cholesterol increased gradually. The HDL-cholesterol increased gradually, the LDL-cholesterol level decreased in both groups, However, the Triglyceride level decreased in the obese group and increased in normal group, but the difference was not significant. 2) The effects of the exercise therapy on serum antioxidants ; Before exercise therapy, the serum FR and GSSG levels of the obese group were significantly highier than those of the normal-weight group(p=0.00, p=0.04). The serum GSH level of the normal-weight group was highier than that of the obese group, and the serum MDA level of the obese group was highier than that of the normal-weight group. Again these differences were not significant. With exercise therapy, serum FR level was reduced and serum GSSG level significantly increased in both group(obese group p=0.01, normal-weight group p=0.01), The serum GSH level of the obese group significantly increased (p=0.01), and serum MDA level significantly increased in the obese group(p=0.01), but the difference in the normal-weight group was not significant. These results show that regular exercise therapy reduces serum FR levels and activation of antioxidant systems, and suppress oxidative stress. These effects were slightly highier in the obese group than in the normal-weight group. The regular exercise therapy decreased the serum Triglyceride levels more in the obese group than in the normal-weight group. However the improvement of the serum lipid profile may require a longer exercise period than eight weeks. The results show that the exercise therapy was overall more effective in the obese group than the normal-weight group.

The Effect of Dietary Modified Potato Starch By Chemically Denatured Treatment and Potato Starch on the Weight Loss, Lipid Metabolism and Redox Antioxidant System in High Fat Diet-Induced Obese Rats (화학적 변성 및 생감자 전분이 고지방식이로부터 유도된 비만 흰쥐의 지질대사 및 항산화계에 미치는 영향)

  • For the first 42 days, we made rats obese by feeding them potato starch instead of corn starch and after that we fed them transformed potato starch by 4 groups for 70 days. The 4 groups are GPS group, SPS group, EZ group and H40 groups and each were fed normal potato, small potato, enzyme treated potato, and acid treated potato starches, respectively. We determined body weight and feeding efficiency, lipid profiles in serum, lipid peroxidation in tissues and redox antioxidant system as GSH and GP-x in vivo. As a result, there was no difference in the increment of body weight in the groups of GPS, EZ and H40. Therefore EZ group showed lower body weight increment than other groups. While GPS group and SPS group did not show significant difference in blood glucose, cholesterol level, LDL-cholesterol and TC, and their measured values were lower than those of EZ and H40 groups. No significant difference was found in HDL-cholesterol level except for GPS group. Furthermore, when calculating atherogenic index (AI) by HDL-cholesterol and TC contents, H40 group showed higher measured value than other groups. When measuring the lipid peroxidation in serum, kidney and liver tissues, the serum lipid peroxidation in H40 group was higher than others. In the tissue of liver and kidney, EZ and H40 groups showed significantly lower contents than others. The content of GSH showed different tendency in each tissue, but the measured value of GP-x activity was lower in SPS group.

Effects of Antioxidant Nutrient Supplementation on the Lipid Peroxidation and Antioxidative Enzyme Activities in Patients with Coronary Heart Disease

  • This study was carried out to evaluate whether antioxidant nutrient suppplementation with $\alpha$-tocopherol, vitamin C, $\beta$-carotene, and selenium reduces the lipid peroxide levels and increases the antioxidative enzyme activities in patients with coronary hart disease. Eighty nine patients participated in a randomized, double-blind, placebo-controlled trial. The antioxidant group (45 patients) was given daily doses of $\alpha$-tocopherol (400 IU), vitamin C (50 mg), $\beta$-carotene (15 mg), and selenium (50 $\mu\textrm{g}$) and forty four patients received a placebo. Thirty eight subjects (84.4%) of the antioxidant group and thirty nine subjects (88.6%) of the placebo group completed the three-month supplementation. Serum levels of tocopherol, vitamin C and $\beta$-carotene significantly increased in the antioxidant group compared with the baseline (p<0.05). Thiobarbituric acid-reactive substances(TBARS) decreased significantly (0.6 nmol MDA/mL) in the antioxidant group compared with that (0.09 nmol MDA/mL) in the placebo group (p=0.03). However, antioxidant supplementation did not affect the level of oxidized-LDL measured as autoantibodies against oxidized-LDL. The superoxide dimutase activity in red blood cells increased in the antioxidant group compared with the baseline (p<0.05). However, glutathione peroxidase activities did not change after supplementation in both groups, and catalase activity significantly decreased in the placebo group (p<0.05). These results suggest that antioxidant supplementation for 3 months with $\alpha$-tocopherol, vitamin C, $\beta$-carotene and selenium in patients with coronary heat disease may be partially protective against oxidative stress.

Investigation of the Protective Effect of Kefir against Isoproterenol Induced Myocardial Infarction in Rats

  • This study aims to investigate the protective effects of kefir against myocardial infarction induced by isoproterenol (ISO). The rats were randomly divided into 4 groups, each group consisting of 8 rats. The control group, the kefir group (5 mL/kg/d kefir administered to rats as intra-gastric gavage for 60 d), the ISO group (100 mg/kg ISO was administered to rats, s.c. on 61. and 62. d), and kefir+ISO group (5 mL/kg/d kefir was administered to rats intra gastric gavage for 60 days prior to ISO, 100 mg/kg in two doses on day 61 and 62). 12 h after the last ISO dose, all rats were decapitated and their blood samples were collected. Cardiac tissue was reserved for histopathological examination. creatine kinase (CK), alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), triglycerides, total cholesterol,very low density lipoprotein (VLDL), low density lipoprotein (LDL), high density lipoprotein (HDL) and glucose were measured by autoanalyzer, whole blood malondialdehyde (MDA), glutathione (GSH) and plasma advanced oxidation protein products (AOPP) levels were measured spectrophotometrically. It was determined that in the group of kefir+ISO, the levels of AST (p<0.001), CK (p<0.001), LDH (p<0.001), MDA (p<0.001) and AOPP (p<0.001) were decreased, while the GSH (p<0.05) increased, compared to ISO group. There were no significant changes in lipid profile and glucose levels between these two groups. In conclusion, by examining cardiac enzymes and histopathological changes in cardiac tissue, it can be concluded that the administration of kefir in myocardial infarction induced by ISO can protect the heart with its antioxidant characteristic and minimize the toxic damage created by ISO.

Hypoglycemic and Antioxidative Effects of Dietary Sea-Tangle Extracts Supplementation in Streptozotocin-Induced Diabetic Rats (Streptozotocin-유발 당뇨쥐에서 다시마 추출물 첨가식이의 항당뇨 및 항산화 효과)

  • The purpose of this study was to investigate the effect of dietary sea-tangle extracts on blood glucose levels, serum lipid levels, thiobarbituric acid reactive substance (TBARS) and glutathione enzymes in diabetic rats treated with streptozotocin (STZ) Four groups of rats (Sprague-Dawley male rats, 180 - 200g) were consisted of normal rats fed control diet (C), diabetic rats fed control diet (CD), normal rats fed sea-tangl extracts diet (E), and diabetic rats fed sea-tangle extracts diet (ED). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W.). After 7 weeks, rats were sacrificed, serum glucose, serum total cholesterol, triglyceride levels and glutathione enzymes were measured. Urine was significantly higher in CD and ED groups than those of others (p < 0.05). Levels of amylase, calcium, uric acid, hemoglobin, cholesterol and low density lipoprotein (LDL)-cholesterol were different among four groups. But high density cholesterol (HDL)-cholesterol of ED group was significantly higher (p < 0.05) than other groups (C and E group) And the weekly change of serum glucose was decreased in the 3th,4th and 5th weeks. But serum triglyceride (TG) of diabetic rats fed sea-tangle extracts diet (ED) was lower than diabetic rats fed control diet (CD). Activity of hepatic microsomal G6Pase was significantly increased CD and ED groups higher than C and E group, but kidney was decreased ED group. Hepateic glutathione S-transferase (GST) of CD and ED group were significantly lower than C and E group (p<0.05), glutathione peroxidase (GPX) of E and ED group were significantly higher than C and CD group (p<0.05), glutathione reductase (GR) activities of ED group was significantly lower than other groups, malondialdehyde (MDA) of ED was lower than E and CD group, but kidney was increased significant in ED group compared to liver. These results suggested that dietary sea-tangle extracts reduce .hepatic disorders such as oxidant than kidney. In conclusion, dietary sea-tangle extracts groups reduced blood TG and hepatic MDA levels in STZ-induced diabetic rats.

The Anti-oxidative and Immune-regulatory Effect of Chungsimyeonja-tang in Aged Rat (태음인(太陰人) 청심연자탕(淸心蓮子湯)이 SD rat 의 항산화(抗酸化) 및 면역조절효과(免疫調節效果)에 대한 실험적 연구)

  • 1. Objectives The purpose of this study is to investigate the anti-oxidative and immune regulatory effect of oral administration of Chungsimyeonja-tang decoction in aged rats. 2. Methods The rats in the normal group were 14 weeks old, and those in the control and Chungsimyeonja-tang groups were 40 weeks old. The rats in Chungsimyeonja-tang group were orally treated with chungsimyeonjatang decoction daily for 2 months. To observe the anti-oxidative effect and immune regulatory effect of Chungsimyeonja-tang, we performed blood chemistry analysis, histological analysis. 3. Results and Conclusions 1) The number of WBC in blood was significantly increased and the number of PLT in blood was significantly decreased by oral administration of Chungsimyeonja-tang decoction. 2) The levels of albumin, LDL cholesterol, ALP and GOT in serum were significantly decreased, and the level of glucose in serum was significantly increased by oral administration of Chungsimyeonja-tang decoction. 3) The concentrations of glutathione and catalase were significantly increased and the concentrations of NO and MDA were significantly decreased in the liver of early aged rats by oral administration of Chungsimyeonja-tang decoction. 4) The concentrations of SOD and catalase were significantly increased and the concentration of MDA was significantly decreased in the lung of aged rats by oral administration of Chungsimyeonja-tang decoction. 5) The levels of IL-4 and IL-6 were significantly decreased while the levels of IFN-${\gamma}$ were significantly increased in the spleen of early aged rats by oral administration of Chungsimyeonja-tang decoction.

A Study on Nutrient Intakes and Hematological Status in Women of Child-Bearing Age Comparison between Non-Pregnant and Pregnant Women (가임기 여성의 영양섭취상태 및 혈액성상 : 비임신여성과 임신여성의 비교)

  • This study was conducted to evaluate nutritional status of Korean women of child-bearing age. A comparison was made between 113 non-pregnant and 200 pregnant women. Pregnant women of 24-28 weeks of gestation were recruited from prenatal clinic in Seoul. Nonpregnant women were included college students, nurses, office employers, and sales women. General characteristics, anthropometric measurements, dietary intakes, and hematological values were assessed. There was no difference in mean BMI for nonpregnant and pregnant (pre-gravid) women, which were 20.2 kg/m$^2$and 20.6 kg/m$^2$respectively. The pregnant women showed a tendency of better health-caring behavior, evidenced by stop smoking and drinking, and in taking nutrient supplements. The mean intakes of energy, vitamin A, vitamin B$_2$, calcium, and iron did not meet Korean RDA for adult women. In particular, calcium and iron intakes of both nonpregnant and pregnant women were under 60% of the Korean RDA. Judging by MDA score, dietary quality was better in pregnant than in nonpregnant women. Mean serum values of albumin, total cholesterol, LDL-cholesterol, HDL-cholesterol, Hb, IgA, IgG were in normal range. However, more than 13% of the nonpregnant subjects showed mild hypercholesterolemia (cholesterol > 200 mg/dl) and anemia (Hb < 12.0 g/dl) . The percentage of anemia in pregnant women were much higher; the subject with Hb < 11.0 g/dl were approximately 30%. The pregnant subjects showed significantly higher serum concentrations of triglycerides and cholesterol, and significantly lower concentrations of albumin, IgA, and IgG compared to nonpregnant women. Correlation analysis showed that Hb concentrations were correlated with the intakes of iron- and protein- containing foods such as meat and vegetables. This study strongly suggest that iron-deficient anemia is a major nutrition problem in Korean child-bearing women and this condition is correlated with dietary intakes.

Antioxidative Activity and Lipid Composition from Different Part and Supplement of Codonopsis lanceolata in Rat (더덕 부위별, 첨가수준이 실험쥐의 항산화활성과 지질조성에 미치는 효과)

  • This study was conducted to examine antioxidative activity and lipid composition from different parts and supplement flesh and skin of Codonopsis lanceolata in vivo. Forty six-week-old white Sprague Dawley rats were divided into 5 groups and fed with experimental diet for six weeks to measure antioxidant enzymes activities and lipid composition in blood and liver microsome. The activity of glutathione peroxidase in blood was high in all groups supplemented with Condonopsis lanceolata and the difference was observed in accordance with the supplemented part rather than the supplemented level. However, glutathione reductase activity and the content of malondialdehyde (MDA) in blood showed difference depending on the level of supplementation rather than the supplemented part. The content of liver MDA in all groups supplemented with Condonopsis lanceolata was lower than that in the control group. As the level of skin supplementation increased, an increase in glutathione peroxidase activity was also observed. Only in the group that 5% of Condonopsis lanceolata skin was supplemented, the glutathione reductase activity was higher than in the control group. Total cholesterol and LDL-cholesterol of blood in the group supplemented with Condonopsis lanceolata flesh or skin were significantly lower than those in control group. HDL-cholesterol in blood was high when the flesh of Condonopsis lanceolata was supplemented. Total cholesterol and triglyceride in liver of the group supplemented with Condonopsis lanceolata flesh or skin were significantly lower than those in control group. In summary, this animal test showed that the supplementation of Condonopsis lanceolata, flesh or skin, generally improved the antioxidative effect of diet and lipid composition.

Effects of Chunglijagam-tang Aqueous Extracts on Streptozotocin-induced Rat Diabetes and Related Complications (청리자감탕이 Streptozotocin으로 유발(誘發)된 흰쥐의 당뇨병(糖尿病) 및 당뇨병합병증(糖尿病合倂症)에 미치는 영향)

  • Objective : The object of this study was to observe the effects of aqueous extracts of Chunglijagam-tang(CLJGT), which has traditionally been used in Korean medicine for treating various diseases, on streptozotocin(STZ)-induced rat diabetes and related complications: diabetic nephropathy, hepatopathy and hyperlipemia. Methods : CLJGT extracts were orally administered once a day for 28 days at a dosage 50, 100 and 200mg/kg from 21 days after STZ treatment, and the changes on body weights, blood glucose levels, kidney and liver weights, serum BUN(blood urea nitrogen), creatinine, AST(aspartate transaminase), ALT(alanine aminotransferase), HDL(high-density lipoprotein), LDL (low-density lipoprotein), total cholesterol and triglyceride levels were observed with pancreatic malondialdehyde(MDA) and glutathione(GSH) contents. The results were compared with silymarin 100mg/kg. Results : Significant decrease of blood glucose levels, kidney and liver weights, serum BUN, creatinine, AST, ALT, LDL, total cholesterol, triglyceride levels, pancreatic malondialdehyde contents and significant increase of body weights, serum HDL levels, pancreatic glutathione contents were detected in CLJGT extracts 50, 100 and 200mg/kg administered groups as compared to the STZ control group. Conclusion : CLJGT extracts showed favorable effects on the STZ-induced diabetes and related complications mediated by their antioxidant effects as similar to silymarin. Therefore, it is expected that DBEH has potential for use in the management of diabetes and various diabetic complications.
