Malondialdehyde, Glutathione, and Nitric Oxide Levels in Toxoplasma gondii Seropositive Patients

  • The aim of this study was to investigate the difference in the serum malondialdehyde (MDA), glutathione (GSH), and nitric oxide (NO) levels between normal and T. gondii-infected patients. To this end, MDA, GSH, and NO levels in the sera of 37 seropositive patients and 40 participants in the control group were evaluated. In Toxoplasma ELISA, IgG results of the patient group were $1,013.0{\pm}543.8$ in optical density (mean ${\pm}$ SD). A statistically significant difference was found between patients and the control group in terms of MDA, GSH, and NO levels. A decrease in GSH activity was detected, while MDA and NO levels increased significantly. Consequently, it is suggested that the use of antioxidant vitamins in addition to a parasite treatment shall prove useful. The high infection vs control ratio of MDA and NO levels probably suggests the occurrence as a mechanism of tissue damage in cases of chronic toxoplasmosis. Moreover, it is recommended that the patient levels of MDA, GSH, and NO should be evaluated in toxoplasmosis.

조경수목(造景樹木)의 대기오염물질(大氣汚染物質)에 대한 피해반응(被害反應)(III) - Malondialdehyde 함량(含量)을 중심으로 - (Injury Responses of Landscape Woody Plants to Air Pollutants - Malondialdehyde content -)

  • 대기오염물질(大氣汚染物質)에 의한 조경수목(造景樹木)들의 피해(被害)의 감수성(感受性) 정도를 조사하기 위하여 서울, 대전 등 대도시(大都市) 지역과 광릉(光陵)지역을 선정하고 소나무, 잣나무, 은행나무, 튜립나무 및 양버즘나무 잎을 채취하여 MDA 함량(含量)을 측정하였다. 또한 소나무, 잣나무, 스트로브잣나무, 은행나무 및 튜립나무에 무처리(無處理), 0.5, 1.5 및 2.5 ppm의 $SO_2$ 가스를 각각 하루에 4시간(時間)씩 6일간(日間) 처리(處理)한 후, 잎의 MDA 함량(含量)을 측정(測定), 분석(分析)하였다. 은행나무, 양버즘나무 및 튜립나무의 MDA 함량(含量)은 비오염지역(非汚染地域)보다 오염지역(汚染地域)에서 높았으며, 조사대상(調査對象) 수종중(樹種中) 튜립나무의 MDA 함량(含量)이 제일 높았다. 소나무와 잣나무에서는 엽령(葉令)이 높아질수록 MDA 함량(含量)이 증가하였다. $SO_2$, 처리시(處理時) 튜립나무는 처리농도(處理濃度)가 높아질수록 MDA 함량(含量)이 증가하였으나 다른 수종에서는 처리농도(處理濃度)에 따라 큰 차이(差異)가 나타나지 않았다. $SO_2$를 6일간(日間) 처리(處理)했을 때 5수종(樹種) 모두 처리일(處理日)이 경과함에 따라 MDA 생성(生成)이 증가하였으며, 특히 튜립나무는 MDA 생성(生成)이 뚜렷하게 높았으며, 처리(處理)개시 후 점정 증가하여 처리(處理) 4일 만에 최대치(最大値)를 나타내다가 그 후에는 감소하였다.

일산화탄소 폭로후 고압산소 투여가 흰쥐 신장에서의 malondialdehyde 함량과 catalase 및 superoxide dismutase 활성에 미치는 영향 (Effects of Hyperbaric Oxygen Treatment on the Malondialdehyde Level and Activities of Catalase and Superoxide Dismutase in the Kidney of the Rats Exposed to Carbon Monoxide)

  • In an attempt to define the effects of hyperbaric oxygen treatment on the lipid peroxidation and oxygen free radical reactions in rats exposed to carbon monoxide, we studied malondialdehyde (MDA) level and activities of catalase and superoxide dismutase in the kidney of the rats exposed to carbon monoxide. Male Sprague-Dawley albino rats weighing 240 to 260 gm were used. Experimental groups consist of Control group (=breathing with air), HBO group (=exposed to hyperbaric oxygen 〔HBO, 3ATA, 100%〕 after air breath), CO group (=exposed to CO〔3,970 ppm〕after air breath), CO-Air group (=exposed to CO after air breath followed by air breath) and CO-HBO group (=exposed to CO after air breath followed HBO treatment). The CO group showed significantly higher MDA level, catalase activity and SOD activity as compared to that of control group. The CO-HBO group showed significantly lower MDA level as compared to that of CO group, and did not show significantly lower catalase activity and SOD activity as compared to that of CO group. These results suggest that the excessive oxygen free radicals is an important determinant in pathogenesis of CO-induced nephrotoxicity and HBO inhibits the lipid peroxidation caused by excessive oxygen free radicals in the kidney of the rats exposed to carbon monoxide.

Stability and Intra-Individual Variation of Urinary Malondialdehyde and 2-Naphthol

  • Objectives : Malondialdehyde (MDA), a lipid peroxidation by-product, has been widely used as an indicator of oxidative stress. Urinary 2-naphthol, a urinary PAH metabolite, is used as a marker of ambient particulate exposure and is associated with lung cancer and chronic obstructive pulmonary disease. However, the stability and intra-individual variation associated with urinary MDA and 2-naphthol have not been thoroughly addressed. The objective of this study was to assess the stability and intra-individual variation associated with urinary MDA and 2-naphthol. Methods : Urine samples were collected from 10 healthy volunteers (mean age 34, range $27{\sim}42$ years old). Each sample was divided into three aliquots and stored under three different conditions. The levels of urinary MDA and 2-naphthol were analyzed 1) just after sampling, 2) after storage at room temperature ($21^{\circ}C$) for 16 hours, and 3) after storage in a $-20^{\circ}C$ freezer for 16 hours. In addition, an epidemiological study was conducted in 44 Chinese subjects over a period of 3 weeks. The urinary MDA and 2-naphthol were measured by HPLC three times. Results : There was no difference in the levels of urinary MDA and 2-naphthol between the triplicate measurements (n=10, p=0.84 and p=0.83, respectively). The intra-class correlation coefficients (ICC) for urinary MDA and 2-naphthol were 0.74 and 0.42, respectively. However, the levels of PM2.5 in the air were well correlated with the levels of both MDA and 2-naphthol in the epidemiological study. Conclusions : These results suggest that urinary MDA and 2-naphthol remain stable under variable storage conditions, even at room temperature for 16 hours, and indicate that these markers can be used in epidemiological studies involving various sample storage conditions. The intra-CC of urinary 2-naphthol and MDA were acceptable for application to epidemiological studies.

고지방식이에서 서로 다른 불포화 지방산 섭취가 쥐의 혈청과 조직의 Tocopherol함량과 지질과산화물 형성에 미치는 영향 (Effects of Dietary Polyunsaturated Fatty Acids on Tocopherol Contents and Lipid Peroxidation of Plasma and Tissues in Rats Fed High Fat Diet)

  • This study was observed the effect of n6 and n3 polyunsaturated fatty acids of dietary corn oil and fish oil which was supplemented with similar levels of tocopherol in high fat diet on the levels of tocopherol, malondialdehyde ( MDA) productions of plasma and tissues of rats. Also RBC hemolysis, superoxide dismutase (SOD) and glutathione peroxidase (GSH Px) activities In liver were determined. Male Sprague Dawley rats were fed high fat (40%Cal) diet which was different only In fatty acid composition for 6 weeks. Dietary (arts were beef tallow(BT) as a source of saturated fatty acid, corn oil(CO) for n6 linoleic acid (LA) and fish oil(FO) for n3 eicosapentaenoic acid(EPA) + docosahexaenoic acid (DHA). Plasma and liver tocopherol levels were lowered by n3 PUy4 but there was no difference in malondialdehyde(MDA) level by different dietary PUFA. However, MDA content of RBC and hemolysis were increased by n3 PUFA. MDA content, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in liver were increased in more unsaturated dietary fat groups. Especially, SOD activity was increased in proportion to the degree of fat unsaturation.

녹차 건분이 급성 알코올 투여받은 9개월령 흰쥐의 뇌 부위별 항산화능에 미치는 영향 (Antioxidative Effects of Green Tea Powder Diet Against Ethanol-Induced Oxidative Damage in 9 Month Old Rat Brain Regions)

  • 본 연구에서는 녹차 건분 식이가 뇌 조직에서 알코올에 의한 산화적 스트레스 감소효과를 나타내는 지를 규명하고 자, 생후 9개월령 Sprague-Dawley에게 1% 녹차 건분식이를 4주간 공급하고 희생 12시간 전메 급성으로 알코올을 투여한 후 뇌 조직을 cortex, cerebellum, striatum, hippocampus로 나누어 malondialdehyde(MDA)의 함량과 catalase. superoxide dismutase(SOD), glutathione peroxidase(GSH-Px)의 활성을 측정하였다. 본 실험 결과를 종합하여 볼 때. 1%녹차 건분 식이의 공급은 striatum과 hippocampus 내 MDA 함량을 유의적으로 낮추고 알코올에 따른 hippocampus 내 catalase 활성 증가를 유의적으로 억제하였다. 이러한 곁과는 녹차 건분의 공급이 알코올로 산화적 stress를 가한 동물에서 일부 뇌 조직의 지질 산화를 감소시키고 알코올에 의한 뇌 조직 손상으로부터 보호할 수 있음을 보여준다.

층층나무의 온존 내성 개체 선발 (Selection of Ozone Tolerant Individuals of Cornus controversa)

  • 본 연구는 층층나무를 대상으로, 엽 내에서 측정한 MDA 함량을 기준으로 오존 내성 개체를 선발하고자 수행하였다. 또한 각종 오염물질에 대한 내성을 평가 하기 위해 MDA 함량을 이용하는 것이 타당한지를 검토하고자 하였다. 실험에 사용된 층층나무는 오대산(5본), 치악산(5본), 점봉산(5본), 주왕산(1본), 태백산(1본), 지리산(5본), 소리봉(5본), 속리산(4본), 소백산(4본), 계방산(4본) 등 9개 지역에서 채취한 종자를 이용하였으며, 오존 처리는 100 ppb에서 하루 8시간 씩 10주 동안 실시하였다. 오존 처리가 종료된 후, 층층나무의 수고와 MDA 함량을 측정하였고, 수고와 MDA 함량간의 관계를 분석 하였으여, 수고와 MDA 함량의 표준화 지수를 이용하여 오존에 대한 내성그룹, 중간그룹, 민감성그룹을 각각 30개체씩 선발하였다. 오존 처리된 층층나무의 수고와 MDA 함량은 가계간, 지역간 차이를 보여주었으며, MDA 함량은 수고생장과 역상관(r=-0.531, p$\leq$0.001)을 나타냈다. 개체별 수고와 MDA 함량을 기준으로 선발한 층층나무는 내성그룹과 민감성 그룹간 수고와 MDA 함량 차이가 뚜렷하였다. 따라서 MDA 함량은 오존에 대한 내성 및 민감성을 구분하는데 유용하게 이용될 수 있을 것으로 판단된다.

Malondialdehyde and 3-Nitrotyrosine in Exhaled Breath Condensate in Retired Elderly Coal Miners with Chronic Obstructive Pulmonary Disease

  • Background: Chronic obstructive pulmonary disease (COPD) is an important cause of occupational mortality in miners exposed to coal mine dust. Although the inflammatory mediators involved in COPD have not been defined, many studies have shown that inflammatory mediators such as reactive oxygen and nitrogen species are involved in orchestrating the complex inflammatory process in COPD. Methods: To investigate the relevance of exhaled biomarkers of oxidative and nitrosative stress in participants with COPD, we determined the levels of hydrogen peroxide, malondialdehyde (MDA), and 3-nitrotyrosine (3-NT) in exhaled breath condensate (EBC) in 90 retired elderly coal miners (53 non-COPD and 37 COPD participants). Results: Mean levels of MDA (4.64 nMvs. 6.46 nM, p = 0.005) and 3-NT (3.51 nMvs. 5.50 nM, p = 0.039) in EBC were significantly higher in participants with COPD. The median level of MDA did show statistical difference among the COPD severities (p = 0.017), and the area under the receiver operating characteristic curve forMDA (0.67) for the diagnostic discrimination of COPD indicated the biomarker. The optimal cutoff values were 5.34 nM (64.9% sensitivity and 64.2% specificity) and 5.58 nM (62.2% sensitivity and 62.3% specificity) forMDA and 3-NT, respectively. The results suggest that high levels ofMDA and 3-NT in EBC are associated with COPD in retired elderly miners. Conclusion: These results showed that the elevated levels of EBC MDA and EBC 3-NT in individuals with COPD are biomarkers of oxidative or nitrosative stress.

수용성 계에서 Phenolic Acid류와 인삼 추출물이 갖는 산화방지 효과 (Antioxidant Effects of Phenolic Acids and Ginseng Extract in Aqueous System)

  • Linoleic acid 1% 완충용액에 3종의 phenolic acid류, 즉 caffeic acid, ferulic acid, vanillic acid 및 인삼 추출물에 대한 산화방지 효과를 비교물질로 ${\alpha}-tocopherol$을 사용하여 MDA (malondialdehyde) 정량법과 headspace oxygen analysis로 측정 비교 하였다. MDA 생성량으로 비교한 산화방지 효과는 $caffeic\;acid{\geq}ferulic\;acid>{\alpha}-tocopherol>$인삼 추출물>vanillic acid 순으로 나타났으며, 이때 산화저해율은 각각 63.5, 62.9, 52.3, 51.2, 5.6% 이었다. 첨가된 인삼 추출물의 농도는 700 ppm, 그외 phenolic aicd류 및 ${\alpha}-tocopherol$의 농도는 각각 100 ppm 이었다. 또한 인삼 추출물에 caffeic acid와 ${\alpha}-tocopherol$을 각각 100 ppm 첨가한 결과 91.2%의 산화저해율을 나타내었다. Headspace oxygen analysis 에 따른 결과에서는 MDA 생성량으로 비교한 산화방지 효과와 유사한 경향으로 나타났으며, ferulic acid>caffeic acid>${\alpha}-tocopherol$>인삼 추출물>vanillic acid의 순 이었고, 이때의 headspace 중 산소 함량은 18.56, 17.78, 17.17, 16.65, 15.95%로 각각 나타났다.
