  • In this paper, we first study some characterizations of left MC2 rings. Next, by introducing left nil-injective modules, we discuss and generalize some well known results for a ring whose simple singular left modules are Y J-injective. Finally, as a byproduct of these results we are able to show that if R is a left MC2 left Goldie ring whose every simple singular left R-module is nil-injective and GJcp-injective, then R is a finite product of simple left Goldie rings.

Karyotype Analysis and Physical Mapping of rDNAs Using McFISH in Jeffersonia dubia Benth (깽깽이풀의 핵형분석과 McFISH를 이용한 rDNA의 물리지도 작성)

  • Karyotype analysis and chromosomal locailization of 45S and 5S rDNAs using McFISH (multi-color fluorescence in situ hybridization) were carried out in Jeffersonia dubia Benth., which is one of medicinal plants belonging to Berberidaceae. The somatic metaphase chromosome number was 2n=2x=12 and the size of chromosomes ranged $1.95{\sim}3.50\;{\mu}m$. The chromosome complement consisted of two pairs of metacentrics (chromosomes 1 and 3), two pairs of submetacentrics (chromosomes 2 and 4) and two pairs of subtelocentrics (chromosomes 5 and 6). In McFISH, one pair of 45S rDNA site was detected on the centromeric region of chromosome 2 and three pairs of 5S rDNA sites were detected on the short arm of chromosomes 4, 5 and 6, respectively.

Association of MC4R Gene Polymorphisms with Growth and Body Composition Traits in Chicken

  • Genetic and pharmacological studies in mice have demonstrated a complementary role for the melanocortin 4 receptor (MC4R) in the control of food intake, energy balance and body weight. This study was designed to investigate the associations of a MC4R gene polymorphism on chicken growth and body composition traits in broiler lines divergently selected for abdominal fat. A SNP (G54C) was found in CDS region of chicken MC4R gene. The analysis of the least squares and variance revealed a significant association between the G54C SNP and BW, CW and SL at 7 wk of age, and there were significant differences in different genotypes (p<0.05). The results from protein secondary structure prediction and tertiary structure prediction showed that it appeared a helix in $13^{th}$ amino acid and two strands at $14^{th}$ and $15^{th}$ amino acid in mutant protein, respectively. It maybe induce the change of the activity or function of MC4R gene in poultry.

Effects of Genotype Mutation and Coat Color Phenotype on the Offspring from Mating System of MC1R Genotype Patterns in Korean Brindle Cattle (칡소의 MC1R의 유전자형에 따른 교배 조합이 자손의 모색과 유전자형 변이에 미치는 영향)

  • Bovine coat color is decided by the melanocortin receptor 1 (MC1R) genotype mutation and melanogenesis. Specially, in the various cattle breeds, dominant black coat color is expressed by dominant genotype of $E^D$, red or brown is expressed in the frame shift mutation of recessive homozygous e by base pair deletion and wild type of $E^+$ is expressed in various coat colors. However, not very well known about the effected of MC1R genotype mutation on the coat color through family lines in KBC. Therefore, this study were to investigate effect of MC1R genotype mutation on the coat color, and to suggest mating breed system in accordance with of MC1R genotype for increased on brindle coat color appearance. Parents (sire 2 heads and dam 3 heads) and offspring (total : 54 heads) from crossbreeding in KBC family line with the MC1R genotype and phenotype records were selected as experimental animals. The relationship between melanocortin 1 receptor (MC1R) genotypes expression verified by PCR-RFLP, and brindle coat color appearance to the family line of the cross mating breed from MC1R genotype pattern was determined. As a result, 4MC1R genetic variations, $E^+/E^+$ (sire 1), $E^+/e$ (sire 2 and dam 3), $E^+/e$ with 4 bands of 174, 207 and 328 bp (dam 1) and $E^+/e$ with 3 bands of 174, 207, 328 and 535 bp (dam 2) from parents (sire and dam) of KBC. However, 3 genetic variations, e/e (24%), $E^+/E^+$ (22%) and $E^+/e$ (56%) were identified in offspring. Also, brindle coat color expressrated was the e/e with the 0%, $E^+/E^+$ with 67% and $E^+/e$ with 77% from MC1R genotype in offspring on the cross mating of KBC. Furthermore, when the sire had $E^+/e$ genotype and the dam had $E^+/E^+$ with the 3 bands or $E^+/e$ genotype, and both had whole body-brindle coat color, 62% of the offspring had whole body-brindle coat color. Therefore, the seresults, the mating system from MC1R genotype patterns of the sires ($E^+/e$) and dams ($E^+/E^+$ with the 3 bands or $E^+/e$) with brindle coat color may have the highest whole body-brindle coat color expression in their offspring.

Relationships between solar/interplanetary (IP) parameters and Dst index, according to IP sources

  • We have investigated interplanetary (IP) structures of 82 intense geomagnetic storms (Dst $\leq$ -100 nT) that occurred from 1998 to 2006. According to their interplanetary origins, we classified them as four groups: 20 sMC events (IP shock and MC), 19 SH events (sheath field), 12 SH+MC events (Sheath field and MC), and 8 nonMC events (non-MC type ICME). For each group, we examined the relationships between Dst index and solar/IP parameters, namely, direction parameter (DP), CME speed ($V_{CME}$), solar wind speed ($V_{SW}$), minimum of IMF $B_z$ component($Bz_{min}$), and maximum of $E_y$ component ($Ey_{max}$).We found that the relationships strongly depend on their IP source. Our main results can be summarized as follows: 1) The correlation between Dst and DP is the best for the SH+MC events (r = -0.61). 2) The relationship between Dst and $V_{CME}$ gives the best correlation for the sMC events (r = -0.56). 3) There is the best correlation between Dst and $V_{SW}$ for the sMC events (r = -0.61), while there is a very weak correlation (r=-0.17) for the SH events. 4) The relationship between Dst and $Bz_{min}$ gives the best correlation (r = -0.87) for the SH+MC events. 5) The correlation between Dst and $Ey_{max}$ is the best for the SH+MC events (r = -0.87). Summing up, the sMC and SH+MC events give us good correlations, but the SH events, weak correlations. From this study, we suggest that this tendency should be caused by the characteristics of IMF southward components, e.g., smooth field rotations for the MC events and highly IMF fluctuations for the SH events.

New Evidences of Effect of Melanocortin-4 Receptor and Insulin-like Growth Factor 2 Genes on Fat Deposition and Carcass Traits in Different Pig Populations

  • The Melanocortin-4 Receptor (MC4R) and Insulin-like Growth Factor 2 (IGF2) are two important candidate genes related to fat deposition and carcass traits. MC4R was found on study on human obesity and then was studied as candidate gene affecting food intake and fat deposition traits in mice and pigs. Insulin-like Growth Factor 2 (IGF2) gene plays an important role on tumor cell proliferation and muscle growth. It also affects fat traits and live weight in pigs. In this paper, MC4R and IGF2 were studied as two candidate genes associated with important economic traits such as fat deposition and carcass traits in five different pig populations. Taq I-PCR-RFLP and Bcn I-PCR-RFLP were respectively used to detect the polymorphism of genotypes of MC4R and IGF2 genes. Different MC4R genotype frequencies were observed in four populations. IGF2 genotype frequencies were also different in two populations. The results of association analysis show both MC4R and IGF2 genes were significantly associated with fat deposition and carcass traits in about 300 pigs. This work will add new evidence of MC4R and IGF2 affecting fat deposition and carcass traits in pigs and show that two genes can be used as important candidate genes for marker assistant selection (MAS) of growth and lean meat percentage in pigs.

Effects of ADCYP1R1, FABP3, FABP4, MC4R, MYL2 Genotypes on Growth Traits in F2 Population Between Landrace and Jeju Native Black Pig (제주재래흑돼지와 Landrace의 F2 집단에서 ADCYP1R1, FABP3, MC4R, MYL2 유전자형이 성장형질에 미치는 효과)

  • Genetic polymorphisms was investigated at five single nucleotide polymorphisms(SNP) sites in four porcine genes(ADCYAP1R1, FABP3, MC4R, and MYL2) and analyzed their statistical association with growth traits in F2 reciprocal-crossbred population between Landrace and Jeju native black pig(JNP). All populations, JNP, Landrace and their F2 were polymorphic for all five SNP loci tested, however, the homozygote T/T of FABP3 g.-158T>C and the homozygote G/G of ADCYAP1R1 intron 2 337A>G were not found in JNP and Landrace, respectively. The genotypes of ADCYAP1R1 were significantly associated with body weights(BW) at 3 weeks and at 20 weeks(P<0.05), respectivley, those of FABP3 g.-135delT were associated with late average daily gain(LADG) (P<0.01), and those of FABP3 g.-158T>G were associated with body weights during late growth period such as, BW20 and LADG(P<0.01). Those of MC4R were also significantly associated with BW10 suggesting by the difference of early average daily gain(EADG) (P<0.05), and with LADG(P<0.01). The body length of F2 animals was affected by the genotypes of ADCYAP1R1, MC4R, and MYL2(P<0.05), respectively. Among these, MC4R A/A homozygotes showed over 3 cm longer in body length than those of other genotypes. As the useful basic information, these results suggested that SNP markers showing statistical association with growth traits and the results help to select the sires of JNP for improving the productivity in JNP-related crossbreeding system in pig industry and also to construct the molecular breeding system for breed improvement of JNP itself.

Analysis of Melanocortin receptor 1 (MC1R) gene differential test for beef species between Hanwoo and Holstein using polmerase chain reaction -restriction fragment length polymorphism (PCR-RFLP) (MC1R gene의 PCR-RFLP를 이용한 한우.젖소고기 감별)

  • The objective of this study was to differentiate the beef species between Hanwoo and Holstein from a total of 1,081 beef samples using PCR-RFLP of MC1R gene. When a PCR product of 403 bp specific band amplified from bovine MC1R gene sequence was digested with restriction enzyme MspA1I, Hanwoo type showed 2 bands, 220 bp and 183 bp size bands. Holstein type, however, showed three bands, 220 bp, 138 bp and 45 bp size band, respectively. The results of the differential test for beef species were as following; 7 samples (0.64%) were determined to Holstein type, of which 4 were submitted from administrative authorities, other 3 from self-collection planing, and none from civilian clients including school.

  • ${\alpha}$-Melanotropin (Ac-Ser-Tyr- Ser-Met-Glu$\^$5/-His-Phe-Arg-Trp-Gly$\^$10/-Lys-Pro-Val-NH$_2$) is one of the first peptide hormones to be isolated and have its structure determined. It was early recognized to have essentially the same N-terminal tridecapeptide sequence as adrenocorticotropic hormone (ACTH) except that the N-terminal was acetylated in the case of ${\alpha}$-MSH but not in the case of ACTH, indicating that their biosyntheses were different (Figure 1). Subsequently it was discovered that ${\alpha}$-MSH and ACTH were derived from the same gene, currently referred to as proopiomelanocortin (POMC). Its original bioactivity was pigmentation, but it also was recognized that it may have activity in the central nervous system, though the precise nature of these central activities have been controversial. The recent cloning and expression of five melanocortin receptors, with the MC3 and MC4 receptors found primarily in the brain and the MC5 receptor (MC5-R) found throughout the body, has provided new impetus to understand the structure-activity relationships of ${\alpha}$-MSH at these receptors. The effects of ${\alpha}$-MSH on pigmentation are mediated by the MC1-R expressed specifically on the surface of melanocytes. Similarly the MC2-R is involved in the regulation of adrenal steroidogenesis by ACTH. However, given the complexity of expression of the MC3, MC4, and MC5 receptors, it has not been possible to identify any simple correlations between these receptors and the reported biological activities of the melanocortin peptides. Consequently, potent and receptor specific agonists and especially antagonists would be extremely valuable tools for the determination of the physiological roles of the MC3, MC4, and MC5 receptors. Though the extensive structure-activity relationships have provided much information on agonist activity related to pigmentary effects, only recently has it been possible to begin to systematically develop potent and selective antagonists.

Identification of Beef Breed using DNA Marker of Coat Color Genes (모색 발현 유전자의 DNA Marker를 이용한 쇠고기 품종 판별)

  • In Korean beef market, one of the major problems is mislabeling or fraudulent distribution of Holstein dairy meat or imported beef as domestic Hanwoo meat. Therefore, there has been a great need for a development of technology to identify beef breeds in meat and meat products. This study was carried out to develop the accurate and reliable method for the identification of beef breed using PCR-RFLP marker of MC1R, MGF and TYRPl genes affecting coat colors in cattle. A single base substitution (G\longrightarrowT transition) at the codon for amino acid position 104 of MC1R gene was identified between Hanwoo and Holstein and Angus breeds. The change at this position creates Msp I restriction site in Holstein and Angus, but not in Hanwoo. When the DNA amplified products (537 bp) was digested with Msp I, Hanwoo meat showed a single band of 537bp, while two fragments of 329bp and 208 bp were observed in Holstein meat and Angus breed, respectively. Thus, breed-specific RFLP marker in the MC1R gene can be used to distinguish between Hanwoo meat and Holstein and Angus meats. In the RFLP genotype of MGF gene, the frequency of r/r type was 75% in Manwoo, whereas the frequency of R/R was 80% in Hereford breed. Holstein and Angus breeds showed 100% for R/r type. Therefore, Hanwoo meat showed significant difference in the MGF genotype frequencies compared with those of Holstein meat and imported beef cattle breeds. However, TYRP1 gene showed the same genotype in all breeds examined. Thus, this TYRP1 gene can not be used as a molecular marker for breed identification. As a consequence, we suggest that RFLP markers of the MC1R and MGF coat color genes could be used as DNA marker for identification of Hanwoo meat from Holstein and imported meats.
