Bactericidal Activity of DW286, a New Fluoroquinolone Antibiotic (새로운 퀴놀론 항균제 Dw286의 살균작용)

  • The bactericidal activities of DW286, a new fluoroquinolone were investigated by comparing the minimal bactericidal concentrations (MBCs) and the time-kill curve of it against some Gram-positive and Gram-negative bacterial strains. The MBCs of DW286 for the strains tested were either equal to or two-fold higher than the MICs, as were observed for the other fluoroquinolones. And DW286 exhibited rapid killing curves against the strains. Accordingly, it could be said that DW286 has bactericidal activity comparable to other fluoroquinolones.

Antimicrobial Susceptiblity of Brucella canis Isolated from Korea (국내 분리 Brucella canis의 항균제 감수성)

  • Little is known to data about the in vitro activity of antimicrobial agents aganist Brucella cams (B cams) isolated from Korea. Our study aimed at determining the in vitro activities of 15 antimicrobial agents against 3 isolates and 52 isolates of B cams from dogs in 1994 and 2002, respectively. In minimal inhibitory concentration (MIC) study, minocycline and doxycycline showed the lowest MICs ( < 0.06-0.5 ug/ml). Gentamicin, streptomycin, ciprofloxacin, norfloxacin and rifampin showed MICs in the range of less than 1 ug/ml. Lincomycin and sulfisox azole showed the highest MICs ( > 32 ug/ml). Interestingly, MICs of macrolides (erythromycin, spiramycin, tylosin) against 52 isolates in 2002 were 16-64 times higher than that of 3 isolates in 1994. In minimal bactericidal concentration (MBC) study, gentamicin, streptomycin, ciprofloxacin and norfloxacin showed the lowest MBCs [0.12-1 ug/ml (1-2 times higher than MIC)], but minocycline and doxycycline showed the highest MBCs [8-32 ug/ml (128 times higher than MIC)]. Rifampin showed the MBCs in the range from 2 to 4 ug/ml (2-4 times higher than MIC).

Bactericidal Activities of DW-116, a New Quinolone (새로운 퀴놀론 항균제 DW-116의 살균 작용)

  • The bactericidal activities of DW-116, a new fluoroquinolone was estimated by comparing the minimal bactericidal concentrations (MBCs) of it against some Gram-positive and -negative bacterial strains with the minimal inhibitory concentrations (MICs). The MBCs against the test organisms were equal to or two times higher than MICs. The results support that the antibacterial activity of DW-116 is bactericidal.

Opening Status of the Korea Midwifery Birthing Centers and Development of Midwifery Practice Guideline (한국 조산원 개원실태 및 조산직무지침 개발)

  • Purpose: This study was to investigate the operational status of the midwifery birthing centers (MBCs) and midwives' job status (Phase 1) and to develop midwifery practice guidelines (MPG) (Phase 2) in Korea. Methods: In the first phase, the subjects were 15 midwives who operated 11 of 14 MBCs that were opened as of August 2018. The questionnaire consisted of items to measure the operational status of the MBC and midwives' job status. In the second phase, the MPG was developed from literature review, interviews with five midwives opening their MBCs, surveys with 74 midwives, and a validity evaluation conducted by seven experts. Results: The distribution of operating MBCs was five in Gyunggi-do, two each in Seoul and Incheon, one each in Busan, Chungcheongbuk-do, Gyeongsangbuk-do, Gyeongsangnam-do and Jeju-do. The mean age of midwives was 54.3 and all were female. In 2017, a total of 762 births including 81 homebirths were performed by midwives. The job performance was highest in the order of neonatal care 3.81, childbirth care 3.56, and postpartal care 3.53, respectively. The MPG included seven areas of prenatal care, childbirth care, postpartal care, neonatal care, primary health care, law/ethics, and administration, with 56 tasks and 166 task elements. Conclusion: This study provides the valid basic data for the operational status of the MBC and the midwives' job status. The MPG describes the midwife's job and may be used as basic data for preparing policies for the development of midwifery practice in Korea.

A Study of P/2010 A2 Dust Cloud : Possibly Impact Triggered Dust Particles

  • Main-belt comets (hereafter MBCs) are one of the hottest topics in the solar system astronomy. They are objects orbiting in the main asteroid belt which show cometary activity. Unlike most comets, which spend most of their orbit beyond 5AU from the Sun, MBCs follow near-circular orbits within the asteroid belt that are indistinguishable from the orbits of major population of the asteroids. P/2010 A2, the fifth MBC, was discovered by on January 6, 2010 by Lincoln Near-Earth Asteroid Research. It passed its perihelion at 2.01AU on December 3, 2009, about a month before it was discovered. With an aphelion of only 2.6 AU, P/2010 A2 spends all of its time inside of the frostline ~2.7 AU. We made observations of P/2010 A2 with Nishi-Harima Astronomical Observatory 2-m telescope only a week after the discovery. From the observed images, we found that the dust cloud was composed of large particles (>1mm) impulsively ejected between March and June, 2009. No coma was detected by our observations, suggesting that this object was no longer active. Consequently, we conjecture that these dust particles could be released by the impact collision among asteroids.

Antimicrobial Activity of Continentalic Acid from Aralia cordata Against Enterococcus Strains

  • Continentalic acid (CA, (-)-pimara-8(14), 15-diene-19-oic acid) was isolated from the roots of Aralia cordata (Araliaceae) using bioassay-guided fractionation of a crude chloroform extract. The antibacterial activity of CA against Enterococcus faecalis and Enterococcus gallinarium was estimated by determining minimum inhibitory concentrations (MICs) and minimum bactericidal concentrations (MBCs). CA exhibited potent activity against standard vancomycin-resistant enterococci (VRE) and vancomycin-susceptible enterococci (VSE), with MICs and MBCs values between 4 and $8{\mu}g/mL$ and 4 and $16{\mu}g/mL$, respectively. This compound exhibited potent activity against strains of VRE, which are highly resistant to clinically useful antibiotics. These findings suggest that continentalic acid may be useful in controlling enterococcal infection.

A Study on Changes in Antibacterial Activity of Pepsin-hydrolyzed Bovine Apo-lactoferrin at Various Method for Pasteurizations and pH Values (살균방법 및 pH 조건에 따른 Pepsin-hydrolyzed Bovine Apo-lactoferrin의 항균성 변화에 관한 연구)

  • This study was carried out to examine that pepsin-hydrolyzed bovine lactoferrin has applicabilities which are market milk and dairy products. The stability of pepsin-hydrolyzed bovine apo-lactoferrin and the change of its antibacterial character has been studied under various method for pasteurization (LTLT; 65$^{\circ}C$ / 30min., HTST ; 75$^{\circ}C$ / 15sec., UHT ; 135$^{\circ}C$ / 3sec.) and pH Values (pH 2.0, pH 4.0, pH 6.8). The ehated samples were assayed for minimal bacteriocidal concentrations (MBCs) and bacteriocidal effect against E. coli. The results obtained were summarized as follows: After fractionation of pepsin-hydrolyzed bovine lactofeerin by gel filtration. several peptide fractions were found that had strong antibacterial activity. SDS-PAGE showed that the one of these fractions with strong antibacterial activity, which had a molecular mass a range of 30∼33KDa. The MBCs for pepsin-hydrolyzed bovine lactoferrin fraction No. 2 against E. coli required to cause complete inhibition of growth varied within the range of 200∼400 $\mu\textrm{g}$/ml, depending on heat treatments and pH conditions. The peptide fraction No. 2 showed strong bacteriocidal activity against E. coli at LTLT and HTST treatments under acidic pH conditions. and was reduced activity at UHT treatment under pH 6.8 condition.

  • LB20304 is a novel fluoroquinolone that exhibits a potent broad spectrum antibacterial activity against both gram-positve and gram-negative bacteria. The MICs (Minimal Inhibitory Concentration) of LB20304 were determined against both gram-positve and gram-negative bacteria under various conditions including several media, pHs, and inoculum concentrations. The in vitro activity of LB20304 was not significantly affected by the changes in testing conditions such as components of media and inoculum concentrations, but it was slightly reduced by acid condition. The MICs and MBCs (Minimal Bactericidal Concentration) of LB20304 against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa were hardly affected by the presence of 50 % human serum, mouse serum, guinea pig serum or horse serum, and the MBCs were equal to or at most four-times higher than the MiCs. The activities of LB20304 were decreased by the presence of high concentraion of $Mg^{++}$ or human urine (pH, 5.5) in the test media. The frequencies of mutants resistant to LB20304 were similar to or lower than those found in ciprofloxacin and sparfloxacin.

Evaluation of Bactericidal Capacity of Silver Ion against Bovine Mastitis Pathogens (소 유방염 원인균에 대한 은이온의 향균력 평가)

  • Bovine mastitis, defined as an inflammation of the mammary gland, is usually associated with bacterial infection. Thus, treatment and control of mastitis relies primarily on antimicrobial therapy. This study investigated the bactericidal actions of silver ion against causing various bovine mastitis pathogens. Minimal inhibitory concentrations (MICs), minimum bactericidal concentrations (MBCs) and bactericidal activity times and concentrations of silver ion against pathogens were determined. The effect of silver ion on bacterial morphology was studied by scanning electron microscopy (SEM). The MICs and MBCs of silver ion for various bacteria strains ranged from 1.9-15.6 ${\mu}g$/ml. SEM images indicated formation of a pit, distortion and disruption of cell walls in silver treated Staphylococcus aureus and Escherichia coli. The results demonstrate that silver ion has a bactericidal capacity against causing various pathogens of bovine mastitis and suggest that silver ions may be exploitable as a therapeutic/preventative tool of bovine mastitis.

Synergistic Effect of Methanol Extract of Salvia Miltiorrhiza and Antibiotics against Dental Caries Pathogens (치아우식증유발세균에 대한 단삼 메탄올추출물과 항생제와의 병용효과)

  • Salvia miltiorrhiza Bunge (S. miltiorrhiza) is a traditional Korean medicine that is commonly used for the treatment of inflammatory diseases such as edema, arthritis, and hepatitis. The present study investigated the antimicrobial activity of methanol (MeOH) extract of S. miltiorrhiza roots against oral bacteria using broth the microdilution method and the checkerboard and time-kill methods evaluated the synergistic effects of treatment with antibiotics. The MeOH extract was demonstrated as a higher antibacterial activity (MICs, 8 to $64\;{\mu}g/mL$; MBCs, 16 to $64\;{\mu}g/mL$) against all tested oral bacteria. Additionally, the extract was observed to have a synergistic effect with ampicillin or gentamicin. A time-kill study evaluating the effects of the extract indicated that the extract treatment in combination with ampicillin or gentamicin showed rapid bactericidal activity. The results suggest that MeOH extract of S. miltiorrhiza could be employed as a natural antibacterial agent against dental caries.
