Embryo transfer (ET) in the animal is an important procedure to generate genetically engineered animals and conserve genetic resources. For ET experiments in mice, pseudopregnant recipients are usually prepared with proestrus stage of females and vasectomized males. However, this conventional method is inefficient because the size of female colonies should be large to select only the proestrus stage in the estrous cycle and the surgical procedures are required to generate vasectomized males. In this study, we established a simple and efficient protocol to prepare ET recipients using the estrous synchronization with hormone injection and the mating with wild male mice. The delivery rate of ET recipients tended to be increased with estrous synchronization using hormone injection (100%) compared to the conventional method (71%). Further, natural pregnancy of the recipients, induced by mating with a wild male, significantly enhanced the birth rate of ET offspring than the conventional method (33% vs. 13%). Based on the results, we concluded that our new protocol using hormone injection to ET recipients and mating with wild males could be more efficient and simpler compared to the conventional method.
This work was carried out to study physiological characteristics of Rhodosporidium toru[oides cells having two different mating types. The mating type A produces internal. cell wall-bound, and external invertases while type a produces only two invertases except external invertase. Comparing their characteristics after partial purification of internal invertases from both mating type cells, invertase from type a has decreased 15% of invertase activity only by $Mn^{2+}$ I while invertase from type A has been increased 11% of invertase activity by $Zn^{2+}$ and decreased 15% of invertase activity by $Mn^{2+}$ On the effect of enzyme inhibitor, invertase of type a was inhibited from 12% to 57% by 2-mercaptoethanol, sodium dodecyl sulfate, phenol. but invertase of type A was slightly inhibited only by phenol. The thermal stability of both invertases has showed steep inactivation at above $80^{\circ}C$ and their optimal temperatures were similar at $60^{\circ}C$ . Invertase from type A showed stability only on condition of acid from pH 3 to 6 and its opimal pH was 5.0, while invertase from type a showed stability at the wide range of pH 3-10 and its optimal pH was 4.0. And the $K_m$ values of invertases from type A and type a were $2.5{\times}10^3$M and$3.4{\times}10^3$M, respectively.
Rare mating was used to select a respiratory deficient mutant of Saccharomyces cerevisiae HDC52 strain. Glucoamylase gene of S. diastaticus K114 was developed into the RD mutant which could uptake maximum amount of non-fermentable sugars through the expression of glu- coamplyase gene and the fermentation characteristics of the developed strain HCS were investigated. The size of HCS yeast and HBD52 yeast strain were 13 $\mu\textrm{m}$ and 10$\mu\textrm{m}$ respectively. HCS strain which can uptake maximum amount of non-fermentable sugar through the expression of glucoamylase gene was developed. By karyotype anal- ysis. HCS stain but not RD mutant HBC52 showed a band of 1150 kb chromosome DNA This band should include glcoamylase gene from Saccharomyces diataticus K114 THis strain has glucoamylase which can degrade starch By transduction and contrnuance of glucoamylase gene HCS strain gegraded strach and formed halo. Also, HCS strain maintained the character after 50 generations. Glucoamylase activities of Saccharomyces diastaticus K114 and HCS yeast strains are 9.5 and 2.7~3.4(unit/ml) HCS and HBC52 strain showed similar sugar fermentation patterns and low flocculation In spore and film forming test, HCS and HBC52 strain formed neither spores nor films. In the limit fermentation test, HBC52 strain showed fermentation level of 68% and HCS strain showed 76~78% As the limit attenuation of HBC52 and HCS were ($2.00^{\circ}$P) and ($0.7~0.93^{\circ}$P) This study demon- strates and HCS strain may be used for low carbohydrate beer fermentation.
The ussur brown katydid, Paratlanticus ussuriensis (Orthoptera: Tettigoniidae), causes substantial damage in orchard areas in South Korea. Daily rhythm and mating behaviors were investigated in last instar nymphs, immature adults and mature adults of P. ussuriensis. The daily rhythm was repeated upon resting, moving, and feeding. The daily rhythm of the movement behavior similar in both last instar nymphs and immature adults. Last instar nymphs becoming active at 10:06 and ceasing activity at 21:12 in average. Immature adults becoming active at 11:30 and ceasing activity at 20:08 in average of totally 10 individuals. However, mature adults showed a different active time of 15:30. Generally, behavior was affected by the conditions of their environment, especially temperature. Mature adults activated later than nymphs and immature adults. Mature adults display mating behavior at a particular time of day (approximately 20:00). P. ussuriensis showed a mating behavioral sequence which was similar to related species in completely retaining the spermatophylax. Females took 270 min to 360 min to completely retain the spermatophylax. The daily rhythm of P. ussuriensis, indicating its occurrence and ecology, could be useful as basic data for managing field populations of P. ussuriensis.
Five to seven day old pupae were irradiated with gamma rays, the mating ability, and competability of the irradiated males were examined. The results obtained are summarized as follows: 1. There is no difference in percentage of males that mated, nor in the maximum number of matings of the males, when individual matings were made, and fresh females were provided every day for his life time. 2. The average days for the first mating of the emerged were 1.5 days for the control, and 2.4 days for the males irradiated with 25 and 30 Krad. The average days between first and second mating were 3.0 days for the irradiated males, and 1.3 days for the control. 3. The average matings of a male were 1.2 and 2.5 times when 5 and 10 females were provided to a normal male respectively. No significant different in average numbers of mated female per male were observed between the normal and irradiated males. 4. To test the mating competitiveness of the irradiated and normal males (1:0, 1:1, 2:1, 1:2, 5:1, 1:5, 0:1) were confined together with five virgin females. The greatest reduction in hatching percentage was found with 5 : 1 : 1 (30 Krad) where $26.6\%$ was hatched.5, The results showed that the irradiated males were not fully competitive with normal males, but if the reduced number of eggs produced by the female mated with irradiated male is considered, it might be used sterile male techniques in rice stem borer control.

Bovine coat color is decided by the melanocortin receptor 1 (MC1R) genotype mutation and melanogenesis. Specially, in the various cattle breeds, dominant black coat color is expressed by dominant genotype of $E^D$, red or brown is expressed in the frame shift mutation of recessive homozygous e by base pair deletion and wild type of $E^+$ is expressed in various coat colors. However, not very well known about the effected of MC1R genotype mutation on the coat color through family lines in KBC. Therefore, this study were to investigate effect of MC1R genotype mutation on the coat color, and to suggest mating breed system in accordance with of MC1R genotype for increased on brindle coat color appearance. Parents (sire 2 heads and dam 3 heads) and offspring (total : 54 heads) from crossbreeding in KBC family line with the MC1R genotype and phenotype records were selected as experimental animals. The relationship between melanocortin 1 receptor (MC1R) genotypes expression verified by PCR-RFLP, and brindle coat color appearance to the family line of the cross mating breed from MC1R genotype pattern was determined. As a result, 4MC1R genetic variations, $E^+/E^+$ (sire 1), $E^+/e$ (sire 2 and dam 3), $E^+/e$ with 4 bands of 174, 207 and 328 bp (dam 1) and $E^+/e$ with 3 bands of 174, 207, 328 and 535 bp (dam 2) from parents (sire and dam) of KBC. However, 3 genetic variations, e/e (24%), $E^+/E^+$ (22%) and $E^+/e$ (56%) were identified in offspring. Also, brindle coat color expressrated was the e/e with the 0%, $E^+/E^+$ with 67% and $E^+/e$ with 77% from MC1R genotype in offspring on the cross mating of KBC. Furthermore, when the sire had $E^+/e$ genotype and the dam had $E^+/E^+$ with the 3 bands or $E^+/e$ genotype, and both had whole body-brindle coat color, 62% of the offspring had whole body-brindle coat color. Therefore, the seresults, the mating system from MC1R genotype patterns of the sires ($E^+/e$) and dams ($E^+/E^+$ with the 3 bands or $E^+/e$) with brindle coat color may have the highest whole body-brindle coat color expression in their offspring.
This study investigated the potential effects of amitraz on the pre- and postnatal development, behavior, and reproductive performance of offspring of parent rats given amitraz during pre-mating, gestation, and lactation. The test chemical was administered via the drinking water containing 0, 40, 120, and 360 ppm to male rats from 2 weeks before mating to the end of 14-day mating period and to females from 2 weeks before mating, throughout mating, gestation and lactation up to weaning. Based on fluid consumption, the male rats received an average of $0,\;5.7{\pm}1.33,\;13.2{\pm}2.08,$ and $35.8{\pm}3.42$ mg/kg/day amitraz, and the female rats received an average of $0,8.7{\pm}4.42,\;20.1{\pm}9.60,\;and\;47.6{\pm}22.38$ mg/kg/day amitraz, respectively. At 360 ppm, an increase in the incidence of abnormal clinical signs, a suppression in the body weight gain, a decrease in the food consumption and litter size, an increase in the post-implantation loss, and a decrease in the seminal vesicle weight were observed in the parent animals. In addition, a suppression in the body weight gain, a decrease in the grip strength, a delay in the negative geotaxis, an increase in the pre- and post-implantation loss, and a decrease in the number of live embryos were observed in the offspring. At 120 ppm, suppressed body weight gain and reduced food consumption were observed in the parent rats. Suppressed body weight gain and decreased grip strength were also observed in the offspring. There were no signs of either reproductive or developmental toxicity at 40 ppm. Under these experimental conditions, the no-observed-adverse-effect level of amitraz for parent rats and their offspring was estimated to be 40 ppm in rats.
Esterase isozyme band patterns were compared between the wild strain and commercial strain of Flammulina velutipes. Monospores were isolated from wild strain. ASI4019 and their mating types were determined. We investigated the relationship between pigmentation on the plate and fruitbody color to understand genetic relationship among F. velutipes strains. Dikaryotic strains mated between nonpigmenting strains produced white fruitbodies. However dikaryon obtained from mating between nonpigmenting monokaryon and brown pigmenting monokaryon produced brown fruitbody as the dikaryon obtained from mating of brown pigmenting monokaryons. The white fruitbody from wild strain was distinguished from that of commercial strain. When the nonpigmenting wild monokaryon was mated with commercial monokaryon, pale brown mushroom was produced. The BC1F1 was obtained by mating the above mentioned $F_1$ with commercial monokaryon. Fruitbody color of BC1F1 shared two types; one strain with all pale brown fruitbodies, and the other strain with separated eight pale brown and two mixed type involving pale brown and white fruitbodies.
The gene CDC70 encoding the${\alpha}-subunit$ of G protein has been known to be a component involved in mating pheromone signalling in the yeast, Saccharomyces cerevisiae. To isolate mutations of the genes involved in the signal transduction, Saccharomyces cerevisiae the strain bearing the cdc70-5 mutation was mutagenized to be forced to recover the ability of colony-formation at restrictive temperature, which means the new mutation can suppress the temperature sensitivity of the cdc70-5 phenotypes. Among these suppressors, $sir^-$ and $mat{\alpha}2^{-}$ mutations are excluded because of no relationship to signal transducer. And the selected suppressors were analyzed for the linkage relationships by the tetrad analysis. Out of fifteen suppressors isolated, twelve were classified into four linkage groups, designated as sga1, sga2, sga3, sga4 by the tetrad analysis. The other three genes were determined for the linkage.
To investigate the mating system of clones in the seed orchard of Japanese red pine, parameters of mating system, such as outcrossing rates, number of potential pollen contributors, and degree of pollen contamination, were estimated in the seed orchard of Japanese red pines on the basis of DNA data including 4 nSSR and 6 cpSSR markers. Estimates of outcrossing rates were ranged from 94.9 to 100% with an average of 98.9% on the basis of the analysis of cpSSR haplotypes. They were ranged from 90.3% to 100% with an average of 95.9% on the basis of the analysis of nSSR genotypes. However, cross checking of both DNA markers revealed that the seeds presumed to be products of self pollination were turned out to be generated by pollination between mother tree and other tree (i.e., 100% of cumulative outcrossing rate). Estimates of pollen contamination ranged from 43.6% (Gangwon-10) to 56.4% (Gangwon-12) with the average of 48.9%. On the basis of pooled cpSSR haplotype of each seed, maximum number of 21 pollen contributors were verified from the seeds reproduced by Kyungbuk-38. Minimum number of 13 pollen contributors were verified in Gangwon-10. Mean of 16.2 pollen contributors were verified from a total of 5 mother trees. In conclusion, considering pretty high outcrossing rates between clones within a seed orchard, it may be expected that a fairly good genetic potential of the seeds, produced in '77 plot of the seed orchard of Japanese red pines at Anmyeon island, may be guaranteed. Observed results from the analysis of mating system of Japanese red pines in a '77 plot of the seed orchard may also provide useful information for the establishment and management of the seed orchard of the progressive generation.
