Calcium Signaling-mediated and Differential Induction of Calmodulin Gene Expression by Stress in Oryza sativa L.

  • $Ca^{2+}$/calmodulin transduction pathways have been implicated in mediating stress response and tolerance in plants. Here, three genes encoding calmodulin (Cam) members of the EF-hand family of $Ca^{2+}$-binding proteins were identified from Oryza sativa L. databases. Complementary DNA for each of the calmodulin genes, OsCam1, OsCam2, and OsCam3 were sequenced. OsCam1 and OsCam2 encode a conventional 148-amino acid calmodulin protein that contains four characteristic $Ca^{2+}$-binding motifs. OsCam3 encode a similar protein with a 38-amino-acid extension containing a putative prenylation site (CVIL) at the carboxyl terminus. RT-PCR showed that each of the genes is expressed in leaves and roots of 2-week old rice seedlings. By RNA gel blot analysis, OsCam1 mRNA levels strongly increased in response to NaCl, mannitol and wounding treatments. In contrast, OsCam2 mRNA levels were relatively unchanged under all conditions investigated. NaCl treatment and wounding also increased the OsCam3 mRNA level, but in a more transient manner. Our results indicate that although the expression of genes encoding different calmodulin isoforms is ubiquitous, they are differentially regulated by various stress signals. In addition, we have demonstrated that the calcium-channel blocker lanthanum chloride inhibited the induction of OsCam1 gene expression by both NaCl and mannitol treatments. These results suggest that osmotic stress induced expression of OsCam1 gene requires the $[Ca^{2+}]_{cyt}$ elevation that is known to occur in response to these stimuli.

Genetic Variation Analysis of Early-heading Plant (Oryza sativa L.) Lines Derived from Gamma-ray Irradiation (방사선조사 유래의 조기출수 식물(Oryza sativa L.) 계통의 유전적 변이 분석)

  • This study was carried out to evaluate genetic variation of early-heading rice (Oryza sativa L. cv. Dongjin 1) lines derived from gamma-ray ($^{60}Co$, 300 Gy) irradiation. The average heading date of the 5 early-heading lines in $M_7$ and $M_8$ generation was faster than that of untreated control as 11 (line ${\gamma}$-2), 10 (line ${\gamma}$-5), 6 (${\gamma}$-1 line), 5 (${\gamma}$-3) and 4 days (line ${\gamma}$-4), respectively. According to ISSR analysis, polymorphic rate of the early-heading lines (from 5.9% to 23.4%) was higher than that of control (4.3%). The result indicates that the gamma-ray promote variation at DNA level. When genetic variations of rps16-trnK region were evaluated by nucleotide analysis, nucleotide length of the rps16-trnK region was 664 bp in all the early-heading lines and control. Out of 5 sites of nucleotide transposition detected in the region, however, 2 sites were appeared only in the early-heading lines.

Black Rice (Oryza sativa L. var. japonica) Hydrolyzed Peptides Induce Expression of Hyaluronan Synthase 2 Gene in HaCaT Keratinocytes

  • Black rice (Oryza sativa L. var. japonica) has been used in folk medicine in Asia. To understand the effects of black rice hydrolyzed peptides (BRP) from germinated black rice, we assessed the expression levels of about 20,000 transcripts in BRP-treated HaCaT keratinocytes using human 1A oligo microarray analysis. As a result, the BRP treatment showed a differential expression ratio of more than 2-fold: 745 were activated and 1,011 were repressed. One of the most interesting findings was a 2-fold increase in hyaluronan synthase 2 (HAS2) gene expression by BRP. Semiquantitative RT-PCR showed that BRP increased HAS2 mRNA in dose-dependent manners. ELISA showed that BRP effectively increased hyaluronan (HA) production in HaCaT keratinocytes.

Enzymatic preparation and antioxidant activities of protein hydrolysates from hemp (Cannabis sativa L.) seeds

  • Hemp (Cannabis sativa L.) seeds have recently been attracting attention as a new high-value-added food material owing to their excellent nutritional properties, and research on the development of functional food materials using hemp seeds is actively progressing. This study aimed to evaluate the antioxidant properties of hemp seed protein hydrolysates. Protein hydrolysates were prepared from defatted hemp seed powder (HS) by enzymatic hydrolysis using five different proteases (alcalase, bromelain, flavourzyme, neutrase, and papain). 2,4,6-trinitrobenzene sulfonic acid (TNBS) assay and SDS-PAGE analysis revealed that HS showed a high degree of hydrolysis after treatment with each enzyme except papain. The total polyphenol content of the protein hydrolysates (<3 kDa) and the RC50 values obtained from two different antioxidant tests showed that alcalase hydrolysate (HSA) had a relatively high level of antioxidant capacity. In addition, treatment with HSA (25-100 ㎍/mL) significantly inhibited linoleic acid peroxidation. These results suggest that hemp seed protein hydrolysates are potential sources of natural antioxidants. Future studies will focus on the identification of active peptides from HSA.

Light dependent arsenic uptake and growth in Lactuca sativa L.

  • Along with other heavy metals, arsenic (As) is one among the substances most harmful to living organisms including humans. Owing to its morphological similarity to phosphorus, the uptake of As is influenced by photosynthesis and the phosphorus uptake pathway. In this study, we varied arsenic exposure and light intensity during nutrient solution cultivation of lettuce (Lactuca sativa L.) to determine the effect of these two factors on arsenic uptake, lettuce growth, and electron transfer in photosystem II. In the treatment exposed to 30 μmol L-1 of arsenic, the shoot arsenic concentration increased from 4.73 mg kg-1 to 18.97 mg kg-1 as the light intensity increased from 22 to 122 μmol m-2 s-1. The water content and ET2o/RC of the shoots were not affected by arsenic at low light intensity; however, at optimal light intensity, they decreased progressively with arsenic exposure. Increased light intensity stimulated the growth of plant roots and shoots; contrarily, the difference in growth decreased as the concentration of As exposure increased. The results of this study suggest that the effect of As on plant growth is dependent on light intensity; in particular, an increase in light intensity can increase the uptake of As, thereby affecting plant growth and As toxicity.

  • The effect of alachlor treatment on peroxidase synthesis in oat root tips was studied. Alachlor caused increase in the amount of soluble peroxidase in oat root tips, peroxidase activity increase as the rate of alachlor application increased, Alachlor treatment of oats with $1{\times}10^{-6}M$, peroxidase activity increased 0.20 unit higher than that of nontreatment. After 12hr, 65mM of peroxide treatment of oats inhibited 16% root growth, and 130 mM peroxide treatment caused 59% inhibition. With PAGE of peroxidase extracted from normal root tips, PAGE give 4 species($P_2$, $P_3$, $P_4$, and $P_5$ band) of peroxidase. Alachlor significantly activated isoperoxidase. Three isoperoxidase($P_1$, $P_6$, and $P_7$) are synthesized at a increased concentration of alachlor, SDS-PAGE analysis of proteins extracted from oat root tips showed that they were made up of subunits blow 100 kD polypeptide.

Salt Stress Responses of an Alanine Aminotransferase Knock-out Mutant of Rice (Oryza sativa L.) (알라닌 아미노기전이효소가 상실된 벼(Oryza sativa L.) 돌연변이체의 고염 스트레스에 대한 반응)

  • An AlaAT knock-out mutant (alaat) of rice (Oryza sativa L.) was isolated from T-DNA tagging lines and the genotypes of its progeny were determined with AlaAT1-specific primers. The alaat phenotypes showed decreased growth and grain yield when compared with control plants. The activity of AlaAT1 in the mutant plants was practically undetectable. The responses of alaat plants to growth under salt stress were compared with those of control plants by measuring chlorophyll fluorescence and the activities and mRNA expression of antioxidant enzymes. All abiotic stresses tested (salt, drought, and chilling) caused a similar decrease in chlorophyll fluorescence in both alaat and wild type plants. The activity of peroxidase (POX), an antioxidant enzyme, decreased following salt treatment of alaat plants, while control plant showed an increased activity. The mRNA levels for cAPX (cytosolic ascorbate peroxidase), POX2, and AlaAT were determined by RT-PCR following salt stress. No AlaAT1 mRNA was detected in alaat plants. The POX2 mRNA showed a slightly increased level in the wild type but was not detected in alaat plants, in agreement with the activity assays. The levels of cAPX mRNA were greatly increased in both the wild type and alaat plants. The salt stress effects on rice plant growth are therefore proposed to reflect a loss of function of AlaAT, which alters the activity and synthesis of antioxidant enzymes (especially peroxidases), rather than a direct effect on photosynthesis.

Comparative Morphology of Plastids on Vegitative Tissue of Cannabis sativa L. (대마(Cannabis sativa L.) 영양조직의 색소체 비교)

  • Various plastids in leaf and bracteal tissues of Cannabis sativa L. were examined by electron microscopy. Young chloroplasts without starch grain in mesophyll cells were ellipsoidal, and osmiophilic globules within them were common in stroma. During the plastid differentiation, the mature chloroplasts in mesophyll were changed in shape depending on the numbers and sizes of starch grain in stroma. Electron-dense granular substances were occurred along the outer membrane of chloroplasts in mesophyll. Typical plastids with reticulate body were present in the glandular trichomes. Electron-grey material appeared along the surface of a plastid. A light area in reticulate body is considered to represent junction point of thylakoids.

  • To evaluate allelopathic effect of volatile chemicals and phytohormones, seed germination and seedling growth test of Lactuca sativa have performed in laboratory experiments. Among used chemicals terpienen-4-ol was the most inhibitory to seed germination of lettuce. ABA and GA inhibited seed germination at $5{\times}10^{-6}M$ concentration but promoted germination at $2.5{\times}10^{-5}M$ and $5{\times}10^{-5}M$. ABA and GA alleviated volatile chemical-induced inhibition of seed germination and seedling elongation of lettuce.

Characterization of a QTL associated with chlorophyll content using progeny from an interspecific cross in rice (Oryza Sativa L.)

  • Rice (Oryza sativa L.) is the world's most important cereal crop. In crop plant, chlorophyll content and leaf senescence could affect grain filling and yield. We analyzed a QTL associated with chlorophyll content and delayed leaf senescence using high chlorophyll near isogenic line (HC-NIL). HC-NIL derived from a cross between Oryza sativa cv. Hwaseong as a recurrent parent and wild species O. grandiglumis as a donor parent showed higher chlorophyll content than Hwaseong. To identify QTL associated with chlorophyll content, 58 $F_3$ and 38 $F_4$ lines were developed from a cross between HC-NIL and Hwaseong. For QTL analysis, simple sequence repeat (SSR) markers were used for genotyping and one-way ANOVA was conducted. A QTL for chlorophyll content (qCC2) was detected in chromosome 2 and explained 24.63% of phenotypic variation. The senescence effect of the qCC2 was examined in dark-induced incubation (DII). Detached leaves from Hwaseong and HC-NIL were incubated on 3mM MES buffer (pH 5.8) at $27^{\circ}C$ under complete dark condition. After 3 days of incubation, the Hwaseong leaves turned yellow, but the HC-NIL leaves were green. HC-NIL has higher chlorophyll content with delayed senescence than Hwaseong. These results indicated that qCC2 is associated with stay-green phenotype. To know whether the qCC2 is responsible for leaf functionality, ion leakage test and Fv/Fm measurement were performed. Both experiment results showed that differences were observed between Hwaseong and HC-NIL but it was not statistically significant. These results might suggest that the qCC2 is possibly related to chlorophyll content and non-functional stay-green phenotype.
