Growth of Bioenergy Crop Miscanthus sacchariflorus cv. Geodae 1 on Barren Reclaimed Land Applied with Solidified Sewage Sludge in Landfill Sites (하수슬러지 고화물을 처리한 유휴 간척지토양에서 바이오에너지작물 거대 1호의 생육특성 연구)

  • This study firstly provides basic data for selection of cultivatable bioenergy grass in barren reclaimed lands applied with solidified sewage sludge. The experimental plots consisted of a plot containing reclaimed land mixed with solidified sewage sludge (MSS 50), a plot covered by solidified sewage sludge (CSS 100), and an original reclaimed soil plot (ORS). The growth, biomass production of bioenergy grasses and soil chemical properties were investigated in each experimental plot for 5 years. The organic matter (OM) and total nitrogen (T-N) content in both MSS 50 and CSS 100 were considerably higher than those in ORS. In bioenergy grasses, M. sacchariflorus cv. Geodae 1 showed an excellent growth and adaptability on reclaimed land applied with solidified sewage sludge. The application of solidified sewage sludge may provided soil nutrition in the reclaimed land due to the fact that bioenergy crops grew better in soils applied with solidified sewage sludge than in untreated soils, and treated soils had higher OM and T-N content than untreated soils. This study suggests that M. sacchariflorus cv. Geodae 1 is the most suitable biomass feedstock crop for biomass production and that solidified sewage sludge may be used as a soil material for cultivation of bioenergy grass on reclaimed lands.

Studies on the Constituents of Korean Higher Fungi (III) -Alkaloids of Korean Ergots- (한국산(韓國産) 고등균류(高等菌類)의 성분연구(成分硏究)(III) -한국산(韓國産) 맥각(麥角)의 Alkaloid 성분(成分)-)

  • Korean ergot sclerotia which were collected from Agropyron semicostatum $N_{EES}$, Miscanthus sinensis $A_NDERS$ var. purpurascens $R_{END}$, M. sacchariflorus $B_{ENTH}$. et HOOK,. M. sinensis $A_{NDERS}$., Eulalia speciosa $O.{\;}K_{UNTZ}$, Themeda triandra $F_{ORSK}$. var. japonica $M_{AKINO}$, and Phalaris arundinacea L. were investigated for their alkaloid constituents. Quantitative determinations of alkaloids were carried out using a modified procedure of van $U_{RK}$ and $M_{ICHELON}-K_{ELLEHER}$ methods. The results showed that the two ergots from Agropyron semicostatum and Phalaris arundinacea contained total alkaloids up to 0.137 per cent. The alkaloids of these two ergots are mostly water-insoluble alkaloids. The ergots parasitizing Reed Canary grass was found to contain both water-soluble and water-insoluble alkaloids. The TLC and PPC tests for the ergot alkaloids from Agropyron semicostatum and Phalaris arundinacea revealed that they contain at least two alkaloids, respectively, and that the ergot from Themeda triandra $F_{ORSK}$. var. japonica has one. The lipid contents of the seven ergots ranged from 13 to 29 per cent.

  • The plant community of water`s edge was investigated using the methodology of the Z-M school of phytosociology, from March to October, 2008, 2011. The results were obtained as follow ; hygrophyte hydrophye of 2008 year of this area consist of 38 taxa : 1 varieties, 37 species, 31 genera, 21 families and 14 orders, hygrophyte hydrophye of 2011 year of this area consist of hygrophyte 15 species, 13 species were reduced by approximately 13.33%. And emerged hydrophyres did not show a change. 9 species, 4 species of Floating-Leaved hydrophyres 55.56%, Submerged hydrophyres of the 4 species, 1 species, 75%, Free-Floating hydrophyres remarkable I the 3 species was reduced to 33.33%, respectively. Plants Community of the water's edge was classified into 16 communities as follows; (Leersia japonica community, Spirodela polyrhiza community, Miscanthus sacchariflorus community, Hydrocharis dubia community, Zizania latifolia community, Paspalum distichum var. indutum community, Leersia japonica-Hydrocharis dubia community, Salix glandulosa community, Trapa japonica community, Humulus japonicus community, Nelumbo nucifera community, Carex dimorpholepis community, Salix koreensis community, Trapa japonica-Spirodela polyrhiza community, Persicaria thunbergii). Higher in 2011 than in 2008, the ratio was reduced to Leersia japonica community. Between July-August by a sudden change in the rate of water storage changes in vegetation are thought to have appeared.

Characteristics of Nutrients Release by Submerged Plants in Flood Control Reservoirs within Juam Lake (주암호 홍수조절용지내 침수 식물체의 영양염류 용출 특성)

  • To improve the quality of water from water supply source and to establish the management plan of dead plants in flood control reservoir around Juam Lake, the effect of water quality by dead plant in column with passing time was investigated. In column test, the amounts of release by Carex dimorpholepis were $7,893-7,917mg\;m^{-2}\;month^{-1}$ COD, $2,711-2,816mg\;m^{-2}\;month^{-1}$ T-N and $342-547mg\;m^{-2}\;month^{-1}$ T-P. The amounts of release by Miscanthus sacchariflorus were $6,487-6,507mg\;m^{-2}$ COD, $1,813-1,868mg\;m^{-2}$ T-N and $226-405mg\;m^{-2}\;month^{-1}$ T-P in column. Therefore, the release of COD, T-N and T-P by Carex dimorpholepis were more than those by Miscanthus sacchariflorus Benth in column.

Occurrence of Streamside Flora in relation to Environmental Condition at Ansungchon Creek (안성천 하천변 식물상 구성과 환경 조건과의 관계)

  • According to the survey map, the altitudes of up and down the Ansungchon were 242m and 49m accordingly. The width of the river was 4m at upperstream and became wider, lam at midstream and 114m at downstream. Water pH of the Ansungchon was measured variously in the range of 6.0-7.9 according to the surveyed times and areas. The water pH measured during June through August decreased continually and suddenly increased at the end of October. Phragmites iaponica, a major species of the fresh water emergent anchored hydrophytes, was surveyed mostly in the area of upperstream, Phragmites communis, Miscanthus sacchariflorus and Typha orientalis were surveyed in the areas of midstream and downstream, and Zizania latifolia was found in all areas of the river. The total of 101 taxa dividing into 32 families, 71 genera, 88 species, and 13 varieties have been inhabited in these areas. Among the inhabited plants, Poaceae was 21.8%, which appeared in the highest percent. Compositae was 20.8%, and Polygonaceae was 11.9%. In the surveyed areas, herbaceous plants were 95.1% and mostly occupied, and woody plants were 4.9%. Among the herbaceous plants, perennial plants were 52.5%, and annual and biannual plants were 28.7% and 13.9% accordingly. Naturalized plants dividing into 8 families and 18 genera have been found in the surveyed areas. The number of the naturalized plants increased from upperstream to downstream as the environmental disruption became worse. Only 2.5% of the total naturalized plants were appeared in up, 17.7% in midstream, and 28.6% in downstream. Shannon-Weiner's index for the degree of diversity was 2.898-3.666 in the area of upperstream, but 3.708-4.135 in downstream, which was little higher.

Losses of Biomass and Mineral Nutrients during Decomposition of Herbaceous Plants in Riverine Wetlands

  • The composition changes of litters were investigated to figure out the effects of the decomposition of Humulus japonicus on nutrient circulation and decomposition process in the riverine wetlands: Tan stream and Amsa-dong. Litterbags (mesh size 1 mm and 5 mm) were installed to monitor mass and nutrient changes of 5 types of litters: H. japanicus only, Miscanthus sacchariflarus only, Phragmites communis only, mixed litters including H. japonicus, and mixed litters without H. japonicus for 7 months. It was shown that k (decay rate) of the H. japanicus ($2.68{\sim}3.12$) was higher than that of M. sacchariflorus ($1.83{\sim}2.16$) and P. communis ($0.02{\sim}1.18$). The mass and organic remainings of the mixed litters including H. japonicus at Tan stream were $47.0{\sim}55.1%\;and\;47.0{\sim}54.9%$ and those of the litterbags without H. japanicus were $49.2{\sim}65.4%\;and\;47.1{\sim}57.5%$, respectively. This result indicated that the nutrient circulation was faster at H. japanicus community than others. Ca, Na, Mg, K, P, C, N and H contents reduced to around $40{\sim}80%$ of original. However, Na concentration increased up to $407{\sim}584%$ at 100 days and decreased to $248{\sim}498%$ at the end of the experiment. Decomposition rates were similar between 1 mm and 5mm mesh size litterbags and this implies that plant litters in studied areas decomposed mainly by microbes rather than small animals. This study revealed that the fast growth of H. japonicus was resulted from fast decomposition in part: positive feedback of nutrient cycling.

  • 억새(Miscanthus spp.)는 우리나라 등 동아시아가 원산이며 바이오매스량이 많아 바이오에너지 원료작물로서 잠재성이 크기 때문에 2세대 바이오에탄올 생산 원료로 주목을 받고 있고, 축산깔개 및 토양개량제 등으로도 이용되고 있다. 미국과 유럽 등에서는 생태계 교란 방지를 위해 4배체 물억새(M. sacchariflorus)와 2배체 참억새(M. sinensis)의 종간 교잡 이질 3배체인 불임성 억새(M. x giganteous)을 주로 재배하고 있으나, 단일 유전형의 품종으로 병해충과 자연재해에 취약하여 다양한 억새 품종의 개발이 시급한 실정이다. 본 연구는 억새를 재료로 하여 반수체 및 배수체 확보를 통한 배수성 별 특성 평가와 함께 기내배양을 통해 탈분화 및 재분화 시스템을 구축하여 억새의 육종 효율을 높이기 위해서 실시하였다. 억새 종자로부터 캘러스의 유도는 MS배지와 N6배지에 1mg/L 2.4-D를 첨가하였을 때 비배발생 캘러스(nonembryogenic callus)가 유도되었고, N6배지에 3~5 mg/L 2,4-D를 첨가하였을 때 배발생 캘러스(embryogenic callus)가 발생하였다. MS배지보다는 N6배지에서 캘러스 유도율이 높았으며, 식물생장조절제로 2,4-D와 BA 조합처리 보다 2,4-D 단용 처리하였을 때 캘러스 유도율이 더 높았다. 억새 종에 따른 캘러스 유도율은 물억새가 25.2~49.3%, 참억새는 30.3~52.0%였고 거대억새는 62.6~81.1%로 나타났다. 억새 신초 및 줄기로부터의 캘러스 유도율은 물억새가 4.4~17.2%, 참억새는 1.8~7.7%, 거대억새는 15.3~19.9%로 나타나 종자에 비해 매우 낮았다. 미성숙화기로부터의 캘러스 유도는 억새 종에 따른 차이가 없었으며, 3mg/L 2,4-D 첨가 배지에서 캘러스 유도율이 비교적 낮았고(60~80%), 1mg/L와 5mg/L의 2,4-D가 첨가된 배지에서 캘러스 유도율이 높게(90~95%) 나타났다.

The Patterns of Inorganic Cations, Nitrogen and Phosphorus of Plants in Moojechi Moor on Mt. Jeongjok. (정족산 무제치늪 식물의 무기이온, 질소 및 인의 양상)

  • To investigate ecophysiological characteristics of plants species adapted to moor habitat, we selected 22 species plants and analyzed inorganic cations (K, Ca, Mg), heavy metals (Al, Fe, Mn) and total nitrogen and phosphorus quantitatively. Moojechi moor indicated typical acidic and oligotrophic conditions with pH of 5.0∼5.6 (pH 4.3∼5.1 in soil) and EC of 15∼30μ S/cm, and contained very low contents of soil divalent cation such as Ca and Mg but high contents of heavy metals (esp. Al). With respect to inorganic cation contents, investigated plants species showed remarkable interspecific difference. Plant species belonging to J. effusus var. decipiens, M. japonica, I. globosa, M. sacchariflorus, R. mucronulatum, R. yedoense var. poukhanense, H. micrantha, D. rotundifolia showed very low contents of inorganic cation below 400 μ M/g DW, but plant species of C. palustris var. spontanea, L. sessilifolia, P. mandarinorum, C. lineare, S. austriaca sub. glabra, V. mandshurica, A. decursiva showed high cation contents in leaves. Especially, S. austriaca sub. glabra (Compositae) and V. mandshurica (Violaceae) showed pattern accumulating Ca and Mg with plant growth, but I. ensata var. spontanea (Iridaceae) and S. officinalis (Rosaceae) showed decreasing tendency. Meanwhile, most plant species showed low contents of soluble metal ions in leaves in spite of high heavy metal contents on soil, and indicated remarkable interspecific differences in the total contents and composition of heavy metals accumulated. Despite low contents of N and P on soil, most plant species indicated relatively high contents of N and P in leaves at the early stage of growth, and showed slowly decreasing pattern according to growth. Consequently, it seems that plant species inhabited on Moojechi moor cope with acidic-oligotrophic conditions, accumulating inorganic cations and nitrogen at the early growing stage and reutilizing them in the course of growth, and developing heavy metal excluding mechanism.

Effects of Application of Solidified Sewage Sludge on the Growth of Bioenergy Crops in Reclaimed Land (간척지토양에서 하수슬러지 고화물 처리가 에너지작물의 생육에 미치는 영향)

  • This study was carried out to obtain the basic data for selecting the cultivatable bioenergy crops through application of solidified sewage sludge in reclaimed lands. The experimental plots consisted of the mixing with solidified sewage sludge plot (SS50), the covering with solidified sewage sludge plot (SS100), and the original reclaimed land plot (ORL) on reclaimed land for the intended landfill in Sudokwon Landfill Site Management Corporation (SLC). The growth of energy crops (Geodae-Uksae 1, Miscanthus sacchariflorus, and Phragmites australis) were investigated from May to October, 2010 in each experimental plot. The soil from ORL showed higher salinity with high contents of exchangeable $Na^+$ cation than that of SS50 and SS100. Soil properties on reclaimed land used in this study must be improved by increasing the buffering capacity of saline with the treatment of solidified sewage sludge due to the fact that the contents of organic matter (OM) in both of SS50 and SS100 were higher than that of the ORL. Thus the growth of energy crops cultivated in the solidified sewage sludge plots were better than in ORL. Geodae-Uksae 1 which showed an excellent adaptability on reclaimed land treated with the solidified sewage sludge has considerably higher biomass than those of other energy crops (M. sacchariflorus and P. australis). This study suggested that Geodae-Uksae 1 is the most suitable biomass feedstock crop for bioenergy productions, and the solidified sewage sludge may be possible to utilize as a soil cover materials for cultivation of bioenergy crops in reclaimed land.

Miscanthus EST-originated Transcription Factor WRKY Expression in Response to Low Temperature in Warm-season Turfgrasses (억새 EST 정보 유래 전사요소 WRKY의 난지형 잔디의 저온 발현 반응성)

  • Whole genome transcriptomes from Miscanthus species were sequenced and analyzed, which provided 50 different types of transcription factor (TF) involving various developmental processes or environmental stresses. Among the explored TF, WRKY gene family was the major type and one of the WRKY genes, MSIR7180_WRKY4, induced under low temperature environment was selected to investigate how the Miscanthus-originated MSIR7180_WRKY4 TF responds when exposed to low temperature in four warm-season turfgrasses (Z. matrella 'Semil', bermudagrass, St. Augustinegrass, and seashore paspalum). The MSIR7180_WRKY4 was expressed higher during low temperature period in Bermudagrass, but the expression was enhanced in St. Augustinegrass. In contrast, the gene in 'Semil' cultivar was barely expressed and relatively less expressed, but repressed gradually in seashore paspalum, which seems to allow two turfgrasses stay-green longer in the fall season. The results indicate that bermudagrass and St. Augustinegrass adapt to low temperature quickly, but relative tolerance to low or cold temperature at the molecular level needs to be further investigated at different physiological stages and the corresponding genes systematically.
